Abstract The racing season is considered a critical period for racing pigeons due to the susceptibility to stress and weakening of the birds. One of the ways how to support their health and avoid problems involves supplementation of mineral-vitamin preparations based on selenium and vitamin E, which act as important antioxidants and protect cells against damage. This study investigated the influence of peroral supplementation of selenium at a dose of 0.3 mg Se.kg−1 feed dry matter (DM) in the form of Na2SeO3 and vitamin E 300 mg.ml−1 added to water at a rate of 4 ml.l−1 during 60 days. The supplemented group comprised 14 pigeons and their results were compared with a control group of 14 pigeons fed non-supplemented commercial feed. Blood samples were collected and examined at the beginning of the supplementation period, one day before a 300 km race and after the race. Pigeons from the supplemented group exhibited increased plasma levels of Se and vitamin E, as well as the activity of glutathione peroxidase (GSH-Px) before and after the 300 km race in comparison with the controls. Comparison of the activity of the enzymes: alanine aminotransferase (ALT), aspartate aminotransferase (ASP), alkaline phosphatase (ALP), and creatinine kinase (CK) at the beginning of the supplementation and one day before the race showed no changes. A significant (P < 0.05) increase in the activity of all investigated enzymes were observed after the race in both groups.

Abstract This study focuses on the effect of parenteral administration of Selenium (Se) and vitamin E on concentration of Se in plasma and the activity of glutathione peroxidase (GPx) in the blood of dairy cows during peripartal period and their effect on the reduction of clinical mastitis. From a 220 individuals Holstein herd in a two-four lactation-gestation cycle the control group (C), 1st (D1) and 2nd (D2) experimental group were selected. Every group consisted of 15 cows in the last phase of the pregnancy. All cows were fed with the diet containing 0.1 mg of Se per kg/DM. The blood samples from vena jugularis were collected approximately 21 days before calving (control sampling), 3 days, 12 days and 21 days after calving. On the day of control sampling and 12 days before calving in D1 group, cows were injected subcutaneously in the sprescapular region with preparation Selevit inj. a.u.v. at the doses of 48.4 mg/head of Se, and 550 IU/head of α-Tocoferol (α-Toc). In D2 group, cows were injected by the same preparation only on 21th day before calving with the same doses of Se and α-Toc. The increase in the concentration of Se in the plasma and activity GPx in blood in D1 group on the 3rd day and 12th day after calving were determined. Increase in plasmatic concentrations α-Toc on 3rd day after calving and reduction of occurrence of clinical mastitis (13.3%) as compared with control group were found

In acute myocardial infarction (AMI), generation of free radicals contribute to its pathogenesis. We evaluated the extent of oxidative stress and its amelioration in AMI patients after regular treatment and assessed its association with risk factors like dyslipidemia and diabetes mellitus (DM). Plasma MDA level increased (p<0.001) and erythrocyte GSH and plasma vitamin E levels decreased (p<0.001) in AMI patients. On the 5th day of regular treatment MDA level reduced (p<0.001) and erythrocyte GSH and vitamin E levels increased (p<0.001). The rise in MDA and fall in vitamin E levels were significant (p<0.001) in patients with both dyslipidemia and DM in comparison to patients without dyslipidemia or DM. It is concluded that an imbalance exists between oxidant and antioxidant molecules in AMI patients which shifts towards oxidative side and treatment restores this balance. There may be some association between oxidative stress in AMI and risk factors like dyslipidemia and diabetes mellitus.University Heart Journal Vol. 10, No. 2, July 2014; 60-65

&nbsp;A 2 &times; 3 factorial design experiment was conducted to evaluate the effect of adding lycopene <br />(0 and 75 mg/kg) and vitamin E (0.50 and 100 mg/kg) to the diet of chickens. Moreover, the study investigated growth traits, oxidative stability and chemical composition of leg meat and the vitamin content of meat and liver. The study was conducted using five hundred and forty Ross 308 male broilers that were assigned to one of the six dietary treatments. Significant interactions between lycopene and vitamin E additions affected the body weight of 21-days-old chickens (P = 0.005), the malondialdehyde content in fresh leg meat (P &lt; 0.001) and leg meat stored for 3 days at temperatures of 2.5 to 4&deg;C (P = 0.032), the cholesterol content in leg meat (P &lt; 0.001) and the lycopene content in liver (P = 0.006). The chickens with the highest body weight were fed 75 mg/kg of lycopene and 50 mg/kg of vitamin E. The vitamin E supplement increased the oxidative stability of fresh and stored leg muscle (P &lt; 0.001). The lowest mean cholesterol value (3.49 g/kg of dry matter) was found out in the meat from broilers that were fed 75 mg/kg of lycopene in contrast to broilers fed the control treatment without lycopene (3.93 g/kg of dry matter). Dietary vitamin E significantly reduced the fat content (P = 0.033) and increased the ash content of&nbsp;leg meat. The highest lycopene concentration in liver (2.82 mg/kg of dry matter) was in chickens that were fed the highest levels of vitamin E and lycopene in contrast with the control group (0.28 mg/kg of dry matter). &nbsp;

Abstract Background Inflammaging is a persistent, low−grade, sterile, nonresolving inflammatory state, associated with the senescence of the immune system. Such condition downregulates both innate and adaptive immune responses during chronic disorders as type II diabetes, cancer and hemodialysis, accounting for their susceptibility to infections, malignancy and resistance to vaccination. Aim of this study was to investigate hemodialysis inflammaging, by evaluating changes of several hemodialysis treatments on indoleamine 2,3-dioxygenase-1 activity and nitric oxide formation. Methods We conducted a randomized controlled observational crossover trial. Eighteen hemodialysis patients were treated with 3 different hemodialysis procedures respectively: 1) Low−flux bicarbonate hemodialysis, 2) Low−flux bicarbonate hemodialysis with vitamin E − loaded dialyzers, and 3) Hemodialfitration. The control group consisted of 14 hospital staff healthy volunteers. Blood samples were collected from all 18 hemodialysis patients just after the long interdialytic interval, at the end of each hemodialysis treatment period. Results Hemodialysis kynurenine and kynurenine/L − tryptophan blood ratio levels were significantly higher, when compared to the control group, indicating an increased indoleamine 2,3-dioxygenase-1 activity in hemodialysis patients. At the end of the low−flux bicarbonate hemodialysis with vitamin E − loaded dialyzers period, L − tryptophan serum levels remained unchanged vs both low−flux bicarbonate hemodialysis and hemodialfitration. Kynurenine levels instead decreased, resulting in a significant reduction of kynurenine/L − tryptophan blood ratio and indoleamine 2,3-dioxygenase-1 activity, when matched to both low−flux bicarbonate hemodialysis and HDF respectively. Serum nitric oxide control group levels, were significantly lower when compared to all hemodialysis patient groups. Interestingly, low−flux bicarbonate hemodialysis with vitamin E − loaded dialyzers nitric oxide serum levels from venous line blood samples taken 60 min after starting the hemodialysis session were significantly lower vs serum taken simultaneously from the arterial blood line. Conclusions The treatment with more biocompatible hemodialysis procedure as low−flux bicarbonate hemodialysis with vitamin E − loaded dialyzers, reduced indoleamine 2,3-dioxygenase-1 activity and nitric oxide formation when compared to both low−flux bicarbonate hemodialysis and hemodialfitration. These data suggest that low−flux bicarbonate hemodialysis with vitamin E − loaded dialyzers lowering hemodialysis inflammaging, could be associated to changes of proinflammatory signalling a regulated molecular level. The aim of the study was to investigate the influence of an increased intake of anthocyanins, contained in chokeberry juice, on the redox parameters in rowers performing a physical exercise during a 1-month training camp. The athletes were randomly assigned to receive 150 mL of chokeberry juice daily, containing 23 mg/100 mL of anthocyanins (supplemented group), or placebo (control group). Before and after the supplementation period, the subjects performed an incremental rowing exercise test. Blood samples were taken from the antecubital vein before each exercise test, 1 min after the test, and following a 24-h recovery period. After the supplementation period, TBARS concentrations in the samples collected 1 min after the exercise test and following a 24-h recovery period were significantly lower in the subjects receiving chokeberry juice than in the control group. In the supplemented group, glutathione peroxidase activity was lower in the samples collected 1 min after the exercise test, and superoxide dismutase activity was lower in the samples taken following a 24-h recovery, as compared to the subjects receiving placebo. These findings indicate that an increased intake of anthocyanins limits the exercise-induced oxidative damage to red blood cells, most probably by enhancing the endogenous antioxidant defense system.

Abstract Abstract 3362 The five channel computerized Whole Blood Aggregation instrument (Multiple Platelet Function Analyzer or Multiplate®), assesses platelet aggregation based on a modified whole blood impedance aggregation method. It permits platelet aggregation to be measured after adding commonly used agonists as arachidonic acid (ASPItest), ADP (ADPtest), collagen (COLtest), ristocetin (RISTOtest) and TRAP (TRAPtest), by detecting changes in electrical resistance in whole blood. Instrument handling is easy. Results are available within 9 minutes. Our objective was to evaluate the effect of aspirin (irreversible inhibitor of COX-1) and/or clopidogrel (irreversible inhibitor of the platelet P2Y12 receptor) on whole blood platelet aggregations induced by the 5 agonists using the Multiplate® in patients treated by aspirin and/or clopidogrel. Patients and controls. Two hundred and twenty two consecutive patients were recruited: 83 treated daily by 75 or 100 mg aspirin (group A); 42 treated daily by 75 mg clopidogrel (group C); 70 treated daily by 75 or 100 mg aspirin plus 75mg clopidogrel (group AC) and 27 who were daily on 100 mg aspirin before coronary intervention were tested 12 h after dual loading dose of aspirin between 75 et 500 mg and 75 to 900 mg clopidogrel according to cardiologists' recommendations: group loading aspirin-clopidogrel (LAC). Among group AC, 23 consecutive patients requiring intracranial stent placement of supra-aortic vessel were tested first at preoperative, without antiplatelet therapy, then 1 month after initiation of daily continuous dual antiplatelet therapy by 100 mg aspirin + 75mg clopidogrel. Ninety six volunteers without pathology or drugs influencing platelet functions constitute the normal control group (N). Blood samples. All patient and controls gave informed consent prior to blood sampling. Blood samples were collected by venipuncture or obtained from the arterial sheath directly into vacutainer Becton Dickinson tube containing 0.129M sodium citrate. Results. Patients under medication showing lower aggregation values than the arbitrary cutoff (fifth percentile of the aggregation in the normal control group was selected for each agonist) were classified as abnormal and having biological sensitivity to the agonist tested. Aggregation values above the cutoff with ASPItest or ADPtest for patients on antiplatelet agents were considered as a persistent platelet aggregation and as a biological resistance. According to the literature, resistance to aspirin was found in 8.6% of patients under aspirin alone or in combination and in 25.1% of patients under clopidogrel alone or in combination. Our main result shows an inhibition in platelet aggregation using ristocetin as agonist for 73.9% of patients taking aspirin alone, for 27.8% on clopidogrel and in 94% of patients receiving combination of the 2 drugs. This inhibition appears after aspirin + clopidogrel intake as we could observe it among patients candidates for intracranial stent placement tested before and after one month of treatment by dual antiplatelet therapy. This effect is not related to von Willebrand Factor (vWF) deficiency since the measurement of ristocetin cofactor activity, and vWF antigen carried out among 14 patients exhibiting an inhibition in whole blood platelet aggregation using RISTOtest were normal and unchanged before and after antiplatelet treatment. VWF is essential platelet-to-platelet interactions which is promoted by the binding of VWF with platelet-receptor glycoprotein IbIX (GPIbIX). Our results suggest: 1) aspirin inhibits the interaction of vWF to GP IbIX. This inhibition appears increased by the association of clopidogrel to aspirin. 2) a new mechanism of inhibition of the platelet function GPIbIX-vWF dependant conjointly to inhibition of cyclooxygenase by aspirin and P2Y12 receptor by clopidogrel.Table I:Biological sensibility according to the five tests (%) in the 4 groups testedGroup (n)ASPItestADPtestCOLtestRISTOtestTRAPtestA (83)84.312.038.373.98.4C (42)38.176.219.227.816.7AC (70)90.074.348.288.222.9LAC (27)100.074.163.0100.029.6 Disclosures: No relevant conflicts of interest to declare.

Vitamin E is familiar for its antioxidative properties for neutralizing oxide radicals and plays a role to keep cells safe from physiological damage and Selenium being an integral part of Glutathione peroxidase assists Vit.E and help chicks grow normal and healthy. In current study four sets of chicken were given doses of vit. E and Selenium (0, 1 ml /10L; 1 ml /7.5L; 1 ml /5L) in water. The Chicken were grouped in four sets as A,B,C and D. The experiment was continued sixty days and5ml blood samples were collected from all the groups A, B, C and D at interval of 15 days. In Fayoumi chicks a decrease was noted in the value of ALAT in all experimental sets excluding D on 3rd and 4th sampling. A significant increase (p>0.05) was observed in the amount of ASAT in all treated groups excluding group C which showed decreased (p less than 0.05) levels of ASAT on 2nd and 3rd sampling. The level of Alkaline Phosphatase reduced considerably in all experimental sets except group C where a slight increase (p>0.05) was observed on 2nd sampling. A considerable decrease was also observed in the amount of Bilirubin in all experimental sets except group C where an insignificant increase (p>0.05) was observed on 2nd sampling. The research suggested that vitamin E increases organism’s ability to cope infections besides hepatoprotective activity in desi layer bird Fayoumi.

The purpose of this study was to examine the effect of a 5-day creatine (CR) supplementation period on red blood cell (RBC) CR uptake in vegetarian and nonvegetarian young women. Blood samples were collected from lacto-ovo vegetarians (VG, n = 6, age 21.8 ± 1.9 yrs) and nonvegetarians (NV, n = 6, age 21.7 ± 1.9 yrs) before and after a 5-day CR loading period (0. 3 g CR/kg lean body mass/day), and from a control group of nonvegetarians (NV, n = 5, age 22.0 ± 0.7 yrs) who did not supplement with creatine. RBC and plasma samples were analyzed for the presence of creatine. Significant increases (p < .05) in RBC and plasma CR levels were found for vegetarians and nonvegetarians following supplementation. The initial RBC CR content was significantly lower (p < .05) in the vegetarian group. There was no significant difference between vegetarians and nonvegetarians in final RBC CR content, suggesting that a ceiling had been reached. As the uptake into both muscle and RBC is moderated by creatine transporter proteins, analysis of the uptake of CR into RBC may reflect the uptake of CR into muscle, offering an alternative to biopsies. This study aimed to determine the effect of fermented kelor leaves (Moringa oleifera) flour as additive in animal feed to increase the weight of boiler carcass. The samples used were 28 DOC strain Cobb CP-707 aged 1 day. This study was conducted using one way pattern complete randomized design with 4 treatments and 7 replications. Control group (P0) was fed with commercial feed; P1 was given 5% fermented kelor leaves flour;  P2 was given 7,5% fermented kelor leaves flour; and P3 was given 10% concentration of fermented kelor leaves flour. Feed replacement was conducted on 08.00 am and 5.00 pm everyday, and water was given ad libitum. Chemical content of flour before and after fermented was analyzed at Research Centre of Biotechnology and Biological Resources, IPB. Body weight measurement was carried out at Veterinary Public Health Laboratorium. Data were analyzed using ANOVA with SPSS 16.0. Mean value (±SD) of carcass weight for P0, P1, P2, and P3 were 938,71±180,83, 1011,86±101,64, 1010,71±126,69, and 907,43±190,35, respectively. This study showed that adddition of fermented kelor leaves flour for 21 days did not affect (P0,05) the weight of broilers carcass.

To investigate the role of certain neurohormones in agonistic behaviour, fights were staged between pairs of size-matched male shore crabs Carcinus maenas, and blood samples were taken immediately after the contests had been resolved. Samples were also taken from these crabs at rest (before and after fighting) and after walking on a treadmill. A control group of crabs also had samples taken on each experimental day. Concentrations of tyramine, dopamine, octopamine, serotonin (5-HT) and norepinephrine were determined in each blood sample using a gas chromatography/mass spectrometry (GC-MS) system. Norepinephrine was not detectable in any of the samples, but the standards were recovered. Tyramine values were not significantly different between the control group and the fought group, so tyramine does not appear to be important in agonistic behaviour. A comparison between the control and fought groups shows that fighting had an effect on the concentrations of octopamine, dopamine and 5-HT, but exercise only had an effect on octopamine levels, which showed a reduction from resting values in both winners and losers. Resting and post-fight concentrations of octopamine, dopamine and 5-HT were higher in winners than in losers. 5-HT concentration increased in the blood of fought crabs from resting values, whereas dopamine concentration decreased. In winners, octopamine concentrations decreased from resting values, but in losers octopamine levels increased from resting concentrations. The escalatory behaviour or intensity of fighting performed by winners and losers was related to dopamine levels but not to those of octopamine or 5-HT. Therefore, there appears to be a link between relative concentrations of these three amines (dopamine, octopamine and 5-HT) and fighting ability; the effects are not simply a result of activity. The better competitors have higher concentrations of these three amines at rest and after fighting.
