Early seedling establishment in rice (Oryza sativa L.), which is measured by primary/secondary tiller, shoot length, biomass, root-related traits, and leaf area index, is an important trait because it helps to compete for light, air, and water for better tolerating various abiotic stresses. Consequently, it can affect the yield. However, there are not many research studies on this subject. Furthermore, previous studies have only measured the target traits once. However, this does not reflect the variation of growth rate during the seedling stage. Thus, two data points, two weeks and four weeks after planting, were used in the current study. As a result, two QTL regions were detected for the growth differences via plant height and green area (reflecting tillering). We expect that these results can be utilized by breeders to evaluate and select vigorous seedlings for their breeding programs.

AbstractSeed priming significantly affects germination and post germination emergence in seedling of plants. Okra is an important vegetable plays significant role in human nutrition. However, little information is available regarding effect of seed priming on germination and post germination early vegetative growth. Therefore, a pot experiment was conducted to find out the effect of seed priming on the emergence and seedling vigor in different okra varieties. Six local Bangladeshi okra varieties were primed in water for 24 hours and evaluated their germination and post germination emergence. The varieties were named as V1-V6 (V1 = Flash power, V2 = Durga, V3 = BARI dheros-2, V4 =Okra pornota, V5 =Dandy all green, V6 = Boishakhi). Post priming evaluation was done based on germination percentage, root-shoot length of seedlings, seedling height, and dry weight of seedlings, germination index and speed of germination at different days after sowing (5-15 DAS). Highest germination was observed in V3 (88.00%) and lowest was in V5 (25.33%). Root length was highest in V3 and V4 (8.77cm) and lowest was observed in V2 (5.15cm). Shoot length of the seedlings was highest in V3 (7.94cm) followed by V1 (7.33). Shoot length of the seedlings was lowest in V5 (5.12cm). Seedling height was highest in V3 (16.71cm) which was followed by V6 (16.26cm). The lowest seedling height was observed in V1 (12.50cm). Dry weight of the seedlings was highest in V3 (2.32g) which was followed in V1 (2.26g). The lowest dry weight was observed in V5 (1.99g). The variety V3 performed better in most of the evaluated parameters and suggested for commercial cultivation.

Four greenhouse experiments were conducted to study the influence of seed size, protein content and cultivar on seedling vigor of wheat measured as seedling dry weight at 20 days. The simple and partial correlation coefficients among the variables were all positive and significant. No significant interaction occurred between seed size and genotype. The genotypic differences in seedling vigor may lead to its use as a selection criterion in wheat breeding programs.

A nondestructive method of screening for seedling vigor is described. The technique involves the measurement of green shoot weight and the subsequent regeneration of the plants from transplanted shoots or from the seedling base. The method is quick and can be adapted to breeding programs.

ABSTRACT: Cultivars of the genus Nopalea are known in Brazil for being tolerant to cochineal carmine attacks, thus making the cultivation of this genus a promising alternative for mitigating the negative effects of this insect on the production of biomass. With the objectives of characterizing morphologically spineless forage cactus varieties and identify morphological characteristics that may be the focus in spineless forage cactus breeding programs, an experiment was conducted in a completely randomized block design with 11 treatments and four replications. The variety Alagoas showed the highest values of weight, area and volume of cladodes. The varieties Negro Michoacan F7 and V7, Tamazunchale V12 showed the highest values of the cladode area index, the total volume of cladodes and total fresh mass production. The varieties Negro Michoacan V7 and F7 presented the highest water use efficiency and dry mass yield. Cladode volume showed the highest correlation coefficients with the fresh weight of cladodes. Aiming the release of varieties for biomass production, varieties Negro Michoacan F7, V7 and Tamazunchale V12 may substitute the Miúda variety. The number and cladode area index may be used as criteria for selection of superior varieties in breeding programs.

Abstract BackgroundEnvironmental adaptation and expanding harvest seasons are primary goals of most peach [Prunus persica (L.) Batsch] breeding programs. Breeding perennial crops is a challenging task due to their long breeding cycles and large tree size. Pedigree-based analysis using pedigreed families followed by haplotype construction creates a platform for QTL and marker identification, validation, and the use of marker-assisted selection in breeding programs.ResultsPhenotypic data of seven F1 low to medium chill full-sib families were collected over two years at two locations and genotyped using the 9K SNP Illumina array. Three QTLs were discovered for bloom date (BD) and mapped on linkage group 1 (LG1) (172 – 182 cM), LG4 (48 – 54 cM), and LG7 (62 – 70 cM), explaining 17-54%, 11-55%, and 11-18% of the phenotypic variance, respectively. The QTL for ripening date (RD) and fruit development period (FDP) on LG4 was co-localized at the central part of LG4 (40 - 46 cM) and explained between 40-75% of the phenotypic variance. Haplotype analyses revealed SNP haplotypes and predictive SNP marker(s) associated with desired QTL alleles and the presence of multiple functional alleles with different effects for a single locus for RD and FDP.ConclusionsA multiple pedigree-linked families approach validated major QTLs for the three key phenological traits which were reported in previous studies across diverse materials, geographical distributions, and QTL mapping methods. Haplotype characterization of these genomic regions differentiates this study from the previous QTL studies. Our results will provide the peach breeder with the haplotypes for three BD QTLs and one RD/FDP QTL for the creation of predictive DNA-based molecular marker tests to select parents and/or seedlings that have desired QTL alleles and cull unwanted genotypes in early seedling stages.

Abstract Soybean (Glycine max (L.) Merr) is a crop of global importance for both human and animal consumption, which was domesticated in China more than 6000 years ago. A concern about losing genetic diversity as a result of decades of breeding has been expressed by soybean researchers. In order to develop new cultivars, it is critical for breeders to understand the genetic variability present for traits of interest in their program germplasm. Sucrose concentration is becoming an increasingly important trait for the production of soy-food products. The objective of this study was to use a genome-wide association study (GWAS) to identify putative QTL for sucrose concentration in soybean seed. A GWAS panel consisting of 266 historic and current soybean accessions was genotyped with 76k genotype-by-sequencing (GBS) SNP data and phenotyped in four field locations in Ontario (Canada) from 2015 to 2017. Seven putative QTL were identified on chromosomes 1, 6, 8, 9, 10, 13 and 14. A key gene related to sucrose synthase (Glyma.06g182700) was found to be associated with the QTL found on chromosome 6. This information will facilitate efforts to increase the available genetic variability for sucrose concentration in soybean breeding programs and develop new and improved high-sucrose soybean cultivars suitable for the soy-food industry.
