Dependence of the content of individual molecules in agranucocytes of whole blood at coronary heart disease from the level of phosphorylation of protein kinase r 38 in terms of low intensity microwave radiation

Molecular indicators reflecting the states of stress-limiting systems of mononuclear leucocytes in the blood, as well as the effects of low-intensive microwave radiation in patients with coronary artery disease were studied. The work it was evaluated the content in mononuclear leucocytes whole blood of components PI3P/AKT/mTOR/p70S6K1 of signaling pathway, heat shock proteins (HSP27, HSP70, HSP90), the concentra-tion of antioxidants and peroxides depending on the level of phosphorylation of the terminal protein kinase MAPK/SAPK of signaling pathway – p38. The results of the study. It was revealed the dependence of a level of studied factors from the degree of phosphorylation p38 in patients with coronary heart disease. It was defined the 38 p sensitivity to the effects of low-intensity microwave radiation, it is manifested by increased level of phosphorylation in the irradiated cul-tures. This study revealed the sensitivity to low-intensity microwave irradiation of content in the mononuclear cells phosphorylated forms of the protein kinases AMPK, AKT1, p70S6K1, as well as the antioxidant status and protein of p53-dependent initial content in the cell phosphorylated form p38. It was shown a possibility of microwave radiation to reduce the content in the cells of the protein kinase and p70 ACT more pronounced at high levels of phosphorylation p38.

The concentration of cytokines of the family IL-10 (IL-10, IL-22, IL-24), interferon-gamma, soluble forms of co-stimulatory molecules CD28, CD80, CD152, as well as NO was investigated in patients with coronary heart disease in interstitial fluid. In addition, the level of caspase-1, inducible and endothelial forms of NO synthasis (eNOS, iNOS), protein kinase AKT1 and AMPK and the total antioxidant activity from the cell culture supernantat were evaluatedin the agranulocytic patients. In the current study (in vitro) the impact on production of these mediators of low-intensity microwave radiation frequency of 1000 MHz, generated by an apparatus of low-intensity physiotherapy "Aquaton" was evaluated. Material of this study was whole venous blood of patients with angina pectoris and acute coronary syndrome. The analysis of the results revealed a reduced level of NO, IL-24, as well as increased production of IL-10, IL-22 and the concentration of soluble forms of CD28, CD80, CD152 in patients with coronary artery disease in the interstitial fluid in comparison with practically healthy persons. At a single irradiation of cell cultures whole blood by low-intensity microwave radiation frequency of 1000 MHz in patients with angina pectoris and acute coronary syndrome there were an increasing of NO production by 20,6 and 25,5‰, IL-24 by 18,6 and 33,6‰, and a reduction in intracellular caspase-1 by 32,5 and 25,8‰, as well as increasing of the levels of IL-10 by 3,5 and 3,1‰, IL-22 by 28,7 and 26,5‰ and reduction of the levels of CD28 by 3,6 and 3,9‰, CD80 6,8 and 5,7‰, CD152 by 9,4 and 11,3‰, respectively. In addition, in terms of irradiation there is an increase in interstitial fluid levels of antioxidants by 37,1 and 49,2‰.

Abstract ObjectiveOur study aimed to investigate the potential mechanisms of the herb pair Zhizi-Danshen (ZD) for coronary heart disease (CHD) using network pharmacological data mining technology.MethodsThe Traditional Chinese Medicine System Pharmacology (TCMSP) database was used to collect the active ingredients of ZD and predict ZD-related target proteins. Afterwards, we identified CHD-related targets from DisGeNET database, NCBI gene database, and TTD database. The common targets both from ZD and CHD were screened by Venny2.1, which were then imported into the String database for protein-protein interaction (PPI) analysis. Finally, the GO and KEGG enrichment analysis were performed by R software, and the network construction was established using Cytoscape3.7.2.ResultsWe obtained 199 possible targets from 62 candidate ingredients of ZD and 1033 CHD-ralated targets, with 83 overlapping common target genes. Then, 11 core targets were acquired from PPI network analysis. Further, GO analysis showed that these common targets mainly influenced receptor ligand activity,cytokine activity,cytokine receptor binding,steroid hormone receptor activity, and peptide binding. KEGG pathway analysis indicated that ZD affected CHD through seven important pathways linked to vascular endothelial function regulation (fluid shear stress and atherosclerosis,AGE-RAGE signaling pathway in diabetic complications, HIF-1 signaling pathway), imflammatory effects (IL-17 signaling pathway, TNF signaling pathway,Toll-like receptor signaling pathway),and hormone regulation (relaxin signaling pathway). ConclusionsThis study revealed the potential pharmacological mechanisms of ZD against CHD, which were mainly associated with regulation of vascular endothelial function and inflammatory effects, promotion of vasodilatation, and prevention of cardiac fibrosis. Moreover, it provided a novel conception for the development of alternative therapies on CHD.

Aim: We aimed to identify the expression profile and role of circular RNAs (circRNAs) in coronary heart disease (CHD). Materials & methods: We performed sequence analysis of circRNAs in peripheral blood mononuclear cells of 70 CHD patients and 30 controls. Eight selected circRNAs were validated using quantitative real-time polymerase chain reaction (qRT-PCR) in human atherosclerotic coronary arteries. Results: In total, 2283 downregulated and 85 upregulated circRNAs were identified in CHD. Parental genes of top 100 dysregulated-circRNAs are related to metabolism and protein modification, and 12 circRNAs might upregulate their CHD-related parental genes through miRNA sponges. Of the eight circRNAs validated in atherosclerotic coronary arteries by qRT-PCR, six were consistent with sequencing results of peripheral blood mononuclear cells. Conclusion: As potential ceRNAs, dysregulated circRNAs may be involved in CHD pathophysiology.

Abstract Population-based studies suggest moderate and regular consumption of alcoholic beverages and especially of red wine to reduce morbidity and mortality from coronary heart disease. These beverages may interfere with immune activation cascades crucial in the pathogenesis of coronary heart diseases. Neopterin concentrations in human body tluids were found to increase in the course of coronary heart disease indicating the activity of the atherogenetic process. In this study, human peripheral blood mononuclear cells (PBMC) stimulated with mitogens phytohaemagglutinin and concanavalin A were exposed to red and white wines, to ethanol and to grape juice as a non-alcoholic control in vitro. Neopterin production and tryptophan degradation were measured in supcrnalants. Both biochemical effects are induced by Th 1-type cytokine interferon-y and allow monitoring of immune actnation. In stimulated PBMC increased production of neopterin and degradation of tryptophan was observed Red and white wines, as well as grape juice inhibited these stimulation-induced effects, higher concentrations being more cffective. Ethanol had comparably small if any effect, Red and white wines as well as grape juice down regulate cytokine-mediated effects in PBMC. Most likely, antioxidant ingredients of wine and grape juice such as resveratrol are capable of interfering with immunologic pathways which appear to be of relevance, e g.. in the pathogenesis of cardiovascular disease.

In this paper, by examining the toxicity of nano-silica to coronary heart disease cells, we explored the apoptosis of rat myocardial cells induced by nano-silica, and explored the effect of apoptosis on cells during the process of myocardial cytotoxicity induced by nano-silica. This article selects rat cardiomyocytes as the research object and conducts a group control experiment. A control group is set up with cells that are not stained with nano-silica. Different concentrations of nanosilica suspensions are applied to rat cells and detected by CCK-8 method. Cell survival rate after exposure to different concentrations of cells is used to determine the most stable exposure time and concentration. We used flow cytometry to detect intracellular reactive oxygen species and apoptotic rates, and used Western Blot to detect the expression of proteins that affect apoptosis. Finally, we investigated the effect of the Wnt signaling pathway on coronary heart disease. The Wnt signaling pathway regulates the development of the heart and blood vessels. In the treatment of cardiovascular disease, this pathway will be activated again to play a regulatory role. We conclude that nano-silica can induce cytotoxicity in rat myocardial cells through the Wnt-1 pathway, and nanosilica can induce myocardial cell apoptosis through the Wnt-1 pathway.

The study discusses the effect on the functional state of the whole blood cells of low-intensity microwave radiation. On the model of intercellular interactions in the conditions of spontaneous cell activity and by stimulating cells complex mitogen, the influence of microwave radiation 1000 MHz on production by whole blood cells interleukins: IL-1β, 2, 6, 8, 10, 12, 13, 17A, TNF-alpha, IFN-g, RAIL-1, and soluble forms of toll-like receptor 1, 2, 4, 6 types was studied. The research results show that a single 45 minutes of exposure of microwave radiation of 0.05 PPM microwatt/cm2 has the ability to enhance mitogen-stimulated production of RAIL-1 by 38,7% (p=0.03), to reduce the induced production of IL-1β by 26,3% (p=0,037), IL-8 by 56,2% (p=0.022) and to increase oppressed segment complex mitogen production of IL-10 by 27,8% (p=0,041). The microwave radiation increased spontaneous and mitogen-stimulated expression of TLR, especially TLR1 during the initial low level.
