Implementation of BioMEMS for Determining Mechanical Properties of Biological Cells

ABSTRACTThis paper describes the implementation of a custom-made bio-microelectromechanical system for determining mechanical properties of biological cells, which is used for the measurement of mechanical properties of fibroblasts. Our system consists of several subcomponents: (a) actuator which deforms the cell in pre-determined, step-wise fashion, (b) force sensor that measures force applied onto the cell, (c) set of dielectrophoretic (DEP) electrodes for positioning cells in the desired position, (d) temperature sensors and (e) heater. Preliminary results of the mechanical properties of NIH3T3 cells have been determined using this tool and our cell compression techniques.

Appropriate mechanical properties and fast endothelialization of synthetic grafts are key to ensure long-term functionality of implants. We used a newly developed biostable polyurethane elastomer (TPCU) to engineer electrospun vascular scaffolds with promising mechanical properties (E-modulus: 4.8 ± 0.6 MPa, burst pressure: 3326 ± 78 mmHg), which were biofunctionalized with fibronectin (FN) and decorin (DCN). Neither uncoated nor biofunctionalized TPCU scaffolds induced major adverse immune responses except for minor signs of polymorph nuclear cell activation. The in vivo endothelial progenitor cell homing potential of the biofunctionalized scaffolds was simulated in vitro by attracting endothelial colony-forming cells (ECFCs). Although DCN coating did attract ECFCs in combination with FN (FN + DCN), DCN-coated TPCU scaffolds showed a cell-repellent effect in the absence of FN. In a tissue-engineering approach, the electrospun and biofunctionalized tubular grafts were cultured with primary-isolated vascular endothelial cells in a custom-made bioreactor under dynamic conditions with the aim to engineer an advanced therapy medicinal product. Both FN and FN + DCN functionalization supported the formation of a confluent and functional endothelial layer.

Textile temperature sensors are highly in demanded keep a real-time and accurate track of human body temperature for identification of healthy conditions or clinical diagnosis. Among various materials for textile temperature sensors, temperature-sensitive metal fibers have highest precision. However, those metal fibers are mechanically too weak, and break constantly during the weaving process. To enhance the mechanical strength of the metal fibers, this paper proposes to make wrapped metal fibers using wrapping technology, and characterize the effect of wrapped metal yarns on both mechanical properties and sensing behaviors. The wrapped yarns were woven into fabrics, forming the fabric temperature sensors. Results show that strength and maximum strain of the wrapped yarns are 2.69 and 1.82 times of pure Pt fibers. The response time of fabric temperature sensors using wrapped yarns was observed as 0.78 s and 1.1 s longer compared to that using Pt fibers when front and back sides contacted heat source, respectively. It is recommended that the wrapping method should be implemented for the protection of Pt fibers in fabric temperature sensors.

Nano-micro grippers are able to pick-transport-place the micro or nanometer–sized materials, such as manipulation of biological cells or DNA molecules in a liquid medium. This paper proposes a novel monolithic nano-micro gripper structure with two axis piezoresistive force sensor which its resolution is under nanoNewton. The results of the study have been obtained by the simulation of the proposed gripper structure in Matlab software. Motion of the gripper arm is produced by a voice coil actuator. The behavior of the cell has been derived using the assumptions in the literatures. Moreover, two simple PID controllers, one for control of the gripper motion and another for control of the force during manipulation of a biologic cell, have been implemented. Although the proposed gripper has not been fabricated, since the geometrical dimensions of the proposed gripper is the same as previously developed electrothermally actuated micro-nano gripper, the results of force control have been also compared with it. The simulated results with the very simple PID force controller which has a more rapid response than previously developed electrothermally actuated micro-nano gripper show that the designed gripper has the potential to be considered and fabricated for manipulation of biological cells in the future.

AbstractIn this work, we have investigated the structural and mechanical properties of a new class of soft and superelastic materials, called schwarzynes. These materials are obtained by inserting sp carbon atoms (acetylenic groups) into the schwarzite framework. Using fully atomistic molecular dynamics simulations with the AIREBO force field, our results show that schwarzynes are stable materials up to high temperatures (1000K). Schwarzynes exhibit a very wide elastic regime, some of them up to 70% strain without structural fractures. Our preliminary results show that the elastic properties can be easily engineered by tuning the number of acetylenic groups and the crystallographic directions where they are inserted.
