The practical use of knowledge on the diagnostic-prognostic role of polysaccharide components of mucins in colorectal cancer (CRC) has been difficult, due to the number of histochemical (HC) reaction types, as well as lack of standard methods of computer-assisted analysis of tissue expression of these molecules. Using two algorithms of digital image analysis (by application of Image-Pro Premier and our originally designed program Filter HSV), we evaluated the expression of polysaccharides in tissue samples of CRC patients (n = 33), and fragments of normal colorectal tissue from the same patients (control) using periodic acid Schiff reaction (PAS) (neutral mucins) and alcian blue staining (AB) (acidic mucins). Our results indicate lower expression of the PAS+ and AB+ mucins in CRC, as compared to the control samples. The higher expression of PAS+ polysaccharides was detected in flat tumors than in protruded CRC, while higher AB+ mucins expression was a feature of mucinous CRC subtypes. Positive correlation between mutual PAS+ and AB+ expression, as well as correlations with glucose concentration (PAS+ mucins), and hemoglobin level (AB+ mucins) were observed exclusively in unchanged colorectal samples (control). Both algorithms of digital image analysis (smart segmentation and Filter HSV) work properly and can be used interchangeably in daily practice of pathologists, as useful tools of quantitative evaluation of HC reaction in both normal and cancerous tissues.

Parathyroid extract, a total of 860 units, has been administered to rats in small, progressively increasing doses over a period of 12 days. The effects on serum protein, total glycoprotein, glycoprotein/protein ratio, calcium, individual protein and glycoprotein fractions, and renal calcification have been presented. Kidney sections from each rat were studied by histochemical techniques for calcification, neutral polysaccharide, and acid mucopolysaccharide. The serum total glycoprotein, glycoprotein/protein ratio, and calcium were found to be elevated. The serum albumin was decreased, whereas the globulin fractions were essentially unchanged. The globulin glycoprotein hexose, mostly α1-globulin, was increased; albumin glycoprotein hexose showed a somewhat less consistent increase. A "precalcification" periodic acid-Schiff-staining intraluminal material was demonstrated in the kidney after parathyroid extract treatment for 4 days. This was followed approximately 2 days later by Alcian blue-staining material and calcification demonstrable by hematoxylin and eosin and by Kóssa staining techniques. This has been interpreted as suggesting a glycoprotein-containing lesion which precedes calcification.

A severe, progressive myopathy developed in an 11-year-old, phosphofructokinase (PFK)-deficient, male, English Springer Spaniel dog. Results from a routine neurological examination were normal. Examination of histologic sections of skeletal muscle revealed large accumulations of material in some myofibers. These deposits were pale, basophilic, somewhat flocculent, and slightly granular with hematoxylin and eosin stain. Most fascicles examined in sections of limb and trunk muscles were affected to some degree, with up to 10% of muscle fibers being involved. Deposits stained strongly with periodic acid-Schiff and were resistant to digestion by alpha amylase but were removed by incubation with gamma amylase. Deposits were faintly positive with Gomori's methenamine silver technique and alcian blue (pH 2.5) and were brown-gray with Lugol's iodine solution but were negative with other stains. Based on staining characteristics, the deposits seemed to consist primarily of an amylopectin-like polysaccharide(s). Alcian blue staining (pH 2.5) was removed by treatment with neuraminidase but not with hyaluronidase, indicating that some sialic acid residues were also present. Electron microscopically, the deposits were composed of short granular filaments, small granules and amorphous material. They were not membrane bound. The morphologic appearance and staining characteristics of the deposits were remarkably similar to deposits previously described in human PFK-deficient myopathy. As expected, total PFK activities were markedly reduced when assayed in skeletal muscles of this dog. In contrast with other PFK-deficient dogs, muscle glycogen in this animal was not increased above that of normal dogs.

Traditionally, vegetative cover has been subjectively assessed by visual assessment. However, visual assessment is thought to overestimate percent vegetative cover. Thus, a repeatable method to objectively quantify percent cover is desirable. In two vineyards near Prosser, Wash., the percentage of ground surface covered by up to 15 different cover crops was assessed both visually and by computer-assisted digital image analysis. Quadrats in the cover crop were photographed digitally and the images analyzed with commercially available software. Areas of green vegetation in each image were identified and measured. Weeds in some images were differentiated from the cover crop by user-defined thresholds. Subjective visual estimates of percent vegetative cover were generally higher than those digitally estimated. Values for the visual estimates ranged from 5% to 70% in 1998 (mean = 52.4%) and 7.5% to 55% in 1999 (mean = 30.7%), compared to digital readings ranging from 0.5% to 24% (mean = 11.1%) and 10.3% to 36.6% cover (mean = 20.1%), respectively. The visual assessments had lower coefficients of variability in 1998 (cv 28.1) than the digital image analysis (cv 52.3), but in 1999, the values for the two techniques were similar (cv 41.2 vs. cv 45.7). Despite initial variations between the two methods, the accuracy of digital image analysis for measuring percentage vegetative cover is superior.

The early infection and colonization processes of Colletotrichum acutatum on leaves and petals of two almond cultivars with different susceptibility to anthracnose (i.e., cvs. Carmel and Nonpareil) were examined using digital image analysis of light micrographs and histological techniques. Inoculated tissue surfaces were evaluated at selected times after inoculation and incubation at 20°C. Depth maps and line profiles of the digital image analysis allowed rapid depth quantification of fungal colonization in numerous tissue samples. The results showed that the early development of C. acutatum on petals was different from that on leaf tissue. On petals, conidia germinated more rapidly, germ tubes were longer, and fewer appressoria developed than on leaves. On both tissues, penetration by the pathogen occurred from appressoria and host colonization was first subcuticular and then intracellular. On petals, colonizing hyphae were first observed 24 h after inoculation and incubation at 20°C, whereas on leaves they were seen 48 to 72 h after inoculation. Intercellular hyphae were formed before host cells became necrotic and macroscopic lesions developed on petals ≥48 h and on leaves ≥96 h after inoculation. Histological studies complemented data obtained by digital image analysis and showed that the fungus produced infection vesicles and broad hyphae below the cuticle and in epidermal cells. In both tissues, during the first 24 to 48 h after penetration fungal colonization was biotrophic based on the presence of healthy host cells adjacent to fungal hyphae. Later, during intercellular growth, the host-pathogen interaction became necrotrophic with collapsed host cells. Quantitative differences in appressorium formation and host colonization were found between the two almond cultivars studied. Thus, on the less susceptible cv. Nonpareil fewer appressoria developed and host colonization was reduced compared with that on cv. Carmel.

BACKGROUND Detection of mucins has been shown to correlate with several clinicopathological characteristics in patients. Currently, periodic acid-Schiff (PAS) and alcian blue staining methods are the histochemistry staining techniques that are frequently used to detect mucin. This study was aimed to evaluate PAS and alcian blue staining in differentiating mucin characteristics between invasive carcinoma of no special type (ICNST) with mucinous degeneration and mucinous carcinoma. METHODS This cross-sectional study of 33 cases included biopsies of mucinous breast carcinoma and ICNST with mucin degeneration that were histologically verified using hematoxylin and eosin (H&E) staining. The PAS and alcian blue staining were conducted in the Laboratory of Histochemistry, Department of Anatomical Pathology, Cipto Mangunkusumo Hospital. Data were recorded using SPSS software, version 21 (IBM Corp, USA). RESULTS There were 17 cases of ICNST with mucinous degeneration and 16 cases of mucinous carcinoma with age varied from 27 to 75 years. PAS had sensitivity of 87.5% and specificity of 41.2%. Alcian blue had sensitivity of 43.8% and specificity of 82.4%. CONCLUSIONS PAS staining method is better than the alcian blue staining method in distinguishing between ICNST with mucinous degeneration and mucinous carcinoma. In the limited setting laboratory, PAS staining alone can be considered to detect mucin.
