The presence of biogenic ferrihydrite nanoparticles at callusogenesis stage decreases the level of necrosis at proliferation stage. Ferrihydrite doesn`t influence on callus growth at the second stage, but decreases the percentage of samples with chlorophyll.

A high-performance protocol for callus induction was devised using germinating mature embryos of two local wheat (Triticum aestivum L.) landraces as explant. The results showed that callus development from germinating embryos was rapid starting one day after culture with an induction rate 20 to 25% higher than those of soaked embryos. In addition, the mean rate of growth of callus developed from germinating embryos was 60 to 70% higher than those cultured from soaked embryos. This study also demonstrated a higher frequency of green spots formation(48 to 56%)on callus derived from germinating embryos compared to their soaked counterpart (24 to 28%), suggesting a better differentiation potential of callus cultures derived from germinating embryo. These findings indicate that germinating mature embryo is more suitable explant for wheat callus induction and regeneration than the soaked mature embryo commonly employed for wheat callus culture.

INDUCTION MUTATION AND SELECTION OF IN VITRO PLANT OF WHEAT (Triticum aestivum L.)The goal of this research was to produce wheat crop which is tolerant to lowland condition.Six varieties were used, Dewata, Selayar, Alibey, Oasis, Rabe and HP1744. This research consisted of 4 stages: production of the best callus on MS medium containing 3 g/L 2.4-D, induced mutation of embryogenic callus using EMS, in vitro selection of callus at temperature of 27–35°C, and callus regeneration. The best result for callus production was 76% for Dewata and 70% for Selayar varieties. Higher concentration of EMS and longer soaking time decreased the percentage of callus growth. LC50 for Dewata was 0.3% EMS at 30 minutes and that for Selayar was 0.1% EMS at 60 minutes. The higher the temperature was, the lower was the adaptation tolerant of the plants, and callus growth was inhibited. At the highest temperature (35°C) the callus did not grow at all.Keywords: Induced mutation, Triticum aestivum, EMS, in vitro selection, callusABSTRAKTujuan penelitian ini adalah untuk merakit tanaman gandum yang toleran pada dataran rendah. Varietas yang digunakan ada 6 yaitu Dewata, Selayar, Alibey, Oasis, Rabe dan HP-1744. Penelitian terdiri atas empat tahap yaitu induksi pembentukan kalus terbaik menggunakan media MS + 3 g/L 2,4-D (dipilih dua varietas yang terbaik), induksi mutasi kalus embriogenik menggunakan EMS, seleksi kalus in vitro pada suhu 27–35°C, dan regenerasi. Hasil induksi kalus terbaik terdapat pada varietas Dewata sebesar 76% dan Selayar sebesar 70%. Semakin tinggi konsentrasi EMS dan semakin lama waktu perendaman yang digunakan maka semakin menurun persentase pertumbuhan kalus. LC50 varietas Dewata adalah EMS 0,3% waktu 30 menit sedangkan LC50 varietas Selayar adalah EMS 0,1% waktu 60 menit.Semakin tinggi suhunya maka semakin berkurang toleran adaptasi tanaman tersebut, dan pertumbuhan kalus semakin sedikit. Bahkan pada suhu tertinggi yaitu suhu 35°C tidak ada pertumbuhan kalus sama sekali.Kata Kunci: Induksi mutasi, Triticum aestivum, EMS, seleksi in vitro, kalus

Penelitian deskripsi jenis-jenis kontaminan dari kultur kalus Catharanthus roseus telah dilakukan. Penelitian ini bertujuan untuk mendeskripsikan jenis-jenis kontaminan yang terdapat pada kultur kalus C.roseus. Penelitian ini dilaksanakan dua tahap. Tahap pertama dilakukan kultur kalus C. roseus. Kultur kalus menggunakan media MS (Murashige dan Skoog) dengan zpt 2,4 D 2 mg/L dan kinetin 0,2 mg/L. Tahap kedua dilakukan pengamatan kultur kalus yang mengalami kontaminasi. Kontaminan-kontaminan yang sama secara kasat mata dikelompokan menjadi satu kelompok. Setiap kelompok yang sama diamati lebih lanjut dibawah mikroskop. Penentuan jenis kontaminan dibandingkan dengan morfologi dari Pustaka (rujukan). Pengamatan kontaminan dilakukan mulai dari pembuatan media, inokulasi, subkultur kalus C.roseus. Pengamatan dilakukan dengan 3 botol kontaminan dari setiap kelompok. Hasil yang diperoleh adalah kontaminan kultur kalus C. roseus sekitar 40 %. Jenis kontaminan yang diamati umumnya golongan jamur. Jenis-jenis kontaminan kultur kalus C. roseus adalah Rhizopus, Mucor, Aspergillus, dan Sacharomyces.Research description of the types of contaminant for cultured callus Catharanthus roseus  has been done. This research aimed describe the types of contaminant found in the cultured kalus C. roseus. This research is carried out in two stages. The first stage in callus culture C. roseus using media MS (Murashige and Skoog) with zpt 2,4 D mg/L and kinetin 0,2 mg/L. The second stage in the observation of contaminated callus culture. The same of contaminants are visible into one group. Each of the same group was observed further under a microscope. Determining the types of contaminant in comparison with reference from the literature. Observation are starting from media or inoculation, subcallus culture C. roseus. Observation  done by taking 3 bottles of contaminat from each group then observed for 7 days. The result  obtained are culture contaminants C. roseus about 40 %. The types of contaminant observed are generally of fungi. The types of contaminant callus culture C. roseus are Rhizopus, Mucor, Aspergillus, and Sacharomyces.

Explants derived from widely divergent tissues within the body of the basswood, Tilia americana L. (ovary wall, root and shoot cambia, perimedullary region, petiole), have the same general appearance when growing as a callus culture on identical media. The cambial-derived calli will meld into a single unit when placed in contact in the same culture bottle. Similarly, calli from several sources of wheat, Triticum aestivum L., are indistinguishable in vitro.From the standpoint of purely observational criteria, parenchyma explanted from several organs within the same organism responds in a seemingly identical way to the cultural environment.

Response of two genotypes of bread wheat (Triticum aestivum), Mahon-Demias (MD) and Hidhab (HD1220), to mature embryo culture, callus production, and in vitro salt and heat tolerance was evaluated. For assessment of genotypes to salt and heat tolerance, growing morphogenic calli were exposed to different concentrations of NaCl (0, 5, 10, and 15 g·L−1) and under different thermal stress intensities (25, 30, 35, and 40°C). Comparison of the two genotypes was reported for callus induction efficiency from mature embryo. While, for salt and heat tolerance, the proliferation efficiency, embryonic efficiency, and regeneration efficiency were used. The results show significant medium and genotype effects for the embryogenesis capacity of calluses induction and plantlets regeneration under saline and thermal stresses. Mahon-Demias showed good callus induction and ability to proliferate and regenerate seedling under heat and salt stress conditions compared to Hidhab. No sizeable differences were observed between the two genotypes at higher salt stress rates. This study will serve as a base line for in vitro screening of several elite wheat cultivars for their ability to induce callus and regenerate plants from mature embryos, and to start selection for tolerance to salinity.
