AbstractMicroalgae hold great potential as a source for renewable energy due to their high photosynthetic efficiency, high growth rates and independence from fertile agricultural lands. However, large-scale cultivation systems of microalgae biomass are still not economically viable mainly due to the difficulties with maintaining optimum growth conditions of microalgae in open pond systems and high costs of biomass cultivation and harvesting. Here we propose the Novel Stacked Modular Open Raceway Ponds (SMORPs) system for microalgae biomass cultivation to be integrated in biogas production plant. The proposed technological solution will eliminate the drawbacks of current microalgae cultivation technologies, mainly, will reduce the land use, improve lighting conditions and reduce the cost of cultivation as a result of the application of waste products from biogas production, i.e. anaerobic digestion effluent and flue gas. In this study we propose the initial design of the SMORP concept and a microalgae biomass kinetic model as a simple approach to screen microalgae strains potentially applicable for large-scale ponds. The developed tool is also useful to evaluate the potential benefit of additional artificial LED light sources and to assess the maximum biomass growth rate with minimal light intensity.

In the paper experiments and theory of biogas production using industrial waste from paper production as a co-substrate are described. The main aim of the experiments was to evaluate the sensitivity and applicability of the biochemical conversion using the anaerobic digestion of the mixed biomass in the pilot fermentor (5 m3), where the mesophillic temperature was maintained. It was in parallel operation with a large scale fermentor (100 m3). The research was carried out at the biogas plant in Kolíňany, which is a demonstration facility of the Slovak University of Agriculture in Nitra. The experiments proved that the waste arising from the paper production can be used in case of its appropriate dosing as an input substrate for biogas production, and thus it can improve the economic balance of the biogas plant.

Abstract Background Dendrobium catenatum belongs to the Orchidaceae, and is a precious Chinese herbal medicine. In the past 20 years, D. catenatum industry has developed from an endangered medicinal plant to multi-billion dollar grade industry. The necrotrophic pathogen Sclerotium delphinii has a devastating effection on over 500 plant species, especially resulting in widespread infection and severe yield loss in the process of large-scale cultivation of D. catenatum. It has been widely reported that Jasmonate (JA) is involved in plant immunity to pathogens, but the mechanisms of JA-induced plant resistance to S. delphinii are unclear. Results In the present study, the role of JA in enhancing D. catenatum resistance to S. delphinii was investigated. We identified 2 COI1, 13 JAZ, and 12 MYC proteins in D. catenatum genome. Subsequently, systematic analyses containing phylogenetic relationship, gene structure, protein domain, and motif architecture of core JA pathway proteins were conducted in D. catenatum and the newly characterized homologs from its closely related orchid species Phalaenopsis equestris and Apostasia shenzhenica, along with the well-investigated homologs from Arabidopsis thaliana and Oryza sativa. Public RNA-seq data were investigated to analyze the expression patterns of D. catenatum core JA pathway genes in various tissues and organs. Transcriptome analysis of MeJA and S. delphinii treatment showed exogenous MeJA changed most of the expression of the above genes, and several key members, including DcJAZ1/2/5 and DcMYC2b, are involved in enhancing defense ability to S. delphinii in D. catenatum. Conclusions The findings indicate exogenous MeJA treatment affects the expression level of DcJAZ1/2/5 and DcMYC2b, thereby enhancing D. catenatum resistance to S. delphinii. This research would be helpful for future functional identification of core JA pathway genes involved in breeding for disease resistance in D. catenatum.

The use of formal methods and artificial intelligence has made it possible to automatically design outdoor lighting. Quick design for large cities, in a matter of hours instead of weeks, and analysis of various optimization criteria enables to save energy and tune profit stream from lighting retrofit. Since outdoor lighting is of a large scale, having luminaires on every street in urban areas, and since it needs to be retrofitted every 10 to 15 years, choosing proper parameters and light sources leads to significant energy savings. This paper presents the concept and calculations of Levelized Cost of Electricity for outdoor lighting retrofit. It is understood as cost of energy savings, it is in the range from 23.06 to 54.64 EUR/MWh, based on real-world cases. This makes street and road lighting modernization process the best green “energy source” if compared with the 2018 Fraunhofer Institute cost of electricity renewable energy technologies ranking. This indicates that investment in lighting retrofit is more economically and ecologically viable than investment in new renewable energy sources.

Abstract Background Predatory flagellates and ciliates are two common bio-contaminants which frequently cause biomass losses in Chlorella mass culture. Efficient and targeted ways are required to control these contaminations in Chlorella mass cultivation aiming for biofuel production especially. Results Five surfactants were tested for its ability to control bio-contaminations in Chlorella culture. All five surfactants were able to eliminate the contaminants at a proper concentration. Particularly the minimal effective concentrations of sodium dodecyl benzene sulfonate (SDBS) to completely eliminate Poterioochromonas sp. and Hemiurosomoida sp. were 8 and 10 mg L−1, respectively, yet the photosynthesis and viability of Chlorella was not significantly affected. These results were further validated in Chlorella mass cultures in 5, 20, and 200 m2 raceway ponds. Conclusions A chemical method using 10 mg L−1 SDBS as pesticide to control predatory flagellate or ciliate contamination in Chlorella mass culture was proposed. The method helps for a sustained microalgae biomass production and utilization, especially for biofuel production.

Staphylotrichum longicolleum FW57 (DSM105789) is a prolific chitinolytic fungus isolated from wood, with a chitinase activity of 0.11 ± 0.01 U/mg. We selected this strain for genome sequencing and annotation, and compiled its growth characteristics on four different chitinous substrates as well as two agro-industrial waste products. We found that the enzymatic mixture secreted by FW57 was not only able to digest pre-treated sugarcane bagasse, but also untreated sugarcane bagasse and maize leaves. The efficiency was comparable to a commercial enzymatic cocktail, highlighting the potential of the S. longicolleum enzyme mixture as an alternative pretreatment method. To further characterize the enzymes, which efficiently digested polymers such as cellulose, hemicellulose, pectin, starch, and lignin, we performed in-depth mass spectrometry-based secretome analysis using tryptic peptides from in-gel and in-solution digestions. Depending on the growth conditions, we were able to detect from 442 to 1092 proteins, which were annotated to identify from 134 to 224 putative carbohydrate-active enzymes (CAZymes) in five different families: glycoside hydrolases, auxiliary activities, carbohydrate esterases, polysaccharide lyases, glycosyl transferases, and proteins containing a carbohydrate-binding module, as well as combinations thereof. The FW57 enzyme mixture could be used to replace commercial enzyme cocktails for the digestion of agro-residual substrates.

The hairy root clones of Gentiana dinarica cl-B, cl-D, cl-3, and cl-14 were cultivated in parallel in diverse simple bioreactors, including temporary immersion systems RITA® (TIS RITA®), bubble column bioreactors (BCB), and Erlenmeyer flasks (EF), and evaluated for biomass production and xanthone content. The obtained results showed that TIS RITA® and BCB containing ½ MS medium with 4% sucrose provided equally good growth conditions in which the majority of the clones displayed the higher percentage of dry matter (DM%), and xanthones norswertianin-1-O-primeveroside (nor-1-O-prim) and norswertianin production than those cultivated in EF. Thin and well branched hairy root clone cl-B grown in BCB for 7 weeks was superior regarding all growth parameters tested, including growth index (19.97), dry weight (2.88 g), and DM% (25.70%) compared to all other clones. Cl-B cultured in TIS RITA® contained the highest amount of nor-1-O-prim (56.82 mg per vessel). In BCB with constant aeration, cl-B accumulated the highest norswertianin content reaching 18.08 mg/vessel. The optimized conditions for cultivation of selected G. dinarica hairy root clones in highly aerated TIS RITA® and BCB systems contribute to the development of bioreactor technology designed for the large scale commercial production of xanthones nor-1-O-prim and norswertianin.
