Perairan pantai Kabupaten Jeneponto, Bantaeng, dan Bulukumba merupakan sentra produksi rumput laut Kappaphycus alvarezii di Sulawesi Selatan. Pengelolaan budidaya yang dilakukan oleh pembudidaya di daerah tersebut cukup bervariasi sehingga dilakukan penelitian yang bertujuan untuk mengetahui faktor pengelolaan budidaya yang mempengaruhi produksi rumput laut. Metode survai melalui pengajuan kuesioner kepada 62 responden secara terstruktur. Sebagai peubah tidak bebas dalam penelitian ini adalah produksi rumput laut, sedangkan peubah bebas adalah faktor pengelolaan budidaya. Analisis regresi berganda dengan peubah boneka digunakan untuk memprediksi produksi rumput laut. Hasil penelitian menunjukkan bahwa produksi rumput laut di perairan selatan Sulawesi Selatan berkisar antara 463-5.000 dengan rata-rata 1.502,3 kg kering/3.000 m2 yang dibudidayakan dengan tali panjang. Faktor pengelolaan budidaya yang mempengaruhi produksi rumput laut adalah jarak antar tali ris, jarak antar rumpun dalam tali ris, hama baronang, penyakit ice-ice, bobot bibit, asal bibit dan sumber cemaran. Untuk meningkatkan produksi rumput laut di perairan selatan Sulawesi Selatan dapat dilakukan melalui peningkatan bobot bibit antara 36,9 sampai 100,0 g/rumpun, menggunakan bibit yang tidak diangkut terlalu lama, tidak menambah jarak antar tali ris sampai melebihi 1,0 m, tidak menambah jarak antar rumpun dalam tali ris yang melebihi 25 cm serta melakukan penanaman berdasar kalender musim tanam untuk mencegah terjadinya serangan hama dan penyakit serta cemaran.Coastal waters of Jeneponto, Bantaeng, and Bulukumba Regencies are the centre of seaweed Kappaphycus alvarezii production in South Sulawesi. Culture management practices applied by farmers in these areas are highly variable. Therefore, this research was conducted to study culture management practices affect the seaweed production in the area. Field survey was conducted by interviewing 62 respondents using questionnaires. The dependent variable in this research was seaweed production, while the independent variables were culture management factors. Multiple regressions with dummy variable were employed to analyze the data to predict seaweed production. The results show that the seaweed productions per one cycle in the south coastal waters of South Sulawesi were ranging from 463 to 5,000 kg dry/3,000 m2. Using long line method, the average yield production was 1,502.3 kg dry/3,000 m2. Culture management practices that affect the seaweed production were distance between ropes, distance between seaweed seeds along the rope, rabbitfish predatory, ice-ice disease, weight of seed, seed origin, and source of pollution. Increasing seaweed production in the south coastal waters of South Sulawesi could be done through increasing weight of seed from 36.9 to 100.0 g/clump, quicker transport of seed, maintaining the distance between the ropes not more than 1.0 m, not increasing the distance between clump along the ropes more than 25 cm along as well as conducting seed planting based on cultivating season to prevent pest diseases and pollution.

Two different colors (green and brown) of Kappaphycus alvarezii have been farmed in Indonesian waters for many years. This study aimed at comparing two ‘varieties’, i.e. green and brown, both genetically and morphologically. Samples for DNA analysis were collected from a farmer in Pinrang Regency, South Sulawesi. Five universal primers i.e. Ca-01, Ca-02, P-40, P-50, and DALRP were selected to obtain DNA genetic markers in differentiating the green and brown varieties. To compare coloration patterns during cultivation and the growth performance of both varieties, a field experiment was performed in a seaweed farming area in Pinrang Regency, during dry season of August-September 2004. The result of genetic assessment showed that the five selected primers revealed different RAPD banding pattern for both varieties. P-50 and DALRP primers demonstrated the greatest amplification in differentiating RAPD fragment between green and brown varieties. Fragment 900 bp and 1.300 bp were consistently generated in the green variety but were not amplified in the brown variety. The result of the field study confirmed that the coloration pattern of green and brown varieties was fixed; no interchange in color occurred during one crop cultivation.

Demand for this commodity is quite high, as reflected in the increasing volume of exports each year. Fulfillment of these demands is obtained from the production of cultivation. Generally farmers get natural seaweed seedlings, namely cuttings from existing seaweed. The continuous use of seeds from nature can cause deterioration in the quality and quantity. Handling the problem of quality deterioration from seaweed seeds originating from nature, can be overcome by multiplying the seeds through tissue culture methods in vitro. In term of tissue culture techniques method, the most important thing to note is environmental parameters. An environment parameter that changes suddenly such as salinity can inhibit the growth of seaweed. Thus, this work is attampting the different salinity treatment on seaweed explants K. alvarezii. This study aims at determining (weight) the explants of K. alvarezii with a comparison of the different salinity levels in the in vitro tissue culture method. The method used in this study was a completely randomized design (CRD) with the different salinity treatments namely 30, 31, 32, 33, and 34 ppt. The results showed that the different salinities influenced the growth rate of K. alvarezii seaweed explants with the best explant growth at the salinity of 31 ppt, while the lowest growth value was obtained at 34 ppt

Abstract:Epiphytes are organisms that attach to other organisms to support their growth and development. One type of epiphytes that has a negative impact on the host is algae (macroalgae). This algae is a competing organism for cultivation algae (Kappaphycus alvarezii). This research was conducted From May to October 2017 in Gerupuk Bay, Central Lombok. The aim of the research is to find out the types of macroalgae epiphytes on macroalgae kappaphycus alvarezii. This research was conducted using exploratory methods by taking seaweed samples overgrown with epiphytes, the research data obtained were descriptive analisys. As supporting data, environmental parameters are measured including temperature, pH, salinity, DO, phosphate, and NH4. Based on the results of exploration found 13 species of epiphytic algae in kappaphycus alvarezi cultivation, namely: Chaetomorpha linum, Chaetomorpha linum, Cladophora dalmatica, Acanthophora spicifer, Batrachospermum sp, Callithamnion sp., Caulachantus ustulatus, Ceramium sp, Hypnea spinnosa, Hypnea spinella, Hypnea sp, Polysiphonia brodiaei , Spyridia filamentosa. The type of epiphytes found that grows in both cultivation methods and some others are found only in one method. Meanwhile, data on environmental parameters are in the optimal range for macroalgae growth.This data is expected to be used as a consideration for the cultivateddevelopment in the Gerupuk Bay cultivation center. Keywords : Macroalgae, Epiphytes, Aquaculture,  Kappaphycus alvarezii

Kappaphycus seaweed is the primary source of carrageenan, a polysaccharide widely utilised as a thickening and gelling agent in many commercial products. Unlike pure refined carrageenans, semi-refined carrageenans (SRC) are considered more cost-efficient and easier to produce. This study evaluates the quality of SRC extracted from Kappaphycus alvarezii (Doty) Doty ex Silva 1996, and Kappaphycus striatus (F. Schmitz) Doty ex P.C. Silva 1996, seaweeds farmed in the northern part of Java Island, particularly in Serang, Seribu Islands, and Karimunjawa Islands. Using 8 % KOH as a solvent, SRC was extracted from clean dried seaweed. Physical and chemical characteristics of the SRC were measured by assessing the percentage SRC yield, ash content, moisture, sulphate content, viscosity (cp) and gel strength (g.cm-2). Pearson’s correlation analysis was performed to investigate the relationship between SRC quality and environmental factors. Fourier transform infrared spectroscopy (FTIR) analysis showed that all samples had kappa-carrageenan from peak absorbance of 1218–1224 cm-1 indicating ester sulphate bonds, 924–925 cm-1 indicating 3,6–anhydrogalactose, and 843–844 cm-1 suggesting galactose–4–sulphate groups. Furthermore, ash content, moisture, and viscosity of SRC passed the FAO standard, while the sulphate content was below the threshold limit. Several environmental factors had a significant correlation with SRC quality, such as water pH positively correlated with gel strength, salinity level positively correlated with gel strength, and depth positively correlated with ash content. This study concluded that SRC produced from the sampling sites had varying qualities and may be affected by environmental parameters in their cultivation site.

Peremajaan bibit melalui seleksi dapat dilakukan untuk perbaikan kualitas bibit rumput laut. Penelitian ini bertujuan untuk menentukan performa bibit rumput laut K. alvarezii hasil seleksi varietas Takalar, SIRICA, dan Bali. Desain penelitian adalah rancangan acak lengkap (RAL), dengan empat siklus pemeliharaan sebagai ulangan. Seleksi bibit dilakukan pada ketiga varietas dengan bibit non-seleksi sebagai kontrol internal. Pemeliharaan dilakukan di perairan Laikang, Takalar, Sulawesi Selatan selama empat siklus dengan durasi 30 hari/siklus. Pengukuran laju pertumbuhan harian (LPH), kandungan karagenan, kekuatan gel, dan monitoring kualitas air dilakukan setiap 30 hari. Data yang diperoleh dianalisis menggunakan analisis ragam (ANOVA). Hasil penelitian menunjukkan bahwa seleksi bibit berpengaruh signifikan terhadap LPH rumput laut (P<0,05). Rumput laut hasil seleksi memiliki LPH lebih tinggi, yakni: Bali (4,75 ± 0,04%/hari), SIRICA (4,74 ± 0,06%/hari), dan Takalar (4,62 ± 0,12%/hari) dibandingkan dengan kontrol internal Bali (3,94 ± 0,15%/hari), SIRICA (3,74 ± 0,10%/hari), dan Takalar (3,66 ± 0,32%/hari). Rumput laut hasil seleksi varietas Bali dan SIRICA memiliki kandungan karagenan yang relatif sama yakni (37,27 ± 6,68%) dan (37,08 ± 7,17%). Varietas Takalar memiliki kandungan karagenan yang rendah (31,84 ± 5,32%) namun memiliki kekuatan gel yang paling tinggi (570,03 ± 145,72 g/cm²) dibandingkan varietas Bali (444,39 ± 157,44 g/cm²) dan SIRICA (438,48 ± 72,70 g/cm²).Regeneration by selection method can improve the quality of seaweed seed. This study was aimed to determine the performance of selected seaweed seeds of K. alvarezii from Takalar, SIRICA, and Bali varieties. The experiments were arranged in a completely randomized design with four cycles cultivations as replications. In each variety, a batch of non-selected/regular seeds was cultivated serving as internal control. The seaweed was cultivated in Laikang waters, Takalar Regency, South Sulawesi for 30 days for each cycle. The measurements of daily growth rate (DGR), carrageenan yield, gel strength, and water quality were conducted every 30 days. The collected data were statistically analyzed using ANOVA. The results showed that the selected seeds showed faster growth than that of its respective control (P<0.05). The selected seed of three varieties of K. alvarezii had higher DGR: Bali (4.75 ± 0.04%/day), SIRICA (4.74 ± 0.06%/day), and Takalar (4.62 ± 0.12%/day) than that of control varieties, Bali (3.94 ± 0.15%/day), SIRICA (3.74 ± 0.10%/day), and Takalar (3.66 ± 0.32%/day). The selected seeds of Bali and SIRICA had similar carrageenan content (37.27 ± 6.68%) and (37.08 ± 7.17%), respectively. Takalar had the lowest carrageenan yield (31.84 ± 5.32) but had the highest gel strength (570.03 ± 145.72 g/cm²) compared to Bali (444.39 ± 157.44 g/cm²) and SIRICA (438.48 ± 72.70 g/cm²).
