Considering the large number of the multiresistant bacterial strains and the increasing need in new antimicrobial formulation, the aim of this study was to evaluate the antipathogenic effect of E. caryophyllata essential oil on collection and recently isolated clinical strains of P. aeruginosa and S. aureus. Experimental assays were carried out by in vitro and in vivo tests. Qualitative and quantitative assessment of the antimicrobial activity was followed by the analysis of the essential oil influence on enzymatic soluble virulence factors expression. The cytotoxic effect of the vegetal extract was quantified by using HeLa cells. Nanosystem embedded essential oil was used to establish the in vivo antipathogenic effect on mice. Qualitative screening results revealed an early microbicidal effect quantified by low minimum inhibitory concentration values. The in vivo study showed a stabiliziation of the essential oil biological activities when using nanosystem embedding, that could be used for the design of proper formulations for delivery systems with antimicrobial effect.

The aim of this study was to verify the existence of the synergistic antibacterial effect of lavender essential oil and linalool individually combined with the antibacterial drug gentamicin. We investigated in vitro the effectiveness of the combinations of gentamicin and lavender essential oil and of gentamicin and linalool against the following strains: Staphylococus aureus ATCC 25923, Staphylococus aureus MRSA and Pseudomonas aeruginosa ATCC 9027. In order to determine the sensitivity of these microorganisms, we determined the minimum inhibitory concentration (MIC – Minimal Inhibitory Concentration). The study of the interaction of gentamicin with lavender oil and linalool was evaluated by the checkerboard method. Synergistic interaction between lavender essential oil and gentamicin and between linalool and gentamicin was observed against Staphylococcus aureus ATCC 25923 and Staphylococcus aureus MRSA. In particular, a very strong synergistic interaction was observed against Staphyloccocus aureus MRSA (lavender essential oil FIC index = 0.14; linalool FIC index = 0.13). In contrast, combinations of gentamicin and lavender essential oil and of gentamicin and linalool were less effective against Pseudomonas aeruginosa growth.

Essential oils from species of the genus Hyptis are well-known for their significant biological properties, including antimicrobial and acaricidal activities. The essential oil from the aerial parts of H. crenata was obtained by hydrodistillation; borneol (17.8%), 1,8-cineol (15.6%) and p-cimene (7.9%) were characterized by GC-MS as its major constituents. The essential oil was evaluated in vitro for its antimicrobial activities against six fungal and five bacterial strains, by measuring the respective MICs, MFCs and MBCs, using broth microdilution methods. The strongest bactericidal activities were shown against Staphylococcus aureus and Enterococcus faecalis, while the strongest fungicidal activities were against Cryptococcus neoformans, Candida glabrata and Candida tropicalis. The oil was also assessed for its anti-tick properties and, at a concentration of 2.5%, it significantly inhibited in vivo oviposition of engorged females of the cattle tick Rhipicephalus (Boophilus) microplus, using the adult immersion test., with an effectiveness of 94.4%.

Biological activity of the new antimicrobial peptide polyphemusin III from the horseshoe crab Limulus polyphemus was examined against bacterial strains and human cancer, transformed, and normal cell cultures. Polyphemusin III has the amino acid sequence RRGCFRVCYRGFCFQRCR and is homologous to other β-hairpin peptides from the horseshoe crab. Antimicrobial activity of the peptide was evaluated and MIC (minimal inhibitory concentration) values were determined. IC50 (half-maximal inhibitory concentration) values measured toward human cells revealed that polyphemusin III showed a potent cytotoxic activity at concentrations of <10 μM. Polyphemusin III caused fast permeabilization of the cytoplasmic membrane of human leukemia cells HL-60, which was measured with trypan blue exclusion assay and lactate dehydrogenase-release assay. Flow cytometry experiments for annexin V-FITC/ propidium iodide double staining revealed that the caspase inhibitor, Z-VAD-FMK, did not abrogate disruption of the plasma membrane by polyphemusin III. Our data suggest that polyphemusin III disrupts the plasma membrane integrity and induces cell death that is apparently not related to apoptosis. In comparison to known polyphemusins and tachyplesins, polyphemusin III demonstrates a similar or lower antimicrobial effect, but significantly higher cytotoxicity against human cancer and transformed cells in vitro.

Due to renewed interest in the cultivation and production of Italian Cannabis sativa L., we proposed a multi-methodological approach to explore chemically and biologically both the essential oil and the aromatic water of this plant. We reported the chemical composition in terms of cannabinoid content, volatile component, phenolic and flavonoid pattern, and color characteristics. Then, we demonstrated the ethnopharmacological relevance of this plant cultivated in Italy as a source of antioxidant compounds toward a large panel of enzymes (pancreatic lipase, α-amylase, α-glucosidase, and cholinesterases) and selected clinically relevant, multidrug-sensible, and multidrug-resistant microbial strains (Staphylococcus aureus, Helicobacter pylori, Candida, and Malassezia spp.), evaluating the cytotoxic effects against normal and malignant cell lines. Preliminary in vivo cytotoxicity was also performed on Galleria mellonella larvae. The results corroborate the use of this natural product as a rich source of important biologically active molecules with particular emphasis on the role exerted by naringenin, one of the most important secondary metabolites.

The chemical composition of the essential oil of Anredera cordifolia (Ten.) Steenis (Basellaceae), grown in Brazil, was studied by means of GC and GC-MS analysis. In all, 19 compounds were identified, accounting for 91.6% of the total oil; hydrocarbons were the main constituents (67.7%). The essential oil was evaluated for its in vitro potential phytotoxic activity against germination and initial radicle growth of Raphanus sativus L., Sinapis arvensis L., and Phalaris canariensis L. seeds. At 1.25 μg/mL and 0.625 μg/mL, the oil significantly promoted the germination of S. arvensis. Moreover, the antimicrobial activity of the essential oil was assayed against ten bacterial strains. The essential oil showed a weak inhibitory activity against the Gram-positive pathogens.

The procedure of Salvia przewalskii shoot multiplication and the ability of regenerated plants to produce essential oil is reported. The essential oil was obtained by hydrodistillation from leaves and flowering stems of field-grown plants, and their chemical composition was examined by GC, GC-MS and 1H NMR. The differences in yield as well as qualitative and quantitative composition between the oils isolated from in vitro and in vivo plants were observed. S. przewalskii essential oil was tested for its antimicrobial and cytotoxic properties. It was found that cytotoxicity against human leukemia HL-60 cells and antimicrobial activity (especially, against Staphylococcus aureus and S. epidermidis strains) of oils isolated from in vitro plants were higher than those for oils from in vivo S. przewalskii plants.

Objective: Zataria is one of the native plants of Iran which is widely used for the treatment of diseases among Iranians. In this study, we investigated the antimicrobial effects of Zataria essential oil on the skin bacteria in rats. Methods: Bacterial strains were isolated from the skin of 6 wistar rats and the antimicrobial effects of Zataria essential oil were evaluated by disk diffusion and microbroth dilution methods. In-vivo tests were performed to evaluate the antimicrobial effect of the essential oil by microbial culture as well as allergy tests on the skin of experimental rats compared to controls. Results: Three bacterial strains were isolated from the skin of rats identified as Staphylococcus aureus, Corynebacterium and Staphylococcus epidermidis. Minimum Growth Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) for the two strains of Staphylococcus aureus and Corynebacterium were obtained 0.39 and 0.78 mg/ml, while for Staphylococcus epidermidis, they were 0.195 and 0.39 mg/ml, respectively. In-vivo test results showed the antibacterial effect of the essential oil on the skin bacteria and no inflammatory effects were observed under the allergy test. Conclusion: Zataria essential oil has antimicrobial effects on the skin infections in lower concentrations. The use of this essential oil as an antiseptic and preservative in cosmetics is recommended instead of chemical preservatives that generally have skin side effects.

Candida albicans is a yeast belonging to the normal microbiota of the human body, considered the most pathogenic species of the genus. It is the main microorganism related to candidiasis. Essential oils of Origanum vulgare have phenolic compounds, such as thymol and carvacrol, which have an effective antimycobacterial character at certain concentrations, but little is known about its biological activity in artisanal preparation. Thus, we evaluated the resistance of standard strains of opportunistic yeast C. albicans against in vitro antifungal activity of artisanal extract of O. vulgare. An inoculum of the challenger was subjected to different concentrations of the fungicidal agent in solidified Muller Hinton and broth followed by incubation at 35°C. The oil was prepared in a manner similar to a possible homemade procedure, and was further sterilized to ensure homogeneity of the indicator. The readings were performed in two days every 24 hours so that there were different possible moments of growth. The tests, which occurred in triplicate, showed that, under experimental conditions, the yeast was resistant to the essential oil compounds at all observed concentrations. Morphological variation was observed in both colonies and yeast cells. Based on analyzes, the artisanal essential oil is incorporated as a promising candidate for the development of antimycotics for clinical use, although in vivo tests are required.

The purpose of the present work aimed at exploring the potential biochemical components and biological activities of an organic extract of the white truffle Tirmania nivea collected from the Iraqi desert, then test the organic extract against the Cytotoxicity on Human Larynx carcinoma cells and selected strains of pathogenic bacteria. Fourier transform infrared spectroscopy (FTIR) and gas chromatography-mass spectrometry GC/MSS were used to analyze mycochemical compositions. The antibacterial activity and Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) was investigated using a disk diffusion agar method. The truffle extract's cytotoxicity effect against the larynx cell line (Hep-2) was assessed by the MTT assay (in vitro). FTIR results provided the presence of phenol, carboxylic acid, and alkane's functional group, The GC-MS analysis of T. nivea disclose the existence of nineteen compounds that can contribute to the pharmaceutical properties of the truffle. As for antibacterial activity result, A growth inhibition activity of truffle extract at (18-40 mm inhibition zones) against the tested pathogenic bacterial strains was detected, which minimum inhibitory concentration values ranged from 3.12 to 6.25 mg/mL for Escherichia coli (ATCC 25922) and Staphylococcus aureus (ATCC 25923) Respectively. The results of cytotoxicity shown that the organic truffle extract exhibited a high inhibitory rate (52.685%) against cell line (Hep-2) at a concentration of 1.56 ?g/mL. In this work, the results showed that the organic extracts of T. nivea are very promising as cancer cytotoxicity and antibacterial agent for future medical applications.

Aromatic plants can interfere in the Mediterranean ecosystem, mainly by the introduction in the environment of volatile compounds. For this reason, we studied the chemical composition and the possible phytotoxic and antimicrobial activities of the essential oil extracted from leaves of Tunisian Artemisia herba-alba Asso. The chemical composition of the essential oil, obtained by hydrodistillation, was analyzed by GC and GC-MS. In all, 24 compounds were identified. The main components were camphor (39.1.%), chrysanthenone (15.0%) and cis-thujone (7.8%). The essential oil was evaluated for its in vitro phytotoxic activity against germination and initial radical growth of Raphanus sativus L., Lepidium sativum L., Sinapis arvensis L., Triticum durum L. and Phalaris canariensis L. seeds. The radicle elongation of the five seeds was affected to different extents by the oil, while germination was not affected. The oil, when tested against eight selected bacterial strains, showed low antimicrobial activity. The chemical composition of the oil of A. herba-alba can help in the chemosystematics of this complex genus. However, the recorded biological activities seem to be neither ecologically nor medicinally significant.
