The design of morphing structures must combine conflicting structural requirements and multiple load conditions that are related to the aerodynamic shapes aimed at optimizing aircraft performance. This article proposes a multilevel approach for the design of adaptive compliant wing devices. A set of aerodynamic shapes, and associated their loads, is defined by a shape optimization, coupled with a three-dimensional parametric technique, that can identify only feasible shape changes due to the morphing. A topology and sizing multiobjective optimization drives the Pareto-optimal structural design of the compliant structure, which is able to deform itself to match, once actuated, all of the previously defined aerodynamic shapes. Next two design levels produce a more detailed solution which is extended until the definition of the complete device. A 90 pax, twin prop green regional aircraft is used as an innovative aircraft demonstration platform for the design of the morphing droop nose to be installed on the wing. The results show the structural capabilities of this device in terms of the external shape quality and the strain requirements. This work enables the validation of the design method and prove the functionality of compliant structures when accounting for the aeroelastic effects due to the interaction with the wing-box.

Structural design for fire is conceptually similar to structural design conducted under ambient temperature conditions. Such design requires an establishment of clear objectives and determination of the severity of the design fire. In the commonly used prescriptive design method for fire, fire resistance (expressed in hours) is the primary qualification metric. This is an artifact of the standard fire tests that are used to determine this quantity. When conducting a performance-based approach for structural design for fire, it is important to determine structural member temperatures accurately when the members are exposed to a real fire. In order to evaluate the fire resistance of structural members such as structural steels and concrete, both the temporal and spatial variation of temperatures must be accurately determined. The transient temperature profiles in structural members during exposure to a fire can be determined from a heat transfer analysis. There are several models/approaches for analyzing heat transfer that have been used to determine the transient structural temperatures during a fire event. These range from simple models to advanced models involving three-dimensional heat transfer analysis employing finite element or finite difference techniques. This document provides a brief summary of some of the common simple and advanced approaches that have been used for conducting heat transfer analysis of both steel and concrete members when exposed to fire.

ABSTRACTAntisense oligonucleotides (ASOs) are an emerging class of drugs that target RNAs. Current ASO designs strictly follow the rule of Watson-Crick base pairing along target sequences. However, RNAs often fold into structures that interfere with ASO hybridization. Here we developed a structure-based ASO design method and applied it to target severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Our method makes sure that ASO binding is compatible with target structures in three-dimensional (3D) space by employing structural design templates. These 3D-ASOs recognize the shapes and hydrogen bonding patterns of targets via tertiary interactions, achieving enhanced affinity and specificity. We designed 3D-ASOs that bind to the frameshift stimulation element and transcription regulatory sequence of SARS-CoV-2 and identified lead ASOs that strongly inhibit viral replication in human cells. We further optimized the lead sequences and characterized structure-activity relationship. The 3D-ASO technology helps fight coronavirus disease-2019 and is broadly applicable to ASO drug development.

In the field of topology optimization problems, the Evolutionary Structural Optimization (ESO) method is one of the most popular and easy to use. When dealing with problems of reasonable difficulty, the ESO method is able to give very good results in reduced times and with a limited request of computational resources. Generally, main applications of this method are addressed to the definition of the optimal topology of a component subjected to a single load condition. In this work, a new methodology, based on the ESO approach, is introduced for the study of the optimal topology of a component subjected to multiple load conditions. The new procedure, entirely developed in the APDL programming language, has been tested with a holed plate subject to two different load conditions; the obtained results are promising in terms of mass reduction and structural performances.

Receptors are commonly defined in terms of number per cell, affinity for ligand, chemical structure, mode of attachment to the cell surface, and mechanism of signal transduction. We propose to show that knowledge of spatial distribution of receptors on the cell surface can provide additional clues to their function and components of functional control.L-selectin and Mac-1 denote two receptor populations on the neutrophil surface that mediate neutrophil-endothelial cell adherence interactions and provide for targeting of neutrophil recruitment to sites of inflammation. We have studied the spatial distributions of these receptors using LVSEM and backscatter imaging of isolated human neutrophils stained with mouse anti-receptor (primary) antibody and goat anti-mouse (secondary) antibody conjugated to 12 nm colloidal gold. This combination of techniques provides for three-dimensional analysis of the expression of these receptors on different surface membrane domains of the neutrophil: the ruffles and microvilli that project from the cell surface, and the cell body between these projecting structures.

AbstractA flight test campaign for system identification is a costly and time-consuming task. Models derived from wind tunnel experiments and CFD calculations must be validated and/or updated with flight data to match the real aircraft stability and control characteristics. Classical maneuvers for system identification are mostly one-surface-at-a-time inputs and need to be performed several times at each flight condition. Various methods for defining very rich multi-axis maneuvers, for instance based on multisine/sum of sines signals, already exist. A new design method based on the wavelet transform allowing the definition of multi-axis inputs in the time-frequency domain has been developed. The compact representation chosen allows the user to define fairly complex maneuvers with very few parameters. This method is demonstrated using simulated flight test data from a high-quality Airbus A320 dynamic model. System identification is then performed with this data, and the results show that aerodynamic parameters can still be accurately estimated from these fairly simple multi-axis maneuvers.

The energy monitoring of heritage buildings has, to date, been governed by methodologies and standards that have been defined in terms of sensors that record scalar magnitudes and that are placed in specific positions in the scene, thus recording only some of the values sampled in that space. In this paper, however, we present an alternative to the aforementioned technologies in the form of new sensors based on 3D computer vision that are able to record dense thermal information in a three-dimensional space. These thermal computer vision-based technologies (3D-TCV) entail a revision and updating of the current building energy monitoring methodologies. This paper provides a detailed definition of the most significant aspects of this new extended methodology and presents a case study showing the potential of 3D-TCV techniques and how they may complement current techniques. The results obtained lead us to believe that 3D computer vision can provide the field of building monitoring with a decisive boost, particularly in the case of heritage buildings.

Reinforced concrete (RC) frame buildings are a widely used structural system around the world. These buildings are customarily designed through standard code-based procedures, which are well-suited to the workflow of design offices. However, these procedures typically do not aim for or achieve seismic performance higher than code minimum objectives. This article proposes a practical design method that improves the seismic performance of bare RC frame buildings, using only information available from elastic structural analysis conducted in standard code-based design. Four buildings were designed using the proposed method and the prescriptive approach of design codes, and their seismic performance is evaluated using three-dimensional nonlinear (fiber) models. The findings show that the seismic performance is improved with the proposed method, with reductions in the collapse fragility, higher deformation capacity, and greater overstrength. Furthermore, an economic analysis for a six-story building shows that these improvements come with only a 2% increase in the material bill, suggesting that the proposed method is compatible with current project budgets as well as design workflow. The authors also provide mathematical justification of the method.

Three-dimensional integrated woven spacer sandwich composites have been widely used as industrial textiles for many applications due to their superior physical and mechanical properties. In this research, 3D integrated woven spacer sandwich composites of five different specifications were produced, and the mechanical properties and performance were investigated under different load conditions. XR-CT (X-ray computed tomography) images were employed to visualize the microstructural details and analyze the fracture morphologies of fractured specimens under different load conditions. In addition, the effects of warp and weft direction, face sheet thickness, and core pile height on the mechanical properties and performance of the composite materials were analyzed. This investigation can provide significant guidance to help determine the structure of composite materials and design new products according to the required mechanical properties.

This paper presents the innovative use of hot-rolled thickness-tapered mill products, longitudinally profiled (LP) plates, for the seismic performance of bridge bents of single and portal framed piers. The study involves the inelastic cyclic testing and numerical analysis of tested beam-columns and portal frames in order to evaluate the effects of tapering ratios of LP plates, penetration of yielding, and number of locally buckled panels on their structural ductility. A structural design method is proposed for the portal frames having LP panels under cyclic loadings.
