Human mesenchymal stem/stromal cells (hMSCs) have been investigated and proven to be a well-tolerated, safe therapy for a variety of indications, as shown by over 900 registered hMSC-based clinical trials. To meet the commercial demand for clinical manufacturing of hMSCs, production requires a scale that can achieve a lot size of ~100B cells, which requires innovative manufacturing technologies such as 3D bioreactors. A robust suspension bioreactor process that can be scaled-up to the relevant scale is therefore crucial. In this study, we developed a fed-batch, microcarrier-based bioreactor process, which enhances media productivity and drives a cost-effective and less labor-intensive hMSC expansion process. We determined parameter settings for various stages of the culture: inoculation, bioreactor culture, and harvest. Addition of a bioreactor feed, using a fed-batch approach, was necessary to replenish the mitogenic factors that were depleted from the media within the first 3 days of culture. Our study resulted in an optimized hMSC culture protocol that consistently achieved hMSC densities between 2 × 105–6 × 105 cells/mL within 5 days with no media exchange, maintaining the final cell population doubling level (PDL) at 16–20. Using multiple hMSC donors, we showed that this process was robust and yielded hMSCs that maintained expansion, phenotypic characteristic, and functional properties. The developed process in a vertical-wheel suspension bioreactor can be scaled to the levels needed to meet commercial demand of hMSCs.

This article explores cost-effective mass media technologies. The experience of the use of paid access to the media content of Belarus was studied, the author also made the forecast on its future functioning. The paper provides global media industry trends and focuses on the use of messagers to promote content and increase the target audience of mass media. The research used the methods of content analysis and a written survey. During the study the author revealed that the media economically oriented technologies help to make a profit through distribution of content and formation of a loyal mass media audience.

AbstractThe intrinsic properties of Mesenchymal Stem Cells (MSCs) make them ideal candidates for tissue engineering applications as they are regulated by the different signals present in the stem cell niche. Considerable efforts have been made to control stem cell behavior by designing material system approaches to engineer synthetic extracellular matrices and/or include soluble factors in the media. This work proposes a novel and simple approach based on ion-channel stimulation to determine stem cell fate that avoids the use of growth factors (GFs). We used boron ion - essential item in cell metabolism - transported inside cells by the NaBC1-channel. Addition of boron alone enhanced MSC adhesion and contractility, promoted osteogenesis and inhibited adipogenesis. The stimulated NaBC1 promoted osteogenesis via activation of the BMP canonical pathway (comprising Smad1 and YAP nucleus translocation and osteopontin expression) through a mechanism that involves simultaneous NaBC1/BMPR1A and NaBC1/α5β1/αvβ3 co-localization,. We describe a novel function for NaBC1 as a mechanosensitive ion-channel capable of interacting and stimulating GF receptors and fibronectin-binding integrins. Our results open up new biomaterial engineering approaches for biomedical applications by a cost-effective strategy that avoids the use of soluble GFs.

The recent invention of Herd Homes has presented NZ dairy farmers with a flexible facility for feeding and standing-off stock especially during wet periods. Herd Homes are a combination of feeding platform, stand-off facility and animal shelter comprising a "greenhouse" type roof over slatted concrete floors with an underfloor manure bunker. The prototype Herd Home built in Northland for 230 cows was closely monitored during its initial year (2002/03). Different bunker media were evaluated for their ability to absorb and retain manure nutrients. The media tested were natural materials (soil, wood shavings and a combination of soil/wood shavings) that were all effective at capturing nutrients (N and S) from the bunker manure. Benefits of media amendments are greater nutrient enrichment of the final bunker manure making it more useful for subsequent land application. Use of soil as a bunker medium appears to be the most viable and cost effective option. In early 2006, manure samples were collected from Herd Home bunkers from four geographically diverse areas of NZ and analysed for nutrient content. Nutrient concentrations found reflected variability in cow numbers, usage and feed inputs. In November 2004, 10 existing Herd Home owners were surveyed as to their reasons for purchase. The farmers reported ease of farm management, "on-off" grazing, reduced pugging damage, and improved stock condition and production were some of the beneficial effects of Herd Homes. Lignin peroxidase belongs to ligninolytic enzyme group and is one of the industrial important enzymes as it has wide applications in different sectors. Lignin peroxidase is produced by submerged fermentation process which requires optimization of physical and chemical parameters to achieve higher activity and make the process cost effective. The present study aimed at the optimization of physical as well chemical parameters of production medium. The optimization includes physical parameter such as incubation time, inoculum size, temperature, pH, RPM (Rotation per minute) while chemical parameters include carbon source, nitrogen source and different mineral elements. Form the optimization study, it was observed that highest lignin peroxidase production was achieved after 72 hours of incubation at temperature 300C, pH 6 and RPM 120. Optimization of chemical parameters reveals that incorporation of sodium nitrite (9g/L) in the media gave significant increase in enzyme activity. It was found that the maximum productivity achieved after optimization was 2214 U/ml which was four times higher than process without optimized parameters.

Carbon concrete polyacrylonitrile (PAN)/lignin-based carbon fiber (CF) composites are a new promising material class for the building industry. The replacement of the traditional heavy and corroding steel reinforcement by carbon fiber (CF)-based reinforcements offers many significant advantages: a higher protection of environmental resources because of lower CO2 consumption during cement production, a longer lifecycle and thus, much less damage to structural components and a higher degree of design freedom because lightweight solutions can be realized. However, due to cost pressure in civil engineering, completely new process chains are required to manufacture CF-based reinforcement structures for concrete. This article describes the necessary process steps in order to develop CF reinforcement: (1) the production of cost-effective CF using novel carbon fiber lines, and (2) the fabrication of CF rebars with different geometry profiles. It was found that PAN/lignin-based CF is currently the promising material with the most promise to meet future market demands. However, significant research needs to be undertaken in order to improve the properties of lignin-based and PAN/lignin-based CF, respectively. The CF can be manufactured to CF-based rebars using different manufacturing technologies which are developed at a prototype level in this study.

AbstractThe application of TiAl-based alloys as an exhaust valve material would allow automotive engines to operate at higher temperatures with increased efficiencies. Development of these materials at Ford initially concentrated on the Ti-48A1-1V (at%) system. This included; 1) room and elevated temperature fatigue, 2) creep and 3) tensile ductility optimization. Results from this test program in conjunction with other available data, previous ceramic experience and limited engine testing led to the conclusion that the major roadblock to implementation was not optimizing mechanical properties, but rather low cost and reliable valve manufacturing technology. When a cost effective manufacturing technology is developed, then the emphasis will shift to ensuring the product durability. Hence, the focus of the current program is the development of valve manufacturing technologies, in particular valve blank fabrication. Currently, casting appears to be the lowest cost alternative for valve blank fabrication. This paper reviews the technology development process as it pertains to TiAl-based valves.

Surfactin produced by Bacillus subtilis BDCC-TUSA-3 from Maldex-15 was used as a growth-associated product in a conventional batch process. Maldex-15 is a cheap industrial by-product recovered during manufacturing of high fructose syrup from corn starch. Surfactin production was greatly improved in exponential fed-batch fermentation. Maldex-15 and other nutrients were exponentially fed into the culture based on the specific growth rate of the bacterium. In order to maximize surfactin yield and productivity, conversion of different quantities of Maldex-15 into surfactin was investigated in five different fermentation runs. In all runs, most of the Maldex-15 was consumed and converted into surfactin and cell biomass with appreciable efficiencies. The best results were obtained with the fermentation run supplied with 204 g Maldex-15. Up to 36.1 g l−1 of surfactin and cell biomass of 31.8 g l−1 were achieved in 12 h. Also, a marked substrate yield of 0.272 g g−1 and volumetric reactor productivity of 2.58 g 1−1 h−1 were obtained, confirming the establishment of a cost-effective commercial surfactin production.

Murashige and Skoog (MS) medium, a common tissue culture media for orchids which is mostly supplemented with various natural organic substances with high nutritional values can accelerate the growth of plant tissues. Nevertheless, the knowledge of actual compositions of these natural added substances is limited, causing various culture results. In this study, we have investigated the effects of added natural organic nutrient ratios on 2 orchid tissue cultures, Dendrobium, Dendrobium farmeri Paxt. and Dendrobium griffithianum Lindl. Additionally, simple, affordable medium recipes, MS media supplemented with banana were thus economically sterilized by either a simple type steaming vapor boiler or an autoclave for reliability improvement. The results showed that the orchids grew better in medium supplemented with Namwa banana especially at 150 g/L for Dendrobiumfarmeri Paxt. and 75 g/L for Dendrobium griffithianum Lindl. The suitable steaming procedure at 100°C for 60 minutes was adequate for eliminating most pathogens from the media and helped both orchid seedlings cultivated very well. Moreover, the optimized homemade tissue culture protocol provides fast effectiveness, simplicity, consistently high nutritional values, and practically being suitable technique for ready transferring to the community.
