Jaruk tigarun is a traditional fermented food from South Borneo made from tigarun flower that was fermented in water without salt addition. The objectives of the research were to isolate and identify the Lactic Acid Bacteria (LAB) from jaruk tigarun then to determine the DPPH radical scavenging activities of jaruk tigarun extracts. LAB was isolated from jaruk tigarun which was fermented for 7 days at room temperature. Media of MRS+CaCO3 were used to isolate the LAB while API 50 CHL kit was utilised to identify them. Fresh flowers and jaruk tigarun were also freeze-dryed, crushed and extracted using methanol, ethanol and ethyl acetate. The total phenolic and antioxidant activity of each extract were determine with Folin-Ciocalteu method and DPPH. Three isolates of LAB were isolated and identified as Lactobacillus plantarum. The fermentation was able to increase total phenolic of jaruk tigarun which was extracted with methanol, ethanol, and ethyl acetate (53.24 ± 0.73, 44.86 ± 0.90 and 23.95 ± 0.13 mg GAE/g extract, respectively). Similarly, the antioxidant activity of jaruk tigarun that were extracted with methanol, ethanol and ethyl acetate increased to 92.68 ± 0.02 %, 92.43 ± 0.11 %, and 42.94 ± 0.02 %. Methanolic extract of jaruk tigarun has the highest IC50 that was equal to 1.511 μg/mL. UV-Vis spectrum analysis and FT-IR were used to identify the compounds isolated from methanolic extract of jaruk tigarun resulting flavonoid as tentative identified compounds. ABSTRAKJaruk tigarun adalah salah satu makanan fermentasi tradisional dari Kalimantan Selatan yang dibuat dengan cara merendam bunga tigarun dalam air matang selama beberapa hari. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri asam laktat yang terlibat selama fermentasi jaruk serta mengetahui aktivitas penghambatan radikal DPPH pada jaruk tigarun. BAL diisolasi dari bunga tigarun yang difermentasi selama 7 hari pada suhu kamar sampai menjadi jaruk. Isolasi BAL menggunakan media MRS+CaCO3 dan diidentifikasi dengan API 50 CHL Kit. Bunga segar dan jaruk tigarun juga masing-masing dikeringbekukan, dihaluskan dan diekstraksi dengan metanol, etanol dan etil asetat. Total fenolik dan aktivitas antioksidan dari masing masing ekstrak ditentukan dengan metoda Folin Ciocalteu dan DPPH. Tiga isolat BAL berhasil diisolasi dan diidentifikasi sebagai Lactobacillus plantarum. Fermentasi berhasil meningkatkan total fenolik dari jaruk tigarun yang diekstrak dengan metanol, etanol dan etil asetat berturut-turut sebesar 53,24 ± 0,73, 44,86 ± 0,90 dan 23,95 ± 0,13 mg GAE/g ekstrak. Demikian pula halnya dengan aktivitas antioksidan dari jaruk yang diekstrak dengan metanol, etanol dan etil asetat  meningkat menjadi 92,68 ± 0,02 %, 92,43 ± 0,11 %, dan 42,94 ± 0,02 %. Ekstrak metanol dari jaruk tigarun  memiliki IC50  yang paling baik yaitu sebesar 1.511 μg/mL. Hasil isolasi dan identifikasi senyawa yang dimurnikan dari ekstrak metanol jaruk menggunakan analisis spektrum UV-Vis dan FT-IR diperoleh dugaan senyawa adalah golongan flavonoid.Kata kunci: DPPH; flavonoid; jaruk tigarun; L. In order to use lactic acid bacteria isolated from Mulkimchi as a raw material for cosmetics, strain screening was carried out. And we measured the antioxidant activity of the lactic acid bacteria heat-killed cells obtained by heating lactic acid bacteria isolated from Mulkimchi. Among lactic acid bacteria derived from Mulkimchi, 41 strains were selected based on the culture suitability such as growth rate and antibiotic resistance. DPPH assay, ABTS assay, nitrite radical scavenging activity assay, and total phenolic contents were measured for 41 strains. Based on Lactobacillus plantarum LP299v, 1 strain was selected through DPPH assay, 1 strain through ABTS assay, 1 strain through nitrite radical scavenging activity assay, and 1 strain through total phenolic contents measurement. Among these, we isolated Lactobacillus sp. P100, which showed excellent antioxidant activity in all experimental methods, and the result of identification through 16s rRNA showed Lactobacillus plantarum. The strain showed 3-10% better antioxidant activity than Lactobacillus plantarum LP299v. And we measured the cell toxicity and anti-inflammatory activity. Lactobacillus sp. P100 showed the lowest cell toxicity and the highest NO production inhibitory rate. In other words, we could isolate lactic acid bacteria that can be used as an antioxidant ingredient in cosmetics from Mulkimchi.

Using natural products as antioxidant agents has been beneficial to replace synthetic products. Efforts have been made to profile the antioxidant capacities of natural resources, such as medicinal plants. The polyphenol content of Himalayan rhubarb, Rheum emodi wall, was measured and the antioxidant activity was determined using DPPH and ABTS+ assay, and the oxidative stress was assessed using SOD enzymatic assay. Five different solvent fractions, n-hexane, n-butanol, ethyl acetate, dichloromethane, and water, were used for screening the antioxidant capacity in effort to determine the optimum extraction solvent. The total phenolic contents for R. emodi fractions ranged from 27.76 to 209.21 mg of gallic acid equivalents (GAE)/g of dry weight. DPPH and ABTS+ assay results are presented into IC50 values, ranged from 21.52 to 2448.79 μg/mL and 90.25 to 1718.05 μg/mL, respectively. The ethyl acetate fraction had the highest antioxidant activity among other fractions. Also, n-butanol and water fractions showed significantly lower IC50 values than the positive control in DPPH radical scavenging activity. The IC50 values of SOD assay of fractions ranged from 2.31 to 64.78 μg/mL. A similar result was observed with ethyl acetate fraction showing the highest SOD radical scavenging activity. The study suggests that the ethyl acetate fraction of R. emodi possess the strongest antioxidant activity, thus the most efficient in extracting antioxidant contents. Moreover, a highly significant correlation was shown between total polyphenol content and antioxidant activity screening assays. The compounds related to the antioxidant activity of R. emodi were identified to myricitrin, myricetin 3-galloyl rhamnoside, and myricetin, which have not been reported in studies about R. emodi before.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

ABSTRACT Allophylus edulis and Cupania vernalis (Sapindaceae) are Brazilian native trees used as medicinal plants for the treatment of respiratory, digestive, circulatory, and skin diseases. Ubiquitously distributed in the Brazilian territory, these species are indicated for mixed plantations aimed at the recovery of degraded ecosystems. In this study, the total phenolic content (TPC) and total flavonoid content (TFC), and the antioxidant activity of extracts and fractions obtained from A. edulis and C. vernalis leaves were assessed. The TPC and TFC was determined spectrophotometrically. Antioxidant activity was evaluated through radical scavenging activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH). The extracts were obtained by two methods: maceration (method 1) and Soxhlet (method 2). Solvents of increasing polarity (hexane, dichloromethane, ethyl acetate, and n-butanol) were used to obtained the fractions. The results showed that the ethyl acetate fraction from A. edulis, obtained from the maceration method, had the highest TPC (442.0 ± 18.2 mg GAE g-1) and TFC (58.1 ± 0.4 mg RUE g-1), and antioxidant activity (EC50 = 43.6 ± 2.6 µg mL-1). By C. vernalis, superior results were obtained with the n-butanol fraction (TPC = 126.1 ± 5.8 mg GAE g-1, TFC = 37.7 ± 0.6 mg RUE g-1). The highest antioxidant potential was found in the crude hydroalcoholic extract (EC50 = 816.1 ± 50.9 µg mL-1) and butanol fraction (1,156.4 ± 3.8 µg mL-1). The results of this study show that the fractions obtained by maceration and liquid-liquid partition with more polar solvents (ethyl acetate and n-butanol) are the richest in TPC and TFC, and presented the greater antioxidant activity. Comparing the two plants, A. edulis showed the best results, with a high content of TPC, TFC, and antioxidant potential, and therefore may be used to treat diseases related to oxidative stress.

<p><strong>Objective: </strong>To evaluate the <em>in vitro </em>antioxidant activity of liquorice (<em>Glycyrrhiza glabra) </em>against H<sub>2</sub>O<sub>2</sub> induced oxidative stress in HepG2 cell line.</p><p><strong>Methods: </strong>Antioxidant activity of methanolic extracts of <em>Glycyrrhiza glabra</em> was investigated by measuring total phenolic content using folin-ciocalteu reagent (FCR), free radical scavenging activity by DPPH and ferric reducing antioxidant power (FRAP). The presence of phenolic compounds and flavonoids in the extract was confirmed by Liquid Chromatography-Mass Spectrometry (LC-MS) analysis. Furthermore, the protective effect of methanolic extract of <em>Glycyrrhiza glabra</em> against oxidative stress induced by H<sub>2</sub>O<sub>2 </sub>in HepG2 cells was investigated by MTT assay. HepG2 cells were exposed with five different treatments viz. liquorice, H<sub>2</sub>O<sub>2</sub>, ascorbic acid, H<sub>2</sub>O<sub>2</sub>+liquorice and H<sub>2</sub>O<sub>2</sub>+ascorbic acid, to explore the effect of the extract on malondialdehyde (MDA) production, catalase activity, and glutathione reductase levels.<strong></strong></p><p><strong>Results: </strong>The total phenolic content estimated in <em>Glycyrrhiza glabra </em>extract was found to be 241.47 µg per 1000 µg/ml of methanolic extract. It was found that as the concentration of the extract was increased both the free radical scavenging activity and ferric ion reducing power was also found to increase. LC-MS analysis confirmed the presence of eight different phenolic compounds in the methanolic extract which are possibly contributing to the antioxidant activity exhibited by the extract. It was also observed that liquorice treated HepG2 cells showed lower MDA and higher glutathione and catalase levels as compared to only H<sub>2</sub>O<sub>2 </sub>treated HepG2 cells where increased MDA production, decreased glutathione reductase and catalase production was observed.</p><p><strong>Conclusion: </strong>Our results thus conclude that, the methanolic extract of <em>Glycyrrhiza glabra </em>can be used as natural supplements in various disease conditions where oxidative stress has been reported. <strong></strong></p><p> </p>

Bioassay guided fractionation of ethyl acetate soluble fraction of methanolic extract of Bridelia retusa yielded tambulin (1), beta-sitosterol (2), and beta-sitosterol glucoside (3), and their structures were elucidated using modern spectroscopic techniques. Compounds 1-3 were evaluated for their antioxidant activity and compound 1 was found to be potent antioxidant. The DPPH assay showed that the compound 1 has potent antioxidant activity with IC50 166.15±1.92 SEM [muM] and the radical scavenging activity was found to be 86.03% where as the standard butylated hydroxy toluene (BHT) has IC50 value 128.83±2.1 SEM [muM]. The radical scavenging activity of standard butylated hydroxy toluene was about 86% .The total phenolic content in ethyl acetate soluble fraction was found to be 147.20±1.5 mg GAE/gm and the total flavonoid content was found to be 16.64±0.00 mg QE/gm.Journal of Institute of Science and Technology, 2015, 20(1): 97-101

The present study examined the effect of Lactobacillus plantarum ATCC14917 fermentation on the chemical composition and antioxidant activity of apple juice. Apple juice was fermented and examined of its antioxidant activity using chemical models and cellular antioxidant assay. Furthermore, the chemical composition of fermented apple juice was characterized by LC-MS/MS. Lactobacillus plantarum ATCC14917 fermentation showed an increase in DPPH and ABTS radical scavenging activity as well as cellular antioxidant activity of apple juice. However, fermentation decreased the total phenolic and flavonoid content. Subsequent LC-MS/MS analysis of the phenolic profile indicated that the content of 5-O-caffeoylquinic acid (5-CQA), quercetin, and phloretin with strong antioxidant activity was increased significantly after fermentation. The modified phenolic composition may contribute to the increased antioxidant activity of fermented apple juice. Our findings showed that Lactobacillus plantarum ATCC14917 fermentation may be an efficient way to enhance the bioavailability of phenolic compounds and to protect cells from oxidative stress.

The methanolic extract of Artemisia incisa Pamp. was dissolved in distilled water and successively partitioned with n-hexane, chloroform, ethyl acetate and n-butanol. The antioxidant potential of all these fractions and remaining aqueous fraction was evaluated by four methods, i.e., the scavenging activity of the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), the total antioxidant activity, the ferric reducing antioxidant power (FRAP) assay and the ferric thiocyanate assay. In addition, the total phenolics was determined. The obtained results revealed that among the studied fractions the ethyl acetate soluble fraction showed the most potent DPPH-radical scavenging activity with an IC50 value of 5.3 ? 0.71 ?g mL-1, which is even more effective than the standard antioxidant butylated hydroxytoluene (BHT) (IC50 value 0f 12.1? 0.92 ?g mL-1). The ethyl acetate fraction also showed the highest FRAP value (3677.13 ? 27.1 ?g TE mL-1), inhibition of lipid peroxidation (60.93 ? 0.84 % at 500 ?g mL-1) and total phenolic content (95.5 ? 0.05 ?g GAE g-1) as compared to other fractions. However, the remaining aqueous fraction was found to posses the highest antioxidant activity of all the fractions.

The petroleum ether, chloroform and ethyl acetate soluble fractions of methanolic extract of Litsea lancifolia Roxb. leaves were subjected to different pharmacological screenings to explore its potential as anti-oxidant, antimicrobial, peripheral analgesic, hypoglycemic and CNS depressing agent. The ethyl acetate soluble fraction showed highest total phenolic content and free radical scavenging activity compared to the standard, acetyl salicylic acid. Potential antimicrobial activity was shown against P. aeruginosa (23.50 mm), E. coli (22.33 mm), B. cereus (18 mm) and S. paratyphi (18 mm). The crude extract demonstrated significant peripheral analgesic (p < 0.01) activity with 69.45 and 77.96% inhibition of acetic acid-induced writhing at 100 and 200 mg/kg b.w., respectively. The crude methanolic extract also showed significant hypoglycemic activity (p < 0.01) at a dose of 500 mg/kg/day on the 7th day of treatment. All the organic soluble fractions exhibited noteworthy (p < 0.001) CNS depressant activity. Taken together, the plant can be considered as a good material for further chemical investigation to isolate the bioactive constituents.

The aim of this study is to investigate antioxidant activity and total phenolic content of ethyl acetate fraction of Sembung leaves. Sembung (Blumea balsamifera L.) is a plant that has the potential as an antioxidant because it contains secondary metabolites of alkaloids, steroids, flavonoids, saponins, and phenolic compounds. In this study, antioxidant activity was tested by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) for free radical scavenging activity assay. Evaluation of total phenolic content was tested by using the Folin-Ciocalteau method. The results showed that the ethyl acetate fraction of Sembung leaves possess showed medium antioxidant activity with IC50 valueof 221.821 compare with reference standard ascorbic acid with IC50 value 68.25 ppm. The ethyl acetate fraction of Sembung leaves has a total phenolic content of 161.101 mg GAE/g. This study provided that Blumea balsamifera leaves possess antioxidant.
