Tissue cultures have been established from leaves of one anther-derived haploid plant of Atropa belladonna L. Regenerants obtained from callus cultures were transferred to soil and reared to maturity. Callus cells and regenerants exhibited variable degrees of ploidy. The frequency of different ploidy levels in both the systems did not vary significantly during fifth to eighth serial passages.Callus tissue and regenerated plants (at flowering stage) were analyzed to determine the concentration of tropine, atropine, scopolamine, and total alkaloids. While the alkaloid content in callus cultures was very low (0.8 × 10−3%), the regenerated plants contained different levels of individual and total alkaloids. The ploidy of the plant had direct bearing on the individual and total alkaloid contents.

The use of acetyl-3-pyridine and pyridine-3-sulfonic acid as analogues for nicotinic acid has been tested with tissue cultures of embryonic chick heart. Both roller tube and Carrel flask cultures were employed. Cell migration, appearance of the cells, and the uptake of tracer P32 were used as criteria for the action of the analogues. Migration of the cells could be inhibited by both compounds, but at different levels. Both produced abnormal types of cells, but not the same type of abnormality. Uptake of P32 was inhibited by both compounds. Addition of nicotinic acid failed to reverse the effects of the analogues at the concentrations used.

Six single-cross hybrids and their parental lines were inoculated with the FG36 Fusarium graminearum isolate in 2005 and 2006. In both years the degree of infection increased after artificial inoculation for both the hybrids and the inbred lines compared with the level of natural infection. The more severe stalk infection recorded in 2005 than in 2006 could be attributed to the weather conditions during flowering and harvesting.The 18 genotypes examined exhibited different levels of resistance to fusarium stalk rot. It could be concluded from the results that the resistance level of the female parent was decisive in the inheritance of the response to fusarium stalk rot (female component-hybrid r = 0.88, male component-hybrid r = 0.39).Some genotypes may be severely affected in epidemic years, while exhibiting a lower rate of infection in years with lower pathogen pressure. This suggests that successful breeding for resistance can only be carried out efficiently by means of artificial inoculation.

Trypanosomatids are diverse and can infect several host species, including small mammals (rodents and marsupials). Between 2012 and 2014, 91 small mammals were surveyed for trypanosomatid infection in the Estação Biológica FIOCRUZ Mata Atlântica (EFMA), an Atlantic Forest area in Rio de Janeiro that presents different levels of conserved and degraded areas. Blood, skin, liver, and spleen samples were submitted to parasitological, serological, and molecular assays to detect the infection and determine the taxonomic status of their parasites. Sixty-eight individuals (74.7%; n = 91) were infected by trypanosomatids, including fourteen mixed infected by different trypanosomatid parasites. These hosts were infected by: T. cruzi DTU TcI (n = 12), T. cruzi DTU TcIV (n = 2), T. janseni (n = 15), T. dionisii (n = 1), and T. rangeli A (n = 1) detected in blood or tissue cultures, in addition to T. cruzi DTU TcI (n = 9) and Leishmania sp. (n = 1) only by the molecular diagnosis. Serological diagnosis was positive in 38 (71.6%) individuals for T. cruzi, the same amount for Leishmania spp., and 23 (43.3%) individuals were mixed infected. These data indicate a remarkable richness of trypanosomatid species/genotypes infecting small mammals, even in a disturbed area with low mammal species diversity—as is the case of the EFMA—reinforcing the generalist aspect of these parasites.

ABSTRACT The objective of this work was to evaluate the response of sugarcane genotypes to Meloidogyne javanica and Pratylenchus zeae. Sugarcane seedlings of the genotypes RB985944, RB987932, RB877935, RB855156, RB935744, RB996961, RB925345, RB935581 and RB966928 obtained from tissue cultures and maintained in pots with sterilized soil in a greenhouse were inoculated with 5,000 eggs + second- stage juveniles of M. javanica or 1,600 of P. zeae per plant, using six replicates per treatment. Tomato (Rutgers) and sorghum (BRS506) plants were used as control and proof of viability of the inoculum for the M. javanica and P. zeae, respectively. The number of root-knot in the plants inoculated with M. javanica, final nematode population and reproduction factor (RF) of both nematodes were evaluated at 120 days after inoculation. The averages of the different variables were compared by the Scott-Knott's clustering test at 5%. The nematodes M. javanica and P. zeae showed RF>1.00 in all sugarcane genotypes assessed, however, different levels of susceptibility were found. The lowest reproduction of the nematodes was found in the genotypes RB008347, RB877935, RB975944 and RB987932 (M. javanica) and RB987932 and RB966928 (P. zeae).

Recently we have been investigating a class of nickel-base superalloys which possess substantial room temperature ductility. This improvement in ductility is directly related to improvements in grain boundary strength due to increased boundary cohesion through control of detrimental impurities and improved boundary shear strength by controlled grain boundary micros true tures.For these investigations an experimental nickel-base superalloy was doped with different levels of sulphur impurity. The micros tructure after a heat treatment of 1360°C for 2 hr, 1200°C for 16 hr consists of coherent precipitates of γ’ Ni3(Al,X) in a nickel solid solution matrix.

Some authors using immunofluorescent techniques had already suggested that some hepatocytes are able to synthetize several plasma proteins. In vitro studies on normal cells or on cells issued of murine hepatomas raise the same conclusion. These works could be indications of an hepatocyte functionnal non-specialization, meanwhile the authors never give direct topographic proofs suitable with this hypothesis.The use of immunoenzymatic techniques after obtention of monospecific antisera had seemed to us useful to bring forward a better knowledge of this problem. We have studied three carrier proteins (transferrin = Tf, hemopexin = Hx, albumin = Alb) operating at different levels in iron metabolism by demonstrating and localizing the adult rat hepatocytes involved in their synthesis.Immunological, histological and ultrastructural methods have been described in a previous work.

Two different cell lines (Be-Wo and Jar) of human gestational choriocarcinoma have been maintained in continuous tissue culture for a period of four and two years respectively without losing the ability to elaborate human chorionic gonadotropin (HCG). Tissue cultures, as revealed by electron microscopy, consisted of small cells with single nuclei. In some instances cell surfaces were provided with microvilli but more often the intercellular spaces were narrow and bridged by desmosomes. However, syncytium was not formed. Endoplasmic reticulum (ER) was poorly developed in both cell lines, except in some Be-Wo cells it was prominent. Golgi complex, lysosomes and numerous free ribosomes, as well as excessive cytoplasmic glycogen, were present in all cells (Fig. 1). Glycogen depletion and concomitant increase of ER were observed in many cells following a single dose of 10 ugm/ml of adrenalin added to medium (Fig. 2).

The differentiation of phloem in plant tissue cultures offers a unique opportunity to study the development and structure of sieve elements in a manner that avoids the injury responses associated with the processing of similar elements in intact plants. Short segments of sieve elements formed in tissue cultures can be fixed intact while the longer strands occuring in whole plants must be cut into shorter lengths before processing. While iyuch controversy surrounds the question of phloem function in tissue cultures , sieve elements formed in these cultured cells are structurally similar to those of Intact plants. We are particullarly Interested In the structure of the plasma membrane and the peripheral ER in these cells because of their possible role in the energy-dependent active transport of sucrose into the sieve elements.
