The medicinal importance of Nigella sativa seeds for treating various ailments is portrayed by its traditional uses. Owing to its immense pharmacological importance, the thymoquinone phytoconstituent of N. sativa can prove beneficial for the South Asian countries including Pakistan, where this seed is commonly produced and healthcare facilities are limited. In this study, the antibacterial activity of various extracts of N. sativa seeds, extracted thymoquinone, and oil samples have been investigated against Bacillus subtilis and Bacillus licheniformis using well and disc diffusion assay. The inhibition zones ranged between 7 and 44 mm against both the bacterial strains by well diffusion assay, while disc diffusion assay provided inhibition zones in the range of 7–23 mm. Commercial and local Kalonji oil samples were included in the study. Oil samples dissolved in methanol showed increased inhibition of bacteria. However, the extracted thymoquinone showed highest antibacterial activity. Medicine formulated using thymoquinone will prove to be an herbal alternate against the resistant microbiota associated with bacterial infections. Antibacterial activity against some Bacillus species will help signify the effect on normal gut flora when oral therapy is followed. Trying different extraction protocols can help increase extraction efficiency. Study on extraction of thymoquinone in local produce of black seed can be fruitful for conducting the stability studies and can help to gain maximum benefits from the bioactives. The crude extracts from 10 g of these seeds were subjected to preliminary phytochemical investigation. Results showed that although methanol extract had the presence of maximum phytochemicals, hexane extract was the most potent in terms of antibacterial activity. Thymoquinone, a therapeutically important bioactive in N. sativa seed, was extracted employing both solvents. TLC assay and UV spectroscopy were used for its qualitative assessment, while HPLC-UV quantification showed that 250 mg/mL of methanol extract had 368.3 μg/mL thymoquinone, while its successive extraction yielded 32.94 μg/mL thymoquinone.

Penelitian terhadap ekstrak metanol, fraksi etil asetat, dan fraksi butanol dari rumput laut Eucheuma spinosum menggunakan metode Disc Diffusion Test dilakukan untuk mengetahui peranannya sebagai antibakteri Escherichia coli ATCC 25922, dan Staphylococcus aureus ATCC 6538. Sebagai kontrol digunakan ekstrak (negatif) pelarut metanol, dan Amoxicilin. Konsentrasi sampel uji ditetapkan 10.000 ppm. Hasil pengujian menunjukkan adanya  aktivitas antibakteri yang ditandai dengan terbentuknya zona hambat terhadap bakteri Escherichia coli ATCC 25922.  Besarnya zona hambat secara berurutan untuk ekstrak metanol, fraksi  butanol, dan fraksi etil asetat, ialah : 6,98 mm, 7,85 mm, dan 7,88 mm.  Nilai zona hambat tersebut, relatif sama untuk setiap sampel uji. Sampel uji juga menunjukkan terdapat aktivitas antibakteri, ditandai dengan terbentuknya zona hambat terhadap bakteri Staphylococcus aureus ATCC 6538.  Besarnya zona hambat secara berurutan untuk ekstrak metanol, fraksi  butanol, fraksi etil asetat,  dan kontrol negatif pelarut metanol, ialah : 7,42 mm, 14.40 mm, 7,90 mm, dan 6,75 mm.  Zona hambat fraksi butanol lebih tinggi dibanding yang lain, ini menunjukkan bahwa aktivitas antibakteri Staphylococcus aureus pada fraksi butanol lebih tinggi dibanding fraksi lainnya.Kata kunci:  Antibakteri;  Eucheuma spinosum;  Escherichia coli; Staphylococcus aureus Antibacteria Activities of Extract Sea Algae  Eucheuma spinosum Fresh from Nain Island Waters North Sulawesi  ABSTRACTA research on antibacterial activity of methanol extract, ethyl acetate, and butanol fraction from Eucheuma spinosum seaweed using the disc diffusion method to know the role as  antibacterial activity was tested against Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 6538. Methanol was used as negative control while amoxicillin was used as positive control. The concentration of the test sample was set at 10,000 ppm. The antibacterial activity was indicated by the formation of an inhibition zone against the tested microorganisms. The inhibition zones against E.coli for methanol extract, butanol fraction, and ethyl acetate fraction were 6.98 mm, 7.85 mm, and 7.88 mm, respectively.  The value of the zone of inhibition is relatively the same for each sample.  In addition, the inhibition zones of methanol extract, butanol and ethyl acetate fraction against Staphylococcus aureus were 7.42 mm, 14.40 mm, 7.90 mm, and 6.75 mm, respectively. The inhibition zone of the butanol fraction is higher than the others, this indicated that the antibacterial activity for butanol fraction is higher than other fractions. Keywords: Antibacterial activity; Eucheuma spinosum; Escherichia coli; Staphylococcus aureus

The essential oil of Cordia verbenacea D.C. (Boraginaceae) that grows in Mérida-Venezuela was obtained by hydrodistillation from the aerial parts of the plant, yielding 0.21%. The oil was analyzed by GC-FID and GC-MS. Thirty one components which made up 94.3% of the oil were identified. The most abundant constituents found were: tricyclene (23.9%), bicyclogermacrene (11.7%), germacrene D (9.9%) and β-caryophyllene (8.2%). Antibacterial activity determination was carried out according to the disc diffusion assay. Activity against Gram-positive bacteria Staphylococcus aureus ATCC 6538 and Enterococcus faecalis ATCC 29212, at a minimal inhibitory concentration (MIC) of 170 μg/mL and 200 μg/mL, was found.

This initial research was intended to detect antibacterial activity of pliek u oil and pliek u. Pliek u oil consist of minyeuk simplah (MS) and minyeuk brok (MB), Pliek u consist of two kinds of solid waste namely wet pliek u (Ap1) and dry pliek u (Ap2). Pliek u was methanol extracted at concentration 10% (w/v). Pliek u oil and methanol extract of pliek u were evaluated for their antibacterial activity, against Bacillus subtilis and four strains of Enterophatogenic Escherichia coli (EPEC) employing agar disc diffusion method. No antibacterial activity was shown by MS. The MB exhibited a little effect 1-2 mm against bacterial tests. The results demonstrated that the higher antibacterial activity was shown by dry pliek u compare with wet pliek u with the inhibition zones from 6.67-10.33 mm and 6.00-7.33 mm respectively.

A series of cobalt(III) mixed ligand complexes of type [Co(en)2L]+3, where L is bipyridine, 1,10-phenanthroline, imidazole, methylimidazole, ethyleimidazole, dimethylimidazole, urea, thiourea, acetamide, thioacetamide, semicarbazide, thiosemicarbazide, or pyrazole, have been isolated and characterized. The structural elucidation of these complexes has been explored by using absorption, infrared, and 1H NMR nuclear magnetic resonance spectral methods. The infrared spectral data of all these complexes exhibit a band at 1450/cm and 1560–1590/cm, which correspond to C = C and C = N, a band at 575/cm for Co-N (en), and a band at 480/cm for Co-L (ligand). All these complexes were found to be potent antimicrobial agents. The antibacterial activity was studied in detail in terms of zone inhibition, minimum bactericidal, and time period of lethal action. Among all, complexes bipyridine, 1,10-phenanthroline, dimethylimidazole, and pyrazole, possess the highest antibacterial activity. Antifungal activity was done by disc-diffusion assay and 50% inhibitory concentrations that possess high antifungal activity.Key words: cobalt(III) complexes, ethylenediamine, antimicrobial, antifungal.

A series of pyrazoles derived from the substituted enaminones were synthesized and were evaluated for antimicrobial activity. All the compounds were characterized by the spectral data and elemental analysis. The synthesized compounds were initially screened for their antimicrobial activity against ATCC 6538, NCTC 10400, NCTC 10418, and ATCC 27853. During initial screening, compounds (P1, P6, and P11) presented significant antimicrobial activity through disc diffusion assay. These compounds were further evaluated for antimicrobial activity at different time points against Gram-positive and Gram-negative bacteria and presented significant activity for 6 hours. The activity was found to be greater against Gram-positive bacteria. In contrast at 24 hours, the activity was found only against Gram-positive bacteria except compound (P11), showing activity against both types of bacteria. Compound (P11) was found to have highest activity against both Gram-positive and Gram-negative bacteria.

Scytonema schmidtii,a cyanobacterium was isolated from the collected soil samples from different locations of Ahmednagar district of Maharashtra state (India). Identification was carried out using morphological variation and taxonomical approaches according to Desikachary (1959) and Prescott (1962) .The axenic culture of Scytonema schmidtii was obtained by using the method recommended by Bolch and Blackburn (1996). The isolated Scytonema schmidtii was grown autotropically in BG-11 medium as described by Rippka et al.,(1979) and incubated at 30±2°c.After 25 days, biomass was harvested by filtration through double layered muslin cloth and dried using air blower. The biomass of this Scytonema schmidtii species was used for the assessment of antibacterial activity against Escherichia coli, Bacillus subtilis, Staphylococcus aureus, Pseudomonas aeruginosa, Micrococcus flavus and Proteus mirabilis. The antibacterial activity was studied by disc diffusion method (Anonymous,1996).Methanol extract of Scytonema schmidtii showed the activity against all the tested bacterial strains.Maximum zone of inhibition (23±1.9 mm) was recorded with methanol extract of Scytonema schmidtii.

The antibacterial activity of the extracts of Aframomum melegueta including n-hexane extract (NHE), nondefatted methanol extract (NDME), and defatted methanol extract (DME) was investigated in this study. The NHE exhibited no antibacterial activity. The DME showed higher antibacterial activity than the NDME against the different isolates. At the highest concentration of 10 mg/mL in agar diffusion, NDME produced inhibition zones ranging from 11 to 29 mm against the microorganisms while DME produced inhibition zones ranging from 20 to 40 mm with the concentration of 10 mg/mL against the microorganisms. 0.1 mg/mL of the DME produced inhibition zones ranging between 12 and 14 mm in Aeromonas hydrophila ATCC 35654 and Pseudomonas aeruginosa ATCC 15442, respectively, while none of the isolates were inhibited by the NDME at a concentration of 1 mg/mL or less. In the agar dilution assay, the MICs of the NDME and DME ranged between 0.31 and 10 mg/mL, but more isolates were inhibited at 0.31 mg/mL of DME than those in NDME. In macrobroth assay, the MICs of the NDME ranged between 0.15 and 5.0 mg/mL and the MBCs ranged between 0.63 and 5.0 mg/mL, and the MICs of the DME ranged between 0.08 and 5.0 mg/mL and the MBCs were between 0.31 and 5.0 mg/mL. This study indicated that DME was more active with higher antibacterial activity than the NDME of this plant, and extracting the fatty portion of plant materials prior susceptibility testing would allow plant extracts to be more effective as well as justifying the use of Aframomum melegueta in traditional medicine for the treatment of bacterial infections.

Objective: The objective of this research was to investigate the preliminary phytochemical screening and antibacterial activity of Bawang Dayak(Eleutherine sp.) and Hati Tanah (Angiopteris sp.) and their combination against Propionibacterium acnes.Methods: The extracts were used for phytochemical screening. Antibacterial activity was performed using disc diffusion technique, with twovariations of the concentration of 5% and 10% for each extracts, and combination of both extracts with three combinations: (1) 5%: 5%, (2) 5%: 10%, and (3) 10%: 5%.Results: Both extracts contained tannins, saponins, and steroids. The antibacterial activity against P. acnes showed that the inhibition zones ofBawang Dayak ethanol extract were 6.1±1.5 mm (5%) and 8.7±1.3 mm (10%). On the other hand, the inhibition zones of Hati Tanah ethanol extract were 4.0±1.6 mm (5%) and 9.2±2.5 mm (10%). The inhibition zones produced in combinations 1, 2, and 3 were 5.8±0.3 mm, 10.8±2.0 mm, and 15.5±2.8 mm, respectively.Conclusion: In this study showed the presence of tannins, saponins, and steroids that might be responsible for antibacterial activity in both extract. The best antibacterial activity was produced by combining the two extracts (combination 3).

Objective: The objective of this research was to investigate the antibacterial activity of bawang dayak from Central Kalimantan against acne-causingbacteria namely Propionibacterium acnes, Staphylococcus epidermidis, and Staphylococcus aureus.Methods: The preliminary phytochemical constituents were qualitatively analyzed. Antibacterial activity of bawang dayak ethanol extract wasperformed using disc-diffusion technique, with five variations of concentration of 1.25%, 2.5%, 5%, 10%, and 20%.Results: Bawang dayak extract contained flavonoids, alkaloids, saponins, and tannins. The inhibition zones of bawang dayak ethanol extract in fivevarious concentrations (1.25%, 2.5%, 5%, 10%, and 20%) were 7.0±1.3 mm, 8.5±0.6 mm, 9.9±0.3 mm, 11.2±0.1 mm, and 11.9±0.3 mm against P. acnes;18.8±3.3 mm, 21.9±3.3 mm, 20.8±0.6 mm, 22.0±0.2 mm, and 23.1±0.6 mm against S. epidermidis; and 14.3±3.1 mm, 13.5±0.9 mm, 14.7±1.5 mm,16.1±1.0 mm, and 20.1±0.6 mm against S. aureus, respectively.Conclusion: This present study showed that bawang dayak ethanol extract was active against all the tested acne-causing bacteria. The highestantibacterial activity was produced by 20% of bawang dayak ethanol extract against S. epidermidis.
