In many countries, bulk tank milk (BTM) has been used for examining milk and analyzed as an important part of milk quality assurance programs. The objectives of this study were to investigate milk quality and the presence of major mastitis pathogens in BTM, and to compare the characteristics of BTM by dairy factory or company. A total of 1588 batches of BTM samples were collected from 396 dairy farms of seven dairy factories owned by four companies in Korea. The means of individual bacterial counts (IBC) and somatic cell count (SCC) were 3.7 × 104 cells/mL and 1.1 × 105 cells/mL, respectively, and no significant differences among dairy factories were observed. The most common pathogen was Staphylococcus spp. (60.1%), followed by E. faecalis (53.8%), E. coli (37.6%) and Streptococcus spp. (22.5%). Enterococcus spp. showed the highest resistance to tetracyclines (51.1% to 73.9%) and macrolides (46.5%). S. aureus and coagulase-negative staphylococci (CNS) showed the highest resistance to penicillin (28.4% and 40.2%, respectively), and three (3.2%) S. aureus and seven (3.3%) CNS were also methicillin-resistant. These data show the diverse prevalence and characteristics of major mastitis pathogens among factories, and support the development of strong monitoring and prevention programs of mastitis pathogens by commercial dairy operations.

The somatic cell count (SCC) in bovine bulk tank milk is presently used as an indicator of raw milk quality, reflecting the udder health status of the herd. During mastitis, SCC increases, mostly owing to an influx of polymorphonuclear leucocytes (PMN) from blood into milk, with a concomitant change in milk composition. Bulk tank milk samples were categorized according to their SCC, as well as polymorphonuclear leucocyte count (PMNC), to study relationships between SCC, PMNC and various raw milk quality traits, i.e. contents of total protein, whey protein, casein, fat and lactose, casein number, proteolysis and rheological properties. The proportion of PMN, obtained by direct microscopy, was significantly higher in samples with high SCC compared with low SCC samples. SCC and PMNC were strongly correlated, yielding a correlation coefficient of 0·85. High SCC samples had lower lactose and casein contents, lower casein number and more proteolysis than low SCC samples. Samples with high PMNC had a lower casein number than low PMNC samples. Samples with high and low SCC or PMNC did not differ in respect to rheological properties. Our results do not indicate that PMNC is a better biomarker than SCC for raw bulk tank milk quality, as previously proposed.

The objective was to determine the proportions and bacterial counts of major mastitis pathogens in samples of bulk tank milk (BTM), as well as to clarify the relationship between these bacteria counts and bulk tank somatic cell count (BTSCC). The purpose was to judge the importance of the counts of mammary pathogens for BTSCC at the population level. Samples of BTM were collected from 268 randomly selected anonymous dairy herds (with approximately 29 000 dairy cows). Staphylococcus aureus, other coagulase-positive staphylococci, Streptococcus agalactiae and Streptococcus dysgalactiae were grouped as contagious pathogens, and Streptococcus uberis, Escherichia coli, Pseudomonas aeruginosa, Enterococcus faecalis and Enterococcus faecium as environmental pathogens. Based on assessment of the dominant pathogen in each herd, environmental pathogens were found to dominate. Counts of specific mammary pathogens in BTM samples did not exceed a geometric mean of 1000 cfu/ml. Significant differences were documented in BTSCC between BTM samples containing Str. agalactiae, Staph. aureus, and Str. dysgalactiae and BTM samples that were pathogen-free. Geometric means of BTSCC associated with these pathogens were noticeably higher than the overall BTSCC (Str. agalactiae 243 628; Staph. aureus 205 610; Str. dysgalactiae 203 978; overall 173 000 cells/ml). It follows that the somatic cell count (SCC) associated with these pathogens contributed substantially to the overall BTSCC. Environmental pathogens predominated in samples of BTM with SCC <300×103/ml and contagious mastitis pathogens predominated in BTM samples with SCC >300×103/ml. No correlation was detected between bacterial counts of specific pathogens and BTSCC. This study revealed that the assessment of bacterial counts of mammary pathogens in samples of BTM in relation to BTSCC is applicable for the monitoring of changes in the occurrence of major mastitis pathogens in dairy herds at a national level.

AbstractThis research communication explores the value of routinely collected bulk tank milk quality data for estimating dairy cattle welfare at herd level. Selected bulk tank milk quality parameters (somatic cell count, total bacterial count, urea, protein and fat contents) recorded during the years 2014–2016 in 287 Italian dairy farms were compared with the animal welfare data of each farm. The welfare assessment data were extracted from the database of the Italian Reference Centre for Animal Welfare (CReNBA), which includes the outputs of the application of the CReNBA welfare assessment protocol for dairy cows, used at national level for on-farm controls. The statistical analysis was carried out using the correlation coefficient for Kendall's Tau ranks, in order to investigate the presence of a categoric relationship between the selected bulk tank milk quality parameters and the overall animal welfare score or the scores of the single areas A (farm management and staff training), B (housing) and C (animal-based measures). Somatic cell count, total bacterial count, urea and proteins demonstrated only a few statistically significant and very weak correlations with farm animal welfare data, while no significant correlations were obtained for milk fat content. Given the weak correlations found, the selected bulk tank milk parameters seems to be able to provide only limited information about the welfare level of the herd, thus it could be difficult to use them for drawing up a pre-screening model for identifying herds at risk of poor welfare.

Environmental mastitis represents a major challenge on dairy farms where contagious pathogens are controlled by improved milking procedures. Therefore, research focused on the environment is important to improve udder health programs. The objectives of this prospective and descriptive study were to (1) describe bedding bacterial counts, pH, and dry matter (DM) of five different bedding types (organic: manure solids, straw, paper fiber; inorganic: sand, recycled sand) and (2) explore the association between bedding bacterial counts with bulk tank milk quality. This study took place within five conveniently selected commercial dairy herds, each with a predominant bedding material in lactating pens. Bedding samples (used n = 237; fresh n = 53) were collected monthly from July 2018 to July 2019 following a standard operating procedure (SOP) to minimize sampling variability. Additionally, a bulk tank (BT) milk sample (n = 40) was collected on the same day unless milk had been picked up prior to arrival. Both BT and bedding samples were submitted to the laboratory for culture and bacterial identification and quantification of Streptococcus spp, coliforms, and non-coliforms as well as detection of several pathogens of mastitis importance. Somatic cell count was evaluated in BT samples. Within bedding type, the correlation between bedding characteristics and bacterial counts in bedding was evaluated using Pearson correlation. Within bedding type, the correlation between bacterial counts in bedding samples and bacterial counts in BT were determined. The Kruskal–Wallis test was used to evaluate the bacterial count by bedding type and to evaluate BT somatic cell count differences based on bedding type. In fresh bedding, bacterial counts were generally higher for manure solids for all bacterial groups compared with other materials. In used samples, organic materials had the highest levels of all bacterial groups. The proportion of samples with detectable organisms of mastitis importance varied within and among herds in both bedding and BT samples throughout the study period. In bedding samples, a higher DM content had the lowest levels of bacterial growth compared with those with lower DM content. Most bedding samples were on the alkaline side within a pH range of 8–11. No relationship between bacterial counts and pH was observed. No associations between BT bacteria counts and bedding bacterial counts were observed. No association between bulk tank somatic cell counts based on bedding type were observed. Despite using an SOP for bedding sampling in an effort to consistently collect samples, we still observed a large amount of variability both within and among bedding samples. This variability may have obscured any potential association between BT milk quality and bedding type.

This study was performed to investigate the distribution characteristics of airborne bacteria emitted from swine manure composting plants. The types of swine manure composting plants selected for the survey in this study were as follows: screw type, rotary type, and natural dry type. Mean levels of airborne bacteria in swine manure composting plants were 7428 (±1024) CFU m−3 for the screw type, 3246 (±1407) CFU m−3 for the rotary type, and 5232 (±1217) CFU m−3 for the natural dry type, respectively. Based on the results obtained from this study, the swine manure composting plant operated by screw type showed the highest concentration of airborne bacteria, followed by the natural dry type and rotary type. The monthly concentration of airborne bacteria was the highest in August and the lowest in November, regardless of the type of swine manure composting plant. The respirable size of airborne bacteria accounted for about 50% of the total. The ratio of respirable to the total quantity of airborne bacteria was 50%. The correlation relationships between airborne bacteria and environmental factors (temperature, relative humidity, particulate matters, and odor) were not found to be significant in the swine manure composting plants. The predominant genera of airborne bacteria identified were Micrococcus spp., Staphylococcus spp., Escherichia(E-coli) spp., Enterococcus spp., and Enterobacteriaceae spp.

ZusammenfassungZiel: Ermittlung der Prävalenz prädisponierender Begleiterkrankungen (BE) bei Katzen mit bakteriellen Harnwegsinfektionen (HWI) und der Prävalenz von Bakterienspezies bei verschiedenen BE sowie deren Sensibilität gegenüber den bei HWI häufig eingesetzten Antibiotika Doxycyclin, Trimethoprim-Sulfamethoxazol (TMS), Amoxicillin-Clavulansäure (AMC), Cephalothin und Enrofloxacin. Material und Methoden: In die retrospektive Studie wurden Katzen mit positiver Urinkultur im Zeitraum 2003–2009 eingeschlossen. Basierend auf den Daten der Krankenakten erfolgte eine Einteilung in vier Gruppen: Katzen mit systemischen prädisponierenden BE, Katzen mit lokalen prädisponierenden BE, Katzen mit Harnblasendauerkathetern (HBDK) und Katzen ohne dokumentierte BE. Zur Ermittlung der wahrscheinlichen Effektivität der Antibiotika wurden deren antimikrobielle Impact-Faktoren berechnet. Ergebnisse: In die Studie gingen 194 Katzen mit 219 bakteriellen Isolaten ein. Davon wiesen 78,4% (152/194) eine BE auf. 49,5% (96/194) hatten eine systemische BE und 28,9% (56/194) hatten eine lokale BE oder einen HBDK. Katzen mit systemischen BE waren signifikant älter als Katzen der anderen Gruppen und häufiger weiblich als Katzen mit lokaler BE und HBDK. Mehr als 50% der Katzen mit systemischer BE zeigten keine klinischen Symptome einer Erkrankung des unteren Harntrakts. Escherichia (E.) coli, gefolgt von Streptococcus spp., Staphylococcus spp. und Enterococcus spp. waren die am häufigsten vorkommenden Isolate. Dabei lag der Anteil der E.-coli-Isolate bei Katzen mit systemischer BE signifikant höher, während bei Katzen mit HBDK und Katzen mit anderen lokalen BE der Anteil an Streptococcus- und Staphylococcus-spp.-Isolaten signifikant höher war. Bei Katzen mit lokaler BE und Katzen mit HBDK ergaben sich niedrigere antimikrobielle Impact-Faktoren als bei Katzen der anderen Gruppen. Schlussfolgerung und klinische Relevanz: Ein Großteil der Katzen mit HWI leidet unter einer prädisponierenden BE. Katzen mit systemischer BE zeigen häufig keine Symptome einer Erkrankung des unteren Harntrakts. AMC und TMS waren in dieser Katzenpopulation die Antibiotika mit den höchsten antimikrobiellen Impact-Faktoren.

The aim of the study was to investigate the effects of season, cow cleanliness and milking routine on bacterial and somatic cell counts of bulk tank milk. A total of 22 dairy farms in Lombardy (Italy) were visited three times in a year in different seasons. During each visit, samples of bulk tank milk were taken for bacterial and somatic cell counts; swabs from the teat surface of a group of cows were collected after teat cleaning and before milking. Cow cleanliness was assessed by scoring udder, flanks and legs of all milking cows using a 4-point scale system. Season affected cow cleanliness with a significantly higher percentage of non-clean (NC) cows during Cold compared with Mild season. Standard plate count (SPC), laboratory pasteurization count (LPC), coliform count (CC) and somatic cell count, expressed as linear score (LS), in milk significantly increased in Hot compared with Cold season. Coagulase-positive staphylococci on teat swabs showed higher counts in Cold season in comparison with the other ones. The effect of cow cleanliness was significant for SPC, psychrotrophic bacterial count (PBC), CC and Escherichia coli in bulk tank milk. Somatic cell count showed a relationship with udder hygiene score. Milking operation routine strongly affected bacterial counts and LS of bulk tank milk: farms that accomplished a comprehensive milking scheme including two or more operations among forestripping, pre-dipping and post-dipping had lower teat contamination and lower milk SPC, PBC, LPC, CC and LS than farms that did not carry out any operation.

Milk somatic cell count (SCC) is the gold standard in diagnosis of subclinical mastitis, and is also an important parameter in quality programmes of dairy cooperatives. As routine SCC analysis is usually restricted to central laboratories, much effort has been invested in the search for alternative biomarkers of mastitis and milk quality, including the presence in the milk of the acute phase proteins (APP), haptoglobin (Hp) and serum amyloid A (SAA). The aim of this study was to investigate relationships between Hp, SAA and SCC in quarter, cow composite, and bulk tank milk samples. Cows (n=165), without any clinical signs of disease or abnormalities in the milk or udder, from three different dairy farms, were used. Cow composite milk samples from all cows delivering milk at the sampling occasion were taken once in each herd. In one of the farms, representative quarter milk samples (n=103) from 26 cows were also collected. In addition, bulk tank milk samples from 96 dairy farms were included in the study. Samples were analysed for Hp, SAA and SCC, and relationships between the parameters were evaluated at quarter, cow and tank milk levels using Chi-square analysis. Milk samples were categorized according to their SCC, and the presence, or no presence, of SAA and Hp, based on the detection limits of the screening methods (0·3 mg/l and 1·0 mg/l for SAA and Hp, respectively). Hp and SAA were found in milk at quarter, cow composite and bulk tank levels. A large proportion (53%) of the animals had detectable milk concentrations of APP, and SAA was detected more frequently, and at higher concentrations than Hp, regardless of sample type. SAA was detected in as many as 82% of the bulk tank milk samples. Significant relationships were found between Hp, SAA and SCC at quarter and cow composite milk levels, but only between SAA and SCC at bulk tank milk level. Detectable levels of APP were more common at high SCC.
