ABSTRACT Escherichia coli cells contain potential division sites at midcell and adjacent to the cell poles. Selection of the correct division site at midcell is controlled by three proteins: MinC, MinD, and MinE. It has previously been shown (D. Raskin and P. de Boer, Cell 91:685–694, 1997) that MinE-Gfp localizes to the midcell site in an MinD-dependent manner. We use here Gfp-MinD to show that MinD associates with the membrane around the entire periphery of the cell in the absence of the other Min proteins and that MinE is capable of altering the membrane distribution pattern of Gfp-MinD. Studies with the isolated N-terminal and C-terminal MinE domains indicated different roles for the two MinE domains in the redistribution of membrane-associated MinD.

RNA-based stable isotope probing (RNA-SIP) is used in molecular microbial ecology to link the identity of microorganisms in a complex community with the assimilation of a distinct substrate. The technique is highly dependent on a reliable separation of isotopic-labeled RNA from unlabeled RNA by isopycnic density gradient ultracentrifugation. Here we show that 13C-labeled and unlabeled Escherichia coli RNA can be sufficiently separated by isopycnic ultracentrifugation even in the absence of formamide. However, a slightly lower starting density is needed to obtain a distribution pattern similar to that obtained when formamide was used. Hence, the commonly used addition of formamide to the centrifugation solution might not be needed to separate 13C-labeled RNA from unlabeled RNA, but this must be verified for more complex environmental mixtures of RNA. Clearly, an omission of formamide would increase the safety of RNA-SIP analyses.

Abstract A quantitative relationship between tumor cell number and biologic marker concentration has been investigated and characterized by the previously developed discrete-time kinetic model for the study of cell kinetics. Here, this model is further expanded to cope with both cell cycle kinetics and biologic marker synthesis. A synchronized tumor-cell population is examined to determine the time course of the synthesis of markers in relation to cell cycle. The DNA content distributions, measured by flow microfluorometry, are analyzed by use of the model, and the cell age distribution is extracted. The average marker content per cell in cell cycle is measured for each time sequence in synchronized cell populations. The model, incorporating the cell age distribution and the average marker content per cell in cell cycle, enables one to generate the marker content distribution, from which the cell number is estimated from the marker concentration. The model performance is further evaluated with Chinese hamster ovary cells, and their polyamine content and the total polyamine concentration during synchronization is calculated and related to the total cell number.

Asymptomatic bacteruria is the presence of significant count bacteria capable of inducing urinary tract infection in the urine tract of clinically healthy individuals. Four hundred samples of urine were investigated for the presence of Escherichia coli in this study, out of which forty by conventional biochemical tests. The percentage prevalence gender ratio of the isolates was 77.5% females to 22.5% males while the age distribution prevalent ranged between 24 years 30years. Ten conventional biochemical confirmatory tests was carried out on the isolates. The antibiogram in this study elicited the effective potency of floroquinolones while ampicillin, a β –lactams derivatives was recorded to be less effective. Sixteen isolates showed multidrug resistant to 4 antibiotics while 2 isolates elicited multidrug resistance to 6 antibiotics in this study. The findings of this study the prevalence and the potency of selected antibiotics could be useful in the therapeutic management of asymptomatic bacteruria in the study region.
