Trace elements (As, Cd, Cr, Pb, Ba, Fe, Al, Mn and Ba) were uptaken by the leaves of the creosote bush (Larrea tridentata (DC.) Coville) in Nelson, Nevada, although at low concentrations. Samples were collected up-gradient of the mine tailings, the tailings, and down gradient from the source to measure spatial distribution. Data show that trace elements (As, Ba, Cr, Hg, Se) enter L. tridentata through root tissues, migrating to leaf tissue, but at significantly lower levels than that of the source sediments. Metalloid (As and Se) concentrations in the leaf tissues ranged from non-detect to greater than 44 mg kg−1 As and non-detect to over 34 mg kg−1 Se. For trace metals, Hg ranged from non-detect to 0.14 mg kg−1; Ba from 1.74 to 4.12 mg kg−1; and Cr from non-detect to 6.18 mg kg−1 while Ag, Cd, and Pb were not detected in the plant leaves. When comparing the ratio of sediment metal concentration to plant metal concentrations, the Techatticup Wash contained the highest levels of trace elements in the leaves of the L. tridentata, followed by the Carnation Wash, with the Eagle Wash containing the lowest concentrations of trace elements.

Vitis vinifera 'Pinot' clones were analysed at 50 microsatellite loci to assess intravarietal genetic diversity. When analysing leaf tissue DNAs, polymorphism mainly resulted from the appearance of a third allele when two were expected for heterozygous loci in a diploid species. The sequencing of the three microsatellite alleles at two loci has confirmed their simultaneous presence in the leaf tissues. A hypothesis explaining the triallelic profiles at a locus is the presence of a periclinal chimera meristem structure, in which genetically different cell layers coexist. The periclinal chimeric state of two Vitis vinifera 'Pinot gris' clones was confirmed by splitting and analysing the genotypes resulting from L1 and L2 cell layers in progeny derived from self-fertilization, in root tissues, and in plants regenerated from somatic embryogenesis. Prevalence of chimerism in polymorphic clones observed in a collection of 145 accessions belonging to 'Pinot gris', 'Pinot noir', Pinot blanc', 'Pinot meunier', and 'Pinot moure' cultivars was demonstrated. The accumulation of somatic mutations and cell layer rearrangements allowed us to deduce the relationships between the various genotypes and to open a way for understanding the diversification process and the phylogeny in the 'Pinot' group.Key words: microsatellite, diversity, somatic mutation, chimerism, Vitis vinifera L., 'Pinot'.

Selenium and sulfur have similar chemical structures. This allows Se to be absorbed and incorporated in the same assimilation pathways as S. Onions (Allium cepa L.) are a crop with unique S metabolism, responsible for growth and flavor intensity. Because of the antagonistic behavior of the two ions, the effects of Se on S and Se nutrient depletion and tissue accumulation were investigated. `Granex 33' onions were grown in nutrient solutions with one concentration of S and increasing Se concentrations. Selenium was applied as sodium selenate (Na2SeO4) at concentrations of 0, 0.5, 1.0, 1.5, and 2.0 mg·L-1. Selenium depletion from the nutrient solution increased linearly with increasing Na2SeO4 treatment concentrations. Sulfur depletion increased and then decreased with increasing Na2SeO4 treatment concentrations. Selenium and S accumulation were highest in leaf tissues, less in root tissues, and lowest in bulb tissues at plant maturity. Selenium accumulation increased linearly with increasing Na2SeO4 for all tissues analyzed. Sulfur accumulation in leaf and bulb tissues was quadratic in response to increasing SeO4-2, while S in root tissues decreased linearly with increasing Na2SeO4. Low concentrations of Na2SeO4 in our study enhanced S uptake and accumulation. Previously, Se was thought to competitively inhibit S uptake and metabolism.

RNA-binding proteins (RBPs) play a crucial role in regulating RNA function and fate. However, the full complement of RBPs has only recently begun to be uncovered through proteome-wide approaches such as RNA interactome capture (RIC). RIC has been applied to various cell lines and organisms, including plants, greatly expanding the repertoire of RBPs. However, several technical challenges have limited the efficacy of RIC when applied to plant tissues. Here, we report an improved version of RIC that overcomes the difficulties imposed by leaf tissue. Using this improved RIC method in Arabidopsis leaves, we identified 717 RBPs, generating a deep RNA-binding proteome for leaf tissues. While 75% of these RBPs can be linked to RNA biology, the remaining 25% were previously not known to interact with RNA. Interestingly, we observed that a large number of proteins related to photosynthesis associate with RNA in vivo, including proteins from the four major photosynthetic supercomplexes. As has previously been reported for mammals, a large proportion of leaf RBPs lack known RNA-binding domains, suggesting unconventional modes of RNA binding. We anticipate that this improved RIC method will provide critical insights into RNA metabolism in plants, including how cellular RBPs respond to environmental, physiological and pathological cues.

Copper uptake, accumulation in different tissues and organs and biochemical and physiological parameters were studied in Erica australis treated with different Cu concentrations (1, 50, 100 and 200 µM) under hydroponic culture. Copper treatments led to a significant reduction in growth rate, biomass production and water content in shoots, while photosynthetic pigments did not change. Copper treatments led to an increase in catalase and peroxidase activities. Copper accumulation followed the pattern roots > stems ≥ leaves, being roots the prevalent Cu sink. Analysis by scanning electron microscopy coupled with elemental X-ray analysis (SEM–EDX) showed a uniform Cu distribution in root tissues. On the contrary, in leaf tissues, Cu showed preferential storage in abaxial trichomes, suggesting a mechanism of compartmentation to restrict accumulation in mesophyll cells. The results show that the studied species act as a Cu-excluder, and Cu toxicity was avoided to a certain extent by root immobilization, leaf tissue compartmentation and induction of antioxidant enzymes to prevent cell damage.

Six different Japanese Maples (Acer palmatum) cultivars `Water Fall', `Burgundy Lace', `Crimson Queen', `Oshio-Beni', `SangoKaKu', and `Bloodgood' from Monrovia Nursery were planted in a randomized block design on 4 June 1997 at the The Morton Arboretum. Leaf heat tolerance was evaluated by measuring ion leakage of the leaf tissue at 25–60 °C in July, Aug., and Sept. 1997. The LT50 (the temperature at which 50% of the tissues were injured) of all the cultivars were higher in July (≈53 °C) and were lower in September (≈47 °C). Water content of the leaf tissues were higher in July compare to August and September and were not related to heat tolerance of most cultivars. Stem cold hardiness was performed by artificial freezing tests in Oct., Dec., and Feb. 1997/98. The Lowest Survival Temperature (LST) for the most hardy to least hardy cultivars in October and December were: `Burgundy Lace' (–15, –27 °C), `Bloodgood' (–18, –24 °C), `Oshio-Beni' (–15, –24 °C), `Crimson Queen' (–15, –18 °C), `Water Fall' (–9, –18 °C) and `SangoKaKu' (–9, –12 °C), respectively. Growth, dormancy development, spring budbreak and performance of these cultivars will be compared.

ABSTRACT Significantly fewer thraustochytrid protists (zoosporic fungi) were observed in association with healthy leaf tissue of the marine angiosperm Thalassia testudinum than in association with sterilized samples that were returned to the collection site for 48 h. In support of the hypothesis that sea grass secondary metabolites were responsible for these differences, extracts of healthyT. testudinum leaf tissues inhibited the growth of the co-occurring thraustochytrid Schizochytrium aggregatum and deterred the attachment of S. aggregatum motile zoospores to an extract-impregnated substrate. By using S. aggregatumfor bioassay-guided chemical fractionation, a new flavone glycoside was isolated and structurally characterized as luteolin 7-O-β-d-glucopyranosyl-2"-sulfate. Whole-leaf tissue concentrations of this metabolite (4 mg/ml of wet leaf tissue) inhibited S. aggregatum attachment, and a significantly lower concentration (270 μg/ml) reduced thraustochytrid growth by 50%, suggesting that natural concentrations are at least 15 times greater than that needed for significant microbiological effects. These results offer the first complete chemical characterization of a sea grass sulfated flavone glycoside and provide evidence that a secondary metabolite chemically defends T. testudinum against fouling microorganisms.

A new species of the encyrtid wasp genus Brethesiella Porter, 1920 is described from the Mojave Desert in California, USA. The type series of B. mojave sp. n. was reared from the margarodid scale Steatococcus tabernicolus Ferris on creosote bush, Larrea tridentata. The genus Aztecencyrtus Timberlake, 1926 is synonymized under Brethesiella and its two described species are transferred to the latter as B. flava (Timberlake, 1926) comb. n. and B. iceryae (Howard, 1892) comb. n. A brief review of the encyrtid parasitoids of Margarodidae, a diagnosis of Brethesiella, and an annotated key to its six described species with known females in the New World are provided.

Field studies characterising the forage resources of mixed-breed goats on Chihuahuan Desert rangelands were reviewed and the principal sources of variation in their diet choices discussed. Goats eat a varied diet comprising at least 126 plant species in this landscape. Goats are highly selective feeders, changing their diet from predominantly forbs (8–64%) to browse (35–88%). Graminoids make up only ~5% of the diet, but grasses could be important dietary components on rangelands in good condition. Feed intakes range between 0.8% and 3.4% of bodyweight, depending on the season. DM digestibility of forage selected by goats ranges between 44% and 65%. Annual crude protein (CP) in forages selected by goats varies from 80 to 160 g CP kg–1 DM. Overall, pregnant and lactating goats manage to ingest food that contains higher amounts of CP and lower amounts of cell wall than non-pregnant, non-lactating goats. Goats not adapted to severe shortage of forage select a diet that does not match their nutritional requirements and fetal losses occur. Goat kids select higher quality diets than do mature goats (94.5 vs 88.5 g CP kg–1 DM). Granadina goats eat ~20% more shrubs, including 3-fold more Larrea tridentata (DC) Cav., than Nubian goats. Goats with severely eroded incisors avoid grasses, focusing on tender-leaved shrubs. During the rainy season, bucks select mainly shrubs (78% of the diet) and avoid grasses (1.7% of the diet), whereas does rely heavily on forbs (about one-third of their diet). Alternating use of grazing grounds increases shrub ingestion by 25%. No evidence exists indicating that food choice by goats in this arid environment is biased towards forages with lower secondary compounds. It is concluded that the flexible, broad-scale and opportunistic feeding behaviour of goats make them an effective animal to sustainably exploit the forage resources of the Chihuahuan Desert.

Abstract Cervidellus sonorensis n. sp. is described from sand near the roots of creosote bush (Larrea tridentata) from Anza-Borrego Desert State Park, CA, USA. It is distinguished from others in the genus by the combination of a greatly distended, guitar-shaped corpus and the morphology of the lips and labial probolae. Distinctive features include a large pharyngeal metacorpus with a double swelling, a knob-like ledge at the base of each labial probola and lips with five filamentous tines, the most apical of which are those closest to the primary axil. One male individual was discovered in this parthenogenetic species. Characters used to diagnose C. sonorensis n. sp. are not definitive with respect to other genera and are probably plesiomorphic or convergent in light of DNA-based phylogenetic hypotheses. The position of C. sonorensis n. sp. and morphologically close congeners in relation to species of Nothacrobeles and Paracrobeles is discussed.
