Esters of thiosulfoacids demonstrate a wide range of biological activity. One of their effects is the influence on the metabolism of proteins and lipids in the body. Therefore, the purpose of our experiment was to study the impact of synthesized thiosulfonates on the total content of proteins and phospholipids, as well as their spectrum in the blood, liver, and kidney of rats. For the experiment, allyl, ethyl, and methyl esters of thiosulfoacid were used. The protein profile of rat tissues was investigated by electrophoresis, and the ratio of different fractions of phospholipids by thin-layer chromatography. Our results have shown that short-term administration of thiosulfonates in a dose of 300 mg/kg of body weight did not cause significant changes in the content of total protein and its fractions in liver tissue, whereas the effect of allyl and ethyl esters of thiosulfoacid was accompanied by an increase in the total protein and albumin in the blood plasma. The decrease in total protein was found in the kidney tissue of rats injected with allyl- and methyl thiosulfonates. The newly synthesized compounds did not lead to significant changes in the total content of phospholipids in blood plasma and tissues of rats, except for methyl thiosulfonate, the effect of which was accompanied by an increase in the total phospholipids in the liver of rats. These data may indicate an adaptive reaction of the rat’s organism. Tissue-specific features of the phospholipid spectrum were detected in rats after short-term exposure to thiosulfonates. The most significant effect on the phospholipid profile in the blood was shown for allyl- and ethyl esters of thiosulfoacid. Their action was accompanied by a decrease in the phosphatidylserine and phosphatidylinositol fractions, while phosphatidylethanolamine and phosphatidylcholine increased, respectively. Esters of thiosulfoacid significantly influenced the ratio of different fractions of phospholipids in the liver and kidney tissues. The phospholipid composition of the liver was more influenced by the allyl and methyl esters of thiosulfoacid, whereas for the kidney tissue a greater effect was observed for ethyl and methyl esters. Thus, the action of allyl ester of thiosulfoacid caused a decrease in the asymmetry coefficient of hepatocyte membranes, indicating an elevation of the lipid bilayer saturation and the increase of membrane microviscosity. Similar changes were found in the kidney of rats treated with allyl- and ethyl thiosulfonates.

Levels of methylenetetrahydrofolate in Krebs ascites cells subsequent to transplantation and the effects of methotrexate on these levels have been measured. To directly measure methylenetetrahydrofolate in tissue extracts, the cofactor was incorporated into a covalent ternary complex with thymidylate synthase and 3H-labelled fluorodeoxyuridine monophosphate. A 3-4-day lag preceded rapid growth of the tumour cells, and this same kinetic behaviour was observed for methylenetetrahydrofolate levels in the tumour cells. Liver and kidney tissue from the same animals also showed an increase in methylenetetrahydrofolate over the same time period. The impact of methotrexate on methylenetetrahydrofolate in the tumour cells depended upon concentration and the post-transplantation time at which treatment was initiated. Levels of methylenetetrahydrofolate in the tumour cells were most sensitive to the drug at the beginning of the rapid growth phase and were more sensitive to a given level of methotrexate in the presence of phospholipids. A slight but significant increase in methylenetetrahydrofolate occurred in some cases in response to the presence of methotrexate.

To delineate the metabolic fate of thyroid hormone in prolonged critically ill rabbits, we investigated the impact of two dose regimes of thyroid hormone on plasma 3,3′-diiodothyronine (T2) and T4S, deiodinase type 1 (D1) and D3 activity, and tissue iodothyronine levels in liver and kidney, as compared with saline and TRH. D2-expressing tissues were ignored. The regimens comprised either substitution dose or a 3- to 5- fold higher dose of T4 and T3, either alone or combined, targeted to achieve plasma thyroid hormone levels obtained by TRH. Compared with healthy animals, saline-treated ill rabbits revealed lower plasma T3 (P = 0.006), hepatic T3 (P = 0.02), and hepatic D1 activity (P = 0.01). Substitution-dosed thyroid hormone therapy did not affect these changes except a further decline in plasma (P = 0.0006) and tissue T4 (P = 0.04). High-dosed thyroid hormone therapy elevated plasma and tissue iodothyronine levels and hepatic D1 activity, as did TRH. Changes in iodothyronine tissue levels mimicked changes in plasma. Tissue T3 and tissue T3/reverse T3 ratio correlated with deiodinase activities. Neither substitution- nor high-dose treatment altered plasma T2. Plasma T4S was increased only by T4 in high dose. We conclude that in prolonged critically ill rabbits, low plasma T3 levels were associated with low liver and kidney T3 levels. Restoration of plasma and liver and kidney tissue iodothyronine levels was not achieved by thyroid hormone in substitution dose but instead required severalfold this dose. This indicates thyroid hormone hypermetabolism, which in this model of critical illness is not entirely explained by deiodination or by sulfoconjugation.

Cadmium (Cd) concentrations in various tissues of Romney sheep and rates of accumulation, as affected by Cd concentrations in pasture and soil, DM intake, and animal age, were determined in a 25-month study. After weaning, 6 groups of 10 wether lambs were grazed on a low Cd (0.l8 � 0.08 8g Cd/g DM) or high Cd (0.52 � 0.17 8g Cd/g DM) pasture in 3 replicates. The rate of accumulation of Cd (8g /day) into the liver and kidney was greatest in sheep aged about 6 months (for the low and high Cd pastures: kidney 0.27 and 0.56; liver 0.35 and 1 I), after which it declined with age (kidney 0.02 and 0.03: liver <0901 and 0.05; at 28 months of age). Total content of Cd in the organs continued to increase. In 6-month-old animals, 0.25% of total Cd ingested was retained by the kidney and liver, but this proportion decreased to about 0.05% of Cd intake with 28-month-old animals. Net retention of Cd in the fleece-free body as a percentage of daily Cd intake for both the control and treatment animals was estimated between 0- 35 and 0 5%. Except for muscle tissue from sheep in the treatment group, for which the mean Cd concentration was lower than that of the control group (2.0 � 0.24 and 3 9 � 0.63 r)g Cd/g fresh tissue, respectively), Cd concentrations in kidney, liver, thymus, muscle, and lung tissue increased over the first 3 months for both groups of animals. After 2 years, animals on the treatment pastures had approximately 3-fold greater Cd concentrations in liver, kidney, and duodenal tissue tissue than those grazing the control pastures (liver, 361 � 58 v. 97 � 11; kidney, l485 � 200 v. 352f48; duodenal tissue, 32f 4 v. 18f 2.5 t)g Cd/g FW, respectively). At higher Cd intakes, the adaptive effect of increased metallothionein synthesis enabled the animal to be more efficient in binding Cd, which restricted accumulation of Cd in muscle. The regression ([Cd] kidney = -205 + 0. 981Cdintake + 0 - 726Time; r = 0.82, P < 0.001) gave the best fit for the observed Cd concentration in the kidney tissue. Daily Cd intake was also a better predictor of the Cd concentration in liver tissue than the concentration of Cd in pasture by itself ([Cd]liver, = 24.7 + 0.353Cdintake; r = 0.81, P < 0.001). Accurate predictions of the potential for Cd accumulation in young grazing animals will enable more effective management strategies to be implemented to reduce Cd accumulation and therefore minimise the potential impact relating to both environment and market.

The aim of the present study was to assess the impact of combined 14-day treatment with mianserin (10 mg/kg) and simvastatin (1 or 10 mg/kg) on selected biochemical liver and kidney parameters in rats (AST and ALT activities and the concentrations of AFP, total protein, urea, creatinine and ß2-M). The results showed the increase in both transaminases activities, creatinine concentration and the decrease of AFP, total protein and ß2-M concentrations. The results indicate that 14-day combined administration of mianserin with simvastatin negatively affects the liver functioning. The observed changes in kidney biochemical parameters may suggest a risk of renal dysfunction during long-term combined treatment with these drugs.

Carbendazim is a systemic broad-spectrum fungicide controlling a wide range of pathogens. It is also used as a preservative in paint, textile, papermaking and leather industry, as well as a preservative of fruits. In the present study, carbendazim was administered at 0, 150, 300 and 600 mg/kg per day doses orally to male rats (Rattus rattus) for 15 weeks. At the end of the experiment, blood samples, liver and kidney tissues of each animal were taken. Serum enzyme activities, and haematological and biochemical parameters were analysed. In toxicological tests, 600 mg/kg per day doses of carbendazim caused an increase of albumin, glucose, creatinine and cholesterol levels. Also, at the same doses, white blood cell and lymphocyte counts decreased. However, mean cell hemoglobin and mean cell hemoglobin concentrations increased. Histopathological examinations revealed congestion, an enlargement of the sinusoids, an increase in the number of Kupffer cells, mononuclear cell infiltration and hydropic degeneration in the liver. At the highest doses, congestion, mononuclear cell infiltration, tubular degeneration and fibrosis were observed in the kidney tissue. These results indicate that 300 and 600 mg/kg per day carbendazim affected the liver and kidney tissue and caused some changes on haematological and biochemical parameters of rats.

Objective. Glyoxalase-1 is an enzyme detoxifying methylglyoxal (MG). MG is a potent precursor of advanced glycation endproducts which are regarded to be a key player in micro- and macrovascular damage. Yet, the role of Glo1 in atherosclerosis remains unclear. In this study, the effect of Glo1 on mouse metabolism and atherosclerosis is evaluated.Methods. Glo1 knockdown mice were fed a high fat or a standard diet for 10 weeks. Body weight and composition were investigated by Echo MRI. The PhenoMaster system was used to measure the energy expenditure. To evaluate the impact of Glo1 on atherosclerosis, Glo1KDmice were crossed with ApoE-knockout mice and fed a high fat diet for 14 weeks.Results. Glo1 activity was significantly reduced in heart, liver, and kidney lysates derived from Glo1KDmice. Yet, there was no increase in methylglyoxal-derived AGEs in all organs analyzed. The Glo1 knockdown did not affect body weight or body composition. Metabolic studies via indirect calorimetry did not show significant effects on energy expenditure. Glo1KDmice crossed to ApoE−/−mice did not show enhanced formation of atherosclerosis.Conclusion. A Glo1 knockdown does not have major short term effects on the energy expenditure or the formation of atherosclerotic plaques.

The evolution of views on tax evasion following the introduction of limited rationality and social and psychological factors into the models of taxpayer behavior has increased the plausibility of the initial assumptions of the models, but it has made it difficult to use classical approaches based on the search for equilibrium states. The variety of behavioral responses of taxpayers due to the many factors that influence their choice has led to the fact that tax evasion has come to be considered as the result of nonlinear and dynamic interactions between the state and taxpayers. In such models, small short-term external influences can act as shocks, which leads to the emergence of a wide range of different long-term trends, the analysis of which within the framework of traditional approaches is difficult. In this regard, the purpose of this review study is to study the evolution of views on the behavior of taxpayers that has led to the emergence of new approaches to modeling tax evasion where the key role is assigned to the analysis of the impact of external shocks of various scales and nature. The research hypothesis is that modern approaches to the study of tax evasion problems make it necessary to consider the interaction of the state and taxpayers within the framework of the theory of non-equilibrium and nonlinear systems in which minor external influences can play the role of shocks, and the most promising direction of their study is the use of agent-based modeling tools. The results of the study confirm that the use of agent-based models is a promising approach for integrating existing approaches in the study of tax evasion processes. The proposed concept of building an agent-based model for analyzing the processes of tax evasion allows us to answer the question of how short-term exogenous shocks will affect the preferences of taxpayers, taking into account their individual characteristics and accepted behavioral patterns in society.

PurposeThis paper investigates stakeholders' perspectives of share buybacks in the context of time-horizons of investment decisions and strategy.Design/methodology/approachWe use in-depth interviews with stakeholders from eight listed UK firms as well as examine their publicly available data.FindingsFindings suggest that share buybacks involve a wide range of stakeholders' rational interests and long-term management perspectives as they enable firms to strategise operational plans towards their long-term corporate goals.Research limitations/implicationsThe findings are based on interviews with a small number of share buyback firms and the findings, therefore, may not be generalised to all firms.Practical implicationsThe results show that share buybacks may be part of the long-term interests of firms and not necessarily used as part of short-term EPS increases as suggested in the extant literature.Originality/valueThe findings contribute to the literature on corporate pay-out policies in the context of short-term financial objectives vs long-term strategic objectives of stakeholders. They show that share buybacks can be an important part of firms' long-term strategic considerations.

Concentrações residuais de antibióticos de uso compartilhado pela terapia médica humana e veterinária são cada vez mais frequentes nos mais variados tipos de matrizes ambientais; no entanto, pouco se sabe sobre o impacto que esses fármacos podem acarretar aos microrganismos do solo. Sendo assim, perturbações relacionadas à exposição da microbiota de um latossolo vermelho-amarelo brasileiro a dois antibacterianos, a amoxicilina (AMOX) e a doxiciclina (DOX), foram investigadas por meio da determinação de atividade (mg CO2) e biomassa (Cmic) microbianas, juntamente com o quociente metabólico (qCO2), em amostras de solo que receberam as seguintes concentrações desses compostos: 0,03, 0,3, 3,0, 30 e 300 mg L-1. Os resultados mostraram diferentes efeitos sobre a microbiota e de forma específica para cada antibiótico. A AMOX mostrou-se mais impactante para os microrganismos do solo, com redução da biomassa e aumento do qCO2, enquanto que a DOX reduziu a atividade microbiana, mas sem efeito na biomassa e qCO2.A B S T R A C TThe residual concentrations of antibiotics used by human and veterinary medical therapy are increasingly common in a wide range of environmental matrices, nevertheless little is known about the impact of these drugs on to the soil microorganisms. Therefore, disturbances related to the exposure of the microbiota of a Brazilian Red-yellow Latosol to two antibacterials, amoxicillin (AMOX) and doxycycline (DOX), were investigated through the determination of the microbial activity (mg CO2) and biomass (Cmic), among with the metabolic quotient (qCO2), using soil samples spiked with: 0,03, 0,3, 3,0, 30 and 300 mg L-1 of each drug. The results showed different effects on the microbiota and in a specific way for each antibiotic. The AMOX showed higher impact impacting for the soil microorganisms, with reduction of the biomass and increase of the qCO2, whereas the DOX reduced the microbial activity, but showed no effect in the biomass and qCO2.Keywords: Antibiotics. Polypropylene (PP) has a wide range of applications in the automobile bumpers due to its many excellent properties. Mechanical properties of PP for automobile bumpers are investigated through an artificial accelerated aging test. The aging rules after different years of normal use and the artificial accelerated aging test are analyzed. The correlation between natural and artificially accelerated aging is also explored. It provides a reference for the study of the aging properties of polymers for automotive applications. Results show that the UV aging test can effectively simulate changes in tensile and bending strengths after natural aging and can be used to evaluate the weathering resistance of PP materials used in automotive bumpers. The tensile and bending strengths of these materials remain good during aging, and elongation is sensitive to aging. The short-term artificial accelerated aging test does not exert a significant influence on the impact strength of materials, and artificial accelerated aging does not completely reproduce the aging process of the material.
