Micropropagation involves the risk of somaclonal variation among the regenerated plants. It is possible that the components of the nutrient medium, especially growth substances, increase the frequency of variation. Elatior Begonia 'Barbara' plants were micropropagatedusing four cytokinins (zeatin 0.5—2.0 mg/l, kinetin 0.5—2.0 mg/l, IPA 0.5—2.0 mg/l or BAP 0.05—0.65 mg/l) in vitro. The number of shoots produced per explant and the subsequent quality of full-sized, flowering plants were studied. The paper presents the results of 2018-2019 research on improving the technology of growing planting material of bog cranberry (Vaccinium oxycoccos L.) of Krasa Severa, Severyanka, Virussaare varieties on the basis of in vitro. Studied was the effect of the concentrations of growth regulators in the composition of the nutrient medium according to Anderson's recipe on the reproduction and subsequent rooting of micro cuttings, as well as the duration of cultivation and adaptation of micro plants depending on partial pruning of shoots. It has been established that at the stage of introduction into in vitro culture, sterilization of explants with 33% hydrogen peroxide in an exposure of 5-8 minutes with washing in 5 portions of sterile distillate gives 60-80 % of viable shoots. The optimum phase of plant development for the successful introduction of in vitro culture is the swelling of buds. Cultivation of micro cuttings was carried out in a light room at a temperature of 25±2 °С, a photoperiod of 16 hours. The duration of each subculturing was 30-60 days. For the stage of actual micropropagation on Anderson's nutrient medium, an increase in the dose of cytokinin 6-benzylaminopurine (6-BAP) from 0.2 to 0.5 mg/l and an increase in the duration of cultivation from 30 to 60 days contributed to a significant increase in the multiplication factor on average for the tested cranberry varieties.According to the efficiency of micropropagation, the varieties Virussaare and Krasa Severa were distinguished – 9.3-12.0 pcs/stalk, respectively. At the rooting stage, the use of a root-forming reagent of indolyl-3-acetic acid (IUK) in doses of 0.2, 0.5 and 1.0 mg/l in the composition of Anderson's nutrient medium did not affect the quality of root formation and the length of shoots of Virussaare micro-plants. No significant varietal differences in the root-forming ability of microcuttings were found. The tendency of better rooting of micro cuttings was observed in the Virussaare variety (90.3 %) compared to the Severyanka (85.7 %) and Krasa Severa (79.3 %) varieties. Micro plants of the Krasa Severa cultivar were characterized by the longest shoots, the total number of roots was less, but their length was longer in comparison with other cultivars. For the adaptation stage, a substrate from a mixture of lowland peat and sphagnum moss was used (1:1). The efficiency of adaptation of micro plants of cranberry varieties when cutting the tip of the shoots was 100 %. Pruning of micro plants shoots contributed to the formation of more side shoots and better development of the aboveground part of the plants.

The growth and development of lingonberry (Vaccinium vitis-idaea L.) plants propagated either by conventional softwood cuttings or by in vitro shoot proliferation from nodal explants and by shoot regeneration from excised leaves of micropropagated shoots, were studied in cultivars `Regal', `Splendor', and `Erntedank'. Significant differences were observed between the treatments. After 3 years of growth, the in vitro-derived plants produced more stems, leaves, and rhizomes than the conventional cuttings which rarely produced rhizomes. In vitro culture on nutrient medium apparently induces the juvenile branching characteristics that favor rhizome production. This increase in vegetative growth and rhizome yield of in vitro-derived plants over stem cuttings varied among genotypes.

Artichoke (Cynara scolymus L.), a medicinal plant with high economic value, contains high levels of phenolic compounds; especially cynarine, which plays an important role in preventing cancer, cardiovascular disease, osteoporosis, diabetes and neurodegeneration, etc. Currently, Artichoke micropropagation has achieved some success; however, the rooting efficiency and plantlet quality are still limited. In this study, improving the quality of Artichoke plantlet related to the shoot quality and suitable substrates in in vitro rooting stage was studied on “Violetta” Artichoke (VA) and “Green Globe” Artichoke (GA). The results showed that shoots (1.5 cm) cultured on MS medium supplemented 0.5 mg/L KIN were most suitable to shoot multiplication of VA with the number of shoots/explant (3.67 shoots), number of shoots ≥ 2 cm (3 shoots); while, 1.0 mg/L BA was suitable to shoot multiplication of GA (5.33 shoots; 5.00 shoots; respectively) after 4 weeks of culture. Besides, the in vitro rooting was improved using 8 g/L commercial agar for VA; meanwwhile, 3 g/L gelrite for GA. In addition, the nylon bag culture system (120 mm × 250 mm) has potential in plantlet production (15 plants/bag) and can be applied for large scale micropropagation. In addition, VA and GA plantlets derived from in vitro culture gave the good acclimatization, growth and development after 8, 12 and 20 weeks cultivating at the green house conditions.

Regeneration competence of pseudobulb segments of Coelogyne flaccida, procured from in vitro grown cultures, was assessed in MS and Mitra (M) medium alone and in combinations with BA (4.44 ?M), Kn (5.65 ?M), NAA (5.37 ?M), and coconut water (CW 10%). Juvenility of the tissues, chemical stimulus, and position of the explants (apical/basal) were the key factors in initiating the response. The explants from pseudobulbs (>0.8 cm in length) did not regenerate whereas those from younger ones (<0.8 cm in length) regenerated. The response frequency and time taken for the development of plantlets also varied with the growth stimulus. Maximum number of shoots was obtained from the basal segments (nodal and extra?nodal regions), cultured in MS + NAA/NAA + BA forming 10.25 ± 0.50 and 10.00 shoots, respectively. The apical segments formed shoots only from apical meristem. Meristem tip explants of Malus robusta Rehd. No. 5 were successfully initiated in vitro on nutrient medium with sorbitol but not with glucose, sucrose, or fructose, each supplied at 30 g/L. Cultures, subsequently subcultured and multiplied on sorbitol medium, produced the highest number of shoots and had the highest shoot fresh and dry weights, followed in descending order by cultures grown on glucose, sucrose, and fructose media. Decreasing the proportion of sorbitol and increasing that of sucrose in the carbon source fraction (30 g/L total) resulted in decreasing shoot number and shoot weight accompanied by increasing leaf/stem ratio and compactness of the shoot cultures. After 32 days on agar medium with 0.5 mg/L indolebutyric acid in the presence of either sorbitol or sucrose, 85% rooting was achieved.

In the work, the study of six different nutrient medium compositions’ influence on the growth and development of a healthy Solnechny potato variety in the growing process in laboratory conditions in vitro was carried out. The influence of different nutrient medium compositions on plant height, number of leaves/internodes, plant weight and total leaf surface area was studied. It was revealed that nutrient media with BAP addition cause a decrease in all of the above indicators. In addition, nutrient media with this growth regulator lead to the appearance of microtubers on the plants. It has been shown that the inclusion of GA and IAA growth regulators in nutrient media leads to a slight increase in the number of internodes. An increase in the total leaf surface area was noted in the above options. It should be noted that the nutrient medium options with the addition of GA and IAA led to a decrease in the mass and length of the root system of Solnechnyi variety potato plants.

Family Orchidaceae has about 800 genera which are already difficult to find and almost extinct, including the orchid genus Grammatophyllum. A species of rare orchids, Grammatophyllum stapeliiflorum, is hard to find in original habitat. The addition of Growmore, BAP and 15% coconut water is expected to increase the growth and development of the G. stapeliiflorum orchid protocorms. This study use a Completely Randomized Design, consisting of 6 treatments (control, 1 and 3 mg/l BAP, 1 and 3 g/l Growmore, and 150 ml/l coconut water) in MS media with 5 replicates with observation for 4 weeks after planting. The results showed did not differ markedly in the number of protocorms, number of shoots and number of browning protocorms, but differ markedly in the parameter protocorm color based on DMRT test. Treatment of 3 mg/l BAP gives the best results on the number of shoots 2,60 protocorms and number of browning protocormss at the least amount 0,80 protocorms. The number of protocorms most widely on the treatment of 1 g/l Growmore 14,40 protocorms. Protocorms color is best found in 3 g/l Growmore with green color. This research managed to multiply the number of protocorms and induces protocorms shoots from Grammatophyllum stapeliiflorum orchid.

Intumescent leaf variants of sugar beet (Beta vulgaris L.) were obtained through callus culture of a monosomic addition that carried resistance to Heterodera schachtii Schm. The frothy pockmarked appearance of the leaf surface was due to hyperplastic growth of the mesophyll and epidermal cells. The epidermis had many malformed stomata. Veins were underdeveloped, but protrusions beneath were pronounced. Intumescence occurred in 20.3% of the regenerated plants and it was heritable to F1 and later progeny. Leaf intumescence is a new phenotype for Beta. About 73.5% of regenerants contained the donor somatic chromosome number, the remainder were doubled or mixoploids, with no chromosome losses apparent. The 38-chromosome intumescent plant represents a dual somaclonal variation, chromosome doubling and leaf intumescence. Progeny of the 19- and 38-chromosome intumescent plants intercrossed or pollinated by diploids or tetraploids had 9, 18, 19, 27, 28, 29, 36, 37, 38, or 39 chromosomes. All intumescent plants were aneuploids with the monosome addition. There were linkages for leaf intumescence (Li), resistance to H. schachtii (Hs), and hypocotyl color (Rpro) on the addition chromosome. The efficacy of Hs remained intact through the in vitro culture and succeeding crosses. The Li-bearing plants manifested depressed growth and markedly reduced seed set. Leaf intumescence was thought to be the alternative expression of galling potential of Beta procumbens Chr. Abstract In vitro culture of twin-scaling explants of Galanthus transcaucasicus with different concentrations of plant growth regulators (PGRs) including 0.5, 1, 2, 3, 4, 6, 8, and 10 mg L-1 naphthaleneacetic acid (NAA) and 0.5, 1, 2, 3, and 4 mg L-1 benzyladenine (BA) was studied. After 18 weeks, the number of regenerated bulblets and intensity of callus was measured. Subsequently, bulblets were transferred to a medium with 0.5, 1, 2, 3, and 4 mg L-1 NAA and 0.5, 1, 2, 3, and 4 mg L-1 BA and, after 15 weeks, the bulblets length and diameter were measured. The highest intensity of callus was obtained on 4 mg L-1 NAA or 8 mg L-1 NAA with 1 mg L-1 BA. The highest number of regenerated bulblets was detected with 6 mg L-1 NAA and 2 mg L-1 BA. The highest diameter of bulblets occurred on four mgL-1 NAA (9.4 mm), while the lowest was observed on 0.5 mg L-1 BA (1.83 mm). The analysis of genetic variation using ISSR revealed that there was no somaclonal variation among the regenerated plants from BA and low level of NAA, but there was a significant somaclonal variation at high concentrations of NAA.

This paper presents the results of the adjustment of 18 modifications of culture medium based on MS medium composition – Murashige and Skoog (1962) for in vitro rhizogenesis of 10 varieties of domestic plum. There were used 4 nutrient media with a full amount of macro- and microelements – MS1 with a different combination of phytohormones, 1 medium without hormones – MSc, 13 media – MS2 with half the amount of macronutrients, differing in hormonal composition. It was found that the maximum number of rooted microshoots from 21.2 to 52.2 % was on MS2–8 medium, from 11.2 to 43.1 % on MS2–5. The analysis showed that into the medium increases the percentage of microplants by 12-17 % comparing with the medium MS2–5 and MS2–7 without FA. The distinctiveness of nutrient media by the type of auxin (IAA, NAA, IBA) made it possible to clarify that IBA is the most optimal of the auxins, and the concentration of 1.7 mg/l is borderline for the growth reactions of plum varieties. It was revealed that in vitro root formation depends not only on the compositions of growth substances in the nutrient medium, but also on the genotype of the variety.
