Children suffering from insulin-dependent diabetes mellitus (IDDM) were examined for unsaturation, that is, total quantity of double bonds in individual fractions of blood serum lipids was assessed. Lipid fractions were isolated by thin-layer chromatography. Unsaturation was assessed by ozonation method. The research was aimed at elucidation of biochemical criteria of the degree of metabolic disorders in children with IDDM and at development of methods for quantitative assessment of such disorders. Thirty-two inpatients with IDDM aged 7 to 15 were examined over time. Control group included 10 healthy age-matched children. Twenty children were examined during compensation stage (group 1), twelve during decompensation with ketoacidosis (group 2). Total unsaturation parameters were found reliably decreased in children with IDDM both during compensation and decompensation stages vs. those in healthy controls. A relative decrease of unsaturated fatty acids in the fraction of uneterified fatty acids (p0.05) was characteristic of group 2, with the content of their fragments in triglyceride fraction being the maximal (p0.05). The pattern of total unsaturation distribution in serum lipid fractions may serve as an indicator for metabolic disorders and decompensation in IDDM.

AbstractEight mid-lactation Holstein-Friesian dairy cows were used in an experiment to investigate whether poor transfer to milk of (n-3) long chain (≥C20) poly-unsaturated fatty acids (n-3 LCPUFA), from diets containing fish oil, is related to their mode of transport in plasma lipoproteins. The cows were split into two groups of four, each of which was housed with 24 other cows which did not provide blood data in this experiment. All cows received a basal diet ad libitum consisting of (kg/t dry matter) silage (539), grain distillers’ grains (148), rolled barley (248) and soya (65). All cows also received a mid-day meal of 2·76 kg per cow per day of molassed sugar-beet shreds. The control group (C) received no oil supplement but those in the treatment group (F) received 300 g per cow per day of fish oil absorbed onto the sugar-beet shreds. Fish oil inclusion progressively depressed milk fat concentration of the four cows from each group over the 3 weeks of the experiment so that levels were approximately 20 g/kg lower for cows in treatment F than for those in C (P < 0·05). Mean concentrations over all cows of the long-chain (≥C20, n-3) poly-unsaturated fatty acids in milk fat (g/100 g) were 0·12 and 0·54 for treatments C and F respectively, whilst corresponding output in milk (g/day) increased from 116 to 3·91, an average efficiency of transfer of 4·6%. The fatty acid compositions of the lipoproteins were measured in plasma samples from the four cows from each group after 3 weeks on the experimental diets The proportions (g/100 g fatty acids) of C20: 5 (n-3) in cholesteryl (CE) and phospholipid (PL) of high-density lipoprotein (HDL) in the plasma were respectively increased from 0·97 to 4·39 (P < 0·05) and from 1·15 to 4·74 (P < 0·01) by the fish-oil supplementation. For C22: 6 (n-3), the proportions in CE and PL of HDL respectively increased (P < 0·001) from 0·02 to 0·23 and from 0·16 to 2·18 as a result of the supplementation. The mean proportions (g/100 g fatty acids) of C20: 5 (n-3) and C22: 6 (n-3) in the triglyceride fraction of very low-density lipoprotein were 1·05 and 2·3 respectively and were not significantly altered by fish-oil supplementation (P > 0·05). It is suggested that the low transfer efficiency of (n-3) LCPUFA to the milk is partly due to their transport in the plasma largely as components of HDL lipids which are unavailable to the mammary gland.

Введение. Сосудистые осложнения сахарного диабета (СД) являются одной из ведущих причин повышения смертности пациентов трудоспособного возраста. Предполагается, что индуцированный гипергликемией окислительный стресс и нарушение антиоксидантной защиты играют роль в патологическом механизме повреждения сосудов, частично за счет влияния оксида азота (NО). Цель исследования: уточнение взаимосвязей в системах асимметричного диметиларгинина (АДМА) и NO у пациентов с цереброваскулярными заболеваниями (ЦВЗ) на фоне СД 2-го типа (СД-2). Материалы и методы. Обследованы 72 пациента с ЦВЗ со стенозирующим поражением внутренней сонной артерии вне острого периода: группу 1 составили 39 пациентов (18 мужчин и 21 женщина) с СД-2 в возрасте 65 [58; 72] лет; в группу 2 вошли 33 больных (15 мужчин и 18 женщин) без СД-2 в возрасте 66 [56; 74] лет. Контрольную группу составили 30 добровольцев (16 мужчин и 14 женщин) без проявлений церебральной ишемии и нарушений углеводного обмена, с нормальными значениями индекса массы тела, некурящие, в возрасте 62 [50; 66] лет. Проводилось клиническое обследование, нейро- и ангиовизуализационное исследование, спектр биохимических исследований крови, в том числе определение содержания АДМА и показателей системы NO. Результаты. В группе 1 содержание нитрата, нитрита и NO составило 62,1 [56; 68] мкмоль/л, 48,5 [26; 52] мкмоль/л и 13,6 [9; 23] мкмоль/л соответственно, что достоверно отличалось от значений этих показателей в группе 2 — 58,3 [45; 64] мкмоль/л, 39,6 [26,0; 42,3] мкмоль/л и 18,7 [16,1; 24,7] мкмоль/л соответственно. Отмечен также более высокий уровень AДМА в крови у пациентов с ЦВЗ в сочетании с СД-2 — 0,42 [0,21; 0,53] ммоль/л. Заключение. Обнаружена взаимосвязь между уровнями AДМА и NO при ЦВЗ на фоне СД-2. Это требует продолжения исследований биомаркеров повреждения сосудистой стенки для определения их места в патогенезе ишемических церебральных осложнений СД-2. Background. Vascular complications of diabetes mellitus (DM) are one of the leading causes of increased mortality in patients of employable age. Hyperglycemia-induced oxidative stress and impaired antioxidant protection have been suggested to play a role in the pathological mechanism of vascular damage, in part due to the effects of nitric oxide (NO). Objectives: clarification of relationships in the systems of asymmetric dimethylarginine (ADMA) and NO in patients with cerebrovascular diseases (CVD) and type 2 diabetes (DM-2). Patients/Methods. We examined 72 CVD patients with stenosing lesions of the internal carotid artery outside the acute period: group 1 consisted of 39 patients (18 men and 21 women; 65 [58; 72] years old) with DM-2; group 2 consisted of 33 patients (15 men and 18 women; 66 [56; 74] years old) without DM-2. The control group consisted of 30 volunteers (16 men and 14 women; 62 [50; 66] years old) without manifestations of cerebral ischemia and carbohydrate metabolism disorders, with normal body mass index, non-smokers. A clinical examination, neuro- and angio-imaging study, a spectrum of biochemical blood tests, including the concentration of asymmetric dimethylarginine (ADMA) and indicators of NO system were carried out. Results. In group 1, the content of nitrate, nitrite and NO was 62.1 [56; 68] μmol/l, 48.5 [26; 52] μmol/l and 13.6 [9; 23] μmol/l, respectively, that significantly differed from the content of these parameters in group 2 — 58.3 [45; 64] μmol/l, 39.6 [26.0; 42.3] μmol/l and 18.7 [16.1; 24.7] μmol/l, respectively. Noted also a higher blood level of ADMA in patients with CVD combined with DM-2 — 0.42 [0.21; 0.53] mmol/l. Conclusions. A relationship was found between ADMA and NO levels in CVD patients with DM-2. This requires further studies of biomarkers of vascular wall damage to determine their place in the pathogenesis of ischemic cerebral complications of DM-2.

The effects of grazing lactating cows in mountainous areas for 12 and 24 h compared with the confined indoor system were evaluated by examining the overall milk fatty acid and cortisol. Twenty-one dairy cows were allocated to three treatment groups: (1) control (confined management system in a free-stall barn; TMR based), (2) grazing for 12 h (12hG; TMR plus grazing pasture), and (3) grazing for 24 h (24hG; pasture-based feeding system). Dry matter intake was higher in the control and 12hG groups than in the 24hG group. The yields of total milk and the 3.5% fat-corrected milk were the lowest in the 24hG group. Milk fat was the highest in the 24hG group and higher in 12hG compared with the control group. Milk protein and lactose levels were the highest in the 12hG group. The highest somatic cell count was observed in the 24hG group. The saturated fatty acid levels were higher in the control group compared with the 12hG and 24hG groups. There was no difference in overall mono-unsaturated fatty acids between 12hG and 24hG groups. Poly-unsaturated fatty acids were higher in the 12hG group compared with the control and 24hG groups. There was no difference in omega-6 (ω-6) fatty acids among the groups, and omega-3 fatty acids were higher in the 12hG group than in the control group. Milk cortisol was the highest in the 24hG group and higher in the control group compared with the 12hG group. Taken together, grazing for 12 h is advisable for farms that have access to mountainous areas to improve the milk fatty acid profile and decrease the stress levels in high-yielding Holstein lactating cows.

This article discusses the effect of the disintegration of the cell wall of the microalgae Chlorella sorokiniana on the output of the lipid fraction. The biomass of the microalgae Chlorella sorokiniana was grown under laboratory conditions in special photobioreactors at a temperature of 25 °C, with a constant aeration of a mixture of carbon dioxide and air at a rate of 1.5 liters/min, illumination 2200-2800 Lx. Nutrient medium for cultivation contained macro – and micronutrients for high-speed growth of microalgae. Selection of optimal cultivation parameters allows obtaining biomass with desired properties. Disintegration was carried out with the homogenization of biomass and under the influence of microwave radiation. Extraction of lipids was carried out on a semi-automatic extractor according to the Randall method, using organic solvents. The output of the lipid fraction without treatment was 10.18% after the destruction of the cell wall 14.45% with the homogenization of biomass and 13.85% under the influence of microwave radiation. A qualitative analysis of the lipid fraction, carried out under gas chromatography, obtained under various conditions showed that there was no significant difference in composition from the disintegration method. Lipid fractions (more than 50%) in both cases consist mainly of unsaturated fatty acids, of which irreplaceable unsaturated fatty acids constitute more than 18% for both samples. The residual biomass formed after the extraction of the lipid fraction can be used as fertilizer in the plant, for the manufacture of sorption materials for the purification of industrial water and as a biofuel. The purpose of this study was to study the effect of cell wall disintegration on the output of the lipid fraction and qualitative composition.

Abstract. INFLUENCE OF HORMONAL DISORDERS ON ENDOTHELIAL DYSFUNCTION IN PATIENTS WITH ARTERIAL HYPERTENSION AND COMORBIDE ENDOCRINOPATHIES Bilovol O.M., Nemtsova V.D., Ilchenko I.A., Zlatkina V.V. Purpose: to investigate the effect of hormonal changes on endothelial dysfunction (ED) in patients with a comorbid course of hypertension (H), type 2 diabetes mellitus (T2DM) and subclinical hypothyroidism (SHT). Methods: 183 patients with  H stage II were divided into 3 groups: Group 1 (n=50) - with isolated H (comparison group); Group 2 (n=63) - with a combined course of H and T2DM; Group 3 (n=70) - with comorbidity of H, T2DM and SHT. Blood pressure levels, carbohydrate, lipid and thyroid metabolism, plasma insulin concentration, insulin resistance (IR) the HOMA-IR index, vascular endothelial growth factor (VEGF-A) plasma levels were investigated. Results: Dyslipidemia was more pronounced in group 2 than in group 1. The addition of SHT was accompanied by a tendency to increase all the atherogenic lipids. IR was observed in all patients groups and was significantly higher than in control group (p<0.05). Significant increase of VEGF-A levels in all patients groups in comparison with the control (p<0.05) was found. In group 2 VEGF-A was lower than in group 1, which is probably due to the protective effect of metformin. Analysis  of the influence of thyroid dysfunction degree on ED revealed significant increase of VEGF-A levels in TSH>6.0 μMU/ml subgroup (352.55±17.64 pg/ml vs 461.74±20.13 pg/ml (p<0.05)). Conclusion: Hormonal disorders contribute to aggravation of endothelial dysfunction in patients with hypertension and comorbid endocrinopathies - type 2 diabetes mellitus and subclinical hypothyroidism. Even minor decrease in thyroid function lead to the progression of endothelial dysfunction. Мета: дослідити вплив гормональних змін на ендотеліальну дисфункцію (ЕД) у пацієнтів з коморбідним перебігом артеріальної гіпертензії (АГ), цукрового діабету 2 типу (ЦД2Т) тасубклінічного гіпотиреозу (СГТ). Матеріали та методи: 183 пацієнта з АГ II стадії були розділені на 3 групи: 1-а група (n=50) - з ізольованою АГ (група порівняння); Група 2 (n=63) - з поєднаним перебігом АГ та ЦД2Т; Група 3 (n 70) – з комбінованим перебігом АГ, ЦД2Т і СГТ. Вивчали рівні артеріального тиску, показники вуглеводного, ліпідного і тиреоїдного обміну, концентрацію інсуліну в плазмі, індекс інсулінорезистентності (ІР) - HOMA-IR, рівні васкулоендотеліального фактора росту (VEGF-A) в плазмі. Результати. Ступінь дисліпідемії у 2-й групі була більш вираженою, ніж в 1-й. Приєднання СГТ супроводжувалося тенденцією до збільшення всіх атерогенних фракцій ліпідів. ІР спостерігалася у всіх групах пацієнтів і була достовірно більше, ніж у контрольній групі (р<0,05). Виявлено достовірне підвищення рівнів VEGF-A у всіх групах пацієнтів в порівнянні з контролем (р<0,05). В 2-й групі рівні VEGF-A були нижче, ніж в 1-й групі, що, ймовірно, пов'язано з протективним ефектом метформіну. Аналіз впливу ступеня гіпофункції щитовидної залози на ЕД виявив значне збільшення рівнів VEGF-A в підгрупі TSH> 6,0 мкМ / мл (352,55 ± 17,64 пг / мл і 461,74 ± 20,13 пг / мл відповідно, р <0,05). Висновки. Гормональні порушення сприяють погіршенню ендотеліальної дисфункції у пацієнтів з артеріальною гіпертензією та супутніми ендокринопатіями - цукровим діабетом 2 типу та субклінічним гіпотиреозом. Навіть незначне зниження функції щитовидної залози призводить до прогресування ендотеліальної дисфункції. Результаты. Степень дислипидемии во 2-й группе была более выраженной, чем в 1-й.  Присоединение СГТ сопровождалось тенденцией к увеличению всех атерогенных фракций липидов. ИР наблюдалась во всех группах пациентов и была достоверно больше, чем в контрольной группе (р<0,05). Выявлено достоверное повышение уровней VEGF-A во всех группах пациентов по сравнению с контролем (р <0,05). Во 2-й группе уровни VEGF-A были ниже, чем в 1-й группе, что, вероятно, связано с протективным эффектом метформина. Анализ влияния степени дисфункции щитовидной железы на ЭД выявил значительное увеличение уровней VEGF-A в подгруппе TSH> 6,0 мкМ/мл (352,55 ± 17,64 пг / мл и 461,74 ± 20,13 пг / мл соответственно, р<0,05). Заключение. Гормональные нарушения способствуют ухудшению эндотелиальной дисфункции у пациентов с артериальной гипертензией и сопутствующими эндокринопатиями - сахарным диабетом 2 типа и субклиническим гипотиреозом. Даже незначительное снижение функции щитовидной железы приводит к прогрессированию эндотелиальной дисфункции. BACKGROUND: Waste cooking oil (WCO) is overused for frying food can be a series of chemical changes, such as rancid, oxidation, and decomposition. Most of the secretion of toxic compounds are a product of oxidizing fatty acids, especially double-unsaturated fatty acids. The intervention of stirrer chamber unit and utilization of chicken egg’s shell as an absorbent is known to be able to improve the quality of WCO, includes reducing the number of peroxide and the free fatty acids (FFA). AIM: The purpose of this study was to analyze the effect of the powdered chicken egg’s shell on the number of peroxide and the WCO’s FFA through the stirrer chamber. METHODS: The research design used a simple experiment type post-test only Control Group Design. The subject was divided randomly into two groups, the treatment was given to one group as a control group and another group (other treatment) as an experimental group. The WCO sample called “Jelantah” for each treatment was 100 ml as much as 48 samples. The replication was carried out twice with 24 treatments of the mass of egg’s shell powder that was 0 g, 5 g, 10 g, and 15 g at a stirring time for 15 min, 30 min, and 45 min before and after heating 35°C using 150 rpm of stirring speed. The sampling technique used purposive sampling and the data were analyzed by Anova statistical test. RESULTS: Based on the result from this research showed that the original WCO contained the number of peroxide and FFA exceeded the quality standard of SNI 3741 (2013) that is 16.7 MekO2/kg and 0.69% in sequence. The average number of peroxide of control group before heating was 15.31 MekO2/kg and after heating was 17.4 MekO2/kg, while the FFA before heating was 0.61%, and after heating was 0.71%. The number of peroxide of the treatment group before heating was 12.83 MekO2/kg and after heating was 6.98 MekO2/kg, while the FFA content before heating was 0.46% and after heating was 0.25%. CONCLUSION: It was concluded that chicken egg’s shell powder could minimize the content of peroxide and WCO’s FFA through the stirrer chamber.
