Background. The modern system of molecular typing of the Russian population of T. pallidum makes it possible to obtain results with a significant dominance of the 14d/f type, which determines the need to increase the differentiating ability of the applied methods of molecular typing of T. pallidum. Aim. Identification and analysis of nucleotide sequence variability of internal gene fragments of the tprII family of Russian T. pallidum subsp. pallidum strains. Material and methods. The study of internal variable fragments of genes of the tprII family was carried out among 240 clinical isolates of T. pallidum obtained from the Central (Kaluga Region, Moscow), North Caucasian (Stavropol Territory), Far East (Republic of Sakha), Volga (Chuvash Republic), Southern (Astrakhan Region) and Siberian (Novosibirsk and Omsk Regions, Republic of Tyva) federal districts in 20142020. The sequence of internal variable fragments of genes of the tprII family was determined using capillary sequencialng technology. Results. The primers allowing both direct amplification of the internal variable region of the tprII genes subfamily and correct sequencing of their internal regions have been proposed. It was found one SNP at positions 1340 of tprG gene. The polymorphism differs the reference Nichols strain from globally distributed Street 14 genogroup variants. Conclusion. The variability of tprII subfamily genes nucleotide sequences in modern Russian strains of T. pallidum subsp. pallidum is an additional fund to increase the efficiency of the modern T. pallidum molecular typing system.

Aim. Analysis of the arp gene internal fragment nucleotide sequences variability in modern russian T. pallidum subsp. pallidum strains. Materials and methods. 57 T. pallidum isolates obtained from specialized dermatovenerologic clinics of the Central (Kaluga), the North Caucasus (Stavropol) and the Siberian (Tyva) regions in 2016 — 2017 were used in the study. The sequensing of the arp gene was performed using capillary electrophoresis technology. Results. A two-round amplification of the arp gene have been proposed, which ensures a correct reading of its internal region. Four variants of 60-nucleotide repeats in the internal arp fragment are described, which differing in 6, 8 and 15 — 17 codons compositions. Various combinations of these repeats, corresponding to the reference Nichols strain, globally distributed Street 14 genogroup, and the firstly described Stavropol regional variant are shown. Conclusion. The prospect of arp gene sequencing as a way to increase T. pallidum molecular typing efficiency is postulated.

Abstract We have determined the lesions in a number of mutant alleles of beta Tub85D, the gene that encodes the testis-specific beta 2-tubulin isoform in Drosophila melanogaster. Mutations responsible for different classes of functional phenotypes are distributed throughout the beta 2-tubulin molecule. There is a telling correlation between the degree of phylogenetic conservation of the altered residues and the number of different microtubule categories disrupted by the lesions. The majority of lesions occur at positions that are evolutionarily highly conserved in all beta-tubulins; these lesions disrupt general functions common to multiple classes of microtubules. However, a single allele B2t6 contains an amino acid substitution within an internal cluster of variable amino acids that has been identified as an isotype-defining domain in vertebrate beta-tubulins. Correspondingly, B2t6 disrupts only a subset of microtubule functions, resulting in misspecification of the morphology of the doublet microtubules of the sperm tail axoneme. We previously demonstrated that beta 3, a developmentally regulated Drosophila beta-tubulin isoform, confers the same restricted morphological phenotype in a dominant way when it is coexpressed in the testis with wild-type beta 2-tubulin. We show here by complementation analysis that beta 3 and the B2t6 product disrupt a common aspect of microtubule assembly. We therefore conclude that the amino acid sequence of the beta 2-tubulin internal variable region is required for generation of correct axoneme morphology but not for general microtubule functions. As we have previously reported, the beta 2-tubulin carboxy terminal isotype-defining domain is required for suprastructural organization of the axoneme. We demonstrate here that the beta 2 variant lacking the carboxy terminus and the B2t6 variant complement each other for mild-to-moderate meiotic defects but do not complement for proper axonemal morphology. Our results are consistent with the hypothesis drawn from comparisons of vertebrate beta-tubulins that the two isotype-defining domains interact in a three-dimensional structure in wild-type beta-tubulins. We propose that the integrity of this structure in the Drosophila testis beta 2-tubulin isoform is required for proper axoneme assembly but not necessarily for general microtubule functions. On the basis of our observations we present a model for regulation of axoneme microtubule morphology as a function of tubulin assembly kinetics.

The retrospective epidemiologic analysis of HFRS morbidity carried out from 2001 to 2007 showed that the highest sickness rate was registered in Privolzhsky federal district with 20.4±2.3 average index that exceeded the similar one countrywide 4-fold and the incidence specific weight was 88.0 % of the total number of cases. The indexes of HFRS incidence in the rest 6 districts were lower than the Russia wide one 3-4-fold and more. During the analyzed period the tendency to increase the HFRS incidence level was observed in Privolzhsky, Ural and Far East districts and in the country.

Objective of the study – assessment of epizootic and epidemic situation on tularemia in 2018 and forecasting the risk of infection in the territory of the Russian Federation in 2019. Analysis of epidemiological situation was carried out on the basis of the data from monitoring activities performed by the Rospotrebnadzor institutions and the data contained in the reports of the Irkutsk Research Anti-Plague Institute of Siberia and Far East, Plague Control Center, Federal Center of Hygiene and Epidemiology, as well as federal statistical survey forms No 5 “Information on preventive vaccination” and No 2 “Information on infectious and parasitic  diseases” over the period of January-December 2018 in eight Federal Districts including 85 constituent entities. Given are the retrospective data on tularemia epidemic situation in the territory of the Russian Federation over the past decade. 1944 human tularemia cases were registered in Russia between 2009 and 2018, 1005 out of which occurred during epidemic outbreak in 2013 in Khanty-Manssiysk Autonomous Region. High sporadic and small cluster incidence was mainly observed in the territories of the North-estern and Siberian Federal Districts over the recent years. In 2018, 71 cases of human infection with tularemia agent were reported. Epizootic manifestations of varying degree of intensity were detected in 52 entities of Russia. Against that background, sporadic cases of human infection were registered in 19 regions of the country. For three years epidemic complications expressed to the maximum were observed in the Omsk Region – 18 cases of tularemia infection, and Karelia – 14 cases, respectively. 15 Francisella tularensis cultures were isolated from ambient  environment objects in Pskov, Leningrad Regions, Altai Territory, Republics of Altai and Tuva. Conclusions have been drawn in relation to the regions where epidemic complications associated with tularemia are most likely to emerge in 2019.

The Article presents an analysis of the role of the media sector in the structure created by the placement of tourist routes in the subjects of the Russian Federation. The sample size was 6000 routes in all federal districts of Russia. The problem of poor service provided by the hotel services in Russia is one of the major problems hindering the development of domestic tourism. Value for money and sometimes do not match. Part of the situation could have saved the increasing competition with the hotels listed in the international circuit, which created its own rules of corporate culture, high standards of service, clearly defined for each category of hotel (in terms of stardom). However, this does not justify a revision of its marketing strategy of independent hoteliers. It is also important to pay attention to accommodation, similar to hotels and other accommodation facilities. They are also well represented in the tourist market. According to the author, the low level of service in accommodation facilities is determined not so much by the reluctance of management to improve it, as the structure of demand from tourists and tour operators, to create a product. Most of the routes, which include the need to accommodate tourists, implemented on average, in the area the day — two. At the same time, tourists do not stay in accommodation facilities for a long time and continue your route on. This circumstance serves as an incentive to change the quality of services. Moreover, the article stipulates that personal and other accommodations are well represented in the routes of major federal districts like Central, Ural, Siberia, Far East. Accommodation in hotels more common routes in the North- West, Volga, the Crimea and North Caucasian Federal District. Accommodation facilities, in this case, are the operators of the individual passive format services. However, this strategy cannot be used by all players of the hospitality industry in all federal districts. The specificity of the regions and their remoteness from each other, the price level in the field must be limiting conditions in which hoteliers could develop. It is therefore proposed that the need to actively offer ideas own hiking tour operators or actively interact with them, attracting all new, including the unorganized tourist flows.
