The risk of degenerative diseases decreases with the consumption of high antioxidant containing fruits. This study aimed to analyze the proximate composition and antioxidant activity of pulp and seed of Syzygium cumini. Proximate composition was determined by following the AOAC method. Minerals and heavy metals were analyzed using a flame photometer and an atomic absorption spectrometer. In-vitro antioxidant activity was evaluated by using phosphomolybdenum, ferrous ion chelating ability assay and reducing power assay methods. In proximate composition analysis pulp contained significantly (p<0.05) greater amount of minerals than seed but seed contained a higher amount of protein and lipid than pulp. In the bioactive compound analysis, seed extracts possessed significantly (p<0.05) greater amount of Chlorophyll A, Chlorophyll B and total carotenoids. Additionally, seed extracts exhibited significantly (p<0.05) more total antioxidant capacity, reducing power and ferrous ion chelating ability compared to pulp extracts. This study revealed that the seed of Syzygium cumini contains significantly (p<0.05) higher amount of minerals, bioactive compounds and showed better antioxidant property. Therefore, Syzygium cumini seed can be a potential source of natural antioxidant in medicine and food production.

<p>Phytotherapy is a discipline which is interested in the design, the preparation and the interpretation of structure activity relationship of the natural bioactive molecules. In this context, ethanolic leaves extract of <em>Melia azedarach</em> L. was phytochemically analysed on the bases of HPLC and by GC–MS. Extract wase tested for his in vitro antioxidant activities by 1,1-diphenyl-2-picrylhydrazyl (DPPH), H<sub>2</sub>O<sub>2</sub>, hydroxyl radical scavenging activity, Ferric Reducing Power (FRP) and Ferrous ion chelating abilities methods. The antioxidant activity of the extract was analyzed simultaneously with their pro-oxidant capacity. The ratio of pro-oxidant to the antioxidant activity (ProAntidex) represents a useful index of the net free radical scavenging potential of the synthesized compounds. Background: Protocatechuic acid (PCA) is a natural phenolic acid widely distributed in plants and is considered as an active component of some traditional Chinese herbal medicines such as Cibotium barometz (L.) J.Sm, Stenoloma chusanum (L.) Ching, Ilex chinensis Sims. PCA was reported to possess various pharmacological effects which may be closely correlated with its antioxidant activities. However, the antioxidant of PCA has not been investigated systematically yet. Methods: In the study, the antioxidant activities of protocatechuic acid were measured in vitro using various antioxidant assays including 1,1-diphenyl-2-picryl-hydrazyl (DPPH•), 2,2’-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS+•), superoxide anion radicals (•O2-) and hydroxyl radical (•OH) scavenging activity, ferric ions (Fe3+) and cupric ions (Cu2+) reducing power, ferrous ions (Fe2+) and cupric ions (Cu2+) chelating activity, compared with the positive controls Trolox or BHT.Results: In all assays, PCA along with positive controls exhibited dose-dependently antioxidant ability. Comparing to a standard antioxidant Trolox, the relative antioxidant activity of PCA (i.e. the ratio of IC50(Trolox)/IC50(PCA) ) was calculated as 2.8, 2.3, 3.7, 6.1, 4.2, 1.0, 2.7, 1.5, respectively, for DPPH, ABTS, reducing power (Fe3+), reducing power (Cu2+), superoxide anion radical-scavenging, hydroxyl radical-scavenging, chelating ability (Fe2+) and chelating ability (Cu2+). Conclusion: Comparing to Trolox, PCA shows much more effective antioxidant activity in vitro in both lipid and aqueous media. Hence, it could therefore be used in pharmacological or food industry as a natural antioxidant. It may exhibit antioxidant activity by both chelating metal transition ions as well as by scavenging free radicals via donating hydrogen atom (H•) or electron (e).Keywords: Protocatechuic acid, antioxidant, reducing power, free radical-scavenging, chelating ability.

Background: Syzygium cumini L., commonly known as Jamun, black-plum, and Indian blackberry, is one of the most widely distributed trees in India with booming medical benefits and possesses antioxidant, anticancer and anti-diabetic properties. It belongs to the family Myrtaceae. Despite countless phytochemicals, seeds are not consumed and are the waste part of Jamun fruit. Objective: The objective of this study was to evaluate the antioxidant capacity of phenolics from Jamun seeds against a bundle of oxidant moieties. Methods: The 50% acetone extract of Jamun seeds was investigated for in-vitro antioxidant profiling. Assays include free radical scavenging activity, metal chelation activity, hydroxyl radical scavenging activity, hydrogen peroxide scavenging activity, total antioxidant activity, total reducing power, nitric oxide scavenging activity, and lipid peroxidation inhibition activity. Results: The extract depicted maximum DPPH radical scavenging activity followed by ABTS radical scavenging activity. Hefty metal chelation and nitric oxide scavenging activity were recorded while lipid peroxidation, H2O2, and OH- scavenging activity was intermediate. Conclusion: Jamun seed showed ample antioxidant activity and certifies that it is the right candidate for exploitation as a source of natural antioxidants to counteract autoxidation-induced pathologies or diseases.

All parts of Syzygium cumini Linn. (duwet) were widely used for medicinal plant in the treatment of various deseases.The seed extract was used to lower blood glucose. Cumini's seed had higher phenolic fractions than others. It were prepared by extracted of duwet seed "Genthong" varieties, using various extractants such as 85% ethyl acetate, 50% methanol and 50% ethanol. Duwet seed extract collected then was determined of phenolics compound and antioxidant activity assayed by measuring 2,2-diphenyl-1-picrylhydrazylradical (DPPH) scavenging activity, reducing Ferric ion (Fe3+) power and inhibition of linoleic acid oxidation. The objective was to select one of the extractant which gave highest yield and polyphenolic content and its stronger antioxidant activity. Extractant 50% methanol gave highest the extract yields was 16.29 % (db.) and phenolics compounds of extract was composed: total phenolic 45.99±0.25 g-GAE/100 g-extract; total flavonoid 2.28±0.07 g-QE/100 g-extract and total tannin 26.9±0.07 g-TAE/100 g-extract. All of extract exhibited strong on behalf of RSA-DPPH assay 87-95% (100  g-mL-l) and reducing power, nevertheless in inhibition of linoleic acid oxidation was moderate 49-55% (400  g-mL-l).Keywords: Syzygium cumini Linn, seed, antioxidant, extraction ABSTRAKTanaman duwet (Syzygium cumini Linn.) pada semua kompartemennya dimanfaatkan masyarakat untuk pengobatansuatu penyakit. Ekstrak bijinya dimanfaatkan untuk penurun gula darah. Bijinya merupakan bagian tanaman yang kaya senyawa polifenol. Fraksi kaya senyawa fenolik dipreparasi dengan mengekstraksi biji duwet varietas "Genthong", dengan tiga jenis ekstraktan; etil acetat 85%, metanol 50% dan etanol 50%. Ekstrak biji duwet (EBD) yang diperoleh dianalisis kelompok senyawa fenolik dan aktivitas antioksidan menggunakan metode uji penangkapan radikal DPPH (2,2-diphenil 1-picrylhydrazyl), uji reduksi ion Feri (ferric reduction antioxidant power-FRAP) dan uji penghambatan peroksidasi asam lemak linoleat. Tujuan penelitian adalah memilih satu dari tiga ekstraktan yang menghasilkaan ekstrak, dengan yield dan kadar senyawa polifenolik terbesar serta sifat antioksidatif terkuat. Yield dengan ekstraktan Met-OH-50%, sebesar 16,29% (db), senyawa fenoliknya sebesar 45,99 ±0,25 g-GAE/100 g-EBD; 2,28±0,07 g-QE/100 g-EBD dan 26,9±0,07 g-TAE/100 g-EBD. Ketiga ekstrak kuat dalam uji penangkapan radikal DPPH antara 87-95 % (100 g-mL-l) dan uji reduksi ion Feri (Fe3+), moderat pada uji penghambatan peroksidasi lipid, 49-52 % pada 400  g- mL-l.Kata kunci: Biji duwet (Syzygium cumini Linn.), antioksidan, ekstraksi

In this study a polyphenolic extract from Cleome arabica leaves (CALE) was investigated for its antioxidant activity in vitro using DPPH•, metal chelating and reducing power methods and for its protective effects against AraC-induced hematological toxicity in vivo using Balb C mice. Results indicated that CALE exhibited a strong and dose-dependent scavenging activity against the DPPH• free radical (IC50 = 4.88 μg/ml) and a high reducing power activity (EC50 = 4.85 μg/ml). Furthermore, it showed a good chelating effects against ferrous ions (IC50 = 377.75 μg/ml). The analysis of blood showed that subcutaneous injection of AraC (50 mg/kg) to mice during three consecutive days caused a significant myelosupression (P < 0.05). The combination of CALE and AraC protected blood cells from a veritable toxicity. Where, the number of the red cells, the amount of hemoglobin and the percentage of the hematocrite were significantly high. On the other hand, AraC cause an elevation of body temperature (39 °C) in mice. However, the temperature of the group treated with CALE and AraC remained normal and did not exceed 37.5 °C. The observed biological effects of CALE, in vitro as well as in vivo, could be due to the high polyphenol and flavonoid contents. In addition, the antioxidant activity of CALE suggested to be responsible for its hematoprotective effect.

Background: Quercus serrata Murray leaves have been used traditionally in the treatment of diabetes, dysmenorrhoea, inflammation and urinary tract infection. So, far no study had been reported on the toxicological profile and antioxidant properties of the plant. Objective: The present study was aimed to investigate the in-vivo toxicological profile and in-vitro antioxidant activities of the methanolic extract of standardized Quercus serrata leaves. Methods: Per-oral sub-acute toxicity study was performed in rats using three dose levels (200, 400 and 800 mg/kg b.w.) of the extract for 28-days. Control group received gum acacia suspended in water. Bodyweight was measured weekly. Biochemical parameters were analysed using the serum, the blood-cell count was done using whole blood. Pathological changes were also checked in highly perfused tissues. Further, in-vitro reducing power assay, nitric oxide scavenging assay, DPPH free-radical scavenging assay were performed to check the antioxidant activity of the extract. Results: There were no significant alterations in the blood-cell count and biochemical parameters analysed in the treatment group when compared with the normal control. Histopathology study of liver, kidney, pancreas, heart and brain revealed normal cellular architecture in the treatment groups alike the control group animals. Quercus serrata also showed a significant reduction of DPPH with IC50 4.48±0.254 µg/mL, in-vitro reducing power activity with IC50121.65±0.320 µg/mL and nitric oxide scavenging activity IC50 106.43±0.338 µg/mL. Conclusion: The above study showed that standardized methanolic extract of Quercus serrata leaves was safe after subacute oral administration in rats and has good antioxidant potential.

Background: Curcumin, as the substantial constituent of the turmeric plant (Curcuma longa), plays a significant role in the prevention of various diseases, including diabetes. It possesses ideal structure features as enzyme inhibitor, including a flexible backbone, hydrophobic nature, and several available hydrogen bond (H-bond) donors and acceptors. Objective: The present study aimed at synthesizing several novel curcumin derivatives and further evaluation of these compounds for possible antioxidant and anti-diabetic properties along with inhibitory effect against two carbohydrate-hydrolyzing enzymes, α-amylase and α-glucosidase, as these enzymes are therapeutic targets for attenuation of postprandial hyperglycemia. Methods: Therefore, curcumin-based pyrido[2,3-d]pyrimidine derivatives were synthesized and identified using an instrumental technique like NMR spectroscopy and then screened for antioxidant and enzyme inhibitory potential. Total antioxidant activity, reducing power assay and 1,1-diphenyl-2-picrylhydrazyl (DPPH• ) radical scavenging activity were done to appraisal the antioxidant potential of these compounds in vitro. Results: Compounds L6-L9 showed higher antioxidant activity while L4, L9, L12 and especially L8 exhibited the best selectivity index (lowest α-amylase/α-glucosidase inhibition ratio). Conclusion: These antioxidant inhibitors may be potential anti-diabetic drugs, not only to reduce glycemic index but also to limit the activity of the major reactive oxygen species (ROS) producing pathways.

Amomum subulatum, commonly known as large or black cardamom, is a commonly used spice in Indian kitchens and is traditionally used to treat various ailments. To add more knowledge about the medicinal values of the fruit, this study was conducted to evaluate the in vitro antioxidant activities of aqueous, methanol, ethanol, hydro alcohol, ethyl acetate, acetone and chloroform extracts of the fruit. Preliminary assessment was done to detect the presence of phytoconstituents using identification tests. The antioxidant activity was measured by employing methods such as diphenylpicrylhydrazyl (DPPH) radical scavenging assay, total antioxidant activity equivalent to ascorbic acid, reducing power assay and superoxide anion scavenging assay. The antioxidant activities were compared with their respective phenol and flavonoid contents. Preliminary assessment revealed that large cardamom fruit is a good source of all the bioactive constituents as well as phenol and flavonoid essential for medicinal values. The extract obtained by polar solvents showed the highest antioxidant efficacy in relation to its phenol content. Also, all the solvent-soluble fractions showed a concentration-dependent antioxidant effect. Results from our study prove that large cardamom can alleviate oxidative stress, suggesting the potential of large cardamom as a functional food

The antioxidant activities of crude extract ofPhaulopsis fascisepalaleaf were evaluated and compared with α-tocopherol and BHT as synthetic antioxidants and ascorbic acid as natural-based antioxidant.In vitro, we studied its antioxidative activities, radical-scavenging effects, Fe2+-chelating ability and reducing power. The total phenolic content was determined and expressed in gallic acid equivalent. The extract showed variable activities in all of thesein vitrotests. The antioxidant effect ofP. fascisepalawas strongly dose dependent, increased with increasing leaf extract dose and then leveled off with further increase in extract dose. Compared to other antioxidants used in the study, α-Tocopherol, ascorbic acid and BHT,P. fascisepalaleaf extract showed less scavenging effect on α,α,-diphenyl-β-picrylhydrazyl (DPPH) radical and less reducing power on Fe3+/ferricyanide complex but better Fe2+-chelating ability. These results revealed thein vitroantioxidant activity ofP.fascisepala.Further investigations are necessary to verify these activitiesin vivo.

The aim of this study was to investigate the biological and chemical activity on two species of fungi of the genus Penicillium isolated from wastewater. On the selected species of fungi the different antioxidant activity assays were carried out: DPPH free-radical scavenging activity, total antioxidant activity, Fe2+- chelating ability and Fe3+- reducing power. Total phenol content was also determinate for ethanolic extract of mycelia. Penicillium chrysogenum ethanolic extract contained higher total phenolic content and better total antioxidant capacity as well as ferrous ion chelating ability. Penicillium fumiculosum ethanolic extract showed higher DPPH free-radical scavenging activity, as well as reducing power. Based on the obtained results it can be concluded that two types of fungi are potential new sources of natural antioxidants.
