The aim of this study was to determine the effect of storage temperature on the antioxidant activity of Sargassum hystrix. Samples were collected from Sepanjang Beach Gunung Kidul, Yogyakarta, and dried, pulverized, and then stored for two months at room temperature (17-33 °C), refrigerator (4 °C) and freezer (-18°C). Furthermore, the samples were tested every two weeks to determine their water content, total phenol, DPPH, and FIC. Also, Gas Chromatography-Mass Spectrometry (GC-MS) analysis was performed for two months before and after storage. The results showed that the water content increased after treatments, along with storage time which ranges from 8.55 to 14.27 %. Total phenol before storage was 175.73 mg GAE/g extract, and freezing has the highest total phenol levels after storage which was 22.07 mg GAE/g extract. Also, DPPH testing showed S. hystrix has anIC50 value of 0.45 mg/mL, and refrigeration has the lowest IC50 value of 3.18 mg/mL. FIC test showed IC50 value of 1.44 mg/mL, and freezing has the lowest FIC IC50 value of 15.36 mg/mL. In addition, GC-MS sample analysis before storage detected 8 compounds with antioxidant activity of 86.96 %. After two months, they were detected at room temperature, refrigerator, and freezing which are 4, 3, and 8 compounds respectively.

Objective: The aim of this study was to investigate the effect of temperature and time of storage on antioxidant activity of probiotics in yogurt drinks. Methods: The samples used were Yakult, Cimory, YoFit, and BioKul marketed in Medan city. The samples were stored at room and refrigerate temperatures for 2 weeks. Various volumes of samples were withdrawn and mixed with 1 ml of 2,2-diphenyl-1-picrylhydrazil solution and homogenized. Then, it was incubated for 30 min and centrifuged at 5000 rpm for 15 min. The supernatant was collected, and the absorbance was measured at 516 nm by spectrophotometer. The absorbance of different volume of supernatant of each sample was used to construct linear regression equation. Then, the IC50 value was determined. Results: The present study shows that the higher the temperature and the longer the time of storage decreased the antioxidant activity of yogurt drinks. Yakult exhibited the highest antioxidant activity among the analyzed samples. Before storage, the IC50 value of Yakult was 0.2763 ml, and after stored for 2 weeks, the IC50 value of Yakult was 0.3867 ml at refrigerated and 0.5393 ml at room temperature. Conclusions: Probiotic in yogurt drinks stored at refrigerated temperature has higher antioxidant activity than stored at room temperature. The IC50 values of yogurt drinks decreased with storage time.

ABSTRACTNanamuha (Bridelia monoica Merr.) is a plant that was used as a cancer and tumor drug by the Sangihe Islands community. This plant contains phenol compounds, flavonoids, alkaloids and tannins. The purpose of this study was to determine the potential of nanamuha leaves that grow in the Sangihe Island to have antioxidant activity and determine the total phenolic ethanol extracts of nanamuha leaf. The extraction method used is maceration with ethanol solvent p.a. The extracts obtained was determined total phenolic and antioxidant activity test using DPPH method. Based on the results obtained, the ethanol extracts of nanamuha leaves has antioxidant activity with an IC50 value of 75,03 µg / mL and the result showed that the total phenol content the ethanol extracts of nanamuha leaves of  53,34 mg / L.  Keywords : Nanamuha leaf, total phenol, antioxidant activity, DPPH  ABSTRAKNanamuha (Bridelia monoica Merr.) merupakan tanaman yang dimanfaatkan sebagai obat kanker dan tumor oleh masyarakat Kepulauan Sangihe. Metode ekstraksi yang digunakan adalah maserasi dengan pelarut etanol p.a. Ekstrak yang diperoleh ditentukan total fenolik dan uji aktivitas antioksidan dengan metode DPPH. Berdasarkan hasil yang didapatkan ekstrak etanol daun nanamuha memiliki aktivitas antioksidan dengan nilai IC50 75,03 µg/mL dan hasil penelitian menunjukan total kandungan fenolik ekstrak etanol daun nanamuha sebesar 53,34 mg/L. Kata kunci : Daun Nanamuha, Total fenol, Aktivitas antioksidan, DPPH

Aquilaria is one of the plant genus-producing agarwood. Aquilaria leaves have pharmacological effects which the potential to be processed into black tea. The aim of this study is to determine the effect of fermentation duration on physicochemical and organoleptic characteristics of Aquilaria leaves tea. This study employed a Completely Randomized Design with two factors; the plant species (A. crassna, A. malaccensis, and A. subintegra) and fermentation duration (0 minutes, 70 minutes, and 90 minutes). Parameters analyzed including water content, total phenol, and antioxidant activity. Organoleptic tests were performed on the color, taste, and aroma of the brewed tea. The results summarized that A. subintegra had the highest total phenol and antioxidant activity, but the taste of the tea was not favored. A. crassna fermented 70 minutes and A. malaccensis fermented 0 minutes were the most preferred, but both have lower total phenol and antioxidant activity than A. subintegra.

Abstrcact Jamu Selokarang is one of the traditional drinks originating from an area, namely Madura, the ingredients used in the manufacture of Jamu Selokarang are turmeric, ginger, ginger, betel leaf, soursop leaves, cloves, nutmeg, coriander, temu ireng and ginger. The ingredients used in the manufacture of herbal medicine selokarang contain a lot of antioxidants and essential oils. Testing is needed to determine the antioxidant activity contained in the herbal medicine selokarang. This study aims to determine the effect of substitution of jamu selokarang on antioxidant compounds, total phenols, flavonoids, ash content and water content. This study used a simple completely randomized design method (simple CRD) with a single factor, namely the black cumin formulation of 0%, 2%, 4%, 6% and 8% which were included in jamu selokarang. The research was repeated 3 times. The parameters observed were: water content, ash content, antioxidants, total phenols, and flavonoids. The results showed that the substitution had a significant effect on water content, ash content and antioxidant activity, while the total phenol and flavonoid analysis had no significant effect. Treatment 0% black cumin had water content of 2.68%; Ash content of 7,90%; DPPH anti-oxidant for 49.08%; antioxidant FRAP 26.25 μgAAE / mL; total phenol was 2.05 μgGAE / mg, and total flavonoids was 0.91 μgQAE / mg.

The purpose of this research to find the effect of the comparison of pregelatinization red rice flour with wheat flour on the characteristics of crackers and also to find the best ratio of red rice flour with wheat flour to the characteristics of crackers. This study used a Compeletely Randomized Design (CRD) with a comparison of red rice flour pregelatinization with wheat flour consisting of 6 treatment namely 0% : 100%;10% : 90%; 20% : 80%; 30% : 70%;40% : 60%; 50% : 50%. The treatment repeated 3 times, so that resulting in 18 experimental units. The resulting data is analyzed with variance and if the inter treatment effect on the observed variance, then continue with Duncan test. The result showed that pregelatinization red rice flour and wheat flour significantly affected to water content, protein content, total phenol, and antioxidant activity in crackers and had no significant effect on ash content. The comparison ratio of 50% pregelatinazation red rice flour with 50% wheat flour produces crackers with the best characteristics, namely: water content of 1.15%, ash content of 1.73%, 10.30% protein content, total phenol 158.83% and 45.17% antioxidant activity colour very like, texture crispy, overall liked, aroma,taste and overall acceptance like. Enzymes are made of protein, that is why they are sensitive molecules and are affected by storage conditions. A small change in enzyme activity during storage may cause a big error in analysis results. The aim of the study was to evaluate the effects of storage time and temperature on urease activity. Urease solutions were prepared at different activities (from 100 to 2000 U/mL) and stored at room temperature, in the refrigerator (4°C), and in the deep freezer (-18°C and -80°C). Activity measurements were made at regular intervals until 28 days by the modified Weatherburn method. The relative activities of 100-1000 U/mL urease solutions stored at room temperature, 4, -18 or -80°C were 75% and below after 4 days. Twenty-eight days later, for 2000 U/mL urease solutions, only at room temperature, the relative activity was reduced to 37%, while at 4, -18 or -80°C, the relative activities were above 80%. Since urease can be maintained at 4°C for 28 days without significant loss of activity, it has practical importance. Low-activity urease solutions (such as 100-1000 U/mL) should not be stored at -18 or -80°C for short or long term storage, they should be stored at 4°C only for one day. The objective of our study is to monitor the oxidative stability of different cultivars of almonds (Australian, American and Iranian) kernels/oil during the 12th month of storage at room temperature. Several physicochemical parameters free fatty acids (FFA), peroxide value (PV), panisidine value (p-AV), total oxidation value (TOTOX), Fourier transforms infrared spectrophotometer (FT-IR) and Gas chromatography Mass spectrometry (GC-MS) were used to check the oxidative stability of almond kernel. According to the results, effects of room temperature in the early stages of oxidation, primary oxidation products remained stable, whereas secondary oxidation product levels continued to rise in the later stages. In general, FFA increased with increasing storage time, the range was observed (0.21-0.97 %), PV (1.31-16.23 meqO2/kg), p-AV (2.21-19.35), TOTOX (4.83-15.81), respectively. During storage at room temperature for up to 12th months, there was no significant shifting of the spectral band in the FT-IR study. The most bounteous fatty acid in the almond oil range was observed oleic acid C18:0 (71.01-79.56 %) followed by linoleic acid C18:2 (13.13–20.65 %), palmitic acid C16:0 (4.86-5.67 %), stearic acid C18:0 (1.20-3.81 %), and palmitoleic acid C16:1(0.21-0.47 %) in all three samples during storage. These results suggest that almond oil during the 12th month of storage keeps its good chemical properties.

Zhumeria majdae (Rech. F. & Wendelbo) is an aromatic herb belonging to the Lamiaceae family, traditionally employed in the Persian medicine for the treatment of a wide number of diseases. In the present study, the chemical composition of Z. majdae essential oil obtained from the plant’s aerial features, and stored at various temperatures (refrigerator temperature 4 °C, freezer temperature −20 °C, and room temperature 20 ± 3 °C) and times (0, 3, 6, and 9 months) was studied. The essential oil was isolated through hydrodistillation, and its composition was evaluated by gas chromatography/mass spectrometry (GC/MS). The results showed that the composition of essential oils changed as a function of the various storage temperatures and times. Linalool (34.85–48.45%), camphor (27.09–39.17%), limonene (1.97–4.88%), and camphene (1.6–4.84%) made up the main volatile compounds which showed differences in their concentrations according to the various storage conditions. Notably, when compared to a non-stored treatment sample (analyzed immediately after essential oil collection), the amount of linalool and camphor increased in all samples stored in all conditions of temperature and time, with the exception of the samples stored for nine months at room temperature. On the other hand, limonene and camphene contents decreased during the storage treatments, showing that the highest content of these compounds occurred in the non-stored treatment. Essential oil storage at the freezer temperature and for three months storage time resulted in the highest average value of the major constituents, highlighting these as the best conditions for obtaining the highest content of the major compounds.

Objective: The aim of this study was to investigate the chemical compound of the antioxidant activity of the extract from andaliman fruit (Zanthoxylum acanthopodium DC.) and the quality parameters and the total flavonoid determination of the highest antioxidant activity. Methods: The andaliman fruits were extracted using a solvent such as n-hexane, ethyl acetate and 70% ethanol obtained extracts, follow the antioxidant activity using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. The quality parameters and total flavonoid determination were performed from highest antioxidant activity. Fractionated was done using vacuum liquid chromatography (VLC) with the mobile phase chloroform-methanol (9:1 ~1:9). Purification with Thin Layer Chromatography (TLC) preparative, identification using UV-spectrophotometry, FTIR, Gas chromatography Mass Spectrometry (GCMS) analysis. Results: The 70% of ethanol has the highest antioxidant activity with IC50 value 84.11±0.47 ppm; the quality parameters in this study meet the quality requirements, both specific and nonspecific parameters. Total flavonoid content of the 70% ethanol extract was 2.37±0.02%. The results of the GCMS studies showed the presence of 1,2-benzenedicarboxylic acid, mono (2-ethylhexyl) ester; squalene, methyl palmitate, methyl isopalmitate, n-tetracosanol-1 and 4-isopropyl chalcone. Conclusion: The results indicate that the 70% ethanol extracts of andaliman fruits contains 1,2-benzenedicarboxylic acid, mono (2-ethylhexyl) ester; squalene, methyl palmitate, methyl isopalmitate, n-tetracosanol-1 and 4-isopropyl chalcone have antioxidant activity with IC50 value 51.35 ppm.

The purpose of this study was to determine the antioxidant activity of fermented tea (black tea) with Ketapang leaves (Terminalia catappa) using two variations of leaf age, namely leaflets 1-3 and 4-5. The parameters observed were water content, pH, antioxidant activity (IC 50) and total phenol content of tea. Results showed that the antioxidant activity of young leaves (shoots 1-3) significantly higher than old leaves (shoots 4-5) but had lower total phenol and pH. In general, the character of Ketapang leaf tea products at shoots 1-5 has water content ranging from 5.55-5.58%, total phenol content of 22.63-28.97 mg / g EAG, and antioxidant activity (IC 50) of 6.27-6 .71 µg/ml and the pH of steeping water from 5.9 to 6.1.
