All parts of Syzygium cumini Linn. (duwet) were widely used for medicinal plant in the treatment of various deseases.The seed extract was used to lower blood glucose. Cumini's seed had higher phenolic fractions than others. It were prepared by extracted of duwet seed "Genthong" varieties, using various extractants such as 85% ethyl acetate, 50% methanol and 50% ethanol. Duwet seed extract collected then was determined of phenolics compound and antioxidant activity assayed by measuring 2,2-diphenyl-1-picrylhydrazylradical (DPPH) scavenging activity, reducing Ferric ion (Fe3+) power and inhibition of linoleic acid oxidation. The objective was to select one of the extractant which gave highest yield and polyphenolic content and its stronger antioxidant activity. Extractant 50% methanol gave highest the extract yields was 16.29 % (db.) and phenolics compounds of extract was composed: total phenolic 45.99±0.25 g-GAE/100 g-extract; total flavonoid 2.28±0.07 g-QE/100 g-extract and total tannin 26.9±0.07 g-TAE/100 g-extract. All of extract exhibited strong on behalf of RSA-DPPH assay 87-95% (100  g-mL-l) and reducing power, nevertheless in inhibition of linoleic acid oxidation was moderate 49-55% (400  g-mL-l).Keywords: Syzygium cumini Linn, seed, antioxidant, extraction ABSTRAKTanaman duwet (Syzygium cumini Linn.) pada semua kompartemennya dimanfaatkan masyarakat untuk pengobatansuatu penyakit. Ekstrak bijinya dimanfaatkan untuk penurun gula darah. Bijinya merupakan bagian tanaman yang kaya senyawa polifenol. Fraksi kaya senyawa fenolik dipreparasi dengan mengekstraksi biji duwet varietas "Genthong", dengan tiga jenis ekstraktan; etil acetat 85%, metanol 50% dan etanol 50%. Ekstrak biji duwet (EBD) yang diperoleh dianalisis kelompok senyawa fenolik dan aktivitas antioksidan menggunakan metode uji penangkapan radikal DPPH (2,2-diphenil 1-picrylhydrazyl), uji reduksi ion Feri (ferric reduction antioxidant power-FRAP) dan uji penghambatan peroksidasi asam lemak linoleat. Tujuan penelitian adalah memilih satu dari tiga ekstraktan yang menghasilkaan ekstrak, dengan yield dan kadar senyawa polifenolik terbesar serta sifat antioksidatif terkuat. Yield dengan ekstraktan Met-OH-50%, sebesar 16,29% (db), senyawa fenoliknya sebesar 45,99 ±0,25 g-GAE/100 g-EBD; 2,28±0,07 g-QE/100 g-EBD dan 26,9±0,07 g-TAE/100 g-EBD. Ketiga ekstrak kuat dalam uji penangkapan radikal DPPH antara 87-95 % (100 g-mL-l) dan uji reduksi ion Feri (Fe3+), moderat pada uji penghambatan peroksidasi lipid, 49-52 % pada 400  g- mL-l.Kata kunci: Biji duwet (Syzygium cumini Linn.), antioksidan, ekstraksi

Glutinous rice-based foods have a long history are consumed worldwide. They are also in great demand for the pursuit of novel sensory and natural health benefits. In this study, we developed a novel fermented glutinous rice product with the supplementation of Fu brick tea. Using in vitro antioxidant evaluation and phenolic compounds analysis, fermentation with Fu brick tea increased the total phenolic content and enhanced the antioxidant activity of glutinous rice, including scavenging of 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) radical, 2,2′-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) radical, and hydroxyl radical, ferric-reducing antioxidant power, and ferric ion reducing power and iron chelating capability. Besides, compared with traditional fermented glutinous rice, this novel functional food exhibited a stronger activity for protecting DNA against hydroxyl radical-induced oxidation damage. Quantitative analysis by HPLC identified 14 compounds covering catechins and phenolic acids, which were considered to be positively related to the enhanced antioxidant capability. Furthermore, we found that 80% ethanol was a suitable extract solvent compared with water, because of its higher extraction efficiency and stronger functional activities. Our results suggested that this novel fermented glutinous rice could serve as a nutraceutical food/ingredient with special sensory and functional activities.

Verbascum L. species have been used since ancient times in traditional medicine thanks to their bioactive compounds. In this study, we evaluated the antioxidant activity and phenolic contents of ethanolic and acetonic extract of Verbascum glomeratum that collected from Denizli, Turkey. The antioxidant activity were evaluated by using various methods (β-carotene/linoleic acid, ferric reducing power assays (FRAP), radical scavenging assays of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2and#39;-azino-bis-3-ethyl benzthiazoline-6-sulphonic acid (ABTS), phoshomolibdenum methods) and total phenolic, flavonoid and tannin contents in the extracts were determined. Antioxidant activity was determined as β-carotene/linoleic acid (%58.5and#177;0.021), DPPH (IC50: 1.220and#177;0.025) and ABTS (IC50:1.552and#177;0.021), ferric reducing antioxidant power (FRAP) (4.151and#177;0.056 mgTE/g), phoshomolibdenum assay (8.218and#177;0.015 mgAE/g) total phenolic, total flavonoid and tannin contents was found as (4.41and#177;0.007 mgGAE/mL), (87.142and#177;0.009 and#181;QE/g), and (30.178and#177;0.008 mgCEs/g) respectively, and then 14 different phenolic compounds in ethanol extracts was carried out by HPLC. Epicatechin (2742.09 and#181;g/g) and 2,5dihidroksi (2544.96 and#181;g/g) were the most abundant phenolic constituents in the extracts. According the results, the extract of V. glomeratum may be considered as a potential source of biological agents.

Objective: Diabetes, the “third killer” of mankind, is now a global burden affecting nearly 10% of the population. The present study aimed at pharmacognostical evaluation of Syzygium cumini seed powder, phytochemical profiling, total phenolic, and flavonoid content determination and to ascertain its antioxidant activity and hypoglycemic potential in alloxan-induced diabetic rats.Methods: Pharmacognostical and physicochemical parameters of the seed powder were determined by using standard methods as per Ayurvedic Pharmacopoeia of India (API). In vitro antioxidant activity was determined by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. Ethanolic extract of Syzygium cumini seed at a dose of 200mg/kg and 400mg/kg body weight (bw) were given orally in alloxan-induced (150mg/kg, i.p) diabetic rats daily for three weeks.Results: physicochemical parameters complied with the API standards. Phytochemical profiling revealed that the extract contains phenolic compounds, flavonoids, glycosides, alkaloids, tannins, and saponins. The extract is rich in phenolic compounds which were 177.33 mg gallic acid equivalent/g (GAE/g dry weight) and showed significant percentage inhibition compared to standard ascorbic acid. The extract reduced the fasting blood sugar (FBS) level by 46.67–52.67% which is very close to the standard drug glibenclamide (reduced FBS by 65.58%). It also improved the bw by 18.20–20.41% after extract treatment where the standard drug increased the bw by 22.95% of the diabetic rats.Conclusion: In conclusion, Syzygium cumini seed extract is rich in phenolic compounds. Results suggested that the extract possesses significant radical (DPPH) scavenging activity and FBS lowering potential in alloxan-induced diabetic rats.

AbstractWe conducted this study to determine and compare the content of phenolic compounds and flavonoids in the argan and olive leaves as well as their antioxidant capacity in aqueous, methanolic, and ethyl acetate extracted fractions. In vitro antioxidant activity was evaluated in comparison with synthetic antioxidants by assessing DPPH• radical scavenging capacity, ferric reducing antioxidant power, scavenging ability by inhibiting the β-carotene/linoleic acid emulsion oxidation, and by the ABTS radical scavenging activity assay. Total phenolic content in argan samples ranged from 221.69 ± 2.07 to 1.32 ± 0.01 mg GAE/g DW and in olive samples from 144.61 ± 0.82 to 1.21 ± 0.02 mg GAE/g DW. Total flavonoids content in argan samples varied from 267.37 ± 1.12 to 25.48 ± 0.02 mg QE/g DW, while in olives from 96.06 ± 0.78 to 10.63 ± 0.05 mg QE/g DW. In vitro antioxidant studies strongly confirmed the antioxidant potency of argan and olive leaves and their richness in secondary metabolites that are effective in free radicals scavenging and metal chelating capacities, indicating their antioxidant power.

Exudates (DE) secreted from two shiitake mushroom mycelia (strains 1358 and L5458) were evaluated for their antioxidative properties and phenolic content. 1358DE and L5458DE showed distinct antioxidant activity in different in vitro assays, including scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, hydroxyl radical, superoxide anions and hydrogen peroxide; the ability to chelate ferrous ions; reducing power; hemolysis inhibition activity in rat erythrocyte; and lipid peroxidation inhibition (IC50 values of 1358DE and L5458DE were 3.3 and 132.6; 44.5 and > 1000; 26.9 and 53.7; 153.6 and >175.0; 176.0 and 521.0; 26.7 and 746.4; 47.8 and 736.9; and 3.1 and > 1000 μg/mL, respectively). Their total phenolic content was 237.33 and 24.08 mg gallic acid equivalent (GAE)/g of dry DE, respectively. Overall, these results show that 1358DE generally possesses better antioxidant properties than L5458DE, possibly due to its larger total phenolic content. Shiitake mushroom mycelial exudates, particularly of 1358DE, could be a good source of natural antioxidants.

A raw extract and four extractive fractions were obtained from Cedrus brevifolia (Cyprus cedar) bark. They were all studied regarding the phenolic content and profile using spectrophotometry and HPLC-DAD-ESI-MS. The antioxidant activity was investigated using in vitro assays: DPPH and ABTS radicals scavenging and reducing power assays. The ethyl acetate fraction had the highest total phenolic and proanthocyanidin contents; a taxifolin-O-hexoside, catechin, epicatechin and procyanidin oligomers (three dimers, two trimers) were identified in this fraction. The ethyl acetate fraction was found to possess the highest DPPH and ABTS radicals scavenging effects (EC50=13.9 ± 0.3 and 2.3 ± 0.0 μg/mL, respectively) and reducing capacity (EC50=9.1 ±0.1 μg/mL). Antioxidant effects were highly correlated with total phenolic and proanthocyanidin contents (r=0.89-0.99). These results suggest that Cedrus brevifolia bark is a new source of antioxidants.

Edible flowers have been used as ingredients because of their biological activities, taste, and overall appearance. This research was aimed to characterize the chemical composition and in vitro antioxidant activity of the marigold flower (Calendula officinalis L.) extracted with different proportions of water and ethyl alcohol, and the lyophilized extract with higher content of antioxidant compounds was incorporated into an organic yogurt. Results showed that the hydroalcoholic extract (50:50 v/v) presented the highest total phenolic content (TPC), flavonoids, and antioxidant activity (ferric reducing antioxidant power (FRAP), total reducing capacity (TRC), and Cu2+/Fe2+ chelating ability). Phenolic acids and flavonoids were quantified in the extract by LC-DAD, while 19 compounds were tentatively identified by ESI-MS/MS. The lyophilized marigold extract (LME) also inhibited 12% of Wistar rat’s brain lipid oxidation in vitro, inhibited α-amylase, and α-glucosidase activities, but showed no cytotoxicity towards cancerous cells (HCT8 and A549). However, marigold flower extract protected human erythrocytes against mechanical stress. When added into an organic yogurt model (0 to 1.5%), LME increased TPC and antioxidant activity (2,2-diphenyl-1-picrylhydrazyl (DPPH) and TRC), and the sensory analysis showed that the organic yogurt had an acceptance of 80.4%. Our results show that the use of LME may be a technological strategy to increase the content of bioactive compounds in yogurts.

Pterygota alata is a large deciduous tree of Malvaceae family. The present study was designed to evaluate in vitro antioxidant activity of stem bark of the plant. Ferric reducing power (FRP) test, 1, 1- diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging test and total antioxidant capacity (TAC) assay were used to detect the antioxidant activity. Total phenolic and flavonoid contents were also determined as they are well known phytochemicals with antioxidant property. The bark of the plant showed efficient reducing power as well as free radical scavenging property (IC50 values 52.25 - 172.05). The bark also found rich in total phenolic and flavonoid content. The highest amount of total phenolic content was found in chloroform soluble fraction (29.898 µg/mL) followed by ethyl acetate soluble fraction (15.88 µg/mL). The highest content of total flavonoid also detected in chloroform soluble fraction (107.56 µg/mL) but followed by crude ethanol extract (98.66 µg/mL). Overall, the bark of the plant possesses significant antioxidant activity, therefore can be used as a good natural source of antioxidant. © 2014 JSR Publications. Crataegusfruit is one of most important fruits in Tunisian flora. Some fruits of this genus are edible. This study was undertaken in order to examine the benefits of these fruits in human health and their composition of antioxidants including total polyphenol, flavonoids, proanthocyanidins content, and total anthocyanins. The antioxidative properties of the ultrasonic methanolic extract were assessed by different in vitro methods such as the FRAP, DPPH, andβ-carotene/linoleic acid assay. We concluded that peel fraction of red fruits possessed relatively high antioxidant activity and might be a rich source of natural antioxidants in comparison with the pulp and seed fruit extract. The results also showed that hawthorn yellow fruit presents lower amounts of phenolic content, absence of anthocyanins, and less antioxidant capacity. Most of peel and seed fractions were stronger than the pulp fractions in antioxidant activity based on their DPPH IC50, FRAP values, and results ofβ-carotene/linoleic acid. The total phenolic compounds contents were also highly correlated with the DPPH method and the FRAP assay.

The present study was designed to confirm the traditional use of the fruits of Ficus hispida Linn. (Moraceae) as an antioxidant agent. Fruits of the plant extracted with methanol and crude methanol extract (CME) were further fractionated with n-hexane, chloroform, and ethyl acetate. All the fractions, n-hexane (NHF), chloroform (CHF), ethyl acetate (EAF), aqueous (AQF) and CME were preliminary screened for in vitro antioxidant activity and total phenolic and total flavonoid content. In DPPH radical scavenging assay, CME exhibited highest scavenging activity (IC50 = 11.20 µg/mL) as compared to other fractions. In this assay, IC50 of reference standard BHT was 5.10 µg/mL. The reducing power of the samples was in the order as AQF > CME > CHF > EAF > NHF. The results for hydrogen peroxide scavenging activity indicated that CME, EAF and AQF had almost the same scavenging activity except NHF. Total antioxidant capacity of CME and other fractions were ranked as CHF > AQF > CME > EAF > NHF.  In the assay of antioxidant constituents (total phenol and total flavonoids content), the CME had highest phenolic and flavonoids content. The results indicate that Ficus hispida fruits could be considered as a potential source of natural antioxidant.
