EFFECT OF POLYDIETHYLSTILBOESTROL PHOSPHATE ON THE PITUITARY CONTENT OF FSH AND ICSH IN MALE RATS

ABSTRACT The effect of a single dose of polydiethylstilboestrol phosphate (PSP) on the pituitary content of follicle stimulating hormone (FSH) and interstitial cell stimulating hormone (ICSH) was studied in intact, adult male rats after 7, 28, 103 and 161 days, respectively. The body weight and different organ weights were also recorded. The results were compared with those obtained in untreated controls and in rats treated with daily doses of diethylstilboestrol (DES) for 7 days. The pituitary FSH and ICSH content in the control rats decreased roughly to about half during a period of approximately 5 months, i. e. from the 2nd to the 7th month of life. PSP (400 μg/100 g body weight) and DES (320 μg/100 g body weight) reduced the pituitary content of FSH and ICSH to approximately 35 and 50% of the control values, respectively, measured on the 7th day of treatment. The gonadotrophin inhibiting effect of PSP lasted for more than 103 days. On the other hand, on Day 28 the pituitary content of ICSH was normal and that of FSH significantly elevated in the DES-treated rats. On Day 161 both PSP- and DES-treated animals had a significantly increased ICSH content, while the FSH content was normal. Body weights, as well as the weights of the testes and accessory glands were reduced both by DES and PSP as early as on the 7th day after the commencement of treatment. Complete restoration of these parameters in DES-treated animals was found on Day 103, while PSP exerted its effect for more than 161 days. Daily injections of 3 IU of FSH and 1.5 IU of ICSH administered in the form of human menopausal gonadotrophin (HMG) to PSP-treated rats from the day of PSP injection for 14 days counteracted the weight loss of the testes and accessory glands, as well as the loss of body weight and the reduced food intake. The hypertrophy of the adrenals and the sterility produced by the administration of PSP were completely prevented by HMG injections.

Abstract. Male rats, body weight 60–75 g, were hypophysectomized. Three days after operation the animals were divided into two groups. Group B received solvent solution and group C 1 mg/kg body weight per day of glibenclamide ip for the following 9 days. Group A consisted on non-operated normal rats. Twenty-four hours after the last injections and after a 12 h overnight fast the body weights of groups B and C were not different, the increase during the 10 days being 10% in both groups. Serum insulin (IRI) was significantly higher in group C than in group B (C: 8.0 ± 0.3 μU/ml, n = 14 vs B: 4.9 ± 1.0 μU/ml, n = 14; P < 0.01, mean ± sem) as was serum somatomedin bioactivity (SM)-porcine cartilage assay — (C: 1.06 ± 0.1 U/ml, n = 14 vs B:0.41 ± 0.01 U/ml, n = 14; P < 0.001). Skeletal growth was determined with the tibia test and by a radiograph of each rat. The width of the proximal epiphyseal growth plate of the tibia was significantly increased in group C compared to group B (C: 204 ± 4.8 μm, n = 12 vs 181 ± 6.5 μm, n = 13; P < 0.005). On the radiograph the area of the right femur was not different between the two groups of animals, while the height and the area of the first lumbar spine were significantly augmented in group C. The results show that glibenclamide stimulates IRI, SM and skeletal growth in hypophysectomized rats. Compared to the glibenclamide treated hypophysectomized animals the normal rats of group A had doubled their body weights. IRI (59 ± 5 μU/ml, n = 4) and skeletal growth (tibia test: 454 ± 5.8 μm) were greatly increased. SM did not differ between group A (1.21 ± 0.35 U/ml and group C. T4 was much lower in group B (0.64 ± 0.09 μg/100 ml, n = 5) than in group A (4.1 ± 0.3 μg/100 ml, n = 6; P < 0.001). It is concluded that a normal SM concentration is not necessarily associated with appropriate growth.

Previously, we have found that the addition of L-ascorbic acid to chitosan enhanced the reduction in body weight gain in guinea pigs fed a high-fat diet. We hypothesized that the addition of L-ascorbic acid to chitosan would accelerate the reduction of body weight in humans, similar to the animal model. Overweight subjects administered chitosan with or without L-ascorbic acid for 8 weeks, were assigned to three groups: Control group (N = 26, placebo, vehicle only), Chito group (N = 27, 3 g/day chitosan), and Chito-vita group (N = 27, 3 g/day chitosan plus 2 g/day L-ascorbic acid). The body weights and body mass index (BMI) of the Chito and Chito-vita groups decreased significantly (p < 0.05) compared to the Control group. The BMI of the Chito-vita group decreased significantly compared to the Chito group (Chito: -1.0 kg/m2 vs. Chito-vita: -1.6 kg/m2, p < 0.05). The results showed that the chitosan enhanced reduction of body weight and BMI was accentuated by the addition of L-ascorbic acid. The fat mass, percentage body fat, body circumference, and skinfold thickness in the Chito and Chito-vita groups decreased more than the Control group; however, these parameters were not significantly different between the three groups. Chitosan combined with L-ascorbic acid may be useful for controlling body weight.

ABSTRACT Two amenorrhoeic women were given a course of 10 injections of human menopausal gonadotrophin (HMG) in daily doses corresponding to 260 IU of follicle stimulating hormone (FSH) activity and 165 IU of interstitial cell stimulating hormone (ICSH) activity. In both patients an extensive ovarian stimulation was observed as indicated by the greatly increased urinary excretion of oestrone, 17β-oestradiol and oestriol. When HMG-treatment was followed subsequently by the administration of human chorionic gonadotrophin (HCG) for 5 days in a total dose of 18 000 and 30 000 IU, respectively, functional corpus luteum tissue was formed in both patients as evidenced by a huge rise in urinary pregnane-3α,20α-diol excretion and by the secretory transformation of a previously atrophic endometrium. At the approximate height of the follicular phase tracer doses of 3H-labelled dehydroepiandrosterone sulphate (DHAS) and 14C-labelled dehydroepiandrosterone (DHA) were administered to both patients in the form of a continuous intravenous infusion of 10 hours' duration. Infusion of the same dose was repeated under identical experimental conditions at the approximate height of the luteal phase. In both patients, very little radioactive material was associated with oestrone and 17β-oestradiol and none with oestriol isolated from 96-hours' urine specimens obtained at both phases of ovarian stimulation. It is concluded that — in contradistinction to the situation in pregnant women — circulating DHAS is not a significant precursor of urinary oestrogens in non-pregnant women.

Background: G protein-coupled bile acid receptor (TGR5) is involved in a number of metabolic diseases. The aim of this study was to identify the role of TGR5 after Roux-en-Y gastric bypass (GBP). Methods: Wild type and TGR5 knockout mice (tgr5-/-) were fed a high-fat diet (HFD) to establish the obesity model. GBP was performed. The changes in body weight and food intake were measured. The levels of TGR5 and peptide YY (PYY) were evaluated by RT-PCR, Western blot, and ELISA. Moreover, the L-cells were separated from wild type and tgr5-/- mice. The levels of PYY in L-cells were evaluated by ELISA. Results: The body weights were significantly decreased after GBP in wild type mice (p<0.05), but not tgr5-/- mice (p>0.05). Food intake was reduced after GBP in wild type mice, but also not significantly affected in tgr5-/- mice (p>0.05). The levels of PYY were significantly increased after GBP compared with the sham group (p<0.05); however, in tgr5-/- mice the expression of PYY was not significantly affected (p>0.05). After INT-777 stimulation in L-cells obtained from murine intestines, the levels of PYY were significantly increased in L-cells tgr5+/+ (p<0.05). Conclusion: Our study suggests that GBP up-regulated the expression of TGR5 in murine intestines, and increased the levels of PYY, which further reduced food intake and decreased the body weight.

Hypothyroidism frequently causes cardiopulmonary dysfunction, such as heart failure and respiratory and metabolic deficiencies. This study investigated the effects of Chinese herbal formula B307 on thyroidectomy-induced cardiopulmonary exercise dysfunction in rats. Twenty male rats were equally divided into four groups: negative control with sham treatment, positive control with oral B307 treatment only, thyroidectomy treatment only, and thyroidectomy with B307 posttreatment groups. The feeding dose of B307 was 50 mg/kg per day for 14 days. We examined and then compared the thyroid-stimulating hormone (TSH), free triiodothyronine (T3), free thyroxine (T4), and reactive oxygen species (ROS) from the blood of these four groups. Also, we compared the body weight, neck subcutaneous blood flow, cardiac ejection function, cardiopulmonary exercise function of oxygen consumption (VO2), carbon dioxide production (VCO2), and respiratory quotient (RQ = VCO2/VO2) among the four groups. Our results indicated that thyroidectomized rats had significantly decreased body weight, neck subcutaneous blood flow, cardiac ejection function, serum T3 and T4, and VO2 and VCO2, but had significantly increased ROS and TSH levels and RQ values compared with sham rats (P<0.01–0.05). In addition, thyroidectomized rats receiving oral B307 treatment had significantly increased body weight, neck subcutaneous blood flow, cardiac ejection function, and VO2, but significantly decreased ROS and TSH levels and VCO2 and RQ values compared with thyroidectomized rats (P<0.01–0.05). We suggest that the B307 could be a protective and beneficial alternative treatment for thyroidectomy-induced cardiopulmonary exercise dysfunction.

Fasting plasma insulin (PI) and glucose (PG) concentrations were measured throughout the body weight cycle of marmots. Animals gained weight during summer, and in late fall body weight peaked, after which they ceased feeding. Each month euthermic animals were injected intra-arterially with either dextrose (500 mg/kg) or porcine insulin (0.1 U/kg), and blood samples were collected over the subsequent 2 h. During weight gain fasting PI concentration and pancreatic B-cell response to injected dextrose increased markedly. Maximal insulin release to a dextrose challenge was measured during peak body weight or when body weight initially began to decline. The PG concentration after exogenous insulin administration was slight (less than 10%) in the fall but increased approximately 25% in the spring after marmots lost weight. Basal PG levels were not significantly different throughout the year. Basal fasting PI concentrations were significantly higher during the fall (P less than 0.01). It is suggested that in the fall, when marmots are obese, hyperinsulinemia and peripheral insulin resistance appear. Furthermore, in two animals with an increase in body weight of approximately 30% or less over the summer, peripheral resistance was demonstrable, albeit not as marked as in animals that appropriately doubled their body weights when given food ad libitum. Thus we hypothesize that factors other than adiposity, i.e., food intake, central nervous system input to the pancreatic B-cell, and/or changes in B-cell sensitivity to PG, may contribute to the observed peripheral insulin resistance and may be involved in body weight regulation.

1. The body composition was determined of forty-one fetuses of different gestational ages born to mothers belonging to a low socio-economic group of the population.2. With increasing gestational age the water content fell from 88% at 28 weeks to 76% at term; the fat content increased from 2.1% to 11.2% and the protein content increased from 6.9 to 9.3%.3. The changes in body composition were more closely related to body-weight than to gestational age.4. The calcium, Phosphorus and magnesium contents of the body per unit fat-free weight progressively increased with gestational age, and at term the values appeared to be considerably lower than those reported in the literature. The Ca:P ratio was constant at different body-weights.5. The body iron content per unit of fat-free weight increased marginally with increasing gestational age. The value was almost 30% lower than the values reported from elsewhere.6. It is suggested that chemical composition and nutrient stores of the developing foetus can be considerably influenced by the state of maternal nutrition.

Goldthioglucose-obese mice cannot adjust their food intake to meet the increased energy requirements due to cold. At all ambient temperatures above 15°C the spontaneous running activity of these animals is less than that observed for nonobese controls. Activity of obese mice is maximal at 19°C and minimal at 15°C or lower. Body weights decrease during exposure to cold. In contrast to that of obese mice, running activity of nonobese controls is maximal at an ambient temperature of 25°C but nearly ceases at 15°C or lower. The food intake of these animals increases in the cold and remains elevated even at temperatures at which activity decreases. The body weight of nonobese controls is either maintained constant or increases during exposure to cold air.

ABSTRACTSeasonal fluctuations in oestrous activity and live weight were studied in ewes of the Marwari breed indigenous to the Rajasthan Desert in north-western India. Of the ewes examined 80 to 100% were in oestrus every month of the year, indicating thereby the non-seasonality of female reproductive activity in this breed. On average, each ewe exhibited heat 18 times a year. The incidence of silent heats was low (4·2 %). The average length of an oestrous cycle was 17 days, while the actual period of oestrus varied from 24 to 48 h with an average of 36 h. No seasonal effect on these activities could be observed. The maximum and minimum body weights of the animals were recorded during the autumn (October) and summer (June) months respectively. The body weights differed significantly (P<0·05) between months. Sexual activity in these animals is, therefore, apparently not affected by changes in body weight. The continuous nature of oestrous activity in Marwari sheep may be economically exploited by regulating the lambing season according o t the prevailing ecological conditions and market demands, without the use of hormones.

Introduction. Historically, pesticides are evaluated more strictly from a medical point of view than other chemicals. Since their features, such as deliberate introduction into the environment, the possibility of contact with them by large masses of the population, and the high biological activity determine their potential danger to humans. Purpose of research - study of the biological effect of a technical product derived from triazoles when it is repeatedly ingested orally in mammals (rats), establishment of inactive and active doses, justification of the permissible daily dose (DSD) for humans. Material and methods. In acute experiments, white rats were used, including 6 animals in the group. Tested dose: 500-4000 mg/kg of body weight. A chronic (12 months) experiment was performed on 80 male rats with a bodyweight of 180-190 g at the beginning of the study. Tested doses: 5.0; 16.0 and 55.0 mg/kg of body weight (1 control and 3 experimental animals, 20 individuals each). In the dynamics of the experiment, we observed the condition and behavior of animals, water, and food consumption, recorded the timing of death, changes in body weight, physiological, biochemical, and hematological indices. Results. Indices of the acute oral toxicity on the studied product LD50 male rats were 2250 ± 483 mg/kg body weight. The dose of 5.0 mg / kg of body weight was not found to cause significant changes in all studied indices. The doses of 16.0 and 55.0 mg/kg of body weight had a polytropic effect on the body in experimental animals. Discussion. The studied product for the acute oral toxicity refers to low-hazard compounds, the doses of 16.0 and 55.0 mg/kg of body weight has a polytropic effect on the mammalian body, causing changes in carbohydrate, lipid, and lipoprotein metabolism in the body of rats - was accepted as acting. The dose of 5.0 mg / kg of body weight, when administered in rats, there are no changes in all the studied parameters throughout the experiment, is accepted as invalid. Based on the inactive dose-5.0 mg/kg of body weight and taking into account the reserve factor of 100, we have scientifically justified DSD for a person at the level of 0.05 mg/kg. Summary. The conducted sanitary and Toxicological studies indicate the need to assess the toxicity of new technical products to the mammalian body, to increase the reliability of the developed hygiene standards in environmental objects and food products.
