Newcastle disease virus {NDV (avian paramyxovirus type 1 [APMV1])} isolates were recovered from imported exotic birds confiscated following importation into the United States, from waterbirds in the United States, and from poultry. The exotic birds probably originated from Central and South America, Asia, and Africa. The NDV isolates were initially characterized as highly virulent because of a short mean death time in embryonated chicken eggs. The isolates were typed as neurotropic or viscerotropic velogenic by intracloacal inoculation of adult chickens. Intracerebral pathogenicity index values for the virulent NDV isolates ranged from 1.54 to 1.90, compared to a possible maximum value of 2.0. These isolates had a dibasic amino acid motif in the fusion protein cleavage site sequence required for host systemic replication. Sequence differences were detected surrounding the fusion protein cleavage site and the matrix protein nuclear localization signal, indicating evolution of highly virulent NDV. Phylogenetically, these isolates were categorized with other highly virulent NDV strains that caused outbreaks in southern California poultry during 1972 and in cormorants in the north central United States and southern Canada during 1990 and 1992. These isolates are related to NDV that may have the APMV1 strain chicken/Australia/AV/32 or a related virus as a possible progenitor. Recent virulent NDV isolates and those recovered during disease outbreaks since the 1970s are phylogenetically distinct from current vaccine viruses and standard challenge strains.

Poultry production in South Africa, a so-called developing country, may be seen as a gradient between two extremes with highly integrated commercial enterprises with world-class facilities on one hand and unimproved rural chickens kept by households and subsistence farmers on the other. Although vaccination against Newcastle disease is widely applied to control this devastating infection, epizootics continue to occur. Since the first official diagnosis in 1945, through the sporadic outbreaks of the 1950s and early 1960s, to serious epizootics caused by genotype VIII (late 1960s–2000), genotype VIIb (1993–1999), genotype VIId (2003–2012) and most recently genotype VIIh (2013 to present), South Africa’s encounters with exotic Newcastle disease follow global trends. Importation – probably illegal – of infected poultry, poultry products or exotic birds and illegal swill dumping are likely routes of entry. Once the commercial sector is affected, the disease spreads rapidly within the region via transportation routes. Each outbreak genotype persisted for about a decade and displaced its predecessor.

Free-range rural chickens (FRCs) dominate the poultry industry in developing countries and chickens are exposed to multi-host infections, including Newcastle disease virus (NDV). The knowledge about the characteristics of NDV from FRCs is limited. This study investigated the persistence, spread and risks of NDV from FRCs. NDV isolates (n = 21) from unvaccinated FRCs in Tanzania were characterised by conventional intracerebral pathogenicity index (ICPI) and sequence analysis of a partial region of the deduced fusion protein encompassing the cleavage site. Results showed that five isolates were screened as lentogenic, nine as mesogenic and six as velogenic. Phylogenetic analysis of the 21 isolates compared to reference sequences revealed three, four, nine and five isolates in genotypes 1, 2, 3c and 4a, respectively. Genotype 3c also included published sequences of Tanzanian isolates obtained from exotic birds and chicken isolates from Uganda. The analysis showed that NDV were persistently present among chicken populations and possibly spread through live chicken markets or migration of wild birds. Differences in amino acid sequences detected around the cleavage site separated the isolates in six types. However, cleavage site pattern could not fully differentiate mesogenic isolates from velogenic isolates.

. Using the method of shifting assay of standardized indirect agglutination (SSIA), the prevalence of Newcastle disease viruses (NDV) and infectious bursal disease viruses (IBDV) in chickens reared in several districts of Thua Thien Hue province in the Spring-Summer and Fall-Winter seasons was determined. In the Spring-Summer season of 2011, about 22.3% of the chickens were infected with NDV, in which A Luoi  accounted for the highest percentage of 25% of the infected chickens and Huong Thuy  the lowest  of 18.2%. Meanwhile, 36% of the same chickens were infected with IBDV, with the highest percentage (46.66%) also in A Luoi and the lowest (30.3%) also in Huong Thuy. The intensity of NDV infection in the Spring-Summer season in A Luoi and Phu Vang was highest (GMT = 1.45), and in Huong Thuy lowest (GMT = 1.31). In addition, in the Fall-Winter season, about 46% of the chickens were infected with NDV and 46.3% with IBDV in Huong Thuy and Phu Vang – two neighbouring districts of Hue City, in which NDV was detected in 54.4% of the chickens in Huong Thuy and 33.9% in Phu Vang. In contrast, IBDV was detected in 41.9% and 52.7% of the chickens respectively in the two districts. The infection was not inter-dependent. Methodically, although the differences in the infection rates were insignificant with the accuracy of 95%, faecal samples showed higher sensitivity in SSIA analyses for both cases of NDV and IBDV infection in comparision with mouth exudates. By SSIA method, results could be read clearly with unaided eyes for a long time after the performance, and it was also proven applicable for cases of haemagglutinating viruses if proper treatments for depletion of animal RBCs’ surface agglutinins could be applied.
