Two possible modes of action of exo-D-galacturonanase from carrot (E.C. 3.2.1.67) were investigated; this enzyme catalyses the sequential hydrolytic cleavage of pectants and oligogalacturonans by a terminal action from the nonreducing end of the molecule. The experiments indicate that the investigated exo-D-galacturonanase degrades these substrates by a predominantly multi-chain mechanism. Distribution of degradation products of oligomeric substrates (hexa- and pentagalacturonide) under an optimal condition for the action of the enzyme (pH and temperature) indicates that a multi-chain enzyme attack with a prevalent simple collision is involved. Results of the enzyme degradation kinetics are in a good agreement with the above-mentioned presumption.

AbstractIn this study, a new α-glucosidase gene from Thermoanaerobacter ethanolicus JW200 was cloned and expressed in Escherichia coli by a novel heat-shock vector pHsh. The recombinant α-glucosidase exhibited its maximum hydrolytic activity at 70°C and pH 5.0∼5.5. With p-nitrophenyl-α-D-glucoside as a substrate and under the optimal condition (70°C, pH 5.5), K m and V max of the enzyme was 1.72 mM and 39 U/mg, respectively. The purified α-glucosidase could hydrolyze oligosaccharides with both α-1,4 and α-1,6 linkages. The enzyme also had strong transglycosylation activity when maltose was used as sugar donor. The transglucosylation products towards maltose are isomaltose, maltotriose, panose, isomaltotriose and tetrasaccharides. The enzyme could convert 400 g/L maltose to oligosaccharides with a conversion rate of 52%, and 83% of the oligosaccharides formed were prebiotic isomaltooligosaccharides (containing isomaltose, panose and isomaltotriose).

The response surface methodology (RSM) and central composite design (CCD) technique were used to optimize the three key process parameters (i.e., pressure, temperature and holding time) of the high-hydrostatic-pressure (HHP) processing either standalone or combined with moderate thermal processing to modulate molecular structures of β-lactoglobulin (β-Lg) and α-lactalbumin (α-La) with reduced human IgE-reactivity. The RSM model derived for HHP-induced molecular changes of β-Lg determined immunochemically showed that temperature (temp), pressure (p2) and the interaction between temperature and time (t) had statistically significant effects (p < 0.05). The optimal condition defined as minimum (β-Lg specific) IgG-binding derived from the model was 505 MPa at 56 °C with a holding time of 102 min (R2 of 0.81 and p-value of 0.01). The validation carried at the optimal condition and its surrounding region showed that the model to be underestimating the β-Lg structure modification. The molecular change of β-Lg was directly correlated with HHP-induced dimerization in this study, which followed a quadratic equation. The β-Lg dimers also resulted in the undetectable human IgE-binding.

AbstractWe consider the equation {-\Delta_{p}u=f(u)} in a smooth bounded domain of {\mathbb{R}^{n}}, where {\Delta_{p}} is the p-Laplace operator. Explicit examples of unbounded stable energy solutions are known if {n\geq p+\frac{4p}{p-1}}. Instead, when {n<p+\frac{4p}{p-1}}, stable solutions have been proved to be bounded only in the radial case or under strong assumptions on f. In this article we solve a long-standing open problem: we prove an interior {C^{\alpha}} bound for stable solutions which holds for every nonnegative {f\in C^{1}} whenever {p\geq 2} and the optimal condition {n<p+\frac{4p}{p-1}} holds. When {p\in(1,2)}, we obtain the same result under the nonsharp assumption {n<5p}. These interior estimates lead to the boundedness of stable and extremal solutions to the associated Dirichlet problem when the domain is strictly convex. Our work extends to the p-Laplacian some of the recent results of Figalli, Ros-Oton, Serra, and the first author for the classical Laplacian, which have established the regularity of stable solutions when {p=2} in the optimal range {n<10}.

AbstractDual implantations of Si+ and P+ into InP:Fe were performed both at 200°C and room temperature. Si+ ions were implanted by 150keV with doses ranging from 5×1013 /cm2 to 1×1015 /cm2, while P+ ions were implanted by 110keV. 160keV and 180keV with doses ranging from 1×l013 /cm2 to 1×1015 /cm2. Hall measurements and photoluminescence spectra were used to characterize the silicon nitride encapsulated annealed samples. It was found that enhanced activation can be obtained by Si+ and P+ dual implantations. The optimal condition for dual implantations is that the atomic distribution of implanted P overlaps that of implanted si with the same implant dose. For a dose of 5×l014 /cm2, the highest activation for dual implants is 70% while the activation for single implant is 40% after annealing at 750°C for 15 minutes. PL spectrum measurement was carried out at temperatures from 11K to 100K. A broad band at about 1.26eV was found in Si+ implanted samples, of which the intensity increased with increasing of the Si dose and decreased with increasing of the co-implant P+ dose. The temperature dependence of the broad band showed that it is a complex (Vp-Sip) related band. All these results indicate that silicon is an amphoteric species in InP.

To produce while pepper using traditional method, the ripe berries are soaked in a river or a pond for 10-14 days prior to decorticating. To improve the traditional method. Research Institute for Spice and Medicinal Crops has designed pepper decorticator to support the white pepper processing. This decorticator has a good function, hut its performance still need to be improved, as it still produces high percentage of white pepper damage. This experiment was aimed to at improving (he performance of the pepper decorticator. The experiment was conducted from July to Ocktobcr 1999 and divided into two steps i.e. preliminary and main experiment. The treatments applied on Ihe main experiment were (I) disc diameter (18, 20, and 22 cm) and (2) rubber elasticity (5.34, 7.74, 9.79, and 15.02 N/mm). The experiment was designed as a completely randomized, arranged factorially in three replications. The pepper variety used for the experiment was Lampung Daun Lebar. The disc distance of pepper decorticator in this experiment was set at 3.5 cm based on the preliminary experiment. Results showed that the optimal disc diameter and nibber elasticity was 20 cm and 7.74 N/mm respectively. Decorticator performance on the optimal condition were : decorticated pepper 97.25%, white pepper damage as pepper defect 2.46% and as broken pepper 1.98% respectively and the capacity of decorticator was 42.27 kg/hours. In general, the quality of while pepper produced met the National Standard of Indonesia.

In this article obtaining complex nitrogen-phosphate fertilizers based on activation of phosphorite powder with partially ammoniated mix of phosphoric and sulphuric acids have been studied. In order to reduce the foaming of the process, for the first time a partially ammoniated mix of acids was used. The optimal technological parameters for activation phosphorite powder with partially neutralized mixes of sulphuric and phosphoric acids are the followings: the mix of sulphuric and phosphoric acids with a ratio of SО3:Р2О5 = 1.2; рН of acids – 2.5; weight ratio of ammonium sulphate-phosphate slurry towards phosphorite powder is 100 : 20; temperature is 60°С; duration is 30 min.  The composition nitrogen-phosphate fertilizers obtained in optimal condition contains (wt., %): N – 11.55; Р2О5total -24.61; Р2О5acceptable by citric acid -21.66; Р2О5 acceptable by EDTA -20.24; Р2О5water-soluble  -13.02; CaOtotal -13.59; СаОacceptable by citric acid -11.43. In that condition, granulated products of nitrogen-phosphate can be produced with high strength. The advantage of offered promising technology concludes in reduction two times expenditure of the most expensive ammonia in comparison with ammonium sulphate production and an increase in gross domestic product.

Crystal bar hafnium prepared by the iodide process was studied. The influences of three important factors (filament temperature, retort materials and feed types) on the removal of iron from hafnium crystal bar under iodide process were investigated. Results show that the impurity contents of iron in the hafnium crystal bar decreased with the rising K value. Both the retort materials and feed types have obvious influence on the iron impurity contents of hafnium crystal bar. It is proposed that the optimal condition was attained when hafnium turnings was used as feed, stainless steel with molybdenum cylinder liner as retort materials, and K value was at a relatively high level.
