The notion “endotoxin” is conditional, for any normal metabolit has a toxic impact of its excessive accumulation in organism. Endotoxins include products of breakdown of tissue proteins, peroxides and other products of free radical oxidation, low molecular weight toxins, toxins of microorganisms. Current knowledge on the mechanism of endotoxin action on the organism of pregnant cows is based on the key role of the immune system: it provides animals with protection against the infections, eliminates extraneous elements of endogenous and exogenous origin. It is defined that under endogenous intoxication, toxic metabolites suppress the activity of immune system of organism, decreases the resistability of an organism to negative effects of environmental factors. Animals under endogenous intoxication had the following clinical symptoms: swelling of mammal gland, edemas of external genitals, anemia of mucous membranes, functional disorders of proventriculuses and intestines, animals were stressed. It was defined that under endogenous intoxication in 8–9 months of pregnancy, blood of highly-productive cows had a tendency of decrease in the number of erythrocytes by 21.0%, in the level of hemoglobin – by 24.3%, increase in the number of leucocytes by 7.6% compared to the value for blood of cows with physiological pregnancy process. The development of endogenous intoxication among pregnant cows causes disorders in protein-synthesizing function of liver, which is indicated by decrease in the level of total protein and its fractions. It was defined that endogenous intoxication decreased the content of total protein in blood by 18.4%. Over the period of study, among pregnant cows under the development of endogenous intoxication, heightened activity of amino transferaces in the blood was observed. In the ninth month of pregnancy, the activity of amino transferaces in blood serum of cows from experimental group increased by 28.6%, and the activity of aspartate aminotransferace increased by 20.1% relatively. The activity of amino transferaces and decrease in the level of total protein and its fractions in blood serum is one of the first biochemical studies on the diagnosing endotoxin manifestations and indicates destructive processes in liver. It was defined that pregnant cows under the development of endogenous intoxication suffer disorders in detoxicating function of liver and disorders in filtrating function of kidneys. The developing endogenous intoxication of pregnant cows suppresses the activity of enzymes in their glutathione system of antioxidant protection, which is indicated by decrease in the activity of glutathione peroxidase by 27.5 %, glutathione reductase by 42.9%, glucose-6-phosphate dehydrogenase by 11.2% in blood serum. The lowest activity of enzymes of glutathione system in blood of highly-productive cows under endogenous intoxication was observed in their ninth month of pregnancy, which is related to increase in activation of processes of lipid peroxidation and misbalance between the activity of antioxidant system and intensity of lipid peroxidation. Reliable increase in the content of lipid hydroperoxides by 70.9% and in concentration of malondialdehide by 54.8% was observed. Misbalance towards generation of active forms of oxygen and their metabolites, exhaustion of antioxidant system and disorders in balance cause oxidative stress.

Mitochondrial free radical formation has been implicated as a potential mechanism underlying degenerative senescence, although human data are lacking. Therefore, the present study was designed to examine if resting and exercise-induced intramuscular free radical-mediated lipid peroxidation is indeed increased across the spectrum of sedentary aging. Biopsies were obtained from the vastus lateralis in six young (26 ± 6 yr) and six aged (71 ± 6 yr) sedentary males at rest and after maximal knee extensor exercise. Aged tissue exhibited greater ( P < 0.05 vs. the young group) electron paramagnetic resonance signal intensity of the mitochondrial ubisemiquinone radical both at rest (+138 ± 62%) and during exercise (+143 ± 40%), and this was further complemented by a greater increase in α-phenyl-tert-butylnitrone adducts identified as a combination of lipid-derived alkoxyl-alkyl radicals (+295 ± 96% and +298 ± 120%). Lipid hydroperoxides were also elevated at rest (0.190 ± 0.169 vs. 0.148 ± 0.071 nmol/mg total protein) and during exercise (0.567 ± 0.259 vs. 0.320 ± 0.263 nmol/mg total protein) despite a more marked depletion of ascorbate and uptake of α/β-carotene, retinol, and lycopene ( P < 0.05 vs. the young group). The impact of senescence was especially apparent when oxidative stress biomarkers were expressed relative to the age-related decline in mitochondrial volume density and absolute power output at maximal exercise. In conclusion, these findings confirm that intramuscular free radical-mediated lipid peroxidation is elevated at rest and during acute exercise in aged humans.

AbstractObjectivesThe main objective of the present study is to evaluate the mitigative effect of hydroalcoholic extract of Momordica cymbalaria fruits against sodium fluoride (NaF) induced hepatotoxicity.MethodsIn this study, Wistar male albino rats were randomly divided into five groups of six rats each. Group I and II served as normal and toxic controls. Group III as plant control received extract at a dose of 400 mg/kg b. wt, p.o and Groups IV and V as treatment groups received extract at a dose 200 and 400 mg/kg b. wt, p.o for 30 days. All groups except Groups I and III received 100 ppm of NaF through drinking water. After completion of the study, blood collected for the estimation of liver blood serum biomarkers such as aspartate aminotransferases (AST), alanine aminotransferases (ALT), alkaline Phosphatase (ALP), direct and total bilirubin, total protein and albumin. The liver tissue homogenate was for estimation of lipid peroxidation, catalase, and reduced glutathione levels.ResultsThe results showed that NaF intoxication caused elevation of liver blood serum levels and lipid peroxidation; decreased levels of serum total protein, albumin and liver reduced glutathione, and catalase observed. The treatment groups showed decreased elevated serum biomarkers (ALT, AST, and ALP), liver lipid peroxidation and increased serum total protein and albumin, liver reduced glutathione and catalase levels in a dose-dependent manner. Histopathological studies also further strongly supported for mitigative effects of the plant.ConclusionsIn conclusion, our findings of the study indicated that M. cymbalaria fruits were a potential drug candidate in the treatment of NaF induced hepatotoxicity.

During modeling of esophageal alkali burns 1 and 2 degrees in immature rats were observed violations of various metabolic systems, melanin administration helped to normalize the biochemical parameters. It was shown that under the esophageal burns there occurred violations of protein (total protein, albumin), nitrogen metabolism (creatinine, urea) metabolism of carbohydrate (glucose) and lipids (cholesterol), changed the activity of enzymes (GOT, GPT, CC), violated fluid and electrolyte balance (K +, Na +, Cl-, Ca2 +, Mg2 +, phosphates), while the introduction of melanin contributed to the normalization of these indicators. The most effective dose were of 0.5 mg/ kg and 1 mg/kg. There were found anti-toxic properties of melanin, which may be a promising drug for the normalization of metabolism in patients with burns of esophagus and prevent its complications.

We observed the processes of lipid peroxidation, characterized by increased content of intermediate and its end products (diene conjugates, lipid hydroperoxides, malonic dialdehyde), in the blood of the pregnant mares. The changes of the data of indicators in the organism of the mares at the 9–11 months of pregnancy were established. The level of diene conjugates in the blood of animals was 3.6 ± 0.12 μmol/l, 33.3 and 44.0%, which was higher compared to non-pregnant mares and mares at 4th month of pregnancy. The concentration of lipid hydroperoxides in the blood of mares increased by 100% and by 42.9% in the blood of non-pregnant mares and mares at 4th month of pregnancy. The concentration of malondialdehyde in the blood of mares increased by 75.0%, 51.2% and 25.0% compared to non-pregnant mares and mares at 4th and 7th month of pregnancy, respectively. The results of our research showed that the mineral-vitamin premix Marmix had a positive influence on the state of lipid peroxidation products, contributed to their lowering in the blood of pregnant mares, which demonstrated antioxidant action and stimulated the function of the immune system. It was established that the bactericidal and lysozyme activity of the blood serum and the concentration of circulating immune complexes increases in the mares with lack of trace elements. It was shown that feeding mares during the 9–11 months of pregnancy with the mineral-vitamin premix Marmix had a positive influence on the state of humoral factors of nonspecific resistance. In particular, the concentration of bactericidal activity of serum increased by 31.0%, lysozyme activity of blood serum – 45.4% and contributed to the decrease of the concentration of circulating immune complexes in serum by 3.8 times. The use of mineral-vitamin premix Marmix during 60 days in feeding pregnant mares caused a restoration of the clinical status, a decrease in the content of lipid peroxidation products.

During modeling of esophageal alkali burns II degree in immature rats were observed violations of various metabolic systems. It was shown that under the esophageal burns there occurred violations of protein (total protein, albumin), nitrogen metabolism (creatinine, urea), changed the activity of enzymes (GOT, GPT), violated fluid and electrolyte balance (Na+, К+, Cl-, Ca2+, Mg2+). In the groups of animals with the alkaline burn of the esophagus, that were injected aqueous extract of pod of bean (Phaseolus vulgaris) there can be traced a change of the indicators in the direction of normalization, indicating about the obvious therapeutic effect of aqueous extract of pod of bean (Phaseolus vulgaris).

The article contains the results of research the impact of Fasciolosis and mycobacteriosis,the indicators of protein synthesizing function of cows liver. Fasciola and mycobacteria inhibit protein synthesizing function of liver, as indicated by the reduction in their blood level of total protein and albumin fraction. The data indicate hepatotropic of the toxin F. Hepatica and mycobacteria, because namely liver serves as a support of dynamic substances equilibrium of blood plasma. Increasing the level of globulin in the blood serum of cows by association of fasciolosis and mycobacteriosis reflects the intensity of inflammatory processes in animal organism caused by infection of these diseases.Application of closaverm A and catosal to research cows promotes restoration protein synthesizing function of liver. It was found that after application to cows from the first research group for the treatment of closaverm A by 14 and 21 days of level of total protein in the blood serum was gradually increased, but did not reach physiological values. Applying both closaverm A and catosal, was set a slightly higher level of total protein, that at the 28th day of the experiment reached physiological values and therefore amounted to 70.30 ± 1.5 g/l. For the study the level of albumin in the blood of research group, which were used drugs combined, was set its increase already from 7 day of experiment. Significant increase of albumin level in the blood of experimental animals is seeing on the 14th day of the experiment, where appropriate it was within 41.90 ± 1.5 g/l. On the 28th day of the experiment the levels of globulin in the blood of experimental group of animals, which were used closaverm A and catosal, was lower by 11% compared to the control group of animals in specified period of the experiment.Cows treated with closaverm A, value of the coefficient A/T was gradually returned to normal. However, at the 28th day of the experiment has remained at 16% less than the initial values, taken before infestation. Application of closaverm A and catosal to experimental animals contributed to increasing the coefficient of A/T. So at 21st and 28th days of the experiment it was higher than in the control group of cows at 60 and 46%.

The activity of enzymes of the glutathione link of the antioxidant system in rats with experimental full-thickness wound has been studied. It is established that in the serum of rats under the experimental model of the cut wound the level of recovered glutathione and glutathione reductase activity decreased, glutathione peroxidase and glutathione transferase activity increased. All investigated parameters normalized in dynamics after treatment with melanin.

ApoD (apolipoprotein D) is up-regulated in AD (Alzheimer's disease) and upon oxidative stress. ApoD inhibits brain lipid peroxidation in vivo, but the mechanism is unknown. Specific methionine residues may inhibit lipid peroxidation by reducing radical-propagating L-OOHs (lipid hydroperoxides) to non-reactive hydroxides via a reaction that generates MetSO (methionine sulfoxide). Since apoD has three conserved methionine residues (Met49, Met93 and Met157), we generated recombinant proteins with either one or all methionine residues replaced by alanine and assessed their capacity to reduce HpETEs (hydroperoxyeicosatetraenoic acids) to their HETE (hydroxyeicosatetraenoic acid) derivatives. ApoD, apoDM49-A and apoDM157-A all catalysed the reduction of HpETEs to their corresponding HETEs. Amino acid analysis of HpETE-treated apoD revealed a loss of one third of the methionine residues accompanied by the formation of MetSO. Additional studies using apoD(M93-A) indicated that Met93 was required for HpETE reduction. We also assessed the impact that apoD MetSO formation has on protein aggregation by Western blotting of HpETE-treated apoD and human brain samples. ApoD methionine oxidation was associated with formation of apoD aggregates that were also detected in the hippocampus of AD patients. In conclusion, conversion of HpETE into HETE is mediated by apoD Met93, a process that may contribute to apoD antioxidant function.

Objective. The aim of the paper is to analyze the impact of plant antioxidant “Dihydroquercetin Baikalsky” on the LPO-AOS system and carbohydrate-lipid metabolism in residents of Khanty-Mansiysk with type 2 diabetes mellitus. Materials and Methods. The study enrolled 132 adult residents of Khanty-Mansiysk, including 78 people with type 2 diabetes mellitus and 54 healthy subjects. The authors examined the indicators of pro- and antioxidant activity in blood samples in all the trial subjects: products of lipid peroxidation (LPO), the state of the antioxidant system (AOS), oxidative stress coefficient. Patients with type 2 diabetes mellitus (n=48) undergoing standard glucose-lowering therapy were taking 1 capsule (60 mg) of Dihydroquercetin Baikalsky post cibum daily. Statistica 10.0 and Microsoft Excel software package were used to process the results obtained. Results. It was found that 12-week dihydroquercetin intake led to a significant decrease in primary and secondary lipid peroxidation products and an increase of AOS activity, which indicated the antioxidant effect of the bioflavonoid. The authors noted a positive trend towards a decrease in the parameters of the carbohydrate-lipid profile. Conclusion. The data obtained indicate the antioxidant properties of dihydroquercetin in persons with type 2 diabetes mellitus. Провести анализ коррекции показателей системы ПОЛ – АОС и углеводно-липидного обмена антиоксидантом растительного происхождения «Дигидрокверцетин Байкальский» у жителей г. Ханты-Мансийск, страдающих сахарным диабетом 2 типа. Материалы и методы. В исследование включены 132 взрослых жителя г. Ханты-Мансийск, в т.ч. 78 чел. с сахарным диабетом 2 типа и 54 условно здоровых добровольца. У обследуемых лиц изучены показатели про- и антиоксидантной активности в образцах крови: продукты перекисного окисления липидов (ПОЛ), состояние антиоксидантной системы (АОС), коэффициент окислительного стресса. Группа больных сахарным диабетом 2 типа (48 чел.) на фоне стандартной сахароснижающей терапии в течение 12 нед. принимала после еды по 1 капсуле (60 мг) в день антиоксиданта «Дигидрокверцетин Байкальский». Полученные результаты статистически обработаны с использованием пакета программ Statistica 10.0 и Microsoft Excel. Результаты. Установлено, что прием дигидрокверцетина в течение 12 нед. способствовал достоверному снижению содержания первичных и вторичных продуктов ПОЛ и повышению активности АОС, что свидетельствует об антиоксидантном действии данного биофлавоноида. Отмечена положительная тенденция к снижению показателей углеводно-липидного профиля. Выводы. Полученные данные указывают на антиокислительные свойства дигидрокверцетина у лиц, страдающих сахарным диабетом 2 типа.
