Alfalfa seeds of Cimarron VR, CW1446, CW2440, C94-118, C94-785, and WL311 were used as explants. BA, zeatin, and TDZ were evaluated on callus initiation, development, and shoot production. Callus initiation and development toward shoot organogenesis were enhanced when BA was added in the culture medium. Calli produced from BA treatments were compact, solid, and dark green. Similar results were obtained when zeatin was added in the culture medium. However, no shoots were produced from such calli. Multiple shoots were produced directly from each individual explant when TDZ was added to culture medium. However, when higher concentration of TDZ was used, number of shoots per explant was decreased, and dwarf shoots were produced. No apparent difference on shoot production was observed among the cultivars tested so far. Data on number of shoots per explant from two of these cultivars need to be statistically analyzed.

Morphogenesis of the terminal shoot was studied in 2-year-old seedlings of California red fir (Abiesmagnifica A. Murr.) and two elevational sources of white fir (Abiesconcolor (Gord. & Glend.) Lindl.). Seedlings were either watered or left unwatered during the growing season in order to produce different shoot morphologies and seedlings with and without a summer shoot. Under favorable soil moisture, the frequency of summer shoot production was 32, 53, and 82% for red fir and high- and low-elevation white fir, respectively. Drought from mid-May to mid-September reduced summer shoot production to less than 1% in both species. Spring shoot morphology was not an indicator of capacity to produce a summer shoot. Rate of primordium production was directly related to apical dome diameter. However, when the normal spring increase in apical dome diameter was arrested by summer shoot elongation, the rate of primordium production appeared to be unaffected. Although the apical and subapical meristems were active at the same time, they did not appear to be antagonistic. The major effects of producing a summer shoot were as follows: (i) elongation of 60–120% more intemodes in the current growing season, (ii) production of 15–40% more needle primordia in the overwintering bud, (iii) production of 30–60% more primordia annually, and (iv) increase in the percentage of total primordium production that developed into needles from 60% to 75–80%.

Papaya is a popular fruit and is grown commercially in many subtropical and tropical countries. Papayas are generally grown from seeds; therefore the offsprings are not true-to-type and could come in three sexes, female, male, and hermaphrodite. Clonal propagation is required to obtain to grow true-to-type hermaphrodite papayas. In this research, we developed an in vitro protocol for shoot multiplication from lateral shoots from in vitro germinated papaya seedlings. The in vitro propagated plant materials could potentially be used as a source of papaya micro cuttings, or as scion for papaya grafting. The experiment was set up as a factorial experiment with NAA at 0, 0.1 and 0.5 mg.L-1, and BAP at 0, 0.1, 0.5, and 1.0 mg.L-1 in a completely randomized block design. BAP interacted with NAA in affecting the shoot production per explant. The optimum BAP and NAA concentration to produce lateral shoots was 0.54 mg. L-1 and 0.1 mg.L-1, respectively. Media without NAA reduced the number of lateral shoots and number of leaf per explant at any BAP concentration.Keywords: hermaphrodite, seeds, true to type, clonal propagation, micro cuttings.

Cavendishia bracteata and Thibaudia floribunda (Ericaceae) are wild fruit shrubs native to the Andes region. This study aimed to characterize the rooting potential of basal and apical cuttings using naphthalene acetic acid (NAA) in different concentrations (0, 200, 500, or 1,000 mg L-1). In C. bracteata, the cutting position on the branch affected its sprouting and rooting, with apical cuttings presenting the best performance with auxin application. The rooting of cuttings was more successful in C. bracteata than in T. floribunda. NAA applications between 200 to 500 mg L-1 were more suitable for the propagation of C. bracteata due to the higher percentages of rooting and shoot production. For propagation of T. floribunda, the use of apical cuttings without the application of auxins is recommended. Adventitious roots in cuttings of both species were originated from parenchyma cells of the stem.

Plants maintain beneficial mutualistic relationships with the mycobiont communities found in their rhizosphere, leading to an increase in plant productivity and health. In nutrient-depleted substrates like mine tailings, mycobiont inoculation is often recommended to help restore a successful plant cover. Our 15-week greenhouse experiment aimed to assess the individual effects of a dark septate endophyte (Phialocephala fortinii #4; KX611529) and two Helotiales strains (Rhizoscyphus ericae #22; EU221877 and Meliniomyces sp #1; KT275679) on the growth of Salix planifolia cuttings on sterilized and unsterilized waste rock. Rhizoscyphus ericae increased cuttings shoot biomass on sterilized waste rock while Meliniomyces sp had a positive effect for cuttings grown on unsterilized waste rock. However, P. fortinii strain had no effect on the survival rate, shoot production, and biomass production of S. planifolia cuttings. This study demonstrates that controlled inoculation with ecologically well-adapted mycobionts could promote plant establishment and productivity on abandoned waste rock and be an efficient and integrated biotechnological approach for ecological restoration of canadian mining boreal ecosystems.

Micropropagation of 11 rare or endangered cacti species belonging to the subtribe Cactinae was achieved by rooting of proliferated axillary shoots. Shoot tip explants were obtained from seedlings of Escobaria missouriensis D.R. Hunt, E. robbinsorum (Earle) D.R. Hunt, Sclerocactus spinosior (Engelm.) Woodruff & L. Benson, and Toumeya papyracantha (Engelm.) Br. & Rose, and from mature plants of Mammillaria wrightii Engelm., Pediocactus bradyi L. Benson, P. despainii Welsh & Goodrich, P. knowltonii L. Benson, P. paradinei B.W. Benson, P. winkleri Heil, and S. mesae-verdae (Boissevain) L. Benson. Three or four species were used in each of a series of experiments investigating the effects of basal media and auxin and cytokinin types and concentrations on axillary shoot proliferation. Low or no auxin but moderate to high cytokinin concentrations were required for axillary shoot production. All species rooted spontaneously on hormone-free media; however, several species rooted better on media containing auxin. All species were re-established in the greenhouse.
