Background/Aim. During exposure to low-level doses (LLD) of ionizing radiation (IR), the most of harmful effects are produced indirectly, through radiolysis of water and formation of reactive oxygen species (ROS). The antioxidant enzymes - superoxide dismutase (SOD): manganese SOD (MnSOD) and copper-zinc SOD (CuZnSOD), as well as glutathione (GSH), are the most important intracellular antioxidants in the metabolism of ROS. Overproduction of ROS challenges antioxidant enzymes. The aim of this study was to examine if previous exposure to low doses of IR induces adaptive response by means of stimulation of intracellular antioxidant defense system. Methods. We investigated a group of medical workers occupationally exposed to IR (n = 44), 29 male and 15 female. The controls (n = 33) consisted of medical workers not exposed to IR, 23 male and 10 female. The examinees from both groups worked in the same environment and matched in crucial characteristics. All measurements were performed by a calibrated thermoluminiscent dosimeter type CaF2:Mn. SOD activity and GSH content were measured spectrophotometrically in the plasma of both groups of medical workers. Half of each blood sample was irradiated by 2Gy of ? radiation, dose-rate 0.45 Gy/min, and the distance from the source of 74 cm. Results. The dosimetry results indicate that occupational doses were very low. Our results confirmed significantly higher SOD activity in the exposed vs. unexposed workers (p < 0.00006). SOD activity after irradiation of blood samples failed to show a significant difference between the exposed workers and the controls (p = 0.905), even the difference in each group before and after the irradiation was significant. In blood samples of the exposed workers expression of enzymes after the irradiation, was not as high as in the controls, or even in the case of the exposed in nuclear medicine personnel, SOD activity was decreased. There were no significant differences in the content of GSH between the groups. Conclusion. Our results pointed out that occupational exposure to low doses of IR compromised mitochondrial function. During occupational exposure, the activity of antioxidant enzymes was increased as a protection against the increased production of ROS. After high-dose irradiation dysfunction of mitochondrial system was noticed, suggesting the break-down of antioxidant defense and failure of an adaptive response. Therefore, the "chronic oxidative stress" might reduce antioxidant defense in the case of accidental exposure to high doses of IR. It could indirectly increase the incidence of some other "free radicals' diseases" in occupationally exposed personnel. .

Aim: The aim of this work was to investigate the relationships between aluminium levels, oxidative status and DNA damage in workers occupationally exposed to aluminium. Subjects and methods: This study was conducted in a secondary aluminium smelter. It included 96 male workers occupationally exposed to aluminium fume and dust compared to 96 male nonexposed individuals. Full history and clinical examination were done for all participants. Laboratory investigations in the form of serum aluminium, total antioxidant capacity (TAC), urinary 8-hydroxy-2′-deoxyguanosine (8-OHdG) and comet assay test were performed. Results: Serum aluminium level ranged from 4 to 30 µg/L of median: 10 µg/L; urinary 8-OHdG ranged from 2.7 to 17.2 ng/mg creatinine of median: 7.6 ng/mg creatinine; comet tail length (CTL) ranged from 19.7 to 50.5 µm of median: 45 µm, were statistically significantly increased in the exposed group compared to nonexposed group. In exposed workers, a statistically significant positive correlations were found between serum aluminium level and urinary 8-OHdG ( r = 0.75, p < 0.001); aluminium level and CTL ( r = 0.71, p < 0.001); and urinary 8-OHdG and CTL ( r = 0.71, p < 0.001). There was a statistically significant negative correlation between serum aluminium and TAC ( r = −0.76, p < 0.001). Conclusion: Occupational exposure to aluminium in secondary aluminium smelters was related to the induction of oxidative stress and DNA damage. This may promote the development of adverse health hazards in the exposed workers

The lung activity of the antioxidant enzymes (AOEs) copper, zinc superoxide dismutase (Cu,Zn SOD), catalase (CAT), and glutathione peroxidase (GP), but not manganese superoxide dismutase (Mn SOD), increases in rats during late gestation; the concentrations of Cu,Zn SOD mRNA and CAT mRNA also rise. During early postnatal exposure to > 95% O2, the lung activity of Cu,Zn SOD, CAT, and GP increases. We now show 1) the lung concentration of Mn SOD mRNA and GP mRNA does not increase in late gestation; 2) Mn SOD activity and the concentration of its mRNA and of GP mRNA increase during exposure of neonatal rats to > 95% O2; and 3) as previously shown for CAT mRNA, the increase in lung concentration of the mRNAs for Cu,Zn SOD, Mn SOD, and GP during early postnatal hyperoxia occurs with a 70–80% prolongation of the half-life of these mRNAs. We conclude that 1) in late gestation the level at which lung AOE gene expression is regulated differs among the enzymes, 2) the level at which lung AOE gene expression is regulated shortly after birth in response to > 95% O2 is uniform among the enzymes, and 3) the lung's AOE response to neonatal hyperoxia is not merely a step-up of its prenatal regulation but involves different regulatory mechanisms based on increased stability of AOE mRNAs

Objective: To compare oxidative stress between primary retinoblastoma and retinoblastoma with distant metastasis. Patients and methods: Forty consecutive patients presented with primary retinoblastoma and the same number of patients presented with distant metastasis, attending the outpatient department of our hospital between August 2002 and April 2005. All the patients with retinoblastoma underwent a standard metastasis workup and were subsequently categorized into two groups (without metastasis and with metastasis).Venous blood samples were drawn from each patient. After proper centrifugation, serum was collected and antioxidant enzymes and reactive oxygen species (ROS) were assayed. Main outcome measures: Serum collected from the patients was subjected to biochemical assay of the antioxidant enzymes (superoxide dismutase, catalase and peroxidise) and ROS to determine any difference in enzyme activity between the two groups. Results: Antioxidant levels were found to be less in the metastasis group as compared to the primary intraocular retinoblastoma group(p<0.05).Mean ROS activity was found to be increased in metastatic group (p<0.05). Conclusion: The decreased antioxidant enzymes level along with increased ROS activity in patients with metastatic retinoblastoma reflect increased oxidative stress as compared to primary intraocular retinoblastoma patients.DOI: http://dx.doi.org/10.3126/nepjoph.v4i2.6543 Nepal J Ophthalmol 2012; 4 (2): 271-276

AbstractThis study examined the effects of chronic restraint stress (CRS: 2 hours × 14 days) on gene expression of three antioxidant enzymes, copper, zinc superoxide dismutase (SOD 1), manganese superoxide dismutase (SOD 2) and catalase (CAT) in the rat hippocampus. Also, we examined changes in the activities of SOD 1, SOD 2 and CAT in the hippocampus of chronically stressed rats. Investigated parameters were quantifi ed by using real-time RT-PCR, Western blot analysis and assay of enzymatic activity. We found that CRS did not change mRNA and protein levels of SOD 1 and CAT, but increased mRNA and protein levels of SOD 2. However, CRS treatment increased the enzyme activities of SOD 1, SOD 2 and CAT. Our fi ndings indicate that the increased activity of antioxidant enzymes (SOD 1, SOD 2 and CAT) in the hippocampus may be an important adaptive phenomenon of the antioxidant defense system in chronically stressed rats.

Purpose. The aim of the work was to study certain morpho-physiological parameters of carp as a result of additional introduction of a mycotoxin sorbent to a feed with signs of spoilage. Methodology. During 2019–2020, two experiments were conducted in the laboratory. The first one included 7 groups-analogues of age-1+ Nyvky carp, 20 fish in each group, with an average weight of 22.5 g. The control group of fish was fed for 24 days with a compound feed with signs of spoilage, «Mykosorb» was additionally added to the diet of the experimental groups in the amounts of 0.05, 0.1; 0.15; 0.2; 0.25 and 0.4%, respectively. The second group included 4 groups of age-1+ fish, 12 fish in each group, with an average weight of 20.0 g. The diet of the control group was identical to that of the first experiment, and the experimental groups with the addition of 0.05, 0.075 and 0.1% of «Mycosorb», respectively. During the study, optimal conditions for keeping fish were provided. Fish after were measured and weighed at the end of the experiment and their weight gains were analyzed according to generally accepted methods in fish farming. The activity of antioxidant enzymes and the content of lipid peroxidation products depending on the composition of the diet were studied. Findings. The studies used compound feed with signs of spoilage as a result of violation of their storage technology, which identified three groups of toxins: Aflatoxin B1, Zearalenone and Deoxynivalenol. As a result of the first stage of the study, the rates of application of mycotoxin sorbent into the diet of carp at amounts of 0.05 - 0.2% were determined. At the same time, there was no clear proportional pattern of changes in the average weight of fish depending on the amount of introduction of «Mycosorb», the value increased in all experimental groups compared to the control by 0.4 – 15.2%. Muscles of age-1+ fish were found to have higher (p<0,001) catalase activity after application of 0.15% of the test drug by 1.4 times, 0.2% — by 46%, 0.25% — by 69 and 0.4% — by 79%. Superoxide dismutase activity also increased, except for the groups of 0.2, 0.25 and 0.4% of «Mycosorb». There was a tendency of 21.0 and 22.6% decrease in the content of diene conjugates as a result of feeding 0.05 and 0.1% sorbent and some increase as a result of feeding 0.15; 0.2; 0.25 and 0.4% additives. Studies of the next stage showed that addition of «Mykosorb» at the rate of 0.05, 0.075 and 0.1% to the feed, the average weight of age-1+ carp in the experimental groups increased by 45.4; 37.2 and 30.3%, the condition factor — by 41.9; 40.8 and 42.7%, the relative growth rate is 4.3; 5.9 and 4.6 times, respectively. The fish body height to length ratio was lower by 10.3, 7.0 and 7.4% relative to the control group. Addition of 0.05, 0.075 and 0.1% of «Mycosorb» increased resistance of fish due to an increase in the hepatopancreatic activity of antioxidant enzymes: superoxide dismutase by 28.3, 30.5 and 5.0% and catalase by 56.5 (p<0.001), 55.5 (p<0.001) and 46.3% (p<0.05). At the same time, addition of 0.05 and 0.1% of «Mykosorb» increased the content of diene conjugates by 11.3 (p<0.01) and 12.3% (p<0.01), TBC-active products by 19.7 (p<0.05) and 13.2%. Originality. For the first time, the feasibility of addition of the mycotoxin sorbent «Mykosorb» into the feed with signs of spoilage was studied. The effect of feeding age-1+ carp with diets of various compositions on some morpho-physiological parameters of carp in the conditions of model tests was carried out. Practical value. Additional weigh gains and improvements of the antioxidant defense system of the carp organism were observed. The use of «Mykosorb» in carp feeding allows reducing the negative impact of low quality feed on fish body. This study was undertaken to investigate the effects of the extracts of buckwheat leaf and flower on the antioxidant status of the brain and liver tissue. The administration of buckwheat extracts (both concentrations were 10%) to mice (at the dose 10 mL/kg of body weight) for 21 days significantly decreased superoxide dismutase (SOD) activity and reduced the amount of glutathione (GSH) and malondialdehyde (MDA) in the mouse brain, while catalase (CAT) activity significantly increased. In the mouse liver, the amount of GSH and activity of SOD increased, while the CAT activity after administering buckwheat leaf and flower extracts was lower in experimental mice than in the control group. However, the administration of 10% ethanol (for 21 days) to control animals also had a significant effect on the antioxidant system in brain and liver cells. Experimental animals demonstrated rather marked changes in the activities of the antioxidant enzymes SOD and CAT in their liver and brain cells, and changes in the levels of GSH and MDA were observed when compared with the control group.

ABSTRACT: This study evaluated the effects of injectable trace minerals (ITM) on antioxidant and immune response, resistance to endoparasites, health and growth of newborn Boer kids. Forty-six Boer kids [24 males and 22 females; 3.94±1.03kg of body weight (BW); 6.2±2.4 d of age] were enrolled in the study. Kids were stratified by type of birth (twins or singlet), sex, and BW and assigned to 1 of 2 treatments: one subcutaneous injection (0.1mL/4.5kg of BW) of (1) saline solution or (2) ITM (60, 10, 5, and 15mg/mL of Zn, Mn, Se and Cu, respectively). Blood samples were collected on d 0, 7, 14, 28 and 56. Feces samples were collected on d 56 and BW on d 0, 28 and 56. Kids were checked daily for signs of diarrhea. ITM kids had greater (P<0.01) plasma concentration of superoxide dismutase and tended (P=0.06) to have greater plasma concentration of glutathione peroxidase. ITM kids had greater (P=0.05) concentration of eosinophils, but no differences (P≥0.11) were observed for other hemogram variables. The ITM application did not affect (P≥0.11) the EPG count. However, ITM kids had less (P=0.02) cumulative incidence of diarhea until d 42 (3.85 vs. 25.93±6.8% for ITM vs. Saline kids, respectively) but no differences (P>0.10) were observed after d 42. The ITM application did not affect (P≥0.40) the growth of kids (0.071 vs. 0.065±0.005kg/day for ITM vs. Saline kids, respectively). Thus, the ITM application, increased the plasma concentration of antioxidant enzymes and eosinophils, decreased the incidence of diarrhea only in the middle of the experiment, but did not affected the EPG count and growth of Boer kids.

Many studies have documented the abnormal concentrations of metals/metalloids in serum or urine of occupational workers, but no works systematically analysed the concentrations of elements in serum or urine of indium-exposed workers. This study was aimed to assess 28 elements in serum and urine from 57 individuals with occupational exposure to indium and its compounds. Control subjects were 63 workers without metal exposure. We collected information on occupation and lifestyle habits by questionnaire. Biological samples were collected to quantify elements by inductive coupled plasma-mass spectrometer. Air in the breathing zones was drawn at flow rates of 1.5–3 L/min for a sampling period of 6 to 8 h, using a Model BFC-35 pump. The average ambient indium level was 0.078 mg/m3. Serum/urine Indium levels were significantly higher in indium-exposed workers than in controls (P < 0.01). Moreover, serum/urine indium concentrations in the group with 6–14 years and ≥15 years of employment were significantly higher than those with ≤5 employment years(P < 0.05). Ten of the other 27 elements/metals measured were higher in serum/urine in indium-exposed workers compared to the controls (aluminum, beryllium, cadmium, cesium, chromium, lithium, manganese, magnesium, molybdenum and vanadium). Zinc levels in serum/urine were significantly decreased in the indium-exposed workers. Additionally, other elements/metals were higher in one specimen (serum or urine) but lower in the other (Selenium was lower in serum but higher in urine in the indium-exposed workers compared with the controls; likewise Thallium and Rubidium were higher in serum but lower in urine). Linear regression analyses, revealed significant correlations between serum and urine for indium, aluminum, arsenic, barium, cadmium, cesium, cobalt, selenium, silver, and zinc (P < 0.05). These data suggest that occupational exposure to indium and its compounds may disturb the homeostasis of trace elements in systemic circulation, indium concentrations in serum or urine appear reflective of workers’ exposure to ambient indium and their years of working, respectively. The serum/urine levels of essential metals are modified by exposure to indium in occupationally exposed workers. Further studies including larger sample size and more kinds of biological sample are needed to validate our findings.
