Tumbuhan bungur (Lagerstroemia loudonii T.B.) termasuk dalam famili Lytrharceae. Famili Lythraceae telah diketahui memiliki aktivitas farmakologi sebagai antidiabetes, antiinflamasi, antimikroba, serta antiobesitas. Daun dan buah bungur (Lagerstroemia loudonii T.B.) memiliki aktivitas dalam menghambat alfa-glukosidase. Berdasarkan teori khemotaksonomi didalam tumbuhan, kemungkinan bagian lain dari tumbuhan bungur memiliki aktivitas dan kandungan kimia yang sama, sehingga dilakukan pengujian aktivitas penghambatan terhadap alfa-glukosidase pada bagian kulit batang bungur. Ekstraksi simplisia dilakukan dengan cara maserasi selama 24 jam menggunakan pelarut etanol 96%. Proses fraksinasi menggunakan cara Ekstraksi Cair-Cair (ECC) dengan pelarut n-heksana, etil asetat dan air. Pengujian aktivitas penghambatan alfa-glukosidase secara in vitro menggunakan metode kolorimetri dengan alat spektrofotometer UV-VIS pada panjang gelombang 400,4 nm dengan substrat p-nitrofenil-α-D-glukopiranosid (PNPG). Akarbose digunakan sebagai pembanding. Hasil penelitian menunjukkan bahwa ekstrak, fraksi air, fraksi etil asetat dan fraksi n-heksana memiliki nilai IC50 berturut-turut sebesar 240,53±0,47 μg/ml, 186,111±1,02 μg/ml, 79,479±0,52 μg/ml dan 113,101±0,46 μg/ml. Nilai IC50 akarbose adalah sebesar 10,457±1,48 μg/ml. Ekstrak dan fraksi-fraksi (air,etil asetat dan n-heksana) kulit batang bungur mampu menghambat aktivitas enzim α-glukosidase. Aktivitas yang paling baik ditunjukan oleh fraksi etil asetat dengan nilai IC50 sebesar 79,479±0,52 μg/ml.   Bungur (Lagerstroemia loudonii T.B.) is included in the Family  Lytrharceae. The Lythraceae has been known to have pharmacological activity as antidiabetic, anti-inflammatory, antimicrobial, and antiobesity. Leaves and fruits of bungur (Lagerstroemia loudonii T.B.) have activities to inhibit alpha-glucosidase. Based on the chemotaxonomy theory in plants, it is possible that other parts of the bungur plant have the same chemical activity and content, Hence the study to evalute the  inhibitory activity against alpha-glucosidase was carried out on its bark stem.The extraction of dried powder material was carried out by maceration for 24 hours using 96% ethanol. The extract was fractionated by Liquid-Liquid Extraction (ECC) method with n-hexane, ethyl acetate and water. The In vitro study of alpha-glucosidase inhibition activity using a colorimetric method with a UV-VIS spectrophotometer at a wavelength of 400.4 nm with a p-nitrophenyl-α-D-glucopiranoside (PNPG) substrate was performed. Akarbose was used as a standard drug.The results showed that extract, water fraction, ethyl acetate fraction and n-hexane fraction showed IC50 values ​​of 240.53 ± 0.47 μg / ml, 186.111 ± 1.02 μg / ml, 79.497 ± 0.52 μg/ ml and 113.101 ± 0.46 μg / ml, respectively. The IC50value of bungur was 10.457 ± 1.48 μg / ml. Extracts and fractions (water, ethyl acetate and n-hexane) of bark stem were able to inhibit the activity of α-glucosidase. Theethyl acetate fraction showed the strongestactivity with IC50 value of 79,479 ± 0.52 μg / ml

Diabetes is a metabolic syndrome characterized by high blood sugar levels or hyperglycemia.Alpha-amylase inhibitors can be used as an antidiabetic that controls postprandial hyperglycemia.Alpha-amylase inhibitors are found in plants as protein and non-protein bioactive content such as in lebuiseed. The purpose of this study was to determine the activity of alpha-amylase inhibition of the fraction ofethanol extract of lebui seed. This research was carried out by extraction of lebui seed and then fractionatedlebui extract to obtain n-hexane fraction (HF), ethyl acetate fraction (EAF), and water fraction (WF). Thefractions then tested to alpha-amylase inhibition activity in vitro by measuring the reducing sugar using3.5-dinitrosalicylic (DNS) reagent. Furthermore, the identification of the bioactive content of each fraction wascarried out using thin layer chromatography. The results of alpha-amylase inhibition activity test showed thatall fractions had the ability to inhibit alpha-amylase. The IC50 value of the n-hexane fraction, ethyl acetatefraction, and water fraction were 173.05 mg/ml, 9.98 mg/ml, and 137.19 mg/ml, respectively. Ethyl acetatefraction has the smallest IC50 value compared to other fractions. Based on the identification of the bioactivecontent, bioactive content that is only found in the ethyl acetate fraction but not in other fractions are tannins.Therefore, the chemical content in lebui seed which acts as an alpha-amylase inhibitor might be tannins.


Dates consisting of 90% flesh have been known to have various benefits, but the remaining 10% of them in the form of seeds has not been utilized optimally. The date palm contains a fairly high polyphenol compound, which can act as an antioxidant. This study aims to determine the antioxidant activity of methanol extract and fractionation of the extract of dates using the in vitro method of DPPH (1,1-diphenyl-2-picrilhidrazil). The date seeds are made into simplicia, macerated with methanol, and then the extracted methanol is gradually fractionated by n-hexane, ethyl acetate, n-butanol, and water. Each fraction and extract is phytochemically screened using the standard methods and its antioxidant activity is determined by DPPH method. Methanol extract, ethyl acetate, n-butanol, and water fractions contain tannins, flavonoids, alkaloids, and saponin, while the n-hexane fraction not contain phytochemical compound based on the phytochemical screening. The ethyl acetate fraction has the strongest antioxidant activity with the lowest IC50 value, that is equal to 3.72 ± 0.44 μg/mL, followed by methanol extract of 4.71 ± 0.64 μg/mL, n-butanol fraction of 6.62 ± 0.08 μg/mL, water fraction of 53.00 ± 18.96 μg/mL, and n-hexane fraction of 250.19 ± 9.69 μg/ mL, while the IC50 value of vitamin C as the positive control is 4.29 ± 0.74 μg/mL. Therefore, date seeds can be a source of natural antioxidants

ABSTRACT Parijoto fruit (Medinilla speciosa B) contains the flavonoid which is one of the phenolic groups compounnd. Flavonoids has biological activities as anti free radical and antionxidants. The aim of this research was to evaluate the potency of ethyl acetat fraction of M.speciosa B. Extract as an antioxidants and antidiabetic. Evaluation of antioxidants activity was carried out by in vitro assay using the ABTS method (2.2 azinobis (3-ethylbenzotiazolin) -6-sulfonic acid), while the antidiabetic assay was carried out using the Nelson-Somogyi method. Research begins with the process of determination, extraction, fravtionation and contiunued by examination of each variable. The parameters of antioxidants activity was determined by IC50 values, while antidiabetic activity was measured by percentage of decreasing of glucoce levels. The results of antioxidants activity showed that ethyl acetate fraction of M. Speciosa B. had antioxidants activity with an IC50 value of 4,246 ppm with a very strong category. In line with these results, ethyl acetate fraction of M. speciosa B. had reduced glucoce levels with an optimal decrease of 50.21% a concentration of 40 ppm.   ABSTRAK Buah Parijoto (Medinilla speciosa B.) mengandung senyawa aktif flavonoid yang merupakan salah satu golongan fenolik. Flavonoid memiliki aktifitas biologis sebagai antiradikal bebas dan antioksidan. Penelitian dilakukan dengan tujuan mengetahui kemampuan fraksi etil asetat M. speciosa B sebagai antioksidan dan antidiabetes. Pengujian aktifitas antioksidan dilakukan secara in vitro dengan metode ABTS (2,2 azinobis (3-etilbenzotiazolin)-6-asam sulfonat), sedangkan uji antidiabetes dilakukan menggunakan metode Nelson-Somogyi. Penelitian diawali dengan proses determinasi, ekstraksi, fraksinasi, dan dilanjutkan dengan pengujian pada masing-masing variabel. Parameter aktifitas antioksidan diwujudkan dengan nilai IC50, sedangkan aktiftas antidiabetes diukur dengan persen penurunan kadar glukosa. Hasil pengujian aktifitas antioksidan menunjukkan bahwa fraksi etil asetat memiliki aktifitas antioksidan dengan nilai IC50 sebesar 4.14±0.08 ppm dengan kategori sangat kuat. Sejalan dengan hasil tersebut, fraksi etil asetat Buah Parijoto (M. speciosa B.) memilili kemampuan dalam menurunkan kadar glukosa dengan penurunan secara optimal sebesar 50.21±0.47% pada konsentrasi 40 ppm.

Objective: The aim of this study is to investigate antioxidant activity and phytochemical screening of ethanol extract, fractions of water, ethyl acetate, and n-hexane from mistletoe tea (Scurrula atropurpurea Bl. Dans).Methods: Simplicia extracted using soxhlet equipment with 96% ethanol. Fractionation was conducted using liquid-liquid extraction using a solvent of water, ethyl acetate and n-hexane. Screening of phytochemical and antioxidant activity was performed against these fractions. Antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl method using ultraviolet-visible spectrophotometry with ascorbic acid as standard. Phytochemical screening was conducted based on the method of Farnsworth.Results: The IC50 values of ethanol extract, water fraction, fraction of ethyl acetate, and n-hexane fraction were 21.92 ppm, 89.57 ppm, 14.08 ppm, and 162.09 ppm, respectively, whereas for ascorbic acid was 4.41 ppm. The ethanol extract and ethyl acetate fraction contained compounds were the same group, polyphenolic, tannins, flavonoids, monoterpenoid, steroids, triterpenoids, and quinones. Fraction of water contained compounds such as polyphenolic group, flavonoids, monoterpenoids, sesquiterpenoids, steroids, and triterpenoids. n-hexane fraction compounds contained steroids and triterpenoids.Conclusion: The ethanol extract, water fraction, ethyl acetate fraction, and n-hexane fraction showed antioxidant activities. The ethanol extract, fractions of water, and ethyl acetate fraction contained flavonoids and polyphenolic potential as antioxidants.Keywords: Mistletoe tea, Scurrula atropurpurea, Antioxidant, 2,2-diphenyl-1-picrylhydrazyl, Ascorbic acid.

Free radical is any molecular species that have unpaired free electrons in their outer orbital shell that make radicals highly reactive, resulting in pathogenesis conditions such as cellular injury, premature aging, cancer, hepatic disorders, neurodegenerative diseases, cardiovascular disease, and kidney disease. One source of natural antioxidant is jackfruit. The purpose of this research was to determine the phenolic and flavonoid contents in the extracts and fractions of jackfruit peel and their potential as antioxidants. Jackfruit peel powder was extracted from maceration. The total phenolic content was determined by the Folin-Ciocalteu method. Meanwhile, flavonoid content was determined using the aluminium chloride complex colorimetric method. Measurements of antioxidant activity were conducted using the 2,2-diphenyl-1-picrylhydrazyl (DPPH). The ethyl acetate fraction had high phenolic and flavonoid contents, which were 49.667±1.508 g GAE/100 g of sample and 70.199±0.374 g of quercertin equivalent/100 g. The ethyl acetate fraction had the strongest antioxidant activity with IC50 value of 4.539±0.201 µg/ mL and correlation value (R2 ) of 0.5881 for phenols and R2 of 0.7241 for flavonoids. Ethyl acetate fraction of jackfruit peel is very potential to be developed as a natural antioxidant and functional food.

Objective: Cervical cancer is a malignant type of cancer, often affects women, particularly in developing countries. Carrisa carandas leaves contained many secondary metabolites that had potency as an anticancer. The purpose of this study was to understand the cytotoxic effect of subfraction of Carrisa carandas leaves against HeLa cells.Methods: Chloroform fraction was separated by VLC gradually with n-hexane–chloroform–ethyl acetate and methanol. The same profiles from eluent chloroform–ethyl acetate composed fraction 18-26 were categorized as Fr4 and ethyl acetate-methanol composed fraction 27-30 as Fr5. The cytotoxic effect was evaluated by MTT assay on HeLa cellsResults: The result showed that the cytotoxic effect of subfraction Fr4 and Fr5 had IC50 values of 177 mg/ml and 98 mg/ml, respectively. Colorless crystal of Subfraction Fr 5-3 had IC50 value of 333 mg/ml. Subfraction Fr 5 showed effective cytotoxic activity than the others. Conclusion: It had chemo-preventive effect against cancer cellsConclusion: This study applied MTT (Microculture Tetrazolium) method by in vitro test. The ethanol-water (7 : 3, v/v) extract of Cotoneaster multiflorus sarcocarp was sequentially fractionated by liquid-liquid partition using n-hexane, diethyl ether, methylene dichloride, and ethyl acetate. The contents of total polyphenols, total flavones, and oligomeric proanthocyanidins in the five parts (including the ethanol-water extract) were determined. In addition, 2,2-diphenyl-1-picrylhydrazyl free radical-scavenging, 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical cation decolorization, reducing power, ferric reducing antioxidant power, and lipid peroxidation inhibition assays were conducted to test the antioxidant activities of Sample 1 (the ethanol-water fraction) and Sample 2 (the ethyl acetate fraction) in vitro. In the above five assays, Sample 2 showed greater antioxidant capacities than Sample 1. Furthermore, Sample 2 was better able to protect low-density lipoproteins from oxidation in a dose-dependent manner. The test results show that C. multiflorus sarcocarp, especially the ethyl acetate-soluble fraction, may be a potential source of natural antioxidants.

A B S T R A C TBackground. The use of plants as anti-fertility in men has been done to reduce sideeffects. The Gendola plant is one of the Basellaceae family plants, which is a naturalmedicinal plant in Indonesia. Gendola leaves contain phenol compounds, flavonoids,tannins, steroids and triterpenoids which can have cytotoxic activity. This studyaims to determine the in vitro anti sperm activity of the ethyl acetate fraction ofGendola leaves on male spermatozoa. Methods. This study is an experimentallaboratory in vitro study using male sperm preparations. The sample used was 20men for each group, both the group was given the gondola leaf fraction and thecontrol group. The treatment group was given ethyl acetate fraction of Gendola leaves(Basella rubra Linn.) With a concentration of 0.05% for 5, 10 and 15 minutes, theviability and motility of sperm were calculated. Data analysis was performed usingthe Mann Whitney test (p <0.05) using SPSS release for Windows software. Results.The results showed that the ethyl acetate fraction affected the motility and viabilityof spermatozoa. When compared with the control group, the motility treatment groupobtained was classified as non-progressive. In the treatment group, the viability ofspermatozoa was significantly different at minute 5, minute 10 and minuted 15 (p<0.05). Conclusion. Ethyl acetate fraction has potential as an anti sperm inreducing sperm motility and viability.

Background. The leaves of Hagenia abyssinica have been used in the management of diabetes mellitus in Ethiopian folk medicine. Thus, this study is aimed at investigating the in vitro α-amylase and α-glucosidase inhibitory and antioxidant activities of the crude extract and solvent fractions of H. abyssinica leaves. Methods. The in vitro α-amylase and α-glucosidase inhibitory and antioxidant activities of the plant extract were assessed using 3,5-dinitrosalicylic acid (DNSA), p-nitro-phenyl-a-D glucopyranoside (p-NPG), and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assays, respectively. Each value of percent inhibition of α-amylase, α-glucosidase, and DPPH scavenging effect was presented as means ± SEM ( n = 3 ). Results. The α-amylase inhibitory activity of the crude extract and solvent fractions was found to be concentration-dependent. The strongest activity was exhibited by the crude extract at the highest concentration with a percentage inhibition of 74.52% (IC50, 14.52 μg/ml) followed by water fraction 68.24% (IC50, 16.31 μg/ml), ethyl acetate fraction 61.57% (IC50, 18.73 μg/ml), and chloroform fraction 56.87% (IC50, 21.57 μg/ml) of H. abyssinica leaves. In the α-glucosidase inhibition assay, the maximum activity was exhibited by the aqueous fraction 62.54% (IC50, 11.67 μg/ml) followed by ethyl acetate fraction 54.97% (IC50, 15.89 μg/ml), crude extract 46.79% (IC50, >16.5 μg/ml), and chloroform fraction 36.44% (IC50, >16.5 μg/ml). In the antioxidant assay, the crude extract exhibited the highest antioxidant activity 86.36% (IC50, 10.25 μg/ml) followed by water fraction 78.59% (IC50, 13.86 μg/ml), ethyl acetate fraction 71.58% (IC50, 16.34 μg/ml), and chloroform fraction 63.65% (IC50, 18.83 μg/ml). Conclusion. This study has revealed that H. abyssinica leaves possess noticeable in vitro α-amylase and α-glucosidase inhibitory and antioxidant activities.
