The aim of this research was to select parameters for supercritical extraction with CO2 of Medicago sativa L., considered as functional food, in quarter-technical plant, providing the highest concentration of bioactive polar constituents and simultaneously maintaining the highest efficiency of the process. For the purpose of optimization, mathematical statistics was used. Qualitative analysis of products was performed with supercritical fluid chromatography (SFC). The SFC analysis revealed a proper separation of flavonoids and phenolics acids for dedicated TFC and TPC optimal parameters. The obtained results have proved that it is a possibility to extract polar compounds with non-polar solvent under higher values of pressure and temperature and to enrich product with desired group of bioactive compounds with proper optimization. The proposed extraction technique allows to obtain on an industrial scale, using an environmentally friendly solvent, a preparation rich in biologically active nutrients that can be implemented in the cosmetics, pharmaceutical and food industries.

The aim of this study was to determine the degradation in the rumen for the dry matter (DM), organic matter (OM) and crude protein (CP) of alfalfa (Medicago sativa L.) harvested at different maturity stages by using an in situ nylon bag method. Maturity and harvesting effects were observed in ruminally degradable DM, OM and CP. High correlations between in situ degradability and maturity were determined. Alfalfa harvested in the bud period exhibited more extensive ruminal DM, OM and CP degradation than that harvested at other maturity stages. The alfalfa hay’s DM, OM and CP degradability were lowest (62.18%, 59.52% and 76.31%, respectively) in its most mature period of seed setting; its degradability decreased with maturity (17%, 14% and 13%, respectively).

This paper deals with probe measurements of low frequency resistance (RLF), high frequency resistance (RHF), and resistance index (= 100 x RLF/RHF), for the determination of cold hardiness or the injury caused by cold treatments. Populations of white clover (Trifolium repens L.) were subjected to different periods of freezing. Discrimination between treatments could be made in terms of RLF, RHF, and resistance index, though discrimination in terms of RHF is poor. The rest of the paper deals with lucerne (Medicago sativa L.). Measurements were made after a cold treatment on field plants consisting of four commercial strains and their F1 crosses, and also on F2 seedlings. RLF and the resistance index appear to be equally good predictors for the recovery of strains or of individual plants from such populations. The Appendix deals with measurements on commercial (tetraploid) plants, diploids, and colchicine-produced tetraploids.

Fermentation is a technology that enhances biologically active ingredients, improves the absorption rate and induces the generation of new functional ingredients by the catalytic action of enzyme systems possessed by microorganisms. In this study, changes in the content of five kinds of bioactive compounds (deacetylasperulosidic acid, asperulosidic acid, scopolin, asperuloside and scopoletin) of Morinda citrifolia L. were confirmed by fermentation, and a high-performance liquid chromatography-photodiode array (HPLC-PDA) analysis method for measuring analytes was developed and validated. HPLC method for the determination of five bioactive compounds in Morinda citrifolia L. extracts (MCE) was validated in terms of sensitivity, linearity, selectivity, limit of detection (LOD) and quantification (LOQ), precision and accuracy. The coefficient of determination of the calibration curve for bioactive compounds (1.56–100 μg/mL) showed linearity (R2 ≥ 0.9999). LOD and LOQ were in the range 0.04–0.97 and 0.13–2.95 μg/mL, respectively. The range of intra- and intraday accuracies values (recovery) were 97.5–121.9% and 98.8–118.1%, respectively, and precision value (RSDs) of the bioactive compounds were <4%. In addition, changes in the content of five bioactive compounds in MCE by fermentation were confirmed. These results indicate that the developed fermentation and analysis method could be applied in the development of potential functional food ingredients.
