The supernatant of Manihot esculenta Crantz rhizome has been used as an alternative remedy of infectious and cancer in Jogjakarta. Cytotoxicity assay showed that  Manihot esculenta Crantz supernatant had cytotoxic effect to cancer cell line, namely Myeloma (LC50 = 180,24 μg/mL) and HeLa (LC50 = 415,55 μg/mL), but have a little cytotoxic effect to SiHa. This research was aimed to identify cytotoxic activity of Manihot esculenta Crantz's supernatant to normal cell, particularly to human mononuclear cell and Vero cell line, and to compare to the cancer cell lines. The result showed that supernatant of M. esculenta had cytotoxic effect to normal mononuclear cell (LC50 = 564,00 μg/mL) and Vero cell line (LC50 = 686,00 μg/mL). The supernatant of M. esculenta had the highest cytotoxic activity to myeloma and relatively toxic to cervix cancer HeLa and normal cell, but less to SiHa.   Keywords: Manihot esculenta Crantz, Ribosome-inactivating Protein (RIP), cytotoxic, normal cell, cancer cell

Background: Hydrazones, one of the important classes of organic molecules, are pharmaceutical agents comprising –CO-NH-N=CH- group in the structure therefore and exhibiting significant biological activity. Methods: 5-Chloro-N’-[(substituted)methylidene] pyrazine-2-carbohydrazide (3a-g) and their Pd(II) complexes (4a-h) were synthesized and investigated in vitro anticancer activity on A549, Caco2 cancer and normal 3T3 fibroblast cell lines, using the MTT assay. Results: Anticancer activity screening results revealed that some compounds showed remarkable cytotoxic effect. Among them, 5-chloro-N'-[(4-hydroxyphenyl)methylidene] pyrazine-2-carbohydrazide (3c) displayed higher cytotoxic activity against A549 cancer cell line than the reference drug cisplatin. Conclusion: Compound 3c showed high cytotoxic activity against A549 cancer cell line but it showed low cytotoxic effect against normal 3T3 fibroblast cell line. Antiproliferative and antimetastatic effects of 3c were determined by the real-time monitoring of cell proliferative system (RTCA DP). The cell proliferation, metastatic and invasive activities of A549 cells were decreased due to increased concentration of 3c.

Objective: The objective of the study was to study the antioxidants and cytotoxic activities of phenolic extracts of some plants against breast cancer and normal cell lines.Methods: Phenolics were extracted from different parts of some plants (15) such as seeds, fruits, leaves, and rhizomes using methanol: ethanol: HCl: Distilled water at a ratio 50:29:1:20 (v:v:v:v). The contents of phenolics and flavonoids were estimated using gallic acid and quercetin as standards, respectively. The antioxidant activity was determined by 2,2ˋ-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging assay. Phenolic flavonoids were partially purified by adsorption chromatography using a silica gel column from selected plants and assayed their cytotoxic activity against breast cancer cell line Michigan Cancer Foundation-7 (MCF-7) and a normal cell line of non-tumorigenic fetal hepatic cell line (WRL-68).Results: The Cinnamomum zeylanicum extract had the highest phenolic and flavonoid contents were followed by Lawsonia inermis, Citrullus colocynthis, Syzygium aromaticum, Peganum nigrum, and Phoenix dactylifera. The antioxidant activity of Curcuma longa, P. dactylifera, C. colocynthis, Solanum melongena, and C. zeylanicum extracts had the highest ability to scavenge the free radicals. The acetone fraction of P. dactylifera and C. colocynthis extracts had the minimum inhibition dose that kills 50% of cells inhibitory concentration 50 values 156.91 μg and 1055.06 μg against MCF-7 and 372.86 μg and 153.8 μg against WRL-68, respectively. While the S. melongena extract had less effect on both cell lines.Conclusions: Phenolics as antioxidant substances had moderate or variable effectiveness on normal and cancer cell lines, and the highest concentrations were cancerous poison impact may be on normal cells over than cancer cells.

Investigations of antibacterial effect of supercritical extraction of thyme (TNKE) and thyme extract obtained by hydrodistillation (THD) covered 18 bacteria strains from the genuses Staphylococcus, Streptococcus and Enterococcus, including the strains MRSA, the strains VRE, as well as the reference strains S.pyogenes ATCC 19615, S. agalactiae ATCC 27959, S. aureus ATCC 11632, and S.aureus ATCC 25923. The anibacterial effect of thyme extracts was examined using the microdillution method in bouillon, and the cytotoxic effect of thyme extract was examined on the VERO cell line. The obtained values for MIC THD extract ranged from 160?g/mL to 1280?g/mL for all bacteria strains covered by the investigations. The obtained values for MIC TNKE extract were 1280 ?g/mL to 2560 ?g/mL for all examined strains except for 1 strain of S. intermedius of 320 ?g/mL. The examined extracts revealed a cytotoxic effect on the VERO cell line in concentrations higher than 40 ?g/mL.

About (20) Pseudomonas rogenosa isolate were experienced for their ability of protease production by calculating the diameter of lysis area after developing on skim milk agar medium (qualitatively ) and the results exhibited that only isolate no (5), was higher isolate for protease making of (26mm) of lysis area. Then, the protein concentration also identified by Bradford method and it was found of 0.16 mg/ ml , then purification was done by using an ion- exchange chromatography with DEAE sephadex G- 100 column and the results showed the presence of 1 peak of enzyme with 50 Kd of molecular weight 2 peaks of other proteins . we tried to investigate the invitro Cytotoxic effect of purified enzyme against two human cancer lines, HeP2 (Human larynx epidermed carcinoma ) , RD ( Rabdo- Sarcoma ) , and one normal cell line Ref ( Rat embryonic fibroblast ) . The cancer and normal cells were treated with different concentrations of protease enzyme ranging from ( 0.05, 0.1, 0.2, 0.4,0.8and 0.16 mg/ml) then incubated for additional 48h at 37C0 and the results showed highest toxicity ( 80.28%) of protease enzyme on RD , moderate cytotoxicity (45.52%) on Hep andslight toxicity ( 37.12% ) on normal cell line (Ref) in a concentration (0.8mg/ml).
