The objective of this work was to investigate the germination of hybrid sunflowers BRS191 and C11 as a means of lowering phytic acid (PA) content by enhancing the activity of endogenous phytase and acid phosphatase. The concentration of PA in hybrid sunflower achenes varied from 2.16 to 2.83g/100g of sample (p < 0.05). The phytase and acid phosphatase activities of sunflowers BRS191 and C11 were the highest on the 4th and 5th days of germination, respectively, with the release of the phosphorus. These results indicated that hybrid sunflower PA reduced and enhance phytase activity at distinct germination periods, which could open up the possibility of applying these enzymes in the control of PA content in cereals, thus improving their nutritional value.

Abstract.  Cereals and legumes used as animal feed containing phytic acid which cannot be absorbed by the digestive tract of monogastric animals because phytic acid (C6H18O24P6) is an antinutrient that binds approximately 80% P in feed, also binds to proteins, vitamins and minerals (Mg++, Fe++, Zn++, Mn++, Ca++). Then, one option to overcome this problem is the application of phytase enzymes from various sources, including those produced by bacteria. Phytase enzymes can hydrolyse phytic acid in the feed. This study was aimed to determine the growth phase of Burkholderia sp. HF.7 and optimisation of production of phytase activity from variations of Phytate Production Media (PPM) media. This research used as a descriptive approach. The study design used a completely randomised design (CRD) with a factorial pattern consisting of 2 factors, each variation in phytate sources: calcium (Ca) phytate, rice bran, corn bran, and soybeans. Nitrogen source: (NH4) 2SO4, yeast extract, and peptone. Burkholderia sp. HF.7 Growth phase as a standard for phytase production is the 62-hour stationary phase with an OD value of 2.060 log/cell. The optimum phytase production in the variation of PPM media is soybean-peptone with a protein content value of 46.5 mg/mL and an activity value of 8.20 U/mL under conditions of pH 7 with incubation of 37oC for 62 hours. So, phytase activity produced by PPM of cereal crops has a higher current value compared to PPM Ca-phytate media with low activity value.Keywords: Burkholderia, Phytate, Phytase, enzymes.

Dairy cows can have different degrees of hypocalcaemia around calving. Lowering dietary Ca availability before calving can prevent it. Rice bran, treated for lower rumen degradability of phytic acid can reduce dietary availability of Ca. During 3 periods of 3 weeks, 113 multiparous cows calved in a single close-up group, which was fed first a control diet, then 140 g/kg DM of rumen-protected rice bran, and at last the control diet again. Cows joined the group 3 weeks before expected calving date and left it at calving. Blood samples were taken weekly before parturition and 0, 6 and 12 h after calving, as well as 3 and 28 d in lactation. Serum was analysed for Ca, Mg, and P. Rice bran introduction produced a transient serum Ca decrease. Rice bran feeding reduced serum P and its withdrawal reduced serum Mg. Serum Ca at calving, nadir of serum Ca and serum Ca the first 3 d after calving was higher in cows calving during rice bran feeding. Serum P decreased less and recovered faster after calving when cows had been fed rice bran. Rumen-protected rice bran reduced dietary availability of Ca and induced adaptation of Ca metabolism resulting in improved Ca and P homoeostasis at calving.
