Catatonia is a psychopathological syndrome displayed as a motor disorder. Catatonia is a sign of many menthal disorders, particularly schizophrenia and depression, with a wide disrtibution in the human population. The GC (“genetic” and “catatonia”) rat strain was obtained from the Wistar rat strain by a long selection (78 generations) for the catatonic type of reaction and is a model of schizophrenic and depressive disorders in humans. It is known that selection for behavior including catatonic reactions results in neuroendocrine, reproductive and morphological changes in animals. However, the influence of selection for a catatonic reaction on the spermatogenic function of testes had not been studied. The aim of this study was to conduct a comparative investigation of sperm quality in rats of the GC and the Wistar strain. The epididymal sperm parameters (sperm count, sperm motility, sperm morphology) were measured, and body, testes and epididymal weight were determined at puberty (50 day of life) and at adulthood (90 day of life). The litter size of the GC and Wistar rats was determined. It was found that adult GC rats had a lower sperm count, sperm motility, testis weight, epydidymal weight and litter size compared to adult Wistar rats. However, at puberty, GC rats had a higher sperm count than the Wistar strain. Interstrain differences in sperm morphology were not found. It has been assumed that the changes of spermatogenic parameters in response to selection for catatonia are caused by changing the ontogenic pattern of testosterone secretion. In conclusion, the hereditary predisposition to catatonic reaction is associated with impaired sperm parameters in adult rats that reduces their chance to reproduction. The GC rat strain can be a perspective model for investigation of the relationship between the hereditary predisposition to catatonia and spermatogenesis

Ruzu bitters black for men (RBBM) is a polyherbal product widely used amongst men in Nigeria to enhance libido, rejuvenate male organs and to manage erectile dysfunctions, prostate anomalies, weak erection, and premature ejaculation. This study was carried out to investigate the toxicological effect from the use of herbal product. Acute toxicity test of RBBM on rats was carried out in two phases; 10 mg/kg, 100 mg/kg and 1000 mg/kg for phase I and 1600mg/kg, 2900 mg/kg and 5000 mg/kg for phase II, were administered respectively. For sub-acute toxicity, two groups of 5 animals each received RBBM (0.87 mg/kg and1.17 mg/kg respectively) and a third group received water orally for 28 days. The study analyzed the median lethal dosage, and sperm morphology, sperm motility, sperm count, sperm viability and histology of the testes as indices for sub-acute toxicity. No death was recorded for the acute and sub-acute studies but there was a moderate physical sign of toxicity. In the sub-acute toxicity study, there was a significant increase (p˂0.05) in testicular weight of Group 1 animals. Also, sperm count, and sperm motility increases significantly (p˂0.05) while there was a decrease in multiple tail sperm across the test groups. RBBM is not toxic to sperm morphology and causes no death at 5000 mg/kg in male albino Wistar rats.

Background: Gentamicin is an aminoglycoside-class antibiotic that can lead to an increase in ROS and decrease antioxidant reserves that cause destruction of testicular cells that are known to inhibit cell division in testicular germ cells and protein synthesis in the testes as well as affect the production of testosterone that affects the quality of spermatogenesis. Vitamin C as an antioxidant can counteract ROS so that testicular cells can be repaired.Methods: This study was an experimental research with post tests only control group design on 25 male rats, 2-3 aged months, weight 160-240 g. Animals were divided into 5 groups. Negative control group was placed in cage without treatment, positive control given induced gentamicin 5mg/kgbb for the first 10 days, and 3 treatment given induced gentamicin 5mg/kgbb for the first 10 days and at days 11-51 were given vitamin C at a dose of 1.6mg; 2.25mg; 4.5mg. On 52 day, all of wistar rats perform terminated for analyzed the testosterone levels were by ELISA, sperm count was calculated with then haemocytometer observed with digital microscope, morphology of spermatozoa was analyzed by eosin staining and was observed with digital microscope.Results: Results of this study showed that the average of testosteron serum on control negatif group of 3,82ng/ml, control positif 3.73ng/ml, treatment 1 is 9.52ng/ml, treatment 2 is 8.29ng/ml, and traetment 3 is 2.28ng/ml. Sperm count on control negatif 41.68 million/ml, control positif 21.06million/ml, treatment 1 is 52.92 million/ml, treatment 2 is 57.12million/ml, treatment 3 is 80.88 million/ml and sperm morphology on control negative  74.00%, control positif 46.96%, treatment 1 is 58.82%, treatment 2 is 68.68%, and treatment 3 is 78.72%.Conclusions: It is concluded that vitamin C has significant effect on increasing testosteron level and sperm quality in gentamicin-induced wistar rats.

Semen hyperviscosity impairs sperm motility and can lead to male infertility. This prospective study aimed at assessing the ability of exogenous DNase in improving sperm quality, taking into consideration that DNase has been found in the seminal plasma of several species and that neutrophils release chromatin in order to trap bacteria. A total of seventy-seven semen samples with high seminal viscosity (HSV) as the study group and sixty-two semen samples with normal seminal viscosity (NSV) as the control group were compared in this analysis. These semen samples were divided into three groups of receiving treatment (a) with DNase I at 37°C for 15 min, (b) by density gradient centrifugation, and (c) with a combination of the above two methods. Following a fifteen-minute treatment of hyperviscous semen, the motility of spermatozoa in 83% of semen samples increased to a statistically significant degree. On the contrary, DNase treatment of semen with normal viscosity had no such effects. The above treatment was also accompanied by a significant increase in the percentage of normal spermatozoa, resulting in a major decrease of the teratozoospermia index. Comparison between semen samples that underwent density gradient centrifugation following DNase I treatment, to those collected after density gradient treatment alone, showed that in the first case the results were more spectacular. The evaluation of each preparation in terms of yield (% total progressively motile sperm count after treatment in relation to the initial total sperm count) revealed that the combined approach resulted in 29.8% vs. 18.5% with density treatment alone (p=0.0121). DNase I treatment results in an improvement of sperm motility and morphology and could be beneficial to men with hyperviscous semen in assisted reproduction protocols.

ObjectiveHigh-fat diet (HFD)-induced metabolic disorders can lead to impaired sperm production. We aim to investigate if HFD-induced gut microbiota dysbiosis can functionally influence spermatogenesis and sperm motility.DesignFaecal microbes derived from the HFD-fed or normal diet (ND)-fed male mice were transplanted to the mice maintained on ND. The gut microbes, sperm count and motility were analysed. Human faecal/semen/blood samples were collected to assess microbiota, sperm quality and endotoxin.ResultsTransplantation of the HFD gut microbes into the ND-maintained (HFD-FMT) mice resulted in a significant decrease in spermatogenesis and sperm motility, whereas similar transplantation with the microbes from the ND-fed mice failed to do so. Analysis of the microbiota showed a profound increase in genus Bacteroides and Prevotella, both of which likely contributed to the metabolic endotoxaemia in the HFD-FMT mice. Interestingly, the gut microbes from clinical subjects revealed a strong negative correlation between the abundance of Bacteroides-Prevotella and sperm motility, and a positive correlation between blood endotoxin and Bacteroides abundance. Transplantation with HFD microbes also led to intestinal infiltration of T cells and macrophages as well as a significant increase of pro-inflammatory cytokines in the epididymis, suggesting that epididymal inflammation have likely contributed to the impairment of sperm motility. RNA-sequencing revealed significant reduction in the expression of those genes involved in gamete meiosis and testicular mitochondrial functions in the HFD-FMT mice.ConclusionWe revealed an intimate linkage between HFD-induced microbiota dysbiosis and defect in spermatogenesis with elevated endotoxin, dysregulation of testicular gene expression and localised epididymal inflammation as the potential causes.Trial registration numberNCT03634644.

We analysed sodium (Na), copper (Cu) and selenium (Se) levels in human semen and glutathione peroxidase activity (GPx) in seminal plasma and examined their relationships with sperm quality. Semen samples were obtained from men (n = 168) undergoing routine infertility evaluation. The study design included two groups based on standard ejaculate parameters: Group I (n = 39) with normal ejaculates (normozoospermia) and Group II (n = 129) with a pathological spermiogram. Se concentration (but not Na or Cu) and GPx activity were significantly higher in normozoospermic males than in those with a pathological spermiogram and also in males with correct sperm motility and normal sperm morphology than in asthenozoospermic and teratozoospermic males. There were significant correlations between sperm motility, Se and GPx, between rapid progressive motility and Cu, between sperm motility and Na, between normal sperm morphology and Se and Cu and between sperm concentration and Cu and GPx. Significant correlations were found between Na and Cu, between Na and Se and between Cu and Se in human semen in relation to alcohol consumption and tobacco use. Na, Cu, Se and GPx are related to sperm characteristics and male fertility and their survey could improve male infertility diagnosis.

Abstract: It is known that Tribulus terrestris can improve sperm quality. One of the Indonesian traditional medicine, jamu, contains this Tribulus terrestris. This study aimed to investigate the effect of jamu containing Tribulus terrestris on sperm quality which consisted of concentration, motility, and morphology of spermatozoa of male wistar rats (Rattus norvegicus). This was an experimental study. Nine male wistar rats were randomly divided into 3 groups of 3 rats each: Group P0 (control), group P1 (given 5 mg of Tribulus terrestris), and group P2 (given 10 mg of Tribulus terrestris). This study was conducted for 52 days. The results showed that the spermatozoa concentration of P1 was 49x106 sperm/ml suspension and of P2 53x106 sperm/ml suspension; both were lower than of P0 (59x106 sperm/ml suspension). The sperm motility category (A+B) of P1 was 33% and of P2 (37%); both were lower than of P0 (45%). The motility category A was not found in P1 and P2. The percentage of spermatozoa with normal morphology of P1 was 45% and the abnormal ones was 55%, meanwhile in P2 there was 49% of normal sperm morphology and 51% of abnormal morphology. Conclusion: Jamu containing Tribulus terrestris had no effect in improvement of the sperm quality.Keywords: jamu, Tribulus terrestris, sperm qualityAbstrak: Tribulus terrestris dikenal sebagai bahan yang dapat memperbaiki kualitas sperma. Penelitian ini menggunakan metode eksperimental. Sampel 9 ekor tikus wistar jantan (Rattus norvegicus) dibagi menjadi 3 kelompok yakni, kelompok P0 (kontrol); kelompok P1 (pemberian Tribulus terrestris 5 mg); dan kelompok P2 (pemberian Tribulus terrestris 10 mg). Penelitian dilakukan selama 52 hari. Hasil penelitian menunjukkan bahwa konsentrasi spermatozoa P1 49x106 sperma/ml suspensi dan P2 53x106 sperma/ml suspensi lebih rendah dibandingkan P0 59x106 sperma/ml suspensi. Motilitas spermatozoa kategori A+B pada P1 (33%) dan P2 (37%) lebih rendah dibandingkan pada P0 (45%). Tidak ditemukan motilitas kategori A pada P1 dan P2. Morfologi spermatozoa normal pada P1 sebesar 45% dan abnormal 55%. P2 memiliki morfologi spermatozoa normal sebesar 49% dan abnormal 51%. Simpulan: Pemberian jamu dengan Tribulus terrestris tidak meningkatkan kualitas sperma yang mencakup konsentrasi, motilitas, dan morfologi spermatozoa.Kata kunci: jamu, Tribulus terrestris, kualitas sperma

Background Infertility remains a highly prevalent global condition in the second decade of the new millennium. Reproductive hormones determine sperm quality as they initiate and maintain spermatogenesis. Hormonal imbalance can cause abnormal sperm quality that can be treated by hormonal replacement therapy. Objective To assess the relationship between sperm quality and male reproductive hormones among male partners with fertility complications attending CHUB. Methods The study was a descriptive cross-sectional, and a convenient sampling strategy was used to recruit subjects at CHUB. Sixty-two male subjects with fertility complications provided both semen and blood sample to analyze sperm quality and reproductive hormones. Descriptive statistics were used to analyze data. Results Both FSH and LH showed a strong negative correlation with sperm count which is more profound with FSH (r= -0.722) than LH (r= -0.545). Testosterone showed a strong positive correlation with sperm count (r= 0.712). FSH and LH showed a negative correlation with sperm motility which is more profound in FSH (r= -0.312) than LH (r= -0.302). Testosterone also showed a positive correlation with sperm motility (r= 0.360). Conclusion Our study found a correlation between sperm quality and male reproductive hormones. We further suggest other studies to investigate predictive power of male reproductive hormones. Rwanda J Med Health Sci 2020;3(3):315-328

Pyrethroids may be related to male reproductive system damage. However, the results of many previous studies are contradictory and uncertain. Therefore, a systematic review and a meta-analysis were performed to assess the relationship between pyrethroid exposure and male reproductive system damage. A total of 72 articles were identified, among which 57 were selected for meta-analysis, and 15 were selected for qualitative analysis. Pyrethroid exposure affected sperm count (SMD= -2.0424; 95% CI, -2.4699 to -1.6149), sperm motility (SMD=-3.606; 95% CI, -4.5172 to -2.6948), sperm morphology (SMD=2.686; 95% CI, 1.9744 to 3.3976), testis weight (SMD=-1.1591; 95% CI, -1.6145 to -0.7038), epididymal weight (SMD=-1.1576; 95% CI, -1.7455 to -0.5697), and serum testosterone level (SMD=-1.9194; 95% CI, -2.4589 to -1.3798) in the studies of rats. We found that gestational and lactational exposure to pyrethroids can reduce sperm count (SMD=1.8469; 95% CI, -2.9010 to -0.7927), sperm motility (SMD=-2.7151; 95% CI, -3.9574 to -1.4728), testis weight (SMD=-1.4361; 95% CI, -1.8873 to -0.9848), and epididymal weight (SMD=-0.6639; 95% CI, -0.9544 to -0.3733) of F1 offspring. Exposure to pyrethroids can increase malondialdehyde (SMD=3.3451; 95% CI 1.9914 to 4.6988) oxide in testes and can reduce the activities of glutathione (SMD=-2.075; 95% CI -3.0651 to -1.0848), superoxide dismutase (SMD=-2.4856; 95% CI -3.9612 to -1.0100), and catalase (SMD=-2.7564; 95% CI -3.9788 to -1.5340). Pyrethroid exposure and oxidative stress could damage male sperm quality. Gestational and lactational pyrethroid exposure affects the reproductive system of F1 offspring.

Introduction. Male infertility is known to contribute about half of all infertility cases. In Ghana, the prevalence of male infertility is higher (15.8%) than in females (11.8%). Sperm quality is associated with the likelihood of pregnancy and known to be the cause of male fertility problems 90% of the time. Exposure to certain environmental factors reduces semen quality in men. The study examined the effects of environmental and lifestyle factors on semen quality in Ghanaian men. Materials and Methods. This was a cross-sectional study involving 80 apparent healthy adult males in their reproductive age. Participants were males referred to the laboratory (Immunology Unit of the Korle-Bu Teaching Hospital) for semen analysis test and/or culture and sensitivity. Participants were made to fill out a questionnaire which entailed selected environmental factors (accidents or trauma, exposure to chemicals, radiation, and heat) and lifestyle habits (including alcohol consumption, smoking, and whether participants sat more or less than 4 hours per day). Semen samples were then collected by masturbation into sterile containers and analysed in accordance with WHO guidance for semen analysis within 60 minutes after ejaculation and collection. Results. About 69% of participants had semen pH within the normal range compared to 15% whose pH were lower than 7.2. There was a significantly high number of immotile sperm cells ( p value = 0.017) in participants who sat for more than 4 hours as compared to those that sat for less than 4 hours in a day. Active sperm motility and viability showed significant increase ( p value = 0.002 and 0.009, respectively) in participants who kept their cell phones in their side pockets. Smoking produced a twofold decrease in sperm count as smokers had a significantly lower sperm count ( 12.28 ± 10.95 × 10 6 /ml) compared to the smoke-free ( 23.85 ± 22.14 × 10 6 /ml). For exposure to STDs, no significant differences were recorded among study groups concerning semen quality. Conclusion. Sperm quality in Ghanaian men is associated with lifestyle habits. Smoking and sitting for long hours influenced sperm motility and count, respectively. Knowledge of the factors that influence sperm quality in this geographical region can contribute to informed decisions on effective management of infertility in Ghanaian men.

Feeding trial was carried out to evaluate sperm quality of Clarias anguillaris broodstock fed varying inclusion levels of goron tula, Azanza garckeana pulp meal. The fish (mean weight 500g) were randomly distributed in concrete tanks (2 x 2 x 1.2m) at nine fish/tank in triplicates. Five isonitrogenous diets (40% CP) were formulated with the pulp meal incorporated at the rate of 0, 5, 10, 15, and 20% of the diet. Fish were fed at 3% of body weight for 56days. At the end of the feeding trial, results showed that dietary A. garckeana significantly (p<0.05) improved the milt quality of C. anguillaris. The broodfish fed 20% A. garckeana inclusion level had the highest (p<0.05) milt volume, sperm motility duration, testes lobe lengths, and whole sperm count while the least values of these variables were observed in group fed with the control diet (0% inclusion level). The findings conclusively revealed that increase in the level of A. garckeana pulp meal in the diet resulted into increase in sperm quality of C. anguillaris. Thus, the pulp meal can be freely utilized in catfish production in terms of sperm quality for mass fingerling production.
