Samarinda woven sarong is typical sarong of Samarinda. This sarong is still using silk spun silk raw material imported from Tiongkok. The development of aquaculture is needed to obtain local silk threads in order to help to reduce production costs in the samarinda silk industry. The high quality mulberry plants are needed to support silkworm cultivation (sericulture). The qualified mulberry plants are obtained by in vitro culture techniques. The objective of this study is to obtain mulberry leaf (Morus Sp) free from disease, uniform, and to obtain quality silk threads through sericulture techniques with feeds that were propagated in vitro. The research stages consist of sterilization, Murashige and Skoog (MS) media, sub culture, observation of data analysis fund. The design used is Completely Randomized Design with single factor, ie BAP concentration (control, 0.5 mg / l, 1 mg / l and 2 mg / l). Each treatment is repeated 8 times. The results showed that administration of ZPT 2 mg / l at age 4 MST gives the best response for all observed variables. The use of ZPT BAP with various concentrations produces the germination rate, shoot height, number of shoots and number of different leaves. All treatments are unsuccessful in inducing roots.Keywords: in vitro; Murbei; SericultureSarung tenun samarinda adalah sarung khas kota Samarinda. Sarung ini masih menggunakan bahan baku sutera jenis spun silk yang diimpor dari Tiongkok. Pengembangan serikultur diperlukan untuk mendapatkan benang sutera lokal. Sehingga membantu mengurangi biaya produksi dalam industri persuteraan samarinda. Tanaman murbei yang berkualitas diperlukan untuk menunjang budidaya ulat sutera (serikultur). Tanaman murbei yang berkualitas diperoleh dengan teknik kultur in vitro. Tujuan penelitian adalah memperoleh daun murbei (Morus Sp) yang bebas penyakit dan seragam serta memperoleh benang sutera berkualitas melalui teknik serikultur dengan pakan yan diperbanyak secara in vitro. Tahapan penelitian terdiri dari sterilisasi, pembuatan media Murashige dan Skoog (MS), sub kultur, Pengamatan dana analisis data. Rancangan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan faktor tunggal yaitu berbagai konsentrasi BAP (kontrol, 0,5 mg/l, 1 mg/l dan 2 mg/l) setiap perlakuan diulang 8 kali. Hasil penelitian menunjukkan bahwa pemberian ZPT 2 mg/l pada umur 4 MST memberikan respon terbaik untuk semua variabel yang diamati, Penggunaan ZPT BAP dengan berbagai konsentrasi menghasilkan waktu kecepatan bertunas, tinggi tunas dan jumlah tunas dan jumlah daun yang berbeda. Salah satu alternatif untuk perbanyakan anggrek hitam (Coelogyne pandurata Lindl.) adalah melalui multiplikasi tunas anggrek secara in vitro. Tujuan penelitian adalah untuk membuktikan pengaruh pemberian NAA (Naphthalene Acetic Acid) dan ekstrak biji jagung (Zea mays) terhadap multiplikasi tunas anggrek hitam. Penelitian dilakukan di Laboratorium Kultur Jaringan Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Tanjungpura Pontianak. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) pola faktorial dengan dua faktor perlakuan. Faktor pertama adalah NAA terdiri dari 5 taraf konsentrasi yaitu A0 (0 M/ kontrol) A1 (10-7 M), A2 (10-6 M), A3 (5x10-7 M) dan A4 (5x10-6 M ) dan faktor ekstrak biji jagung (B) dengan 5 taraf konsentrasi yaitu B0 (0%), B1 (2,5%), B2 (5%); B3 (7,5%) dan B4 (10%). Pemberian kombinasi NAA dan ekstrak biji jagung berpengaruh nyata terhadap semua parameter yaitu jumlah tunas, jumlah daun, dan tinggi tunas. Hasil terbaik rerata jumlah tunas pada perlakuan A4+B4 yaitu 5x10-6M NAA+10% ekstrak biji jagung. Hasil terbaik pada rerata jumlah daun pada perlakuan A2+B2 yaitu 5x10-7M NAA+5% ekstrak biji jagung dan hasil terbaik pada rerata tinggi tunas pada perlakuan A1+B1 yaitu 10-7M NAA+2,5% ekstrak biji jagung.Kata Kunci: multiplikasi; tunas anggrek hitam; ekstrak biji jagung; NAA. ABSTRACTGenerative reproduction of orchid plants it takes a requires the help of mycorriza mushrooms for seed germination, whereas conventional propagation business takes a long time to obtain large quantities of plants. One alternative to the propagation black orchids (Coelogyne pandurata Lindl.) is required through tissue culture techniques. The purpose of this study is to find the influence and concentration corn seed extract (Zea mays) and NAA (Naphthalene Acetic Acid) on the multiplication black orchids. This research was conducted in the tissue culture laboratory Biology Department Faculty of Mathematics and Natural Sciences Tanjungpura University Pontianak. The study used a Complete Randomized Design (RAL) of factorial patterns with two treatment factors. The first factor is that the NAA consists of 5 concentration levels  A0 (0 M) A1 (10-7 M), A2 (10-6 M), A3 (5x10-7 M) and A4 (5x10-6 M ) and the second factor is that corn seed extract of 5 levels concentratio B0(0%), B1 (2,5%), B2 (5%); B3 (7,5%) and B4 (10%). The administration NAA and corn seed extract in combination has a real effect on all parameters namely the number shoots, the number leaves, and the height shoots. The best results where the average number of shoots in the treatment of A2+B2 namely 5x10-6M NAA + 10% corn seed extract. The best results average number of leaves in the treatment  A2+B2 namely 5x10-7M NAA + 5% corn seed extract and in the best results for shoot height in the treatment of A1+B1 namely 10-7M NAA + 2.5% corn seed extract.Keywords: Multiplication; black orchid’s shoot; corn  seed extract; NAA

This study was aimed at determining the response of turmeric shoot explants after the provision of various types and concentrations of cytokinins in vitro. This experiment was conducted at the Tissue Culture Laboratory, Faculty of Agriculture, Padjadjaran University from January to April 2018. The data were analyzed using T-Test. The experimental method used in this research was Completely Randomized Design (CRD). Explant planting was carried out in Laminar Air Flow. The experiment consisted of 7 treatments consisting of 4 replications and each test consisted of 4 units. Observation of this experiment was carried out for 12 MST. The main observations were made on the data that were tested statistically namely the percentage of explant growing shoots, percentage of explant growing roots, shoot height, number of tuns, number of roots and root length. The treatments consisted of Control, 2.5 mg L-1 BAP, 5 mg L-1 BAP, 0.5 mg L-1 TDZ, 1 mg L-1 TDZ, 0.01 mg L-1 Zeatin and 0.1 mg L Zeatin -1. The results show that the treatment of 1 mg L-1 TDZ shows the best response to the growth of turmeric explants by increasing the number of turmeric shoot explants than the others.RESPONS EKSPLAN TUNAS KUNYIT SETELAH SITOKININ SECARA IN VITROTujuan dari penelitian ini yaitu untuk melihat respons eksplan tunas kunyit terhadap pemberian berbagai jenis dan konsentrasi sitokinin secara in vitro. Percobaan ini dilakukan di Laboratorium Kultur Jaringan, Fakultas Pertanian, Universitas Padjadjaran dari bulan Januari sampai April 2018. Hasil percobaan dianalisis dengan Sample T-Test. Metode percobaan yang digunakan dalam penelitian ini yaitu Rancangan Acak Lengkap (RAL). Penanaman eksplan dilakukan di dalam Laminar Air Flow. Percobaan terdiri dari 7 perlakuan sebanyak 4 ulangan dan setiap ulangan terdiri dari 4 unit. Pengamatan percobaan ini dilakukan selama 12 MST. Pengamatan utama dilakukan terhadap data-data yang diuji secara statistik yakni persentase eksplan tumbuh tunas, persentse eksplan tumbuh akar, tinggi tunas, jumlah tunas, jumlah akar dan panjang akar. Perlakuan terdiri dari Kontrol; 2,5 mg L-1 BAP; 5 mg L-1 BAP; 0,5 mg L-1 TDZ; 1 mg L-1 TDZ; 0,01 mg L-1 Zeatin; dan 0,1 mg L-1 Zeatin. Hasil penelitian menunjukkan bahwa perlakuan 1 mg L-1 TDZ menunjukkan respons yang lebih baik terhadap pertumbuhan eksplan kunyit dengan meningkatkan jumlah tunas eksplan tanaman kunyit daripada yang lainnya.

Background: Pineapple fruit used in this study was obtained from Sipahutar sub-district, North Tapanuli regency, North Sumatra which was a superior commodity fruit. Propagation of the Sipahutar pineapple plant was conducted vegetatively using bud stems and crown shoots, but the amount was limited to be planted in large areas. Therefore, propagation through tissue culture techniques could be used as an alternative solution.Objective: Explants source were taken from plantlets in vitro aged 3 months were grown on Murashige and Skoog (MS) basic medium with the addition of Indole Acetic Acid (IAA) 1 ppm.Methods: Sipahutar pineapple tissue culture work has been done in YAHDI Plant Tissue Culture Laboratory, Medan. The study design used a factorial completely randomized design. The first factor is coconut water with concentration of 0%, 10%, 20%, and 30%, second factor is Benzyl Amino Purine (BAP) with concentration 0 ppm, 2 ppm and 4 ppm, so there were 12 combination with 3 repetition.Result: The results showed that the addition of coconut water on MS base medium with IAA of 1 ppm gave a significant effect on the number of leaf yield and shoot height. The provision of BAP on basic MS medium with IAA of 1 ppm gave a significant effect on the number of shoots produced, the number of leaves produced, the length of the leaves, the width ofleaves and the length of shoots. The interaction of coconut water and BAP addition on MS base medium with IAA of 1 ppm gave significant effect on leaf length, leaf width and shoot length as well.Conclusions: MS + IAA basic media combined with coconut water + BAP could stimulate the growth of Sipahutar pineapple in vitro.

Pineapple plants are usually propagated vegetatively by utilizing the base of the fruit (ratoone), stem buds (sucker), fruit buds (slip) and crown (crown). Propagation of plants by utilizing parts of the plant is less effective in meeting the needs because the planting material produced is small and requires a long time. An alternative to the propagation of pineapple to increase the amount of planting material is by tissue culture techniques. This study aims to obtain the best types of complex organic compounds and the right concentration for the growth of pineapple shoots in vitro. The research was conducted in June 2017 until October 2017 at the Laboratory of Agronomy at the Faculty of Agriculture, University of Bengkulu. Complete randomized design with 3 replications was used to allocate 9 treatments of complex organic compounds. The basic media used are the composition of Murashige and Skoog (MS). Treatment consists of 1) Coconut water 100 ml / l, 2) Coconut water 150 ml / l, 3) Coconut water 200 ml / l, 4) Banana porridge 100 gl / l, 5) Banana pulp 150 g / l, 6) Banana slurry 200 g / l, 7) Bean sprout extract 100 g / l, 8) Bean sprout extract 150 g / l, 9) Bean sprout extract 200 g / l. The results showed that at 16 weeks after MS media planting the addition of 100 g / l banana pulp was able to produce the highest plant height with an average of 3.45 cm. MS media with the addition of 200 ml coconut water resulted in the highest shoot height of 1.5 cm and followed by MS with the addition of 100 g / l media of banana pulp with an average of 0.9 cm. MS media with the addition of 100 ml / l of coconut water is suitable for the growth of pineapple roots with the average value of the number of roots and the highest length of roots. MS media with the addition of bean sprout extract with various concentrations is not able to stimulate shoot growth and has a growth response that tends to be slow. Keywords: Pineapple, In Vitro, Complex Organic Compounds.

Abstract Tissue culture techniques can increase the number of garlic seedlings. The purpose of this research is to determine the effect of NAA and coconut water in increasing the number of garlic seeds. This research used a Completely Randomized Design of two factors. The treatment used is NAA with concentrations of 0 ppm, 0.5 ppm, 1 ppm, 1.5 ppm, and coconut water concentrations of 0%, 10%, 20%. The variables observed were shoot emergence time, root emergence time, number of shoots, number of roots, number of leaves, shoot height, root length, and number of plantlets. The results showed that the addition of coconut water 20% without the addition of NAA in 1 bulb can produce 3.33 planlets and the results of explant propagation in 1 bulb can produce the number of shoots as many as 15.33 shoots. Giving coconut water with concentrations of 10% and 20% can increase the number of leaves, shoot height, and some planlets. The concentration of NAA 0.5 ppm can accelerate the root emergence time on garlic explant.

Valeriana officinalis belongs to the Valerianaceae family, is a well-known herb and medicinal plant. Plant roots are commonly used as medicine by the pharmaceutical industry. Observation on morpho-agronomic characteristics of valerian propagated in vitro is needed to determine their morphological characteristics and production in the field. The study aimed to observe the morphological characteristics of valerian derived from in vitro culture. The experiment was conducted at the Indonesian Spices and Medicinal Crops Research Institute (ISMCRI) Bogor, arranged in a Completely Randomized Design with three replications. Each replication consisted of ten individual plants. Valerian plantlets from in vitro culture were acclimatized in the greenhouse and planted in media composed of a mixture of soil, compost, and husk. Afterward, 3 months old valerian was then transferred into a polybag. The plants were harvested at 9 and 12 months after planting (MAP). Valerian plantlets from in vitro propagation were successfully acclimatized in the greenhouse. Furthermore, the morphological characteristics of the plant were similar to the parents. The yield was significantly higher at 12 MAP than 9 MAP. The tissue culture technique was prospective for valerian propagation to support rapid plant material provision for seeds or raw material for the pharmaceutical industry.

An attempt was done to develop a micropropagation protocol for oak using embryo culture. Oak is considered a hard-to-root woody plant by conventional propagation methods, that’s why using tissue culture techniques is a very suitable alternative method. For oak embryo culture, WPM was used and found to be better than MS medium for embryo germination which gave 66.13%. As well as adding of GA3 to the medium improved the germination rate of embryos (43.25% and 82.25 %). At initiation stage, WPM was used and found to be the best medium by giving the highest number of shoots/ explant which was 1.80, the highest number of leaves (15.17 leaves/ explant) and the longest shoots (1.42 cm) followed by MS medium then GD which gave the lowest parameters which gave 0.98 shoots/ explant, 7.20 leaves/ explant and 1.06 cm shoot length. At shoot multiplication stage, BA was better than Kinetin for multiplication of oak explants. The addition of BA at 3 mg.l-l gave the highest number of shoot and leaves which were 3.33 and 26.11 respectively. The longest shoots were achieved when 4.5 mg.l-l of BA was used. Furthermore, kinetin at 3 and 4.5 mg-l gave the lowest parameters which were 1 cm in length and 1.54 leaves/ explant. For rooting stage, NAA was better than IAA in giving better parameters and rooting percentage. The highest number of roots and rooting percentage were achieved when 1 mg.l-l was added by giving 6 roots/ explant and 100% rooting percentage. While the longest roots were achieved when 0.5 mg.l-l of NAA was used (3.67 cm) followed by 1.5 mg.l-l IAA which gave 3.55 roots/ explant with rooting percentage 90%. The produced plantlets were successfully acclimatized and transferred to the open-air conditions with a rate reached 85%.

Tribulus (Tribulus terrestrisL.) is a medicinal plant with considerable implementation such as aphrodisiac and anti‑inflammation drugs. This research was conducted to study the effectivity of coconut water and IAA (Indole‑3‑acetic acid) application on various concentration as the growth regulator for tribulus growth on thein vitroand its effect toward observation variables. The material used were cotyledon from tribulus embryo and MS (Murashige and Skoog) medium. The research method used a Completely Randomized Design with first factor of coconut water concentration of 0 mL L-1, 50 mL L-1, 100 mL L-1, and 150 mL L-1and IAA concentration of 0 ppm, 0.15 ppm, 0.20 ppm, and 0.25 ppm as the second factor. Observed variables were shoots emergence time, number of shoots, shoot height, number of leaves, roots emergence time, number of roots and regression between observation variables. The result showed that the combination of 150 mL L-1coconut water and 0.25 ppm IAA gave the highest shoot height, roots emergence time, and number of roots. Coconut water treatment on 150 mL L-1concentration alone gave the best result on shoots emergence time, number of shoots, and number of leaves, while IAA concentration of 0.25 ppm independently gave the highest number of leaves. Regression analysis result indicate that the number of roots has a positive correlation with shoots height, number of shoots and number of leaves.

Orchids of the Vanda helvola genus are extensively grown as ornamental plants. The method of propagation of plant seeds in tissue culture is an alternative that may be employed in the provision of a short period. Murashige and Skoog (MS) media and activated charcoal can optimize orchid cultivation utilizing tissue culture techniques. One of the keys to success in tissue culture is the application of natural types of growing regulatory substances (ZPT) at the proper concentration, which can lower the cost of orchid tissue culture.The objective of this study was to see how a combination of Murashige and Skoog (MS) media and activated charcoal influenced the Vanda helvola orchid subculture in vitro. The researchers employed a Complete Randomized Design (RAL) with four treatments and three repeats: negative control (K), ¼ MS (K1), ½ MS (K2), and 100% MS (K3) (K3). The observed parameters include the percentage of live explant, number of roots, and height of orchid plants.   The results showed that the explant had a 100% life percentage of over 21 HST and had no noticeable influence on the number of roots and the plant's height.

Demand for this commodity is quite high, as reflected in the increasing volume of exports each year. Fulfillment of these demands is obtained from the production of cultivation. Generally farmers get natural seaweed seedlings, namely cuttings from existing seaweed. The continuous use of seeds from nature can cause deterioration in the quality and quantity. Handling the problem of quality deterioration from seaweed seeds originating from nature, can be overcome by multiplying the seeds through tissue culture methods in vitro. In term of tissue culture techniques method, the most important thing to note is environmental parameters. An environment parameter that changes suddenly such as salinity can inhibit the growth of seaweed. Thus, this work is attampting the different salinity treatment on seaweed explants K. alvarezii. This study aims at determining (weight) the explants of K. alvarezii with a comparison of the different salinity levels in the in vitro tissue culture method. The method used in this study was a completely randomized design (CRD) with the different salinity treatments namely 30, 31, 32, 33, and 34 ppt. The results showed that the different salinities influenced the growth rate of K. alvarezii seaweed explants with the best explant growth at the salinity of 31 ppt, while the lowest growth value was obtained at 34 ppt
