The study discusses the effect on the functional state of the whole blood cells of low-intensity microwave radiation. On the model of intercellular interactions in the conditions of spontaneous cell activity and by stimulating cells complex mitogen, the influence of microwave radiation 1000 MHz on production by whole blood cells interleukins: IL-1β, 2, 6, 8, 10, 12, 13, 17A, TNF-alpha, IFN-g, RAIL-1, and soluble forms of toll-like receptor 1, 2, 4, 6 types was studied. The research results show that a single 45 minutes of exposure of microwave radiation of 0.05 PPM microwatt/cm2 has the ability to enhance mitogen-stimulated production of RAIL-1 by 38,7% (p=0.03), to reduce the induced production of IL-1β by 26,3% (p=0,037), IL-8 by 56,2% (p=0.022) and to increase oppressed segment complex mitogen production of IL-10 by 27,8% (p=0,041). The microwave radiation increased spontaneous and mitogen-stimulated expression of TLR, especially TLR1 during the initial low level.

Back in the 60s-70s of the 20th century, in the experimental studies of Novosibirsk scientists, carried out under the leadership of Academician V.P. Kaznacheev, the role of super weak physical fields in the development of the cytopathological action of biological objects of viral nature, agents of chemical and physical nature was established. In recent years, in experimental and clinical medicine, along with the use of low-intensity extremely high-frequency millimeter radiation (less than 10 mW / cm), microwave generators of super low intensity of the centimeter wavelength range have been successfully used at a power level not exceeding 10 μW/cm. Such super low-power microwave radiation, is realized at the frequency of vibrations of first-order water clusters, contributes to the normalization of metabolic processes. The present study was work out in accordance with the plans of the problem commission on "chronobiology and chronomedicine" of the Russian Academy of Sciences. On the basis of interdisciplinary collaboration, a virtual team of researchers working in different universities in the country was formed and included mathematicians, systems programmers, engineers and medical researchers. At the same time, to implement the goals and objectives of the study, the so-called linear model of the flow of information from the scientific laboratory to clinical practice (from bench to bedside) was used. In this connection, we have implemented a project aimed at developing a modular-type technical system designed to generate microwave radiation with desired properties. In the microcontroller of the microwave generator, a digital analogue of signals reflecting the neurodynamic processes of the brain was programmed. This article shows the electrophysiological efficiency of super-low-intensity radiation in the centimeter range of wavelengths modulated by low-frequency signals similar to the EEG rhythm. In experimental studies in patients with diabetes mellitus II with concomitant hypertension, the transformation of the pathological neurodynamic activity of the brain with the subsequent modification of the patient's functional state has been shown. The results of ineffective influence, effective influence and successful influence with changes in the dynamics of the level of plasticity of neurodynamic activity of the brain are considered

Abstract Objectives Hyperuricemia is a metabolic condition central to gout pathogenesis. Urate exposure primes human monocytes towards a higher capacity to produce and release IL-1β. In this study, we assessed the epigenetic processes associated to urate-mediated hyper-responsiveness. Methods Freshly isolated human peripheral blood mononuclear cells or enriched monocytes were pre-treated with solubilized urate and stimulated with LPS with or without monosodium urate (MSU) crystals. Cytokine production was determined by ELISA. Histone epigenetic marks were assessed by sequencing immunoprecipitated chromatin. Mice were injected intraarticularly with MSU crystals and palmitate after inhibition of uricase and urate administration in the presence or absence of methylthioadenosine. DNA methylation was assessed by methylation array in whole blood of 76 participants with normouricemia or hyperuricemia. Results High concentrations of urate enhanced the inflammatory response in vitro in human cells and in vivo in mice, and broad-spectrum methylation inhibitors reversed this effect. Assessment of histone 3 lysine 4 trimethylation (H3K4me3) and histone 3 lysine 27 acetylation (H3K27ac) revealed differences in urate-primed monocytes compared to controls. Differentially methylated regions (e.g. HLA-G, IFITM3, PRKAB2) were found in people with hyperuricemia compared to normouricemia in genes relevant for inflammatory cytokine signaling. Conclusion Urate alters the epigenetic landscape in selected human monocytes or whole blood of people with hyperuricemia compared to normouricemia. Both histone modifications and DNA methylation show differences depending on urate exposure. Subject to replication and validation, epigenetic changes in myeloid cells may be a therapeutic target in gout.
