Gynecological scientific journals report that traditional therapeutic regimens used to treat vaginal inflammation recommended by global treatment centers and health organizations are not producing satisfactory results. Insufficient duration of the drug’s contact with vaginal mucosa does not provide adequate pH conditioning the physiological biocenosis of the organ. The conducted research and obtained positive results encourage further research possibilities in the field of optimization of pharmaceutical properties of the examined gynecological preparations. Assuming the above assumptions tested impact of polyvinylpyrrolidone K-15, polyvinylpyrrolidone K-30 and polyvinylpyrrolidone K-90 in the properties of the globules. The study was prepared formulations with different pH and rheological properties. The device simulating the conditions in the vagina, studied the adhesion and movement of the gel on the mucosa of the organ. Globules after application of the apparatus simulates the natural conditions in the gel passes and cover the surface. A wide range of pH of the gels allows the selection of the optimum formulation

The investigated powders on a methylcellulose base revealed good adhesion to the vaginal mucosa and were present on it throughout the daily activities of the patient. This property enabled the continuity of the 24-hour action of the drug. The treatment of vagina inflammatory conditions is usually long and difficult because of the frequent relapse. Powders were transformed into gels and were tested for their properties. Formulations were prepared with varying pH and rheological properties. The test showed the work of adhesion of gels. All gels with ratios of lactic acid to chitosan of 1:1 and 2:1 showed a pH in the physiological range at 37°C. Additionally, carboxymethylcellulose sodium salt and excipients allow various formulations with a wide range of pH to be obtained. Rheological investigation revealed an increase in the dynamic viscosity of preparations containing lactic acid complexed with chitosan and carboxymethylcellulose sodium salt in comparison to gels without carboxymethylcellulose sodium salt.

ABSTRACTCRISPR-Cas9 technology has been exploited for the removal or replacement of genetic elements in a wide range of prokaryotes and eukaryotes. Here, we describe the extension of the Cas9 application toolbox to the industrially important dairy speciesLactococcus lactis. The Cas9 expression vector pLABTarget, encoding theStreptocccus pyogenesCas9 under the control of a constitutive promoter, was constructed, allowing plug and play introduction of short guide RNA (sgRNA) sequences to target specific genetic loci. Introduction of apepN-targeting derivative of pLABTarget intoL. lactisstrain MG1363 led to a strong reduction in the number of transformants obtained, which did not occur in apepNdeletion derivative of the same strain, demonstrating the specificity and lethality of the Cas9-mediated double-strand breaks in the lactococcal chromosome. Moreover, the same pLABTarget derivative allowed the selection of apepNdeletion subpopulation from its corresponding single-crossover plasmid integrant precursor, accelerating the construction and selection of gene-specific deletion derivatives inL. lactis. Finally, pLABTarget, which contained sgRNAs designed to target mobile genetic elements, allowed the effective curing of plasmids, prophages, and integrative conjugative elements (ICEs). These results establish that pLABTarget enables the effective exploitation of Cas9 targeting inL. lactis, while the broad-host-range vector used suggests that this toolbox could readily be expanded to other Gram-positive bacteria.IMPORTANCEMobile genetic elements inLactococcus lactisand other lactic acid bacteria (LAB) play an important role in dairy fermentation, having both positive and detrimental effects during the production of fermented dairy products. The pLABTarget vector offers an efficient cloning platform for Cas9 application in lactic acid bacteria. Targeting Cas9 toward mobile genetic elements enabled their effective curing, which is of particular interest in the context of potentially problematic prophages present in a strain. Moreover, Cas9 targeting of other mobile genetic elements enables the deciphering of their contribution to dairy fermentation processes and further establishment of their importance for product characteristics.

The insufficient duration of the drug’s contact with the vaginal mucosa does not provide adequate pH, conditioning the physiological biocenosis of the organ. This effect is hampered by numerous anatomic and physiological conditions which do not facilitate prolonged contact of the drug with the application site. This study has shown the impact of the used excipients and the ratio of lactic acid to chitosan on pH, dynamic viscosity and adhesiveness of methylcellulose gels obtained from powders. The addition of sodium alginate and excipients to these complexes allows various formulations to be obtained over a wide range of pH. Rheological investigations revealed an increase in the dynamic viscosity of preparations containing lactic acid complexed with chitosan and sodium alginate in comparison to gels without sodium alginate. Studies of the work of adhesion showed the effect of glycerol, 1,2-propylene glycol and their concentrations on the value of the work of adhesion.

The construction and selection of a suitable bijective function, that is, substitution, is now becoming an important applied task, particularly for building block encryption systems. Many articles have suggested using different approaches to determining the quality of substitution, but most of them are highly computationally complex. The solution of this problem will significantly expand the range of methods for constructing and analyzing scheme in information protection systems. The purpose of research is to find easily measurable characteristics of substitutions, allowing to evaluate their quality, and also measures of the proximity of a particular substitutions to a random one, or its distance from it. For this purpose, several characteristics were proposed in this work: difference and polynomial, and their mathematical expectation was found, as well as variance for the difference characteristic. This allows us to make a conclusion about its quality by comparing the result of calculating the characteristic for a particular substitution with the calculated mathematical expectation. From a computational point of view, the thesises of the article are of exceptional interest due to the simplicity of the algorithm for quantifying the quality of bijective function substitutions. By its nature, the operation of calculating the difference characteristic carries out a simple summation of integer terms in a fixed and small range. Such an operation, both in the modern and in the prospective element base, is embedded in the logic of a wide range of functional elements, especially when implementing computational actions in the optical range, or on other carriers related to the field of nanotechnology.

Angiogenesis plays an important function in tumor proliferation, one of the main angiogenic promoters being the vascular endothelial growth factor (VEGF) which activates specific receptors, particularly VEGFR-2. Thus, VEGFR-2 has become an essential therapeutic target in the development of new antitumor drugs. 1,2,4-triazoles show a wide range of biological activities, including antitumor effect, which was documented by numerous reports. In the current study the selection of 5-mercapto-1,2,4-triazole structure (1H-3-styryl-5-benzylidenehydrazino-carbonyl-methylsulfanil-1,2,4-triazole, Tz3a.7) was conducted based on molecular docking that emphasized it as suitable ligand for VEGFR-2 and EGFR1 receptors. Compound Tz3a.7 was synthesized and physicochemically and biologically evaluated thus revealing a moderate antiproliferative activity against breast cancer cell line MDA-MB-231.

There are eight columns in the Periodic Table. The eighth column is comprised of the rare gases, so-called because they are the rarest elements on earth. They are also called the inert or noble gases because, like nobility, they do no work. They are colorless, odorless, invisible gases which do not react with anything, and were thought to be unimportant until the early 1960s. Starting in that era, David Fisher has spent roughly fifty years doing research on these gases, publishing nearly a hundred papers in the scientific journals, applying them to problems in geophysics and cosmochemistry, and learning how other scientists have utilized them to change our ideas about the universe, the sun, and our own planet. Much Ado about (Practically) Nothing will cover this spectrum of ideas, interspersed with the author's own work which will serve to introduce each gas and the important work others have done with them. The rare gases have participated in a wide range of scientific advances-even revolutions-but no book has ever recorded the entire story. Fisher will range from the intricacies of the atomic nucleus and the tiniest of elementary particles, the neutrino, to the energy source of the stars; from the age of the earth to its future energies; from life on Mars to cancer here on earth. A whole panoply that has never before been told as an entity.

This paper presents a computer simulation showing how the gear contact ratio affects the dynamic load on a spur gear transmission. The contact ratio can be affected by the tooth addendum, the pressure angle, the tooth size (diametral pitch), and the center distance. The analysis presented in this paper was performed by using the NASA gear dynamics code DANST. In the analysis, the contact ratio was varied over the range 1.20 to 2.40 by changing the length of the tooth addendum. In order to simplify the analysis, other parameters related to contact ratio were held constant. The contact ratio was found to have a significant influence on gear dynamics. Over a wide range of operating speeds, a contact ratio close to 2.0 minimized dynamic load. For low-contact-ratio gears (contact ratio less than two), increasing the contact ratio reduced gear dynamic load. For high-contact-ratio gears (contact ratio equal to or greater than 2.0), the selection of contact ratio should take into consideration the intended operating speeds. In general, high-contact-ratio gears minimized dynamic load better than low-contact-ratio gears.

The Open University is the UK's foremost distance teaching university. For over twenty five years we have been presenting courses to students spanning a wide range of degree level and vocational subjects. Since we have no pre-requisites for entry, a major component of our course profile is a selection of foundation courses comprising one each in the Arts, Social Science, Mathematics, Technology and Science faculties. The Science Faculty's foundation course is currently undergoing a substantial revision. The new course, entitled “S103: Discovering Science”, will be presented to students for the first time in 1998.

Eleven published articles (4 reviews, 7 research papers) are collected in the Special Issue entitled “Organelle Genetics in Plants.” This selection of papers covers a wide range of topics related to chloroplasts and plant mitochondria research: (i) organellar gene expression (OGE) and, more specifically, chloroplast RNA editing in soybean, mitochondria RNA editing, and intron splicing in soybean during nodulation, as well as the study of the roles of transcriptional and posttranscriptional regulation of OGE in plant adaptation to environmental stress; (ii) analysis of the nuclear integrants of mitochondrial DNA (NUMTs) or plastid DNA (NUPTs); (iii) sequencing and characterization of mitochondrial and chloroplast genomes; (iv) recent advances in plastid genome engineering. Here we summarize the main findings of these works, which represent the latest research on the genetics, genomics, and biotechnology of chloroplasts and mitochondria.

Experimental screening of large sets of compounds against macromolecular targets is a key strategy to identify novel bioactivities. However, large-scale screening requires substantial experimental resources and is time-consuming and challenging. Therefore, small to medium-sized compound libraries with a high chance of producing genuine hits on an arbitrary protein of interest would be of great value to fields related to early drug discovery, in particular biochemical and cell research. Here, we present a computational approach that incorporates drug-likeness, predicted bioactivities, biological space coverage, and target novelty, to generate optimized compound libraries with maximized chances of producing genuine hits for a wide range of proteins. The computational approach evaluates drug-likeness with a set of established rules, predicts bioactivities with a validated, similarity-based approach, and optimizes the composition of small sets of compounds towards maximum target coverage and novelty. We found that, in comparison to the random selection of compounds for a library, our approach generates substantially improved compound sets. Quantified as the “fitness” of compound libraries, the calculated improvements ranged from +60% (for a library of 15,000 compounds) to +184% (for a library of 1000 compounds). The best of the optimized compound libraries prepared in this work are available for download as a dataset bundle (“BonMOLière”).
