Biofilm bacteria inactivation by citric acid and resuspension evaluations for drinking water production systems

The study investigates the inactivation of biofilm bacteria colonized on the surface of polyvinyl chloride (PVC) pipes delivering either groundwater or treated wastewater. It does so using a citric acid (C6H8O7) solution. The results of the study showed that the optimal conditions of the biofilm bacteria inactivation were over 10,000 mg/L citric acid concentration and 60 minutes of contact time at least. Under these conditions, the removal efficiency could reach above 99.999% for heterotrophic plate count (HPC) bacteria and 99.95% for coliform bacteria. The study also showed that the biofilm bacteria were the major sources of planktonic bacteria resuspended into water purified by drinking water production systems (DWPS).

AbstractWorldwide, surface waters like lakes and reservoirs are one of the major sources for drinking water production, especially in regions with water scarcity. In the last decades, they have undergone significant changes due to climate change. This includes not only an increase of the water temperature but also microbiological changes. In recent years, increased numbers of coliform bacteria have been observed in these surface waters. In our monitoring study we analyzed two drinking water reservoirs (Klingenberg and Kleine Kinzig Reservoir) over a two-year period in 2018 and 2019. We detected high numbers of coliform bacteria up to 2.4 x 104 bacteria per 100 ml during summer months, representing an increase of four orders of magnitude compared to winter. Diversity decreased to one or two species that dominated the entire water body, namely Enterobacter asburiae and Lelliottia spp., depending on the reservoir. Interestingly, the same, very closely related strains have been found in several reservoirs from different regions. Fecal indicator bacteria Escherichia coli and enterococci could only be detected in low concentrations. Furthermore, fecal marker genes were not detected in the reservoir, indicating that high concentrations of coliform bacteria were not due to fecal contamination. Microbial community revealed Frankiales and Burkholderiales as dominant orders. Enterobacterales, however, only had a frequency of 0.04% within the microbial community, which is not significantly affected by the extreme change in coliform bacteria number. Redundancy analysis revealed water temperature, oxygen as well as nutrients and metals (phosphate, manganese) as factors affecting the dominant species. We conclude that this sudden increase of coliform bacteria is an autochthonic process that can be considered as a mass proliferation or “coliform bloom” within the reservoir. Pre-treatment of seawater plays a critical role in removing colloidal particles, algae, sediment, and microbes, which could adversely affect the desalination process. This study focused on the utilization of the natural process of infiltration by beach wells to pre-treat the intake water for the desalination process. The scope of the study was achieved by drilling two beach wells at Al-Khobar and Jubail sites at the Arabian Gulf of Saudi Arabia at 50 m depth each. In addition to that, a total of eight monitoring wells were drilled for pump testing. Numerical modeling was conducted using SEEP/W to investigate the properties of well water flux at the beach wells. The comprehensive physio-chemical parameters such as cation, silt density index (SDI), total dissolved solids (TDS), total suspended solids (TSS), chemical oxygen demand (COD), total organic carbon (TOC), salinity, and alkalinity were analyzed for a quality assessment concerning the actual seawater. Preliminary experimental results show a reduction of the targeted parameters and indicate that beach well sand filtration in the Eastern Province would be a valuable pre-filtration step in reverse osmosis (RO)-based drinking water production systems. The water flux values for both sites were 0.0197 and 0.0208 m3/s/m2, respectively, which corresponds to 72 m3/h/m2 and 1356.48 m3/h/m2. In terms of the rate of pumping flow, the model suggests production can be increased by 20 and 53 times the measured production of the Jubail and Al-Khobar sites, respectively. The experimental results of water parameters, such as cation, SDI, TDS, TSS, etc., indicate that beach well sand filtration in the Eastern Province would be a valuable pre-filtration step in reverse osmosis drinking water production systems.

Surface waters are a major source for drinking water production. Therefore, it is essential to examine microbial processes within the water bodies, such as mass proliferations of coliform bacteria. Here, we report the draft genome sequences of Enterobacter spp., Lelliottia spp., and Serratia spp. isolated from drinking water reservoirs and lakes.

Membrane fouling by natural organic matter (NOM), namely by humic substances (HS), is a major problem in water treatment for drinking water production using membrane processes. Membrane fouling is dependent on membrane morphology like pore size and on water characteristics namely NOM nature. This work addresses the evaluation of the efficiency of ultrafiltration (UF) and Coagulation/Flocculation/UF performance in terms of permeation fluxes and HS removal, of the water from Tagus River (Valada). The operation of coagulation with chitosan was evaluated as a pretreatment for minimization of membrane fouling. UF experiments were carried out in flat cells of 13.2×10−4 m2 of membrane surface area and at transmembrane pressures from 1 to 4 bar. Five cellulose acetate membranes were laboratory made to cover a wide range of molecular weight cut-off (MWCO): 2,300, 11,000, 28,000, 60,000 and 75,000 Da. Severe fouling is observed for the membranes with the highest cut-off. In the permeation experiments of raw water, coagulation prior to membrane filtration led to a significant improvement of the permeation performance of the membranes with the highest MWCO due to the particles and colloidal matter removal.

The Llobregat is a 156 km long river, which supplies 35% of the Barcelona's drinking water needs from the Sant Joan Despí Water Treatment Plant. Since the establishment of the Salt Mine Works in the Llobregat basin in 1923, a progressive salinization of the water sources has been recorded. The operation of the Brine Collector, as a public work carried out by Aigües de Barcelona (AGBAR), started in 1989; it enabled a very significant improvement in the quality of the surface water used for drinking-water production.

Работа посвящена системному анализу данных показателей исходной воды при производстве питьевой воды в системе центрального водоснабжения крупного населенного пункта. На сегодняшний день на фоне увеличивающегося негативного антропогенного воздействия на окружающую среду наблюдается ухудшение состояния многих источников питьевого водоснабжения в широком спектре показателей, в частности, таких как органолептические свойства воды. Как следствие, возникает проблема и для питьевой воды. В работе приводится процесс подготовки данных о параметрах исходной воды, забираемой из водохранилища, которые ежемесячно (с 2002 по 2014 год) учитывались на предприятии при дезодорации воды. Приведенные параметры оказывают существенное влияние на органолептические свойства конечной воды. Подготовка данных для анализа проводится методом главных компонент К. Пирсона. Данные, полученные в пространстве R9, переводятся в пространство меньшей размерности R3. Понижение размерности позволяет снизить автокорреляцию между компонентами. Отбор компонент в пространство R3 проводится по правилу Парето. В пространстве R3 методом сферической кластеризации данных «Форель» с постоянным радиусом группировки проводится кластеризация. Приводится пошаговое визуальное представление алгоритма кластеризации в пространстве R3. В работе показано, что в данных показателях качества исходной воды имеются кластеры. Проводится корреляционно-регрессионный анализ данных, представленных в главных компонентах. Строятся регрессионные зависимости показателей органолептических свойств от главных компонент из пространства R3.

It was postulated that a phytobiotic preparation containing cinnamon oil and citric acid added to drinking water for chickens in a suitable amount and for a suitable time would beneficially modify the microbiota composition and morphology of the small intestine, thereby improving immunity and growth performance without inducing metabolic disorders. The aim of the study was to establish the dosage and time of administration of such a phytobiotic that would have the most beneficial effect on the intestinal histology and microbiota, production results, and immune and metabolic status of broiler chickens. The experiment was carried out on 980 one-day-old male chickens until the age of 42 days. The chickens were assigned to seven experimental groups of 140 birds each (seven replications of 20 individuals each). The control group (G-C) did not receive the phytobiotic. Groups CT-0.05, CT-0.1, and CT-0.25 received the phytobiotic in their drinking water in the amount of 0.05, 0.1, and 0.2 mL/L, respectively, at days 1–42 of life (continuous application, CT). The birds in groups PT-0.05, PT-0.5, and PT-0.25 received the phytobiotic in the same amounts, but only at days 1–7, 15–21, and 29–35 of life (periodic application, PT). Selected antioxidant and biochemical parameters were determined in the blood of the chickens, as well as parameters of immune status and redox status. The morphology of the intestinal epithelium, composition of the microbiome, and production parameters of chickens receiving the phytobiotic in their drinking water were determined as well. The addition of a phytobiotic containing cinnamon oil and citric acid to the drinking water of broiler chickens at a suitable dosage and for a suitable time can beneficially modify the microbiome composition and morphometry of the small intestine (total number of fungi p < 0.001, total number of aerobic bacteria p < 0.001; and total number of coliform bacteria p < 0.001 was decreased) improving the immunity and growth performance of the chickens (there occurred a villi lengthening p = 0.002 and crypts deepening p = 0.003). Among the three tested dosages (0.05, 0.1, and 0.25 mL/L of water) of the preparation containing cinnamon oil, the dosage of 0.25 mL/L of water administered for 42 days proved to be most beneficial. Chickens receiving the phytobiotic in the amount of 0.25 mL/L had better growth performance, which was linked to the beneficial effect of the preparation on the microbiome of the small intestine, metabolism (the HDL level p = 0.017 was increased; and a decreased level of total cholesterol (TC) p = 0.018 and nonesterified fatty acids (NEFA) p = 0.007, LDL p = 0.041, as well as triacylglycerols (TAG) p = 0.014), and immune (the level of lysozyme p = 0.041 was increased, as well as the percentage of phagocytic cells p = 0.034, phagocytosis index p = 0.038, and Ig-A level p = 0.031) and antioxidant system (the level of LOOH p < 0.001, MDA p = 0.002, and the activity of Catalase (CAT) p < 0.001 were decreased, but the level of ferric reducing ability of plasma (FRAP) p = 0.029, glutathione p = 0.045 and vitamin C p = 0.021 were increased).
