We studied the intensity of lipid peroxidation (LPO) and the status of antioxidant system in blood of 6-60 months ostriches. We proved that specific functioning of the antioxidant system and the accumulation of lipid peroxidation intermediates characterized each period of ostrich life cycle. Thus, the period of puberty was accompanied by intensification of lipid peroxidation in ostrich blood serum and this was confirmed by a significant increase in the amount of TBA-reactive substances. This could be the consequence of the insufficient activity of enzymes of the antioxidant system (catalase, glutathione-S-transferase, glutathione reductase). The egg-laying peak was characterized by the increased body metabolism, which caused the activation of free-radical oxidation. The concentration of lipid hydroperoxides and diene conjugates increased and the TBA-reactive substances did not change significantly. Antioxidant protection of the blood during this period was mainly caused by the significant concentration of ceruloplasmin and reduced glutathione due to the increased activity of glutathione reductase. We supposed that the period of puberty and intensive period of egg laying in ostriches should be referred as the periods of stress of metabolic processes with increased intensity of lipid oxidation, associated with the certain changes in ostrich physiological and functional state.

Aim: To evaluate the intensity of lipid peroxidation and the activity of catalase in patients with laryngeal cancer. Materials and Methods: Samples of blood serum and erythrocytes from 40 patients with laryngeal cancer of stages II and III and 20 healthy volunteers were investigated. Results: The level of TBA-positive products and diene conjugates in blood serum samples of stage II and III patients with laryngeal cancer was increased than that of control. In the same time catalase activity was significantly reduced. In the erythrocytes of patients in both groups the level of diene conjugates and catalase activity was higher than that of control. The level of TBA-positive products increased only in patients with III stage disease. Our results indicate the intensification of free-radical oxidation against the background of the functional stress of the antioxidant defense system in patients with laryngeal cancer, as well as the violation of the structural and functional state of erythrocyte plasma membranes, which is manifested in the activation of lipid peroxidation. This indicates about the formation of a complex of disorders, which can play a significant role in the development and deepening of the pathological process.

Abstract Goat husbandry is a relevant and significant research topic. The goat age is possible to reach 15–18 years and it is important to study not only performance processes, but also to evaluate age-related changes. The purpose of this study was to evaluate antioxidant system characteristics in Saanen goats depending on age. The experiment was conducted in the north-western region of Russian Federation, the Leningrad Region. The five experimental groups included 30 Saanen goats each, selected using matched pairs method. 1st group included 1 year old goats, 2nd group included 2-year-old goats, 3rd group included 3-year-old goats, 4th group included 4-year-old goats and 5th group included 5-year-old animals. The blood samples were taken once –during the summer season. The blood levels of lipid peroxygenation markers (malondialdehyde, dienketone and conjugated dienes) and superoxide dismutase and catalase activity were assessed by standard methods. Results presented as mean±standard error of the mean. Student’s t-test was used after proving normal distribution. Level of significance is labeled as P < 0.05. The results are displayed in table 1 and 2. After analyzing the received data of antioxidant system characteristics there were revealed high intensity of free-radical oxidation in aged 4–5-year-old goats. This is explained by oxidative stress development. Noticeably that anti-oxidizing enzymes activity was reduced by the age of 5 years – the possible reason for lacking compensation of developed oxidative stress. By the age of 4 activity of anti-oxidizing enzymes was elevated marking the compensation stage of oxidative stress, this elevation is statistically significant. Consequently, these data allow taking into consideration exogenous anti-oxidizing agents administration in 5 years and older goats in order to reduce oxidative stress.

Ivanusa

TernopilStateMedicalUniversity

Activation of lipid peroxidation of animals at poisoning by acetaminophen with prolonged usage of estrogen and progestin are investigated. This is evidenced by increased concentration of reactive oxygen species by mononuclear leukocytes, the content of lipid peroxidation products: malondialdehyde, diene and triene conjugates. The ability of the corrective factors inhibits the formation of these products in the plasma and homogenate can indirectly evidenced about the antioxidant properties of the drug. So it is interested to investigate the influence of thiotriazoline and hepadif on activity of these processes in rats, which are poisoned by acetaminophen with prolonged usage of estrogen and progestin.

KEY WORDS: acetaminophen, thiotriazoline, hepadif, free radical oxidation of lipids and proteins, antioxidant system.

INTRODUCTION. Acetaminophen is present in more than 200 drugs at various brand names. Almost all of them are used as analgesic and antipyretics. Poisoning may be at usages a big single dose of pure drug or combined drugs containing acetaminophen. Hepatoprotectors are one of the most widely using drugs for the treatment of liver diseases. These drugs are used for metabolism normalization, increase resistance to pathogenic factors, normalization of functional activity and stimulation of reparative - regenerative processes in the liver.

Therefore, we set a goal to investigate the effect of acetaminophen on parameters of free radical oxidation and antioxidant system of animals at poisoning by acetaminophen with prolonged usage of estrogen and progestin at the correction by Thiotriazoline and Hepadif.

INVESTIGATION METHODS. The experiments were performed on white female rats weighing 200 ±20 g, are kept on a standard diet of vivarium and free access to water.

We did three series of experiments. In the first experiment: toxic damage of rats caused by a single intragastric administration of acetaminophen suspension in 2% starch solution at a dose of 1250 mg/kg body weight (1/2 LD50), the second - a suspension of acetaminophen in 2% starch solution at a dose of 55 mg/kg (highest therapeutic dose) is administered within 7 days. Levonorgestrel in 2% solution of starch is administered both series intragastric at a dose of 1.17 mg/kg, and ethinylestradiol - a dose of 0.23 mg/kg within 40 days. Thiotriazoline was injected intraperitoneally at a dose of 100 mg/kg; Hepadyf - intraperitoneally at a dose of 8.6 mg/kg.

In 1 series of experiments, the experimental rats were divided into 3 groups: 1st - intact (control); 2nd – poisoning by acetaminophen after 40-day administration of levonorgestrel and ethinylestradiol, 3rd - poisoning by acetaminophen after 40-days administration of levonorgestrel, ethinylestradiol and once Thiotriazoline administration.

In 2 series of experiments, the experimental rats were divided into 3 groups: 1st - intact (control); 2nd - rats with acetaminophen administration within 7 days after 40-day administration of levonorgestrel and ethinylestradiol, 3rd - poisoning by acetaminophen after 40-days administration of levonorgestrel, ethinylestradiol and 7-days Thiotriazoline administration.

In 3 series of experiments, the experimental rats were divided into 3 groups: 1st - intact (control); 2nd - rats with acetaminophen administration within 7 days after 40-day administration of levonorgestrel and ethinylestradiol, 3rd - poisoning by acetaminophen after 40-days administration of levonorgestrel, ethinylestradiol and 7-days Hepadif administration.

RESULTS AND DISCUSSION. Concentration of diene conjugates decreased in blood plasma of rats with single and 7-days administration of acetaminophen after 40-days administration of estrogen and progestin and Thiotriazoline administration. Decrease intensity generation of reactive oxygen by blood mononuclear leukocytes is observed at Hepadif action, and Hepadif almost neutralized its toxic effect at seven-times administration of acetaminophen at therapeutic doses. Concentration diene (respectively 2.8 and 2.6 times), and triene conjugates (3.7 and 3.2 times) decreased in plasma of affected animals. Similarly effect was in liver homogenate.

Amount of oxidized modified proteins in blood plasma and liver of affected animals decreased at correction of lesion by Thiotriazoline and Hepadif. The administration of corrective drugs is caused decrease of malonic aldehyde concentration in plasma and liver of affected rats. Peroxidase activity in blood at correction by Thiotriazoline decreased respectively on 159 and 162% compared with affected animals. Greater effect is observed at action of Hepadif, administration of which normalizated of the peroxidase activity of blood and concentration of ceruloplasmin. Administration of Thiotriazoline and Hepadif has protective effects on glutathione system.

CONCLUSIONS. Usage Thiotriazoline and Hepadif in rats with acetaminophen toxic damage against prolonged administration of estrogens and progestins inhibit reactive oxygen species formation by mononuclear leukocytes, decreas intensity of lipid peroxidation and protein, and normalization of enzyme and nonenzyme links of antioxidant system.

To study the influence of hypothyroidism on the course of free-radical processes and state of antioxidant system in rats with acute extensive peritonitis, the production of active forms of oxygen by mononuclear leukocytes, concentration of lipid hydroperoxides, TBA-active products, Schiff bases, SOD and catalase activity were determined. Simulation of extensive peritonitis with associated hypothyroidism led to the less obvious, than in euthyroid rats, increase of active forms of oxygen and lipid peroxidation products. Although the reduction of activity of enzymes of the first line of antioxidant defense (superoxide dismutase and catalase) was more evident.

Background. The injured with polytrauma are classified as extremely severe patients, as their emergency care and intensive care is followed by significant difficulties, frequent development of complications, high mortality and survivors disability.Objective. This study is aimed to evaluate the effect of lipid peroxidation in heart, lungs, liver and blood of rats in early and late periods of combined traumatic injury of chest and hips and to establish their influence on the development of multiple organ failure.Methods. The study was conducted on 70 adult male white nonlinear rats. It was determined the content of lipid hydroperoxides and concentration of TBA-active products in erythrocyte mass, heart, lungs and liver.Results. The data prove activation of free radical oxidation at the first day of post-traumatic period. Analysis of the data evidences the increase in of lipid hydroperoxides (HPL) rate in liver homogenate in group E1 in 1.5 times, in all subsequent periods of the research the increase in rate fluctuated within 1.9-2.0 times. In blood and heart the HPL rate increased twice in group E1, reached maximum in group E2 and gradually decreased till the end of the experiment, data exceeded the control group. The highest level of HPL was determined in lung tissues (r≤0.01). In the post traumatic period the highest TBA-AP was detected in lungs and liver.Conclusions. In case of simulated trauma (injury of chest with fractures of both hips) hyperactivation processes of free radical oxidation is observed in 1 day, reaching a peak in 7-14 days of post-traumatic period in blood, tissues, liver, heart and lungs if compared to the control group causing multiple organ failure.

Purpose of the study: to assess the state of lipid peroxidation processes in patients with kidney cancer in relation to the clinical factors of the prognosis of the disease and against the background of surgical intervention.Material and methods. For the study, we used blood plasma samples from 300 patients with kidney cancer T1–4N 0–1M0–1, obtained before surgery and 8–9 days after surgery. Oxidative stress was assessed by the intensity of free radical processes by the method of induced biochemiluminescence on the BKhL-07 biochemiluminometer apparatus (Medozons Co., Russia), the activity of lipid peroxidation processes was assessed by the accumulation of initial products in the blood plasma – diene conjugates (DC), triene conjugates (TC) and fial Schiff bases (OSh) according to I. A. Volchegorsky et al. method. (1989). Statistical processing of the results was carried out using Statistica 12 and Microsoft Offie Excel 2010 programs.Results. In patients with kidney cancer, there was a decrease in the level of DC, an increase in OSh and the ratio of Osh / (DC + TC) compared with the control (p ≤ 0.05), against the background of depletion of the resources of the antioxidant defense system. The content of DC did not differ from the norm with the size of the primary tumor of the kidney pT1–pT2 and decreased (p ≤ 0.05) in the case of the presence of the tumor pT3–pT4. At the same time, there was an increase in the OSh content (p ≤ 0.05). In patients with thrombi in the inferior vena cava system, a decrease in the level of DC was observed (p ≤ 0.05), the OSh level did not differ from that of healthy individuals. In the postoperative period, there was an increase in the content of DC (p ≤ 0.05) and a decrease in the Osh / (DC + TC) coeffiient (p ≤ 0.05) compared with the initial level.Conclusion. Kidney cancer is accompanied by the activation of free radical oxidation processes against the background of depletion of the resources of the antioxidant defense system. Changes in the pro-antioxidant homeostasis are interrelated with the size of the primary tumor and the presence of a tumor thrombus. In the postoperative period, the activity of free radical oxidation processes in the patient’s body decreases.

The article presents the results of research on the state of the antioxidant defense system of piglets under nitrate-nitrite toxicosis and the action of corrective factors (methionine, phenarone and methyphene). Nitrate-nitrite load in piglets caused inhibition of the activity of enzymes of the antioxidant defense system, that indicates a decrease in the activity of superoxide dismutase, catalase and glutathione peroxidase in the serum of testing animals. The lowest activity of antioxidant enzymes under chronic nitrate-nitrite toxicosis was on the 60th day of the experiment in the serum of piglets of the control group, where accordingly the activity of superoxide dismutase decreased by 14 %, catalase activity – by 18 %, glutathione peroxidase activity – by 12 %. Lipid peroxidation processes are activated in the blood of animals, namely the level of lipid hydroperoxides and diene conjugates increases under the conditions of experimental nitrate-nitrite load. It should be noted that the highest level of lipid peroxidation products in the serum of piglets with chronic nitrate-nitrite toxicosis was on the 60 th day of the testing, and accordingly the content of lipid hydroperoxides increased by 67 % and diene conjugates – by 33 %. Administration of methionine, phenarone and methyphene under nitrate load contributed to the activation of the antioxidant defense system compared to animals in the control group. The use of antioxidant drugs in sick piglets also contributed to the inhibition of lipid peroxidation, as indicated by a decrease in the content of lipid hydroperoxides and diene conjugates in the serum of experimental group. More pronounced antioxidant effect was shown by administrating methyphene. The mechanism of action of phenarone and methyphene is associated with the direct effect of its components on the inhibition of the absorption of metabolites that have a toxic effect on cell membranes of animals. As a result of the adsorbing action of zeolite, which is part of the drugs, in the digestive tract there is a decrease in the concentration of substances that can be substrates for lipid peroxidation, as well as removal of toxic metabolites in the blood, which are prooxidants. This process occurs by osmosis and diffusion of these substances through the capillaries of the microvilli of the small intestine and their subsequent fixation on the sorbent granules.

Investigation of glutathione antioxidant system activity and diene conjugates content in rats liver and blood serum at the influence of melaksen and valdoxan under experimental hyperthyroidism (EG) has been revealed. It has been established that the activities of glutathione reductase (GR), glutathione peroxidase (GP) and glutathione transferase (GT), growing at pathological conditions, change to the side of control value at these substunces introduction. Reduced glutathione content (GSH) at melaxen and valdoxan action increased compared with values under the pathology, that, obviously, could be associated with a reduction of its spending on the detoxication of free radical oxidation (FRO) toxic products. Diene conjugates level in rats liver and blood serum, increasing at experimental hyperthyroidism conditions, under introduction of melatonin level correcting drugs, also approached to the control meaning. Results of the study indicate on positive effect of melaxen and valdoxan on free radical homeostasis, that appears to be accompanied by decrease of load on the glutathione antioxidant system in comparison with the pathology.
