ABSTRACT: A novel cost-effective in vitro regeneration protocol has been evolved for the therapeutically important Ocimum citriodorum Vis. In the present study, table sugar (3%) and isabgol (Psyllium husk) (3.5%) were used as an alternate source of carbon and gelling agent respectively in Murashige and Skoog’s (MS) medium. The explant used in the current study was nodal segment. A significant observation revealed that all the cultures resulted in shoot induction and maximum number of shoots/ culture (6.04) and their average length (2.15 cm) was obtained on modified MS-medium supplemented with table sugar, isabgol and BAP. However, best root induction (95.83%) was obtained on ½ MS-medium augmented with table sugar (3%) , isabgol (3.5%) and NAA. An increase in average number of roots per shoot (6.91%) as well as average root length (2.73 cm) was also observed in the same modified medium. The in vitro regenerated plantlets were successfully transferred to the field and no notable variation was observed in their morphology. The overall cost of culture medium for in vitro propagation of O. citriodorum Vis. was reduced significantly by 92.69% when agar and sucrose were replaced by isabgol and table sugar, respectively.

Introduction: The present research work was undertaken with a view to developing a suitable protocol for in vitro plant regeneration of economically important plant (Glycine max) (Bangladesh Agricultural Research Institute BARI- 5) variety, via both direct and indirect organogenesis from in vitro grown seedlings. Methods: For micropropagation explants were cultured on MS and half strength Murashige and Skoog (MS) medium supplemented with various plant growth regulators (cytokinins and auxins). In the present study for inducting of callus, among 3 different hormone combinations, the suitable medium was 3.32 mg/L 2, 4-D containing MS medium and the callus was deep green in color. Different type of media like MS, 1/2 MS and MS with different (6-Benzyl Amino Purine) BAP concentration was used for seed germination of Glycine max. 100% of seed germination was observed in MS +1 mg/L BAP containing the medium. Results: In the present investigation, different concentration of cytokinins and auxins{BAP, 2, 4-D, and Naphthalene Acetic Acid (NAA)} were used individually or in combinations with MS medium to observe their effect on multiple shoot regeneration from the cotyledonary nodal segment. 100% shoot formation from cotyledonary nodal segment was recorded in 1.5 mg/L BAP and 0.15 mg/L BAP + 0.025 mg/L NAA containing MS medium, the best number of shoot was 10.9±2.0 found in MS + 1.5 mg/L BAP containing medium and highest length of shoot was 2 cm recorded in 1.5 mg/L BAP + 0.3 mg/L (different concentrations of Giberrellic acid) GA3 containing MS medium. In addition, for root induction in vitro raised well developed and elongated shoots were excised and cultured on MS and 1/2 MS medium supplemented with various concentration of Indole-3-Butyric acid (IBA). It was observed that MS medium containing 0.1 mg/L IBA and 1/2 MS medium containing 0.25 mg/L IBA was optimal for root induction. In which 100% shoots rooted well within 13 days of culture. The highest average number of roots per shoot was 6 recorded in MS +0.5 mg/L IBA containing the medium and highest average length of root was 8 cm recorded in 0.1 mg/L IBA containing MS medium. Conclusion: The most effective surface sterilization treatment for explants of Glycine max has been found in 0.1% HgCl2 solution for 15 minutes.

An in vitro multiple shoot regeneration protocol has been developed for Centella asiatica L. Urban from the nodal and shoot tips explants using MS medium with different concentrations and combinations of growth regulators. Multiple shoot regeneration was observed from both the explants on MS containing either BAP alone or in combination with IAA, NAA and Kn. Of the two explants, nodal segment showed comparatively better response towards multiple shoot regeneration. Maximum multiple shoots were found on MS supplemented with 1.0 mg/L BAP and 0.4 mg/L NAA. For root induction, well-developed shoots were excised and cultured on both MS and half strength of MS with various concentrations of IBA, IAA and NAA. Profuse healthy rooting was obtained on MS medium containing 0.2 mg/L IBA. An efficient in vitro regeneration system was developed for Rauvolfia serpentina L. through direct and indirect organogenesis from nodal and leaf explants. Among the different growth regulators, MS medium supplemented with 2.0 mg/l BAP, 0.5mg/l IAA and 0.02mg/l NAA found best for the multiple shoot formation from nodal segments. In this combination 98% explants produced multiple shoots and the average number of shoots per explants is 13∙4. The frequency of callus induction and multiple shoot induction from leaves was highest 88% in MS medium supplemented with 2.0 mg/l BAP, where mean number of shoots/explants was 12.5. The highest frequency of root induction (80%) and mean number of roots/plantlets (10) were obtained on half strength of MS medium containing 0.2 mg/l IBA. In vitro study was carried out in an important medicinal plant Tinospora cordifolia (Willd.) Miers belonging to the family: Menispermaceae. Vegetative parts such as stem, leaf and nodal explants were excised from an elite in vivo grown mature plant and thereafter cultured on Murashige-Skoog (MS) medium supplemented with different hormonal concentrations for callus induction and organogenesis. Callus formation occurred from nodal segments, leaf and inter-node explants when planted on different combinations of hormones. Tinospora cordifolia showed response for in vitro shoot growth from the nodal segment. The best shoot growth was observed on MS medium supplemented with kinetin (1.5 mg/l). Similarly, the best result for root induction was obtained on MS medium supplemented with 6-benzylaminopurine (1.0 mg/l) and naphthaleneacetic acid (2.5 mg/l). A standard protocol was established for rapid in vitro propagation of watermelon (Citrullus lanatus Thumb.) from nodal explants of field grown plant. Multiple shoot proliferation was achieved from nodal explants on MS medium supplemented with 1.0 mg/l BAP + 0.2 mg/l NAA within 30 days of inoculation. The elongation of shoots was obtained on the same medium. Highest percentage of root induction was achieved on MS medium supplement with 1.0 mg/l IBA within 25 days of culture. Well rooted plantlets were transferred to small pots and after proper acclimatization the plantlets were transplanted in the field condition, where 80% plantlets were survived and grew successfully. An efficient rapid in vitro regeneration protocol was described from nodal segment, leaf and petiole explants. MS medium supplemented with 1.0 mg/l BAP and 0.5 mg/l IAA was found best for the multiple shoot formation from nodal segments. In this combination 99% explants produced multiple shoots and the average number of shoots per explants was 20.1 ± 1.96. For petiole and leaf explants best response was observed on MS supplemented with 2.0 mg/l BAP, 1 mg/l IAA and 0.5 mg/l Kn. Petiole explants produced highest mean number of shoots/explant (22.9 ± 1.728) among the three explants when the explants were cultured on MS with 2.0 mg/l BAP, 1 mg/l IAA and 0.5 mg/l Kn. The highest frequency of root induction (100%) and mean number of roots/plantlets (11.75) were obtained on MS. The experiment was conducted to develop an efficient protocol for in vitro regeneration of mungbean (Vignaradiata) on the aspect of regeneration potentiality of two mungbean varieties (BINA mung 5 and BINA mung 7) as influenced by different combinations of growth regulators supplemented with MS medium. Cotyledon explant of both varieties was used for the present study. Data were collected for various characters of callus initiation, shoot regeneration and root proliferation. Initiation of callus (%) and required days for its initiation and weight of callus were influenced significantly due to the effect of varieties where BINA mung 5 produced more callus induction (40.36%) at minimum requiring time (18.27 days) including heavier sizes of callus (1.54 g) than BINA mung 7 when BINA mung 5 further recorded the longest root (2.92 cm) compare to BINA mung 7. Effect of treatments of the present study were significantly influenced the whole characters regarding callus culture, shoot regeneration and root proliferation. The highest percentage of callus (88.44%) within minimum time (12.53 days) including larger sizes callus (3.521 g) were produced in 1.0 mg L–1 BAP + 2.5 mg L–1 NAA among the treatments while the highest percentage of regenerated shoot (83.44%) at minimum requiring time (17.59 days) and more shoots (7.69 callus–1) were obtained in 1.0 mg L–1 BAP + 2.0 mg L– 1 NAA. Root induction (82.50%), number of roots plantlet–1 (8.469) with minimum requiring time for initiation (14.13 days) and root length (5.250 cm) were the highest in 0.2 mg L–1 IAA + 1.0 mg L–1 kinetin + 0.2 mg L–1 BAP. Incase of interaction, percentage of callus initiation (89.38 %) was the highest in BINA mung 5 treated by 1.0 mg L–1 BAP + 2.5 mg L–1 NAA at requiring minimum time (12.38 days) while same treatment produced the larger callus (3.581 g) among the interactions. The highest percentage (84.38%) and number (7.813 callus–1) of shoot with minimum requiring time (17.50 days) were found from BINA mung 5 treated by 1.0 mg L–1 BAP + 2.0 mg L–1 NAA. Similarly, the longest shoot (5.58 cm) was produced from the BINA mung 5 treated by 1.0 mg L–1 BAP + 2.0 mg L–1 NAA. However, root induction (%), roots plantlet–1, days required for root initiation and root length were statistically similar among the whole interaction treatments due to non significant variation. This result mentioned that the variety BINA mung 5 was better than BINA mung 7 for callus induction, shoot regeneration and root initiation while 1.0 mg L–1 BAP + 2.5 mg L–1 NAA, 1.0 mg L–1 BAP + 2.0 mg L–1 NAA and 0.2 mg L–1 IAA + 1.0 mg L–1 kinetin + 0.2 mg L–1 BAP supplemented with MS medium were the best combinations for better callusing, higher ability of shoot regeneration and root proliferation.DOI: http://dx.doi.org/10.3329/jesnr.v7i2.22203 J. An in vitro regeneration protocol of Adhatoda vasica has been developed using excised nodal segments and juvenile leaves for multiple shoots regeneration directly or through callus induction. Explants were cultured on MS medium with different concentrations of IAA, NAA, BAP, GA3 and Kn singly or in combinations. MS medium supplemented with BAP (10.0 mg/l) was found best for multiple shoot formation, in which 93.33% explants produced multiple shoots. After two months, maximum number of multiple shoots were 10.6 ± 1.82, highest length of plantlets was 5.2 ± 2.20 cm. 100% calli formation were observed on MS medium supplemented with IAA (0.05 mg/l) + NAA (0.05 mg/l) + BAP (1.0 mg/l). Callus initiation started after 14 days and gave light green colored callus. Best callus mediated shoot regeneration was found on MS+10.0 mg/l BAP medium. Root induction of in vitro raised shoots was best on ½ MS + IBA (1.0 mg/l). Well rooted plantlets were transferred to plastic pots containing garden soil and compost in a ratio of 2:1 for hardening. Current Status of Saussurea heteromalla was investigated in Galiyat areas of District Abbottabad, viz., Jahaffar, Seri, Beeran Gali, Banj, Haryala, Daryala Gali, Sarbhanna, Barriyan, Akhreela and Broangiala. Saussurea heteromalla was found common in Seri, Sarbhanna and Barriyan; endangered in Haryala, Jahaffar, Banj, Daryala Gali and Beeran Gali and absent in Akhreela and Broangiala. In vitro regeneration of Saussurea heteromalla (D. Don.) Hand-Mazz on MS media was conducted and shoots were developed on full strength MS medium supplemented with 1 mgL-1 GA3. The developed shoots were transferred for root induction to half strength MS medium fortified with various concentrations of Indole butyric acid (IBA) and α-Naphthalene acetic acid (NAA) i.e. T1 (control), T2 (2 mgL-1 IBA), T3 (3 mgL-1 IBA), T4 (4 mgL-1 IBA), T5 (1 mgL-1 NAA), T6 (2 mgL-1 NAA) and T7 (3 mgL-1 NAA). Maximum mean shoot length (6.3 cm), mean number of leaves (7), mean number of nodes (5.25); highest root emergence percentage (71%), means root length (1.5 cm), mean number of roots (3) and highest survival rate (100%) was recorded in treatment T4. However, treatments T5 and T6 also seem to be effective for in vitro rooting of Saussurea heteromalla. Treatments T2, T3 and T7 showed minimum root growth. It was concluded that IBA at higher concentration is more effective for in vitro rooting and better shoot growth of Saussurea heteromalla whereas NAA also initiate rooting but at lower concentration.

An experiment was conducted to develop an efficient protocol for in vitro regeneration of plantlets from hypocotyl segments and cotyledonary explants of Abelmoschus esculentus L. Monech. In this investigation the cultivar OK-285 of Abelmoschus esculentus L. Monech was used. At first somatic embryogenic calli were induced using MS medium supplemented with different concentrations of auxin and cytokinin singly or in combination. The cotyledonary explants showed the best callus induction rate (87.9%) in MS medium containing 1.5mgl-l 2.4-D + 0.1mgl-l BAP hormonal concentration while hypocotyl segments showed 82.6% callus induction rate in the same medium. For shoot formation, calli were subcultured on MS solid medium and the hypocotyl segments showed the best result (72.1%) in MS medium containing 2.0 mgl-l BAP + 0.1 mgl-l IAA and the mean number of shoots per callus was recorded 4.2. For root induction from shoots MS and ½MS media were used. The highest 63.5% microshoots initiated roots in ½ MS + 0.1 mgl-l IBA medium and the highest mean number of root was 4.8. Rooted shoots were acclimated and successfully established into soil under natural condition with 70% survival.
