Kidney histology in streptozotocin-induced diabetic male Wistar rats treated with combined extract of temulawak rhizome and belimbing wuluh fruit. Nusantara Bioscience 9: 312-317. Complications that are occurred in patients with Diabetes Mellitus usually followed by kidney damage. Temulawak (Curcuma xanthorrhiza Roxb.) and belimbing wuluh (Averrhoa bilimbi L.) were traditionally used to decrease blood glucose level. Thus, they were potential as antidiabetic drugs. This study aimed to evaluate the combination of ethanol extracts of temulawak rhizome and belimbing wuluh fruit in repairing kidney damage in diabetic male Wistar rats induced by streptozotocin (STZ). An experimental method using a completely randomized design that consist of seven treatments with three replications. Six treatment groups were injected intraperitoneally with a dose of 60 mg/kg BW STZ, and one group served as a control. The animals which have blood glucose level ≥200 mg/dl were stated as diabetic. Furthermore, the animals were treated orally with single extract i.e. temulawak 17.5 mg/kg BW or belimbing wuluh 750 mg/kg BW and combined extracts 383.75 or 767.5 mg/kg BW, as well as glibenclamide 0.45 mg/kg BW as reference, including diabetic rat as positive control and non-diabetic rat as negative control. The results showed that combine extract at dose of 383.75 mg/kg BW treatment repaired the kidney histology, i.e., glomerular diameter and Bowman space width, as well as significantly decreased the necrosis percentage of proximal tubular in diabetic rat compared with positive control group (p<0.05). In conclusion, the combined extract of temulawak rhizome and belimbing wuluh fruit has potent to cure renal failure in diabetic rats induced by streptozotocin.

Background: The global population of diabetes mellitus tends to increase every year. This is related to the increased of population numbers, lifestyle, increased prevalence of obesity and less physical activity (Smeltzer & Bare, 2002). Olive oil is a functional diet because it is rich of monounsaturated fatty acids (MUFA), especially oleic acid (70% - 80%), contains antioxidants and polyphenol compounds. Polyphenols are α-amylase enzyme inhibitors that have function in the breakdown of carbohydrates, so that the level of blood glucose in hyperglycemia can be lowered (Astrawan made, Wresdiyati Tutik, Nasution Nurlayla Amas, 2015). The goal of this study is to examine the effectiveness of extra virgin olive oil to blood glucose level. Method:This study used experimental laboratory study with post test only control group design. The independent variable was the extra virgin olive oil, while the dependent variable was the blood glucose level. The samples were 32 healthy 8 weeks old white male wistar strain rats with the weigh between 180 mg - 200 mg. It divided into 3 groups, the negative control group was fed with standard diet, the positive control group was fed with standard diet and 1 ml fructose for 4 weeks, the treatment group was fed with standard diet, 1 ml fructose and 0.5 gr extra virgin olive oil for 4 weeks, on the fifth week the fructose was increased into 4 ml/day for the next 4 weeks, on the 52nd day the blood glucose level was checked after it was fasted for 6 hours. The data analysis used t-test and Mann Withney test with α = 0.05. Result: The blood glucose level average of the positive control group was 309.57 ± 120.12, the blood glucose level average of the negative control group was 111.85 ± 10.57, the blood glucose level average of the treatment group was 115.12 ± 12.25. Conclusion: Giving the extra virgin olive oil can effectively lower the blood glucose level on the male wistar rats.

INTRODUCTION Diabetes mellitus is the most common metabolic disease worldwide. Nigella sativa also known as black cumin, is an emerging miracle herb with a rich historical and religious background. In the present study, we have attempted to study the hypoglycemic effect of aqueous extract of Nigella sativa on diabetes-induced rats. MATERIAL AND METHODS This preclinical study was conducted in Department of Pharmacology from August to October 2019. In this study, hyperglycemia was induced by a single intraperitoneal dose of 60 mg/kg streptozotocin in 0.1 M citrate buffer (pH 4.5). Animals were divided into five groups containing normal control, negative control, standard, test-1, and test-2. The aqueous extract of Nigella sativa was administered 400 and 800 mg/kg per oral in both the test groups for 18 days. Blood glucose level was measured at three-days interval by glucometer. Oral glucose tolerance test was done by administrating 2 gm/kg per oral glucose and the blood glucose was recorded every two-hours interval for eight hours. RESULTS An oral glucose tolerance test showed a decrease in blood glucose level at the dose of 400 and 800 mg/kg compared to the negative control group. The administration of aqueous extract of Nigella sativa for 18 days showed significant decrease in blood glucose level (p=0.001) compared to negative control group. The blood glucose level in test 1 (400 mg/kg) was significantly reduced compared to the standard group (p***<0.001). The blood glucose level in test 2 at 18th day was less significant (p*< 0.05) compared with standard group and test 1 (p**<0.01, p***<0.001). CONCLUSION The findings of the present study indicate that the aqueous extract of Nigella sativa has significant hypoglycemic effect in Wistar rats.

Red Palm Oil is one of processed products from palm oil and also a food source rich in carotenoid,around500-800 ppm. Carotenoid concentrate is also a product obtained from the oil removal process and the carotenoid level increases to 6000-7000 ppm. This research aimed to determinethe potential of the red palm oil and the carotenoid concentrate as a preliminary test of anti-oxidant supplement Glucose Tolerance Test on Rat. The anti-oxidant activity of red palm oil and carotenoid concentrate was tested using DPPH method with 60 minutes incubation period. The anti-hyperglycemia activity evaluationwas conducted using animals(n=40) which divided into 8 groups: Negative control group (distilled water), positive control (glibenclamide 10 mg), three groups were given red palm oil with dose of1, 2, and 3 ml , and three groups were given carotenoid concentrate with dose of 1, 2, and 3 ml . The decrease of blood glucose for 180 minutes in anti-hyperglycemia test were in order ofglibenclamide10 mg> MSM 3 ml> MSM 2 ml> MSM 1 ml> KK 3 ml> KK 2 ml> KK 1 ml and distilled water. The red palm oil induction could lower blood glucose level faster than carotenoid concentrate, The anti-oxidative properties of minor componentsin red palm oil prevent the blood glucose oxidation and the pancreas could be actively produce insulin normally. In conclusion, red palm oil could be used synergistically toenhance the effectiveness oforal hyperglycemic agent.
