Madhuca longifolia commonly known as the Mahua tree is a medium to large sized deciduous tree distributed in India, Nepal and Sri Lanka. It is found growing wild and cultivated. The tree is considered as a kalpavriksha by the tribal who are forest dwellers and keenly conserve this tree. The flowers and fruits are edible and the flowers are fermented to prepare mahua drink which is an integral part of their cultural heritage. The seeds are good source of edible fats and used to extract oil. Several pharmacological activities are reported using various parts of the tree. Very little information exists on the molecular aspects of Madhuca longifolia which requires high quality DNA. Optimization of DNA extraction using CTAB and SDS buffer system was standardized. A protocol for extraction of high quality DNA from tree species which are rich in polyphenols is hereby discussed.

The production of tree seedlings in forest nurseries and their use in the replanting of clear-cut forest sites is a common practice in the temperate and boreal forests of Europe. Although conifers dominate on replanted sites, in recent years, deciduous tree species have received more attention due to their often-higher resilience to abiotic and biotic stress factors. The aim of the present study was to assess the belowground fungal communities of bare-root cultivated seedlings of Alnus glutinosa, Betula pendula, Pinus sylvestris, Picea abies and Quercus robur in order to gain a better understanding of the associated fungi and oomycetes, and their potential effects on the seedling performance in forest nurseries and after outplanting. The study sites were at the seven largest bare-root forest nurseries in Lithuania. The sampling included the roots and adjacent soil of 2–3 year old healthy-looking seedlings. Following the isolation of the DNA from the individual root and soil samples, these were amplified using ITS rRNA as a marker, and subjected to high-throughput PacBio sequencing. The results showed the presence of 161,302 high-quality sequences, representing 2003 fungal and oomycete taxa. The most common fungi were Malassezia restricta (6.7% of all of the high-quality sequences), Wilcoxina mikolae (5.0%), Pustularia sp. 3993_4 (4.6%), and Fusarium oxysporum (3.5%). The most common oomycetes were Pythium ultimum var. ultimum (0.6%), Pythium heterothallicum (0.3%), Pythium spiculum (0.3%), and Pythium sylvaticum (0.2%). The coniferous tree species (P. abies and P. sylvestris) generally showed a higher richness of fungal taxa and a rather distinct fungal community composition compared to the deciduous tree species (A. glutinosa, B. pendula, and Q. robur). The results demonstrated that the seedling roots and the rhizosphere soil in forest nurseries support a high richness of fungal taxa. The seedling roots were primarily inhabited by saprotrophic and mycorrhizal fungi, while fungal pathogens and oomycetes were less abundant, showing that the cultivation practices used in forest nurseries secured both the production of high-quality planting stock and disease control.

AbstractIn this article, we present the genomic DNA yield and the microsatellite and single nucleotide polymorphism (SNP) genotyping success rates of genomic DNA extracted from a large number of mouth swab samples. In total, the median yield and quality was determined in 714 individuals and the success rates in 378,480 genotypings of 915 individuals. The median yield of genomic DNA per mouth swab was 4.1 μg (range 0.1–42.2 μg) and was not reduced when mouth swabs were stored for at least 21 months prior to extraction. A maximum of 20 mouth swabs is collected per participant. Mouth swab samples showed in, respectively, 89% for 390 microsatellites and 99% for 24 SNPs a genotyping success rate higher than 75%. A very low success rate of genotyping (0%–10%) was obtained for 3.2% of the 915 mouth swab samples using microsatellite markers. Only 0.005% of the mouth swab samples showed a geno-typing success rate lower than 75% (range 58%–71%) using SNPs. Our results show that mouth swabs can be easily collected, stored by our conditions for months prior to DNA extraction and result in high yield and high-quality DNA appropriate for genotyping with high success rate including whole genome searches using microsatellites or SNPs.

The sapling stage is an important phase due to maintaining plant growth, stability, and survival over the life cycle of trees. However, there are limited investigations in the literature related to both growth and stability of different tree species. This study thus investigated how different tree species at the sapling stage showed different anatomical, morphological, and flexural traits despite being of similar age and growing under the same environmental conditions. The variation of sapling properties was determined in two deciduous tree species: common oak (Quercus robur L.) and Oriental beech (Fagus orientalis Lipsky). The results of anatomical and morphological measurements showed that the highest average values of ray length, ray width, pith radius, pith%, bark%, and node numbers were obtained in oak saplings, whereas average ring width, number of rays, and wood% were found to be higher in beech saplings. Oak also exhibited better functional stability in its saplings. The flexural properties were almost 60% greater in oak stems than beech stems. The variations in flexural properties were explained by the morphological and anatomical traits since stability was positively correlated with pith radius, pith%, and bark% and negatively correlated with the number of rays and wood%.
