There is a lack of studies on the root-associated bacterial microbiome of cassava plants. The identification and characterization of rhizobacteria can contribute to understanding the adaptation of the agriculturally important crop plants to abiotic stress. Rhizobacteria play a significant role in plants, as they can alleviate the drought stress by various mechanisms that enhance the plant growth under these stressor conditions. In this study, Gram-negative bacterial strains from the plant rhizosphere of cassava Manihot esculenta Crantz CIAT MCOL1734 variety subjected to water deprivation were isolated, characterized according to their morphological properties, and then identified by VITEK® 2. An increase in the diversity, abundance, and species richness of Gram-negative rhizobacterial community was found in cassava plants subjected to water-deficit stress. In total, 58 rhizobacterial strains were isolated from cassava plants. The identification process found that the bacteria belonged to 12 genera: Achromobacter, Acinetobacter, Aeromonas, Buttiauxella, Cronobacter, Klebsiella, Ochrobactrum, Pluralibacter, Pseudomonas, Rhizobium, Serratia, and Sphingomonas. Interestingly, Pseudomonas luteola and Ocrhobactrum anthropi were rhizobacteria isolated exclusively from plants submitted to drought conditions. The cassava roots constitute a great reservoir of Gram-negative bacteria with a remarkable potential for biotechnological application to improve the drought tolerance of plant crops under water-deficit conditions.

Abstract Methyl chavicol and methyl eugenol are important phenylpropanoid compounds previously purified from basil. These compounds are significantly enhanced by the water deficit stress-dependent mechanism. Here, for the first time, pObCVOMT and pObEOMT promoters were extracted by the genome walking method. They were then cloned into the upstream of the β-glucuronidase (GUS) reporter gene to identify the pattern of GUS water deficit stress-specific expression. Histochemical GUS assays showed in transgenic tobacco lines bearing the GUS gene driven by pObCVOMT and pObEOMT promoters, GUS was strongly expressed under water deficit stress. qRT-PCR analysis of pObCVOMT and pObEOMT transgenic plants confirmed the histochemical assays, indicating that the GUS expression is also significantly induced and up-regulated by increasing density of water deficit stress. This indicates these promoters are able to drive inducible expression. The cis-acting elements analysis showed that the pObCVOMT and pObEOMT promoters contained dehydration or water deficit-related transcriptional control elements.

A preliminary morphological screening and isolation of bacterial colony from latex industrial wastewater was carried out. Bacteria colonies from latex processing wastewater were isolated from a local latex processing industry. It was found that 17 bacterial isolates had been purified grown on nutrient agar under 35˚C. The colonies were then purified and morphologically indicated via Gram staining and motility test. After morphological observation, it was identified that out of 17 isolates, 9 isolates were Gram positive and 8 isolates were Gram negative. There are 11 out of 17 colonies were rod-shaped bacterial colonies, while the other 6 colonies were cocci-shaped bacteria. There were 11 colonies of gliding bacteria, three colonies were non-motile bacteria and the other three colonies were flagellated bacteria. This study is only limited to morphological observation as the main aim of this study was to investigate the potential occurrence of viable growth in treated latex processing wastewater. The bacterial colonies were classified base on their morphological properties shown. This study has classified several genera such as Staphylococcus, Escherichia, Thiobacillus, Arthrobacter and other Genus. The growth curve of 17 isolates studied and the chemical oxygen demand were determined.
