ObjectivesThis study was conducted to evaluate the dehydration methods of goat meat based in Malawi and the effects on food safety.Materials and MethodsGoat meat was prepared as ground, minced, and whole muscle strips. Samples were treated with 6% lemon juice marinade, 6% vinegar marinade, or salt rub. During phase 1, dehydration of the meat was performed with a solar dehydrator (n = 108), electric oven (n = 108) or drum oven (n = 108). Qualitative data on the three drying methods was collected from a panel of students from Mzuzu University, Malawi, on the practicality of each method in a local rural setting. Additionally, visual observations were conducted 30 d prior to drying for the presence of mold and insects to give an indication of shelf life. Phase 2 was performed at Texas Tech University in Lubbock, Texas where whole muscle strips of lamb were submerged in a five-strain Escherichia coli surrogate cocktail of Escherichia coli for 5 min, allowed 30 min for cell attachment, then dried using an electric and drum oven, replicating the dehydration procedure in Malawi. For each replicate (n = 2), attachment samples (n = 10), samples dried in the electric oven (n = 10) and samples dried in drum oven samples (n = 10) were aseptically plated on MacConkey agar with a TSA overlay and enumerated for E. coli.ResultsIn phase 1, mold growth was observed on 15.7% (34/216) of samples dried in the solar dehydrator and drum oven. Of those positive for mold, 32.4% (n = 11) were minced, and 67.6% (n = 23) were whole muscle strips. No samples dried using the electric oven displayed mold (0/108). No samples displayed insects. Based on qualitative data that was gathered, top reasons to dry goat meat using the drum oven include “not requiring electricity” and “drum ovens are a common piece of equipment in villages”. Top reasons against using a drum oven include “unequal distribution of heat” and “high level of oversight required during drying”. Top reasons to dry goat meat using electric oven include “fast drying time”, “uniform distribution of heat”, and “limited oversight required”. Top reasons against using electric oven to dry goat meat include “requiring electricity” and “low knowledge of electric oven operation in a community setting”. Top reasons to use the solar dehydrator to dry goat meat include “not requiring electricity or firewood” and “limited oversight required”. Top reasons against using the solar dehydrator to dry goat meat include “slow drying time” and “uneven heat distribution due to time of day and shadows”. In phase 2, a 5-log reduction was observed for all electric oven treatment replicates (100%, 2/2) and half drum oven (50%, 1/2) replicates. However, variation in the reduction of E.coli is a direct result of weather and fuel provided to the drum oven.ConclusionElectric drying oven displayed the most consistent results for shelf life and safety. However, in rural Malawi, dehydrating methods should be chosen on a case by case basis.

Background Online information resources and support have been demonstrated to positively influence the well-being of people diagnosed with cancer. This has been explored in past literature for more common cancers; however, for rare cancers, such as neuroendocrine tumors (NETs), there are little to no support or resources available. Despite relatively good prognoses, the quality of life (QoL) of patients with NETs is significantly lower compared with samples of mixed cancer patients and the general population. Patients with NETs also typically report unclear and difficult pathways of disease management and treatment, given the heterogeneity of the diagnosis. There is a vital need to improve the availability of disease-specific information for this patient group and provide supportive care that is tailored to the unique needs of the NET patient population. Objective This study described the protocol of a study aimed to better understand the outcomes and experiences of patients diagnosed with NETs and to develop and pilot test a nurse-led online and phone-based intervention that will provide tailored supportive care targeted to NET subgroups (functioning vs nonfunctioning). Methods This is a multisite cohort with 3 phases, incorporating both quantitative and qualitative data collection. Phase 1 is a mixed methods prospective cohort study of NET patients identifying differences in patient experiences and priority of needs between NET subgroups. Phase 2 utilizes results from phase 1 to develop an online and nurse-led phone-based intervention. Phase 3 is to pilot test and evaluate the intervention’s acceptability, appropriateness, and feasibility. Results Currently, the project is progressing through phase 1 and has completed recruitment. A total of 138 participants have been recruited to the study. To date, patient-reported outcome data from 123 participants at baseline and 87 participants at 6-month follow-up have been collected. Of these, qualitative data from semistructured interviews from 35 participants have also been obtained. Phase 2 and phase 3 of the project are yet to be completed. Conclusions Limited research for patients with NETs suggests that QoL and patient experiences are significantly impaired compared with the general population. Furthermore, past research has failed to delineate how the clinical variability between those with functioning and nonfunctioning NETs impacts patient supportive care needs. This study will improve on the availability of disease-specific information as well as informing the design of a nurse-led online and phone-based supportive care intervention tailored for the unique needs of the NET patient population. The aim of this study is to determine the efficiency of Forsythia suspense extract (FSE) as an antibiotics substitute on performance, nutrient digestibility, serum antioxidant capacity, fecal Escherichia coli concentration and intestinal morphology of weaned piglets. A total of 108 Duroc × (Landrace × Yorkshire) weaned piglets (28 days (d) weaned, average body weight of 8.68 ± 1.36 kg) were randomly assigned into three dietary treatments, six pens per treatment, three barrows and three gilts per pen. The treatments contained a corn-soybean meal basal diet (CTR), an antibiotic diet (basal diet + 75 mg/kg chlortetracycline; CTC), and an FSE diet (basal diet + 200 mg/kg FSE; FSE). The experiment included phase 1 (d 1 to 14), phase 2 (d 15 to 28) and phase 3 (d 29 to 35). Compared with CTR, piglets fed FSE show improved (p < 0.05) average daily gain (ADG) and average daily feed intake in phase 2, as well as enhanced (p < 0.05) ADG from day 15 to 35 and day 1 to 28. Piglets supplemented with CTC and FSE showed a reduced (p < 0.05) diarrhea rate in phase 1, while piglets fed FSE showed enhanced (p < 0.05) apparent total tract digestibility (ATTD) of dry matter, organic matter, crude protein and gross energy, as well as lower (p < 0.05) nitrogen output in phase 2 compared with CTR and CTC. The content in the form of Colony-Forming Units (CFUs) of fecal E. coli on day 14 and 28 was lower (p < 0.05) in piglets fed FSE in comparison with CTR. The contents of total antioxidant capacity, superoxide dismutase and catalase in serum are enhanced (p < 0.05) compared with CTR and CTC, whereas the concentration of malondialdehyde in serum was decreased (p < 0.05) for piglets fed FSE on day 28 compared with CTC. The villus height to crypt depth ratio in ileum was numerically higher (p < 0.05) in piglets fed FSE in comparison with CTR. In conclusion, dietary FSE supplementation could substitute CTC in improving antioxidant capacity, nutrients digestibility and reducing fecal E. coli content, so as to reduce nitrogen output and diarrhea rate, and eventually improve performance in weaned piglets.

This chapter investigates how individuals interpreted and considered the audio-diary technique, understanding the interaction between the subject and the medium and the potential of new technological tools (e.g., smartphone, social network) in producing data. The research is based on a previous study conducted during the COVID-19 lockdown in Italy, more specifically, the transition from phase 1 to phase 2. Each participant—11 female and 6 male, between 28 and 45 years old, and living in the northern part of Italy—was asked to register one audio per day for a week (7-13 May). After this period, the author undertook a final follow-up semi-structured interview to evaluate how much the audio-diary had an impact both on people's daily lives and on their way of expressing information. The data collected suggest a number of advantages and disadvantages to the use of audio-diary to collect individuals' experience. The author will briefly describe the steps of AD technique by using the collected material (interviews) and what has emerged from the analysis of qualitative data.

This study aimed to investigate the responses of university students (late adolescents) about their conceptualization of a child, exploring the characteristics they associate with being a child. The study was conducted in two phases. In phase 1, responses to one open-ended question, what is a child? (N=75), were analyzed using qualitative content analysis. In phase 2, students (N=90) filled in an online closed-ended survey that was derived from the subthemes that emerged from the qualitative data collected in phase 1. Findings revealed multiple interconnected aspects of the conceptualization of the child, making it a complex whole. This study is helpful for understanding the concept of the child grounded in various theoretical and mythological categories that portray the complexities of existing dichotomies that often come up as interconnected in traditional societies.

Samples (10 by 20 by 2.5 cm) of beef carcass tissue were inoculated (104 to 105 CFU/cm2) with Escherichia coli O157: H7 that was either non–acid habituated (prepared by incubating at 15°C for 48 h in inoculated filter-sterilized composite [1:1] of hot and cold water meat decontamination runoff fluids, pH 6.05) or acid habituated (prepared in inoculated water fluids mixed with filter-sterilized 2% lactic acid [LA] runoff fluids in a proportion of 1/99 [vol/vol], pH 4.12). The inoculated surfaces were exposed to conditions simulating carcass chilling (−3°C for 10 h followed by 38 h at 1°C). Treatments applied to samples (between 0 and 10 h) during chilling included the following: (i) no spraying (NT) or spraying (for 30 s every 30 min) with (ii) water, (iii) cetylpyridinium chloride (CPC; 0.1 or 0.5%), (iv) ammonium hydroxide (AH; 0.05%), (v) lactic acid (LA; 2%), (vi) acidified sodium chlorite (ASC; 0.12%), (vii) peroxyacetic acid (PAA; 0.02%), (viii) sodium hydroxide (SH; 0.01%), or (ix) sodium hypochlorite (SC; 0.005%) solutions of 4°C. Samples were taken at 0, 10, 24, 36, and 48 h of the chilling process to determine changes in E. coli O157:H7 populations. Phase 1 tested water, SH, PAA, LA, and 0.5% CPC on meat inoculated with non–acid-habituated pathogen populations, whereas phase 2 tested water, SC, AH, ASC, LA, and 0.1% CPC on meat inoculated with acid- and non–acid-habituated populations. Reductions in non–acid-habituated E. coli O157:H7 populations from phase 1 increased in the order NT = water = SH &lt; PAA &lt; LA &lt; CPC. Reductions from phase 2 for acid-habituated cells increased in the order NT = water = SC &lt; ASC = LA = AH &lt; CPC, whereas on non–acid-habituated cells the order observed was NT = water = SC &lt; AH = ASC &lt; LA &lt; CPC. Previous acid habituation of E. coli O157:H7 inocula rendered the cells more resistant to the effects of spray chilling, especially with acid; however, the trend of reduction remained spray chilling with water = non–spray chilling &lt; spray chilling with chemical solutions.

ABSTRACT Our objectives were to evaluate the prevalence of Escherichia coli O157:H7 in cattle fed diets supplemented with 20 or 40% dried distillers' grains (DG) (DDG) or wet DG (WDG) and assess whether removing DG from diets before slaughter affected fecal shedding of E. coli O157:H7. Eight hundred forty steers were allocated to 70 pens (12 steers/pen). Treatments were no DG (control), 20% DDG or WDG, and 40% DDG or WDG, and each was replicated in 14 pens. In phase 1, eight floor fecal samples were collected from each pen every 2 weeks for 12 weeks for isolation of E. coli O157:H7 and detection of high shedders. In phase 2, half of the pens with DG were transitioned to the no-DG control diet, and pen floor fecal samples were collected weekly from all pens for 4 weeks. During phase 1, prevalence of E. coli O157:H7 was 20.8% and 3.2% for high shedders. The form of DG had no significant effect on fecal E. coli O157:H7 shedding. The prevalence levels of E. coli O157:H7 and the numbers of high shedders were not different between diets with 0 or 20% DG; however, cattle fed 40% DG had a higher prevalence and more high shedders than cattle fed 0 or 20% DG (P ≤ 0.05). During phase 2, overall and high-shedder prevalence estimates were 3.3% and <0.1%, respectively, and there were no differences between those for different DG forms and inclusion levels or when DG was removed from diets. The form of DG had no impact on E. coli O157:H7; however, fecal shedding was associated with the DG inclusion level.

ObjectivesIn the U.S. water immersion chilling (WC) is commonly used to chill poultry, while the E.U. utilizes air chilling (AC). With demand for poultry continuing to rise, poultry products with longer shelf life and less food waste will be needed. Meanwhile, widespread efforts to reduce natural resource and energy expenditures, such as water, as a means of enhancing sustainability, exist across the meat industry, including the poultry industry. Therefore, the objective of this study was to compare the impact of WC and AC on the shelf life and meat quality of bone-in and boneless chicken breast.Materials and MethodsA total of 256 eviscerated non-chilled chicken carcasses were obtained from a commercial processing facility in California and transported to the UC Davis meat laboratory within 2 h. Carcasses were randomly and evenly assigned to either water immersion chilling (WC) or air chilling (AC) and then were evenly assigned to be fabricated into bone-in (BI) or boneless (BL) breast. The breast samples were subsequently packaged onto polystyrene trays, overwrapped, and placed into cardboard boxes for dark storage at 4°C for either 7d (phase 1) or 14d (phase 2). Then breast samples were placed into a retail display case maintained at 4°C for 3d. Instrumental color measurement was performed every 12 h during retail display. Microbial analysis was conducted for samples collected on arrival, post chilling, post-fabrication, after dark storage at 4°C for 7d or 14d and after 3d retail display (n = 10 per sampling point per treatment). A panel of 8 untrained participants were asked to evaluate the color and their willingness to purchase (for example color: desirable, acceptable, unacceptable). Analysis of variance was conducted to evaluate the effect of chilling method and storage time on all dependent variables using Proc Mixed in SAS (version 9.4).ResultsThe WC chicken possessed lower psychrotrophic bacterial counts (1.05 log CFU/g) pre-fabrication than the AC chicken (2.12 log CFU/g), indicating that WC may remove a portion of the psychrotrophic bacteria. However, no difference in mesophilic bacterial counts was observed between the two treatments for pre-fabrication samples. The WC chicken and AC chicken, regardless of fabrication type, reached the end of shelf life (7 log CFU/g) at the 14d. The BL samples, regardless of chilling method, had lower total microbial counts throughout storage and display than the BI samples, since the removal of the skin physically removed the general microbial population as well. In terms of objective color, the a* and b* values were higher for AC breast, suggesting that AC breast was more red and yellow than WC breast through the display time. Chilling method did not have an impact on subjective color measurement. During phase 1, untrained panelist considered the color of BL chicken breasts more desirable than the BI breasts. During phase 2, regardless of chilling method or fabrication type, the desirability of color by untrained panelist decreased as display time increased.ConclusionThe results indicate that chilling method had a minimal impact on the shelf life in terms of the microbial counts. Although AC chicken breast tend to be more yellow based on objective color measurement, consumers did not detect a distinct color difference of chicken treated with air chilling or water chilling.

Abstract A total of 360 barrows (DNA 200×400; initially 5.6 kg) were used to evaluate yeast-based probiotics (Phileo by Lesaffre, Milwaukee, WI) in diets with or without pharmacological levels of Zn on growth and fecal Escherichia coli antimicrobial resistance (AMR). There were 5 pigs/pen and 18 pens/treatment. Dietary treatments were arranged in a 2×2 factorial with main effects of yeast pre- and probiotics (0 vs. 0.10% Actisaf Sc 47 HR+, 0.05% SafMannan, and 0.05% Nucleosaf in phase 1 then concentrations were lowered by 50% in phase 2) and pharmacological levels of Zn (110 vs. 3,000 mg/kg in phase 1 and 2,000 mg/kg in phase 2 provided by zinc oxide). Treatments were fed in two phases from d 0 to 7 and 7 to 21 with a common diet fed from d 21 to 42 post-weaning. There were no probiotics×Zn interactions. From d 0 to 21, pigs fed pharmacological Zn had increased (P &lt; 0.001) ADG and ADFI; however, there were no effects of added pre- and probiotics. Fecal samples were collected on d 4, 21, and 42 from the same three pigs/pen for AMR profiles and fecal dry matter (DM). On d 4, pigs fed pharmacological Zn had greater fecal DM (P = 0.043); however, no differences were observed on d 21 or 42. E. coli was isolated from fecal samples and species confirmation was accomplished by PCR detection of uidA and clpB genes. Microbroth dilution method using SensititreTM CMV3AGNF panel was used to determine antimicrobial susceptibilities of E. coli isolates to 14 different antimicrobials. There was no evidence for differences in AMR of fecal E. coli isolates to antibiotics by added pre- and probiotics or Zn. Results suggest that pharmacological levels of Zn stimulate intake and growth and improve fecal consistency in the nursery with no statistical response from added pre- and probiotics.
