Abstract Background Arsenic (As) toxicity induces a range of metabolic responses in plants, including DNA methylation. The focus of this paper was on the relationship between As-induced long-term stress and plant senescence in the hyperaccumulator Pteris cretica var. Albo-lineata ( Pc -Al).Results We showed that Pc -Al grown in pots of haplic chernozem contaminated with 100 mg As kg -1 (As 100 ) for 122 days could accumulate more than 2000 and 2800 mg As kg -1 dry matter in old and young fronds, respectively. Analysis of 5-methylcytosine (5mC) in Pc -Al confirmed that the overall DNA methylation status in fronds of As 100 ferns was reduced in contrast to control treatment. Compared with controls, the overall DNA methylation status in fronds of As 100 ferns was reduced (by 6% in young and 10% in old fronds); however, the decrease was significant only in old fronds. The significant correlations for 5mC, in contrast to direct As toxicity, showed that decreases of chlorophylls, fluorescence and photosynthetic rate could be affected by epigenetic changes. Photosynthetic processes were determined in As 100 treatment and showed a reduction of gas-exchange parameters, and a decrease in carotenoids and chlorophylls (by 5% and 26%, respectively). Hyperaccumulation of As resulted in a significant elevation of all analysed nutrients (Cu, Mn, Zn, Mg, S) in old fronds, but not in young fronds.Conclusions The results of this paper point to complex changes in the metabolism of the hyperaccumulator plant Pc -Al, upon exposure to As contamination. The most significant impact was found in young fronds. The physiological parameters correlated more significantly with a decrease of DNA methylation than with direct As toxicity. Our analysis of the very low water potential values and lignification of cell walls in roots showed that transports of assimilated metabolites and water between roots and fronds were reduced.

In the present study, free amino acid (AA) regulation in the arsenic (As) hyperaccumulating ferns was evaluated in a pot experiment to determine the relationship between As stress and the characteristic change in metabolism of AAs. The ferns Pteris cretica cv. Albo-lineata (Pc-Al) and cv. Parkerii (Pc-Pa) were exposed to As treatments at 0, 20, 100, and 250 mg As/kg for 90 days. Greater As content, as well as higher biomass production, were identified in Pc-Al compared with Pc-Pa. Ferns showed changes in the stress metabolism of free AA homeostasis. These results indicate a disturbance in nitrogen metabolism and depletion of pool assimilated carbon metabolism. In the fronds and roots, Pc-Pa accumulated higher amounts of free AAs than Pc-Al. The total free AA content, as well as the ratio of the main AA family pathway (glutamate family), were increased by the accumulation of toxic As in the ferns. Results suggest that Pc-Al tolerates higher As doses better due to changes in AA biosynthesis; however, at higher As doses, Pc-Pa upregulated AA biosynthesis due to As toxicity. The most abundant free AAs of ferns was glutamine, which was enhanced by As. Furthermore, the ratios of selected individual free AAs revealed a characteristic phenotype difference between ferns.

So far, the relative importance of the plant and its microbiome in the development of early stages of plant seedling growth under arsenic stress has not been studied. To test the role of endophytic bacteria in increasing plant success under arsenic stress, gnotobiotic seeds of J. montana were inoculated with two endophytic bacteria: Pantoea conspicua MC-K1 (PGPB and As resistant bacteria) and Arthrobacter sp. MC-D3A (non-helper and non-As resistant bacteria) and an endobacteria mixture. In holobiotic seedlings (with seed-vectored microbes intact), neither the capacity of germination nor development of roots and lateral hairs was affected at 125 μM As(V). However, in gnotobiotic seedlings, the plants are negatively impacted by absence of a microbiome and presence of arsenic, resulting in reduced growth of roots and root hairs. The inoculation of a single PGPB (P. conspicua-MCK1) shows a tendency to the recovery of the plant, both in arsenic enriched and arsenic-free media, while the inoculation with Arthrobacter sp. does not help in the recovery of the plants. Inoculation with a bacterial mixture allows recovery of plants in arsenic free media; however, plants did not recover under arsenic stress, probably because of a bacterial interaction in the mixture.

The ability of melatonin to regulate number of physiological and biochemical processes under different environmental stresses has been widely studied in plants. So, this investigation was done to study the protective roles of melatonin on Cicer arietinum L. grown under arsenic stress. Subjecting Cicer arietinum L. seeds to arsenic stress caused significant decreases in germination percentage, radicle growth, biomass accumulation, protein content and activities of antioxidant enzymes. On the other hand, melatonin treatment significantly increased growth parameters and protein quantity via improving antioxidant enzyme systems as compared with their corresponding untreated controls.
