The growth hormone is involved in skin homeostasis and wound healing. We hypothesize whether it is possible to improve pressure ulcer (PU) healing by locally applying the recombinant human growth hormone (rhGH) in a human skin mouse model. Non-obese diabetic/severe combined immunodeficient mice (n = 10) were engrafted with a full-thickness human skin graft. After 60 days with stable grafts, human skin underwent three cycles of ischemia-reperfusion with a compression device to create a PU. Mice were classified into two groups: rhGH treatment group (n = 5) and control group (n = 5). In the rhGH group for local intradermal injections, each had 0.15 mg (0.5IU) applied to the PU edges, once per week for four weeks. Evaluation of the wound healing was conducted with photographic and visual assessments, and histological analysis was performed after complete wound healing. The results showed a healing rate twice as fast in the rhGH group compared to the control group (1.25 ± 0.33 mm2/day versus 0.61 ± 0.27 mm2/day; p-value < 0.05), with a faster healing rate during the first 30 days. The rhGH group showed thicker skin (1953 ± 457 µm versus 1060 ± 208 µm; p-value < 0.05) in the repaired area, with a significant decrease in collagen type I/III ratio at wound closure (62 days, range 60–70). Local administration of the rhGH accelerates PU healing in our model. The rhGH may have a clinical use in pressure ulcer treatment.

Abstract Background A burn is a thermal injury caused by biological, chemical, electrical and physical agents with local and systemic repercussions. There are several ways of classifying burns: Classification by mechanism or cause, depth and extent of burn . Objectives The objective of this study was to determine the safety and efficacy of using recombinant human growth hormone (rhGH) in the treatment of pediatric burn victims and their probable effect on accelerating burn wound healing. Patients and Methods This study was an Interventional randomized controlled Double Blind Study in which Patients subdivided randomly into 2 groups: Group A received somatotropine hormone after their 3 days of resuscitation besides their conventional treatment during their stay in the Burn ICU. Group B received the conventional treatment only in the Burn ICU. Results The comparison between the GH group and the control group showed that that there was statistically significant difference found between the two studied groups regarding TBSA of burn at 3rd week. The mean TBSA in GH group was ( 9.06 ± 7.47 ) while in the control group (13.94 ± 11.96) with P value (0.041). There was highly statistically significant difference found between the two studied groups regarding Insulin like growth factor .the mean Insulin like growth factor in GH group was (16.48 ± 11.40) while in the control group(2.77 ± 0.64) with P value(0.000). Conclusion The use of recombinant Growth hormone with a dose of 0.2 mg/Kg SQ 2 days per week with 3 days time interval in pediatric burn patients after their primary resuscitation from the burn injury, shows a marvelous improvement concerning the total body surface area of burn(TBSA) as the patient received the growth hormone showed a decrease total body surface area of burn(TBSA) than the control group. This may be accounted for the faster wound healing and readiness for grafting .

In the last few decades, the incidence of caesarean section is increasing in the world, especially in Indonesia. One of the way to treat tissue scar is through biologic and synthetic dressing where nowadays, amnion has been used as biologic dressing frequently. This study was conducted to determine the effect of the use of fresh amniotic membrane on wound incision Caesarean section compared with Caesarean section incision wound covered using regular gauze bandages and fixated with plaster in RS. Dr. Reksodiwiryo Padang. The design of this study is an experimen-tal study with Post test design with control group design. Sampling was done using a formula consecutive sampling two different test samples obtained an average of 72 people for each group. The analysis used include univariate and bivariate analyzes. The average wound healing time the difference was statistically significant (p value <0.05) in the treatment and control groups. There was highly significant difference in the proportion of local infection on day 3 between the treatment and control groups (p value <0.05). There were very significant differences in the proportion of local allergic reactions at day 3, and 5 between the treatment and control group (p <0.05). There are significant differences in terms of the cost of care per day between treatment and control groups (p <0.05). Pressure ulcer or pressure sore is one of the complications seen in bedridden patients. Management of these ulcers is often challenging. But there is no well-established method that accelerates the wound healing rate. Various adjunctive methods are used for wound bed preparation before definitive reconstruction plan is made. Here we describe our experience in the role of insulin therapy as an adjunct in the management of pressure sores.

Diabetic foot ulcers (DFUs) are a serious complication in diabetic patients and lead to high morbidity and mortality. Numerous dressings have been developed to facilitate wound healing of DFUs. This study investigated the wound healing efficacy of silver-releasing foam dressings versus silver-containing cream in managing outpatients with DFUs. Sixty patients with Wagner Grade 1 to 2 DFUs were recruited. The treatment group received silver-releasing foam dressing (Biatain® Ag Non-Adhesive Foam dressing; Coloplast, Humlebaek, Denmark). The control group received 1% silver sulfadiazine (SSD) cream. The ulcer area in the silver foam group was significantly reduced compared with that in the SSD group after four weeks of treatment (silver foam group: 76.43 ± 7.41%, SSD group: 27.00 ± 4.95%, p < 0.001). The weekly wound healing rate in the silver foam group was superior to the SSD group during the first three weeks of treatment (p < 0.05). The silver-releasing foam dressing is more effective than SSD in promoting wound healing of DFUs. The effect is more pronounced in the initial three weeks of the treatment. Thus, silver-releasing foam could be an effective wound dressing for DFUs, mainly in the early period of wound management.

Bone is a complex tissue consisting of cells and matrix. Mass and thick bone mass has a dynamic addition and subtraction through the process of remodeling (bone matrix absorbed and formed again). Bone is formatted by osteoblast cell and resorption by osteoclast cell. Osteoblasts produce a matrix of osteoid, which is composed mainly of type I collagen, and osteoclast removes bone tissue by removing its mineralized matrix and breaking up the organic bone. Bone remodeling purpose to defend shape and structure of bone. the purpose of this study is to prove that submaximal-intensity exercise and salmon calcitonin improve bone density in growing rat this research method uses design of the randomize posttest only control group design. We compered femur bone density in 24 male norvegicus rats aged 6 weeks that were divided into 4 groups: controls, calcitonin, exercise, combine. Exercise group swam 3 times a week in submaximal intensity, calcitonin group injected synthetic salmon calcitonin 2 iu /100 gram of rat weight every day and combine group did both of it. After 8 weeks, rat femur bone density measured using ultrasound. the result: there are significant differences in bone density between group 1 (control) and group 4 (combine) with p = 0.001, thus the p value &lt;0.05 indicates that there is a significant difference to the average density in both groups. While comparisons to other groups found no significant difference because the value of p&gt; 0.05. the benefits of this research are calcitonin salmon and submaximal-intensity exercise increase the density bone in the growth period. High bone density is mean the bone is strong and health, not porous and fragile so decrease bone fracture risk. increase the bone density in of growth period make the bone get the best mass, and avoid from early osteoporosis.

Infeksi nifas menjadi salah satu penyebab kematian ibu di negara berkembang termasuk Indonesia. Beberapa penyebab pemicu terjadinya infeksi nifas seperti pelayanan kebidanan, daya tahan tubuh, perawatan nifas, gizi, anemia dan kebersihan genetalia. Pada masa nifas penting untuk merawat luka perineum karena kuman bisa masuk melalui luka bekas jahitan pada perineum agar tidak menyebabkan infeksi. Salah satu alternatif terapi nonfarmakologis yang dapat digunakan untuk penyembuhan luka perineum masa nifas adalah daun binahong. Tujuan penelitian ini menguji pengaruh daun binahong dalam mempercepat penyembuhan luka perineum pada masa nifas. Penelitian ini menggunakan rancangan quasi experimental dengan pendekatan posttest only control group design. Posttest only control group design merupakan pengumpulan data dilakukan sesudah perlakuan. Total sampel yang digunakan dalam penelitian ini adalah 32 orang ibu nifas sesuai dengan kriteria inklusi. Data diobservasi berupa derajat kesembuhan luka perineum dengan menggunakan instrumen lembar observasi penyembuhan luka perineum pada skala REEDA (Redness, Edema, Ecchymosis, Discharge dan Approximation). Hasil penelitian menunjukan terdapat pengaruh pemberian rebusan Daun binahong terhadap penyembuhan luka perineum pada ibu nifas. Rerata hasil uji Uji statistik Mann Whitney, pada kelompok perlakuan adalah 11,83 lebih rendah dibandingkan pada kelompok kontrol, yaitu 21,8 (p-value = 0,001). Penelitian ini menunjukan bahwa ada pengaruh pemberian rebusan daun binahong mampu mempercepat penyembuhan luka perineum dibandingkan iodine povidone 10%. Daun binahong dapat menjadi alternatif terapi komplementer pada ibu nifas yang mengalami luka robekan perineum.   One of the main causes of maternal death in developing countries is puerperal infection. Midwifery care, body immunity, postnatal care, nutrition, anemia and genetalia hygiene are factors that trigger puerperal infections. The perineal rupture if not treated properly will cause an infection in the suture wound in the perineum. Wound care is important because germs can enter through the suture marks on the perineum and cause infection. Non-pharmacological therapies that can be used to accelerate wound healing are Anredera cordifolia. The research design was quasi experimental with posttest approach Only Control Group Design, namely data collection was done after treatment. The total sample was 32 postpartum mothers according to inclusion criteria. The data observed in this study were the degree of healing of perineal wounds using the REEDA scale perineal wound healing observation sheet instrument (Redness, Edema, Ecchymosis, Discharge and Approximation). Based on the results of the Mann Whitney test, the mean rank of the results in the treatment group was 11.83 lower than the control group, namely 21.8 (p-value = 0.001). Effect of giving Anredera cordifolia leaves on the healing of perineal wounds in postpartum mothers significantly. Care with giving Anredera cordifolia leaves can accelerate the healing of perineal wounds compared to 10% iodine povidone. Anredera cordifolia leaves can be used as a complementary therapy for postpartum mothers who experience perineal torn wounds.

AbstractThe purpose of this study was to evaluate whether cryopreserved (frozen) adipose-derived regenerative cells (ADRCs) have a therapeutic effect on burn wound healing as well as freshly isolated (fresh) ADRCs.Full thickness burns were created on dorsum of nude mice and burn wound was excised. The wound was covered by artificial dermis with; (i) fresh ADRCs, (ii) frozen ADRCs, and (iii) PBS (control). The assessment for wound healing was performed by morphological, histopathological and immunohistochemical analyses.In vivo analyses exhibited the significant therapeutic effect of frozen ADRCs on burn wound healing up to the similar or higher level of fresh ADRCs. There were significant differences of wound closure, epithelized tissue thickness, and neovascularization between the treatment groups and control group. Although there was no significant difference of therapeutic efficacy between fresh ADRC group and frozen ADRC group, frozen ADRCs improved burn wound healing process in dermal regeneration with increased great type I collagen synthesis compared with fresh ADRCs.These findings indicate that frozen ADRCs allow us to apply not only quickly but also for multiple times, and the cryopreserved ADRCs could therefore be useful for the treatment of burn wounds in clinical settings.

ABSTRACT Background: Chronic wounds in patients with Diabetes Mellitus often become incurable due to prolonged and excessive production of inflammatory cytokines. The use of probiotics modifies the intestinal microbiota and modulates inflammatory reactions. Aim: To evaluate the influence of perioperative supplementation with probiotics in the cutaneous healing process in diabetic rats. Methods: Forty-six rats were divided into four groups (C3, P3, C10, P10) according to the treatment (P=probiotic or C=control, both orally administered) and day of euthanasia, 3rd or 10th postoperative days. All rats were induced to Diabetes Mellitus 72 h before starting the experiment with alloxan. Supplementation was initiated five days before the incision and maintained until euthanasia. Scalpel incision was guided by a 2x2 cm mold and the wounds were left to heal per second-intention. The wounds were digitally measured. Collagen densitometry was done with Picrosirius Red staining. Histological parameters were analyzed by staining by H&E. Results: The contraction of the wound was faster in the P10 group which resulted in a smaller scar area (p=0.011). There was an increase in type I collagen deposition from the 3rd to the 10th postoperative day in the probiotic groups (p=0.016), which did not occur in the control group (p=0.487). The histological analysis showed a better degree of healing in the P10 group (p=0.005), with fewer polymorphonuclear (p<0.001) and more neovessels (p=0.001). Conclusions: Perioperative supplementation of probiotics stimulates skin wound healing in diabetic rats, possibly due to attenuation of the inflammatory response and increased neovascularization and type I collagen deposition.

The isolated effect of growth hormone on carbohydrate metabolism in rat skeletal muscle was studied in growth hormone-deficient dwarf rats (dw/dw) treated with either recombinant human growth hormone or saline for 10 days. In addition, age-matched heterozygous (DW/dw) (normal weight and plasma IGF-I) control rats were treated with saline. Growth hormone increased weight gain from 0.1+/-0.1 (s.e.m) to 3.6+/-0.1 g/day and plasma IGF-I concentration from 364+/-23 to 451+/-32 ng/ml. Glucose metabolism in skeletal muscle perfused with basal, submaximal and maximal concentrations (0, 600 and 60 000 pmol/l respectively) of insulin was not changed by growth hormone. No change could be detected in the total number of glucose transporters (GLUT1 and GLUT4) in the skeletal muscles, except from a lower amount of GLUT4 in the soleus muscle in the heterozygous control group. However, at submaximal insulin concentrations, skeletal muscle glucose uptake and transport were significantly lower in the heterozygous control group compared with the growth hormone-deficient group. This could indicate either a direct long-term effect of growth hormone or more likely a secondary effect attributable to the difference in body weight (205.2+/-3.1 vs 361. 6+/-5.9 g for dwarf rats and heterozygous controls respectively), and thereby muscle fibre size, between the groups probably resulting in lower average interstitial insulin and glucose concentrations at a given plasma concentration in the heterozygous rats. It is concluded that restoration of subnormal growth hormone concentrations for 10 days has no effect on insulin-stimulated glucose metabolism in skeletal muscle in vitro.

Tooth extraction is a process of removing teeth from the alveolar bone. In wound healing, fibroblast are very important cells. The main purpose of this study was to determine the effect of mobe leaf 1% and 3% extract gel (Artocarpus lakoocha) on fibroblast proliferation in post extraction tooth socket wound healing. This research used 16 samples of wistar rats, divided into 4 groups, a positive control group, a negative control group and a 1% and 3% mobe leaf extract gel group. The left mandibular incisors were extracted, then 1% and 3% gels of mobe leaf extract were applied on day 1 to day 7. Data analysis was calculated using the Kruskal-Wallis test on clinical data and one way ANOVA test for microscopic. The result of the socket wound healing activity test for a good concentration of mobe leaf extract gel was 3%. This research shows significant resultith p-value of 0.018 (< 0.05) on the closure of the socket wound clinically which means the closure of the wound accelerates because of the mobe leaf 3% extract gel treatment. The distance of fibroblast on microscopically shows significant resultith a p-value of 0.002 (< 0.05), which means that there was an enlargement of the distance fibroblast at the socket wound closure with application of mobe leaf 3% extract gel. From the results of the study it can be concluded that mobe leaf 3% extract gel has the best ability to show acceleration the closure of the socket wound either clinically or microscopically.
