A total of 160 piglets, females and castrates, were divided at weight of 25 kg into four groups in order to study the effects of dietary vitamin A supplementation. The groups were balanced according to initial weight, sex and litter origin. The same basic mixture, consisting of barley and commercial protein concentrate with all minerals and vitamins except vitamin A, was used in feeding of all the piglets. The vitamin A activity of the mixture, however, was 646 IU/kg. Special vitamin A premixes were prepared by using a commercial product providing the four final four diets with 0, 1000, 3000 and 50000 IU/kg. Three piglets were sacrificed at the beginning of the trial to determine the content of liver vitamin A. It was found to vary between 7.3 and 10.2 mg/100 g fresh liver. There were no differences between the groups in daily gain,feed: gain ratio or carcass quality. A histological examination ofleg joints did not reveal an increased frequency of osteochondrosis in pigs fed with a high level of vitamin A. Growing finishing pigs, having adequate liver stores of vitamin A at 25 kg of live weight, grew without showing any visible signs of deficiency up to 100 kg weight with no vitamin A additions. However, the liver vitamin A analysis at slaughter showed that the recommendation for vitamin A in practical pig feeds should be higher than 1000 IU/kg. In the present experiment, 3000 HJ/kg was found to be a sufficient level.

SUMMARYGestating female pigs received either a low or a high intake per day of diets containing either a low or high protein concentration. A change in the composition of both diets occurred after 2 years, when the protein quality of the high protein diet was improved and the energy content of both diets was increased. Vitamin A determinations were carried out on 245 piglet livers and 32 pairs of kidneys and lungs at birth from 47 sows. The livers, kidneys and lungs of 16 sows were also analysed for vitamin A after approximately 4 years on experiment. Vitamin A was detected at birth with antimony trichloride in the liver of the piglet, but not in the kidney or lung. The sow's kidney was found to contain only small amounts and lung tissue only traces.A dietary vitamin A level of 4800 i.u./kg during the breeding life of healthy sows, or 8600 i.u./day during gestation, was adequate from the point of view of both a constant storage in piglet livers at birth over eight to ten parities and a relatively high concentration remaining in sow livers after that period. This conclusion is in line with recommendations of the Agricultural Research Council (1966).As a consequence of differences in both the condition of the sows and in their responses in the two periods, the results for each period are presented separately. In the first 2-year period, when the sows received a relatively low intake of dietary protein during gestation (between 248 and 317 g protein/sow/day), and a low energy intake; that is, when protein was used for energy production, the liver vitamin A storage of the piglet at birth was increased by raising either the daily protein intake during gestation to 352 g, or the food intake from 1·8 to 2·3 kg/sow/day. Liver vitamin A and N concentrations were negatively correlated with liver weight, but increasing dietary protein concentration raised liver weight and its vitamin A content. Liver vitamin A per piglet was not affected by litter size.A conclusion may not be drawn concerning the contribution of dietary energy to the differences in response between periods, because in addition to dietary changes other differences occurred between periods. Nevertheless, in the second 2-year period, when energy intake during gestation was adequate for normal growth and development, a difference of 150 g in protein intake/sow/day (363 g against 208 g) had no effect on liver weight or its vitamin A content. Furthermore, there was no significant treatment effect on total protein or albumin concentrations in the serum of the sow.

To determine the effects of dietary vitamin A on the activity of the mixed function oxidase (MFO) system and on the susceptibility to 3-methylindole (3MI)-induced respiratory disease, goats were fed a high (supplemented with 12 000 IU retinyl palmitate/kg) or a low (no supplementation) vitamin A diet for 14 weeks. Four animals in each dietary group were then challenged with [14C]3MI. The remaining four animals served as controls. Enzyme assays were performed and severity of the 3MI-induced lung damage was scored. Consumption of the low vitamin A diet resulted in a significant reduction in the liver and lung contents of vitamin A; however, serum concentration of vitamin A was unaffected. High vitamin A supplementation did not alter the activity of the MFO system. Clinical signs, lung – body weight ratio and wet – dry weight ratio of the lungs of 3MI-infused goats were not significantly different; however, in the low vitamin A goats, the microscopic lesions such as the extent of alveolar epithelial hyperplasia were judged to be significantly more severe than those observed in the high vitamin A goats.

ABSTRACT Background Biochemical vitamin A deficiency (VAD) is believed to be a serious public health problem (low serum retinol prevalence >20%) in Indian children, justifying universal high-dose vitamin A supplementation (VAS). Objective To evaluate in Indian children younger than 5 y the risk of biochemical VAD from the Comprehensive National Nutrition Survey, as well as dietary vitamin A inadequacy and excess over the tolerable upper limit of intake (TUL) from national and subnational surveys, factoring in fortification and VAS. Methods Child serum retinol data, corrected for inflammation, were examined to evaluate national- and state-level prevalence of VAD. Simultaneously, dietary intakes from the National Sample Survey Office and the National Nutrition Monitoring Bureau were examined for risk of dietary vitamin A deficiency against its average requirement (AR) derived for Indian children. Theoretical estimates of risk reduction with oil and milk vitamin A fortification were evaluated along with the risk of exceeding the TUL, as well as when combined with intake from VAS. Results The national prevalence of biochemical VAD measured in 9563 children was 15.7% (95% CI: 15.2%, 16.3%), and only 3 states had prevalence significantly >20%. The AR of vitamin A was 198 and 191 µg/d for boys and girls; the risk of dietary inadequacy was ∼70%, which reduced to 25% with oil and milk fortification. Then, the risk of exceeding the TUL was 2% and 1% in 1- to 3-y-old and 4- to 5-y-old children, respectively, but when the VAS dose was added to this intake in a cumulative 6-mo framework, the risk of exceeding the TUL rose to 30% and 8%, respectively. Conclusion The national prevalence of VAD risk is below 20% in Indian children. Because there is risk of excess intake with food fortification and VAS, serious consideration should be given to a targeted approach in place of the universal VAS program in India.
