Streptococcus mutanspredominantly creates an acidic environment in an oral cavity. This results in dental demineralization and carious lesions. The probiotics are beneficial microorganisms that modulate the bacterial balance in the digestive system. Prebiotics are defined as nondigestible oligosaccharides that are utilized for the selective stimulation of the beneficial microorganisms. The objective of this study was to evaluate the efficacy of the prebiotics, galactooligosaccharides (GOS) and fructooligosaccharides (FOS), for enhancing the probioticLactobacillus acidophilusATCC 4356, for inhibitingStreptococcus mutans(A32-2) for the prevention of dental caries. The growth rate of theS. mutanssignificantly decreased when cocultured withL. acidophilusin the GOS-supplemented medium at 3%, 4%, and 5%. In the FOS-supplemented medium, the growth rate ofS. mutanssignificantly decreased in all concentrations when cocultured withL. acidophilus. There was no significant difference in the growth rate ofL. acidophilusin all concentrations of either GOS or FOS. It can be concluded that the growth rate ofS. mutanswas significantly retarded when cocultured withL. acidophilusand the proper concentration of prebiotics. These prebiotics have potential for a clinical application to activate the function of the naturally intraoralL. acidophilusto inhibitS. mutans.

ABSTRACT The in vitro activities of human β-defensin 3 (hBD-3) alone or combined with lysozyme, metronidazole, amoxicillin, and chlorhexidine were investigated with the oral bacteria Streptococcus mutans, Streptococcus sanguinis, Streptococcus sobrinus, Lactobacillus acidophilus, Actinobacillus actinomycetemcomitans, and Porphyromonas gingivalis. hBD-3 showed bactericidal activity against all of the bacterial species tested. The bactericidal effect was enhanced when the peptide was used in combination with the antimicrobial agents mentioned above.

Pembuatan pasta gigi ini dilatar belakangi oleh keinginan peneliti agar memudahkan masyarakat menggunakan bahan alami katekin teh hijau yang hasilnya langsung di uji efektifitasnya terhadap daya hambat bakteri kariogenik. Penelitian ini bertujuan untuk melakukan pembuatan prodak pasta gigi katekin teh hijau dengan formula yang sesuai dengan hasil uji dan melakukan pengujian daya hambat pasta gigi katekin teh hijau terhadap bakteri streptococcus mutans dan lactobacillus acidophilus. Pembuatan pasta gigi katekin teh hijau didahului dengan pembuatan ekstrak katekin dan uji konsentrasi hambat minimal (KHM) dan uji konsentrasi bunuh minimal (KBH) menggunakan metode dilusi cair.Peracikan formula dan pembuatan pasta gigi katekin teh hijau dilakukan sesuai hasil uji KHM (MIC). Pasta gigi katekin teh hijau ini dilakukan pengukuran kadar total flavonoid dan total polifenol, selanjutnya pengukuran uji daya hambat pasta gigi katekin teh hijau dilakukan dengan metode difusi menggunakan well (sumuran) sebagai reservoar sampel uji terhadap bakteri streptococus mutans dan lactobacilus acidophilus. Hasil diperoleh prodak pasta gigi katekin teh hijau dengan kandungan kadar flavonoid 77% dan kadar polifenol 41%, sedangkan hasil uji diameter hambatan pasta gigi katekin teh hijau terhadap bakteri streptococcus mutans 17,2 mm dan bakteri lactobacillus acidophilus 19,6 mm. Hasil ini menunjukkan bahwa prodak katekin teh hijau ini sangat efektif digunakan sehari-hari sebagai perawatan gigi dan juga sebagai antibakteri kariogenik. Making Green Tea Catechin Toothpaste and Inhibition Test Against Streptococcus Mutans and Acidophilus Lactobaccilus. The manufacture of this toothpaste is motivated by the desire of researchers to facilitate the public to use natural ingredients of green tea catechins whose result effectiveness against cariogenic bacteria was directly tested inhibition. This study is to find the right formula composition for greentea catechins toothpaste based on the result of the inhibition properties of greentea cathecins against tostreptococcus mutans dan lactobacillus acidophilus. The manufacture of greentea catechins toothpaste was after making catechins extracts and testing minimal inhibition concentration (MIC) and minimal bactericidal concentration (MBC) using liquid dilution method. The toothpaste was then made according to the result of MIC. The toothpaste’s flavonoid and polyphenol total contain was measured; then the inhibition property of the toothpaste to was measured using diffusion method with wells as the sample reservoar for streptococus mutans dan lactobacilus acidophilus. It is found that the toothpaste contains 77% flavonoid and 41% polyphenol, and the diameter of the inhibition property of green tea catechyn is 17.2 mm for streptococus mutans and 19.6 mm for lactobacilus acidophilus. This result shows that greentea catechyn toothpaste is very effective for daily oral care which is antibacterial.

Introducción y objetivo: La incorporación de propóleos en los dentífricos tiene como objetivo ayudar de manera más efectiva al control y la prevención de patologías orales a través de la eliminación de los patógenos presentes en la biopelícula. Sin embargo, se sabe poco sobre la eficacia antimicrobiana de diferentes productos en el mercado de microorganismos para estas patologías. El objetivo de este estudio fue investigar la acción antimicrobiana de tres dentífricos que contienen propóleos sobre los patógenos orales. Material y métodos: Se utilizó el método de difusión en agar para analizar tres dentífricos basados en propóleos, que incluyen: Noplak Max®, Protta® y Forever Bright®. Se utilizó un dentífrico sin propóleos (Malvatrikids ®) como control negativo. Los controles positivos fueron 0,2% de clorhexidina diluida adicionalmente al 30% para igualar la concentración de clorhexidina de uno de los dentífricos evaluados, y el extracto de propóleos (Apis Flora®) al 11%. Para la determinación de la actividad antimicrobiana se utilizaron las cepas de Fusobacterium nucleatum y Aggregatibacter actinomycetemcomitans, Enterococcus faecalis, Streptococcus mutans, Lactobacillus acidophilus y Candida albicans. Resultados: De los dentífricos probados, Protta® y Forever Bright® mostraron acción inhibitoria contra S. mutans, E. faecalis y microorganismos de C. albicans. El dentífrico Noplak® mostró baja actividad antimicrobiana, limitándose a S. mutans y E. faecalis. Cuando hubo un efecto antimicrobiano, los diámetros de los halos de inhibición del crecimiento variaron de 9mm a 28,83mm. Conclusión: El uso de un dentífrico que contiene propóleos para su uso eventual como complemento terapéutico en odontología está justificado, considerando las actividades farmacológicas.

Antibacterial compounds are generally produced from many herbal ingredients, but antibacterials may be produced from soil microbes, one of which is Actinomycetes. Actinomycetes are a soil bacterium capable of producing secondary metabolites in the form of antibacterial compounds. The antibacterial compound has the potential as an alternative antibacterial agent against oral microbes. One of the oral microbes that have a major role in the occurrence of caries is Streptococcus mutans and Lactobacillus acidophilus. This study aimed to analyze the antibacterial activity of Actinomycetes against the growth of S. mutans and L. acidophilus Actinomycetes extracts at the concentration of 0.625%, 1.25%, 2.5%, 5%, 10%, 20%, and 40% were respectively exerted as the treatment groups while 0.12% Chlorhexidine Gluconate (CHX) and aquadest as control. The measurement of antibacterial activity is done by looking at MIC and MBC. One Way ANOVA and Post Hoc Dunnets T3 test MIC and MBC values show p=0,000 (p <0.05). Actinomycetes extract at the concentration of 0.625% was able to inhibit the growth of S. mutans and L. acidophilus which was determined as the MIC and at the concentration of 40% as MBC of S.mutans and the concentration of 10% as MBC of L. acidophilus. There was an antibacterial activity of Actinomycetes to the growth of S. mutans and L.acidophilus.
