The tomato, Solanum lycopersicum L. is one of the most consumed vegetables in the world; nevertheless, it is affected by biotic and abiotic factors that reduce its productivity. The whitefly is globally considered as the main pest under protected crop conditions, where biologic control using endophytic fungi emerges as a sustainable alternative. We evaluated the indirect effects of five native endophytic strains of Beauveria bassiana on the reproduction of greenhouse whiteflies and the growth of tomatoes. The plant growth substrate was inoculated with five strains of this endophyte and the resulting plants were then exposed to whiteflies afterwards. The effect that endophytic strains had on phosphate solubilization, iron siderophore production, plant height, and plant biomass were evaluated. The evaluated endophytes reduced the number of eggs per cm2 on leaflets compared to the control and behaved similarly to the commercial synthetic insecticide. Leaflets inoculated with strains RGM-557, RGM-644 and RGM-731 showed fewer nymphs than the control and those treated with insecticide. RGM-557 and RGM-731 produced the greatest plant heights; RGM-731 obtained the greatest plant biomass. Our study provides evidence that native endophytic strains of B. bassiana have a biocontrol effect on whiteflies and could be used to promote tomato growth.

AbstractPhosphate solubilization and growth promotion by Pseudomonas fragi CS11RH1 (MTCC 8984), a psychrotolerant bacterium isolated from a high altitude garlic rhizosphere from the Indian Himalayas, are reported here. The identity of the isolate was arrived on the basis of its biochemical features and sequencing of the 16S rRNA gene. The isolate grew and solubilized phosphate at temperatures ranging from 4 to 30°C. Besides solubilizing P it produced indole acetic acid (IAA) and hydrogen cyanide (HCN). Seed bacterization with the isolate significantly increased the percent germination, rate of germination, plant biomass and nutrient uptake of wheat seedlings. While Pseudomonas fragi is normally associated with the spoilage of dairy products stored at cold temperatures, this is an early report on the plant growth promoting ability of the bacterium.

ABSTRACT Trichoderma fungi are considered relevant plant growth promoters for increasing the efficiency in the use of nutrients, as well as acting as biological control agents. This study aimed to select Trichoderma spp. isolates with potential for phosphate solubilization and their application as growth promoters in interaction with homeophatic preparations, in cherry tomato. Among 16 Trichoderma spp. isolates obtained from soils of organic tomato growing areas tested in vitro, together with a commercial product (Trichodermil®), two of them showed the ability for indole-3-acetic acid production and phosphate solubilization. The Trichoderma “R” had the highest mycelial growth speed index and presented twice as much spores than the commercial product. An in vivo experiment was also conducted in a greenhouse, in order to observe the potential of Trichoderma spp. isolates and homeophatic preparations on the cherry tomato growth promotion, using a randomized block experimental design, in a 4 x 3 factorial arrangement, with three Trichoderma isolates and two homeopathic preparations (Phosphorus 6CH and Carbo vegetabilis 6CH) + treatment without homeopathic preparation. The leaf area and dry mass of leaves and roots were determined. It was possible to observe that the isolate “R”, identified as Trichoderma asperellum, was effective in the cherry tomato growth promotion, while the homeopathic preparations applied did not show any effect.

SummaryThis study aims to develop a biocontrol agent against Fusarium oxysporum f.sp. radicis-lycopersici (FORL) in tomato. For this, a set of 23 bacterial endophytic isolates has been screened for their ability to inhibit in vitro the growth of FORL using the dual plate assay. Three isolates with the most sound antagonistic activity to FORL have been qualitatively screened for siderophore production, phosphates solubilization and indolic acetic acid (IAA) synthesis as growth promotion traits. Antagonistic values of the three candidates against FORL were respectively: 51.51 % (EB4B), 51.18 % (EB22K) and 41.40 % (EB2A). Based on 16S rRNA gene sequence analysis, the isolates EB4B and EB22K were closely related to Enterobacter ludwigii EN-119, while the strain EB2A has been assigned to Leclercia adecarboxylata NBRC 102595. The promotion of tomato growth has been assessed in vitro using the strains EB2A, EB4B and EB22K in presence of the phytopathogen FORL. The treatments with the selected isolates increased significantly the root length and dry weight. Best results were observed in isolate EB4B in terms of growth promotion in the absence of FORL, improving 326.60 % of the root length and 142.70 % of plant dry weight if compared with untreated controls. In the presence of FORL, the strain EB4B improved both root length (180.81 %) and plant dry weight (202.15 %). These results encourage further characterization of the observed beneficial effect of Enterobacter sp. EB4B for a possible use as biofertilizer and biocontrol agent against FORL.

O controle da broca-da-erva-mate, Hedypathes betulinus, limita-se ao emprego de práticas silviculturais e catação manual dos insetos adultos, não sendo permitido o uso de inseticidas químicos na cultura. Esta pesquisa teve por objetivos verificar a presença natural de Beauveria bassiana no solo de plantio em monocultura de erva-mate, determinar a persistência de B. bassiana no solo após duas e três aplicações do fungo e averiguar a influência de fatores abióticos na estabilidade do fungo no solo. O estudo foi conduzido em um erval no município de Campo Alegre, Santa Catarina, Brasil, onde foram coletadas amostras de solo em duas áreas, para avaliação da presença natural de inóculos do fungo B. bassiana, procedendo-se à contagem, isolamento e identificação.Não foi detectada a ocorrência natural do fungo no solo em nenhuma das duas áreas. Após essa confirmação, foram realizadas pulverizações com B. bassiana em ambas as áreas, sendo que em uma ocorreram duas e na outra três aplicações. Após a última pulverização, realizada em fevereiro/2006, verificou-se que os inóculos do fungo persistiram por 90 dias na área com duas aplicações e 120 dias na área com três aplicações. A temperatura afetou a persistência do fungo no solo em ambas as áreas, mas a umidade relativa não mostrou influência sobre os inóculos em campo.Palavras-chave: Broca-da-erva-mate; controle biológico; epizootia; fungo entomopatogênico. AbstractPersistence in soil of inoculum of Beauveria bassiana used to control Hedypathes betulinus in yerba maté plantation, Ilex paraguariensis. Control measures of Hedypathes betulinus are restricted to agricultural practices and hand picking of adult insects. No chemical control is allowed in yerba maté cultivation areas. The objectives of this research were to verify the natural occurrence of the entomopathogenic fungus Beauveria bassiana in the soil of yerba maté in monoculture; to determine the persistence of inoculums in the soil after applications of the fungus; and to evaluate the influence of abiotic factors on fungus stability in the soil. The soil samples were taken from two experimental areas in a yerba maté plantation in the county of Campo Alegre, Santa Catarina, Brazil. The samples were analyzed in order to count, isolate and identify inoculums of B. bassiana and other fungi. After the confirmation that B. bassiana was not present in the soil, a suspension of the fungus was applied, twice in one of the areas and three times in the other. After the last application in February/2006, the inoculum persisted for 90 and 120 days, in the area with two and three applications, respectively. The temperature affected the persistence of B. bassiana in soil; however, the relative humidity did not.Keywords: Biological control; epizooty; entomopathogenic fungus; wood borer.

Sixteen soil samples were collected from wheat, barley and yellow corn rhizosphere in Abu-Ghraib, Aqraqof, Latifieh,Tarmiah, Jadriya and  of Agriculture in Baghdad university/ Baghdad city. The results found nine phosphate solubilizing bacteria (PSB) isolates (Y1, Y2, Y3, Y4, Y5, Y6, Y7, Y8, Y9), formed clear zones on National Botanical Research Institute's (NBRIP) agar. The solubility index (SI) of PSB isolates ranged from 2.00 to 3.66. Y4 have the highest SI (3.66) followed by Y3 and Y6 (3.33). Phosphate solubilization abilities varying from (20.10-39.00 μg.ml-1), Y4 was the highest (39.00 μg.ml-1) followed by Y3 (37.00μg.ml-1). The results of hydrolytic enzymes production showed that almost all nine isolates are able to produce protease and pectinase, while Y1 and Y2 showed negative results in cellulase production. Maximum ability for hydrogen cyanide (HCN) and indole acetic acid (IAA) production were showed byY3 and Y4 isolates. The isolate Y4 was found to be the most efficient isolate, so it was selected identified as Bacillus cereus using biochemical tests confirmed by VITEC 2 compact system. The results of High performance liquid chromatography (HPLC) revealed that Bacillus cereus produce oxalic acid (2.996), citric acid (9.117) and malic acid (3.734). Bacillus cereus (Y4) enhanced the growth of mung bean plants. A significant increase in branches number (12.33), plant length (83.0cm), fresh weight (27.25 g) and dry weight (1.427g) were obtained compared with control treatments. The main objective of this study is to isolate PSB and evaluate their roles in plant growth promotion. The results showed the high phosphate solubilization efficiency of PSB isolates and the identified isolates was found to be good enough for plant growth promoting.

Endophytic fungi (EF) can enhance both plant growth and defense barriers against pests and pathogens, contributing to the reduction of chemical pesticides and fertilizers use in agriculture. Beauveria bassiana is an entomopathogenic fungus showing endophytism in several crops, often associated with a good capacity to limit the development of pests and disease agents. However, the diversity of the protective efficacy and plant response to different strains can be remarkable and needs to be carefully assessed for the successful and predictable use of these beneficial microorganisms. This study aims to select B. bassiana strains able to colonize tomato plants as endophytes as well as to control two important disease agents, Botrytis cinerea and Alternaria alternata, and the pest aphid, Macrosiphum euphorbiae. Nine wild-type isolates and one commercial strain were screened for endophytism, then further characterized for plant-growth promotion plus inhibition of disease development and pest infestation. Four isolates proved to have a good control activity against the biotic stressors tested, but only Bb716 was also able to promote plant growth. This work provides a simple workflow for the selection of beneficial EF, paving the way towards more effective use of B. bassiana in Integrate Pest Management (IPM) of tomato.

Fourteen isolates of soil bacteria, including two known plant growth promoting rhizobacteria (PGPR) strains, Azotobacter vinelandii Mac 259 and Bacillus cereus UW 85, were tested in vitro. Parameters assessed were indoleacetic acid (IAA) production, phosphate solubilization, dinitrogen fixation, and siderophore (Fe-III chelating agent) production. IAA production was assayed colorimetrically using ferric chlorideperchloric acid reagent. Phosphate-solubilization and siderophore production were tested qualitatively by plating the bacteria in Pikovskaya and chrome azurol S agar, respectively. The ability to fix dinitrogen was measured based on nitrogenase activity of the bacteria by gas chromatography. The results showed that twelve isolates produced IAA, ranged from 2.09 to 33.28 µmol ml-1. The ability to solubilize precipitated phosphate was positively exhibited by four isolates (BS 58, BTS, TCaR 61, and BTCaRe 65). Seven isolates including Mac 259 positively produced siderophore. None of the isolates showed nitrogenase activity. Only one isolate (TS 3) did not exhibit any of the traits tested. Isolate TCeRe 60 and reference strain Mac 259 were found to have IAA- and siderophore-producing traits. Four P-solubilizing bacteria (BS 58, BTS, TCaR 61, and BTCaRe 65) were also IAA- and siderophore-producing bacteria. Potential use of these PGPR isolates needs further test in enhancing plant growth.

Background: The plant growth promotion and efficacy against phytopathogens by the endophytic bacteria are being focused now due to their ecofriendly nature. Methods: Endophytic bacteria (24 nos) isolated from the roots, stems and leaves of black gram plants collected from different locations were tested for their potential to inhibit the growth of R. bataticola under in vitro and in vivo conditions. Result: The leaf endophyte BLE 4 exhibited maximum inhibition (79.6%) of R. bataticola followed by BSE 4 (77.4%), BSE 7 (77.0%) and BLE 1 (74.0%). Among these 4 isolates tested as seed treatment and soil application, there was significant increase in dry weight (7.1 g), plant height (37.7 cm), number of branches (13.2) and number of pods (26.2) in BLE 4 treated plots. Whereas, the incidence of dry root rot and yield were insignificant. In the screening study of isolates for their phosphate solubilization potential, protease activity, siderophore and HCN production, no single isolate possessed all the properties, but siderophore production, protease activity and phosphate solubilization were found in BSE 4, BRE 3, BRE 5 and BRE 10 isolates.
